Abstract

Automated vehicles are pictured as the future of transportation, and facilitating safer driving is only one of the many benefits. However, due to the constantly changing role of the human driver, users are easily confused and have little knowledge about their responsibilities. Being the bridge between automation and human, the human-machine interface (HMI) is of great importance to driving safety. This study was conducted in a static driving simulator. Three HMI designs were developed, among which significant differences in mental workload using NASA-TLX and the subjective transparency test were found. An electroencephalogram was applied throughout the study to determine if differences in the mental workload could also be found using EEG's spectral power analysis. Results suggested that more studies are required to determine the effectiveness of the spectral power of EEG on mental workload, but the three interface designs developed in this study could serve as a solid basis for future research to evaluate the effectiveness of psychophysiological measures. Abstract

Photoneutron reactions on $^{208}$Pb in the Giant Dipole Resonance energy region have been investigated at the $\gamma$-ray beam line of the NewSUBARU facility in Japan. The measurements made use of quasi-monochromatic laser Compton backscattering $\gamma$-ray beams in a broad energy range, from the neutron threshold up to 38 MeV, and of a flat-efficiency moderated $^3$He neutron detection system along with associated neutron-multiplicity sorting methods. We report absolute cross sections and mean photoneutron energies for the $^{208}$Pb$(\gamma,\,inX)$ reactions with $i$~=~1 to 4. The fine structure present in the $^{208}$Pb$(\gamma,\,n)$ cross sections at incident energies lower than 13~MeV has been observed. The photoabsorption cross section has been obtained as the sum of the $(\gamma,\,inX)$ reaction cross sections. By reproducing the measured ring-ratio values at excitation energies below the two neutron separation energy, we were able to extract estimations on the $^{208}$Pb$(\gamma,\,n)$ photoneutron energy spectra and on the partial photoneutron cross sections for leaving the residual $^{207}$Pb in its ground and first two excited states. The present results are compared with data from the literature and statistical model calculations.

Abstract

Connected automated vehicles (CAV), which incorporate vehicle-to-vehicle (V2V) communication into their motion planning, are expected to provide a wide range of benefits for individual and overall traffic flow. A frequent constraint or required precondition is that compatible CAVs must already be available in traffic at high penetration rates. Achieving such penetration rates incrementally before providing ample benefits for users presents a chicken-and-egg problem that is common in connected driving development. Based on the example of a cooperative driving function for bottleneck traffic flows (e.g. at a roadblock), we illustrate how such an evolutionary, incremental introduction can be achieved under transparent assumptions and objectives. To this end, we analyze the challenge from the perspectives of automation technology, traffic flow, human factors and market, and present a principle that 1) accounts for individual requirements from each domain; 2) provides benefits for any penetration rate of compatible CAVs between 0 % and 100 % as well as upward-compatibility for expected future developments in traffic; 3) can strictly limit the negative effects of cooperation for any participant and 4) can be implemented with close-to-market technology. Abstract

Bioimaging refers to a collection of methods to visualize the internal structures and mechanisms of living organisms. The fundamental tool, the microscope, has enabled seminal discoveries like that of the cell as the smallest unit of life, and continues to expand our understanding of biological processes. Today, we can follow the interaction of single molecules within nanoseconds in a living cell, and the development of complete small organisms like fish and flies over several days starting from the fertilized egg. Each image pixel encodes multiple spatiotemporal and spectral dimensions, compounding the massive volume and complexity of bioimage data. Proper handling of this data is indispensable for analysis and its lack has become a growing hindrance for the many disciplines of the life and biomedical sciences relying on bioimaging. No single domain has the expertise to tackle this bottleneck alone. As a method-specific consortium, NFDI4BIOMAGE seeks to address these issues, enabling bioimaging data to be shared and re-used like they are acquired, i.e., independently of disciplinary boundaries. We will provide solutions for exploiting the full information content of bioimage data and enable new discoveries through sharing and re-analysis. Our RDM strategy is based on a robust needs analysis that derives not only from a community survey but also from over a decade of experience in German BioImaging, the German Society for Microscopy and Image Analysis. It considers the entire lifecycle of bioimaging data, from acquisition to archiving, including analysis and enabling re-use. A foundational element of this strategy is the definition of a common, cloud-compatible, and interoperable digital object that bundles binary images with their descriptive and provenance metadata. Abstract

Personalized treatment strategies based on non‑invasive biomarkers have potential to improve patient management in patients with newly diagnosed glioblastoma (GBM). The residual tumour burden after surgery in GBM patients is a prognostic imaging biomarker. However, in clinical patient management, its assessment is a manual and time‑consuming process that is at risk of inter‑rater variability. Furthermore, the prediction of patient outcome prior to radiotherapy may identify patient subgroups that could benefit from escalated radiotherapy doses. Therefore, in this study, we investigate the capabilities of traditional radiomics and 3D convolutional neural networks for automatic detection of the residual tumour status and to prognosticate time‑to‑recurrence (TTR) and overall survival (OS) in GBM using postoperative [11C] methionine positron emission tomography (MET‑PET) and gadolinium‑enhanced T1‑w magnetic resonance imaging (MRI). On the independent test data, the 3D‑DenseNet model based on MET‑PET achieved the best performance for residual tumour detection, while the logistic regression model with conventional radiomics features performed best for T1c‑w MRI (AUC: MET‑PET 0.95, T1c‑w MRI 0.78). For the prognosis of TTR and OS, the 3D‑DenseNet model based on MET‑PET integrated with age and MGMT status achieved the best performance (Concordance‑Index: TTR 0.68, OS 0.65). In conclusion, we showed that both deep‑ learning and conventional radiomics have potential value for supporting image‑based assessment and prognosis in GBM. After prospective validation, these models may be considered for treatment personalization.

Abstract

(1) Background: The sensitivity of head and neck squamous cell carcinoma (HNSCC) to ionizing radiation, among others, is determined by the number of cells with high clonogenic potential and stem-like features. These cellular characteristics are dynamically regulated in response to treatment and may lead to an enrichment of radioresistant cells with a cancer stem cell (CSC) phenotype. Epigenetic mechanisms, particularly DNA and histone methylation, are key regulators of gene-specific transcription and cellular plasticity. Therefore, we hypothesized that specific epigenetic targeting may prevent irradiation-induced plasticity and may sensitize HNSCC cells to radiotherapy. (2) Methods: We compared the DNA methylome and intracellular concentrations of tricarboxylic acid cycle metabolites in radioresistant FaDu and Cal33 cell lines with their parental controls, as well as aldehyde dehydrogenase (ALDH)-positive CSCs with negative controls. Moreover, we conducted a screen of a chemical library targeting enzymes involved in epigenetic regulation in combination with irradiation and analyzed the clonogenic potential, sphere formation, and DNA repair capacity to identify compounds with both radiosensitizing and CSC-targeting potential. (3) Results: We identified the histone demethylase inhibitor GSK-J1, which targets UTX (KDM6A) and JMJD3 (KDM6B), leading to increased H3K27 trimethylation, heterochromatin formation, and gene silencing. The clonogenic survival assay after siRNA-mediated knock-down of both genes radiosensitized Cal33 and SAS cell lines. Moreover, high KDM6A expression in tissue sections of patients with HNSCC was associated with improved locoregional control after primary (n = 137) and post-operative (n = 187) radio/chemotherapy. Conversely, high KDM6B expression was a prognostic factor for reduced overall survival. Abstract

Utilizing a novel and exact inversion scheme, we determine the complex linear conductivity $\sigma (\omega )$ from the linear magnetic ac-susceptibility which has been measured from 3\,mHz to 50\,MHz in fields between 0.4\,T and 4\,T applied parallel to the c-axis of a 250\,nm thin disk. The frequency derivative of the phase $\sigma ''/\sigma '$ and the dynamical scaling of $\sigma (\omega)$ above and below $T_g(B)$ provide clear evidence for a continuous phase transition at $T_g$ to a generic superconducting state. Based on the vortex-glass scaling model, the resulting critical exponents $\nu $ and $z$ are close to those frequently obtained on films by other means and associated with an 'isotropic' vortex glass. 47. Post-operative thrombotic thrombocytopenic purpura: A review

Abstract

Post-operative thrombotic thrombocytopenic purpura (TTP) is a recently recognised life-threatening clinical syndrome with considerable similarity to classic TTP in presentation and response to early treatment with plasma exchange. To date, 29 cases of TTP associated with surgery have been reported. The majority of cases have complicated vascular surgeries, with a few cases seen following gastrointestinal or orthopaedic procedures. Characteristically, patients develop microangiopathic haemolytic anaemia and consumptive thrombocytopenia 5 to 9 days following surgery with variable presence of fever, impaired renal function and altered mental status. The pathogenesis of post-operative TTP is speculative but may involve the release of large amounts of high-molecular-weight von Willebrand factor (vWF) multimers due to endothelial damage resulting from surgery in the setting of marginal levels of vWF-cleaving enzyme. The myriad of common post-surgical complications that may present with clinical manifestations similar to TTP may result in confusion with the potential for delay in the initiation of life-saving plasma-exchange therapy. It is important that physicians be alert to the phenomenon of post-operative TTP so that prompt recognition and treatment will prevent serious morbidity or mortality.

Thereafter, an alternating magnetic field generates heat which may be used to release the thermosensitively bound drug and for hyperthermia. Until today, only superficial tumors can be treated with this method. Our aim was to transfer this method into an endoscopic setting to also reach the majority of tumors located inside the body. To find the ideal endoscopic magnetic field trap, which accumulates the most SPION, we first developed a biophysical model considering anatomical as well as physical conditions. Entities of choice were esophageal and prostate cancer. The magnetic susceptibilities of different porcine and rat tissues were measured with a superconducting quantum interference device. All tissues showed diamagnetic behavior. The evaluation of clinical data (computed tomography scan, endosonography, surgical reports, pathological evaluation) of patients gave insight into the topographical relationship between the tumor and its surroundings. Both were used to establish the biophysical model of the tumors and their surroundings, closely mirroring the clinical situation, in which we could virtually design, place and evaluate different electromagnetic coil configurations to find optimized magnetic field traps for each tumor entity. By simulation, we could show that the efficiency of the magnetic field traps can be enhanced by 38-fold for prostate and 8-fold for esophageal cancer. Therefore, our approach of endoscopic targeting is an improvement of the magnetic drug-targeting setups for SPION tumor therapy as it holds the possibility of reaching tumors inside the body in a minimal-invasive way.
