Abstract

Precision absolute gravity measurements are growing in importance, especially in the context of the new definition of the kilogram. For the case of free-fall absolute gravimeters with a Michelson-type interferometer tracking the position of a free falling body, one of the effects that needs to be taken into account is the speed of light perturbation due to the finite speed of propagation of light. This effect has been extensively discussed in the past, and there is at present a disagreement between different studies. In this work, we present the analysis of new data and confirm the result expected from the theoretical analysis applied nowadays in free-fall gravimeters. We also review the standard derivations of this effect (by using phase shift or Doppler effect arguments) and show their equivalence.

Abstract

The microtubule cytoskeleton determines the internal architecture of cells, crucial for their proper functioning. In many vertebrate cells the interphase microtubule cytoskeleton has an astral organisation with stable microtubule ends focused near the cell centre and dynamic microtubule ends emanating outwards towards the plasma membrane. This arrangement is determined by a complex self-organisation process involving the cell boundary and many protein activities within and at the periphery of a cell. This project aimed at the elucidation of the minimal set of activities required for this process to understand the rules governing the self-organisation of microtubule arrays within a boundary. I reconstituted self-organising microtubule arrays inside lipid monolayer surrounded droplets in oil from purified components and hence established a well-controlled system to systematically study the interplay of organising activities by fluorescence microscopy. My studies showed that restriction in space as such has no influence on de novo polymerisation of microtubule polymerisation, however on their arrangement. In presence of a surrounding lipid monolayer microtubule nucleation and polymerisation could be achieved using a lipid composition close to that of a natural plasma membrane and microtubule nucleation enhancing buffer conditions. Without any further organising activity, the arrangement of microtubules depends on the droplet diameter and microtubule nucleation efficiency. The microtubule-crosslinking and organising kinesin 14 forms single microtubules asters in larger droplets, in smaller droplets it bundles microtubules indicating a microtubule length – droplet diameter interdependency influencing the aster formation inside a confined volume. Therefore I investigated whether microtubule shortening by a kinesin-13 or severing by Spastin, either homogenously distributed in the droplet or located to the droplet boundary, allows motor-driven aster formation in small droplets. In order to transfer that basic model system into artificial liposomes and investigate the influence of a pliable boundary, I designed a flow chamber to appropriately image vesicles.

Abstract

We report on the comparison between three absolute gravimeters that took place in April 2010 at Laboratoire National de M\'etrologie et d'Essais. The three instruments (FG5#209 from METAS, Switzerland, IMGC-02 from INRIM, Italy, and CAG from LNE-SYRTE, France) rely on different methods: optical and atomic interferometry. We discuss their differences as well as their similarities. We compare their measurements of the gravitational acceleration in 4 points of the same pillar, in the perspective of an absolute determination of g for a watt balance experiment

Abstract

As part of systemic inflammatory reactions, interleukin 6 (IL-6) induces acute phase protein (APP) genes through the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway. Epidermal growth factor (EGF), which contributes to the regenerative process after liver injury and also activates STATs, does not induce but attenuates IL-6-stimulated expression of several APP genes in primary mouse hepatocytes. The APP-modifying action of EGF receptor (EGFR) was characterized in HepG2 cells. Although EGF less effectively engages STAT proteins in these cells, it reduces expression of fibrinogen and haptoglobin, but stimulates production of alpha(1)-antichymotrypsin and induces transcription through the alpha(1)-antichymotrypsin and C-reactive protein promoter. The stimulatory EGFR signal is insensitive to inhibition of JAKs and appears to involve Src kinases and STAT proteins as shown by inhibition through overexpression of C-terminal Src kinase (Csk) and transdominant negative STAT3, respectively. A mediator role of Src is supported by the ability of c-Src and v-Src to activate STATs and induce transcription through APP promoters. Src kinases have been observed in association with the IL-6 receptor; however, inhibition of Src kinases by Csk enhances IL-6-induced transcription. The Csk effect is attributed to prevention of Src kinases from phosphorylating gp130 at the docking site for the signal-moderating protein tyrosine phosphatase SHP-2. The inhibitory EGFR signal on APP expression correlates with the activation of Erk1 and Erk2. The study shows a dual signaling function for EGFR and suggests that the ratio of receptor-activated STATs and Erks influence the level of stimulated or inhibited expression of individual APPs.

Abstract

This contribution aims to propose a possible integration of science communication, which recognizes three modes of interaction (the deficit, dialogue and participation models) between experts and non-experts to the field of social LCA. The purpose of such an integration stems from the understanding that through the product chain, there are a wide variety of actors that enable the material product to "flow" and that are working with scientific and sustainability-relevant information. We first discuss the key arguments that evolved in science communication debates from one-way communication of scientific knowledge to public to recent discussions on democratizing science and involving the public in the formative stages of science and technology. We then propose reasons why it could be worthwhile to combine these arguments in the growing literature on social LCA. We use Oatly, the Swedish oat drink company, and the types of communication in its product chain, as the case study to argue the relevance of science communication scholarly frameworks in social LCA.
