Biostimulants have been found effective in enhancing plant resistance toward stressful conditions. The aim of the present study was to evaluate the efficacy of selected biostimulants to overcome the negative effects of nutrient limitation on the growth performances and on the fruit quality of soilless cultivated strawberry plants. The condition of nutrient limitation was imposed by supplying the plants with only a single fertilization at transplantation and by excluding any further nutrient supply for the entire duration of the experiment (three months, from May to July). Strawberry plants were treated seven times during the period from preflowering up to berry maturation with different classes of biostimulants (humic acids, alfalfa hydrolysate, macroseaweed extract and microalga hydrolysate, amino acids alone or in combination with zinc, B-group vitamins, chitosan, and a commercial product containing silicon) at commercial dosages. The use of alfalfa hydrolysate, vitamins, chitosan, and silicon was able to promote biomass accumulation in roots (four to seven folds) and fruits (+20%) of treated plants, whereas the total leaf area increased by 15%&ndash, 30%. Nutrient concentrations in leaves and roots showed variations for microelements (e.g., Fe, B, Zn, and Si) in response to biostimulant applications, whereas no significant differences were observed for macronutrient contents among treatments. Final berry yield was found around 20% higher in chitosan- and silicon-treated plants. Chitosan treatment significantly increased pulp firmness (by 20%), while a high nutritional value (e.g., phenolic compounds concentration) was observed in alfalfa- and seaweed-treated fruits (+18%&ndash, 20% as compared to control). The overall outcomes of the present experiment show that selected biostimulants can be considered as a valid agronomic tool able to contrast the negative consequence of growing crops under insufficient nutritional conditions.

Mass spectrometry is a highly versatile and multi faceted analytical technique, which can be successfully applied to the study of structure-property relationships of small molecules, as new drug candidates, and to better characterize macromolecular targets. During the three years of Ph.D. project, mass spectrometry was employed as analytical tool to solve different tasks related to two main research themes: prodrugs of palmitoylethanolamide (PEA) and allosteric inhibitors of the serine hydrolase monoacylglycerol lipase (MGL). Palmitoylethanolamide (PEA), belonging to the family of fatty acid amide lipid mediators, has antinflammatory and antinociceptive properties widely exploited in veterinary and human medicine, despite its poor pharmacokinetics. Looking for prodrugs that could progressively release PEA to maintain effective plasma concentrations, carbonates, esters and carbamates at the hydroxyl group of PEA were tested for their chemical stability (pH=7.4) and stability in rat plasma and liver homogenate by in vitro assays and high performance liquid chromatography/tandem mass spectrometry (HPLC/MS/MS). Carbonates and carbamates resulted too labile and too resistant in plasma, respectively. With ester derivatives, prepared by conjugating PEA with various amino acids, it was possible to modulate the kinetics of PEA release in plasma and their stability in liver homogenate. L-Val-PEA, with suitable PEA release in plasma, and D-Val-PEA, with high resistance to hepatic degradation, were orally administered to rats and plasma levels of prodrugs and PEA were measured at different time points. Both prodrugs showed significant release of PEA, but lower than that obtained with equimolar doses of PEA. Amino-acid esters of PEA are a promising class to develop prodrugs, even if they need further chemical optimization. The second research project involved the characterization of the chemical reactivity for a series of benzisothiazolone-based compounds, designed to allosterically inhibit the serine hydrolase MGL, a chief enzyme in the metabolism of the main endocannabinoid, 2-arachidonoylglycerol, by targeting selected cysteine residues known to regulate MGL enzymatic activity. Starting from a lead compound, a series of chemical modifications were made by introducing at position 5 and 6 of the benzisothiazolone ring a set of substituents endowed with opposite electronic properties. The reactivity of the newly synthesized compounds was tested in vitro versus glutathione setting up HPLC-MS/MS analytical methods to detect all reaction species in order to have a crucial information about the oxidative potential of each benzisothiazolinone. The role of these regulatory cysteine residues in the fine tuning of MGL activity, their sensitivity towards the redox state of the surrounding environment and their capacity to respond to changes in the redox potential were further explored in a series of pharmacological assays on purified human MGL. In these experiments, hMGL was pre-incubated for a certain period of time in buffer with different oxidizing or reducing agents, subsequently with a probe (dimedone) for identifying an hypothesized oxidized species of cysteine, sulfenic acid. Employing MALDI-TOF/TOF mass spectrometry on oxidized hMGL after trypsin digestion, it was possible to identify which cysteine residue was oxidized to sulfenic acid and which cysteine-containing peptide had been covalently modified by dimedone. Finally, during a three months period spent in the laboratory of Prof. Kasper Rand, at the university of Copenhagen, the conformational changes in hMGL following the interaction with a benzisothiazolone inhibitor were investigated, employing the hydrogen/deuterium exchange mass spectrometry technique (H/DX-MS). La spettrometria di massa è una tecnica analitica molto versatile e poliedrica, che può essere applicata con successo allo studio delle relazioni struttura-proprietà di piccole molecole, nuovi candidati al ruolo di farmaci, e per meglio caratterizzare obiettivi macromolecola. Durante i tre anni del dottorato è stata utilizzata la spettrometria di massa quale strumento analitico per compiere diverse analisi correlate a due principali temi di ricerca: i profarmaci della palmitoiletanolamide (PEA) e gli inibitori allosterici della serina idrolasi monogliceride lipase (MGL). La palmitoiletanolamide (PEA), appartenente alla famiglia dei mediatori lipidici a struttura ammidica derivanti dagli acidi grassi, ha proprietà antinfiammatorie e antinocicettive ed è ampiamente utilizzata in medicina veterinaria ed umana, nonostante le sue scarse proprietà farmacocinetiche. Nella ricerca di profarmaci in grado di rilasciare progressivamente PEA per garantirne significative concentrazioni plasmatiche, una serie di profarmaci, carbonati, esteri e carbammati al gruppo ossidrile di PEA sono stati testati per la loro stabilità chimica (pH = 7.4) e per la stabilità in plasma di ratto ed in omogenato di fegato mediante saggi in vitro impiegando la cromatografia liquida accoppiata alla spettrometria di massa/tandem (HPLC-MS/MS). I carbonati ed i carbammati sono risultati profarmaci rispettivamente troppo labili e troppo resistente nel plasma. Con i derivati esterei preparati coniugando la PEA con vari amminoacidi, è stato possibile modulare le cinetiche di rilascio di PEA nel plasma e la loro stabilità in omogenato di fegato. Il profarmaco L-Val-PEA, caratterizzato da un adeguato rilascio di PEA in plasma, e il profarmaco D-Val-PEA, caratterizzato da un’elevata resistenza alla degradazione epatica, sono stati somministrati per via orale ai ratti ed i livelli plasmatici dei profarmaci e della PEA sono stati misurati a diversi tempi. Entrambi i profarmaci hanno mostrato una significativa liberazione di PEA, anche se inferiore rispetto a quella ottenuta con dosi equimolari di PEA. I coniugati a struttura esterea della PEA con aminoacidi sono una promettente classe per sviluppare profarmaci, anche se necessitano di un'ulteriore ottimizzazione chimica. Il secondo progetto di ricerca ha coinvolto la caratterizzazione della reattività chimica per una serie di benzisotiazoloni, progettato per inibire allostericamente la serina idrolasi MGL, il principale enzima coinvolto nel metabolismo dell’endocannabinoide più abbondante nell’organismo, il 2-arachidonoilglicerolo, attraverso l’interazione con residui di cisteina noti per regolare l'attività enzimatica di MGL. Partendo da un lead compound, sono state apportate una serie di modifiche chimiche introducendo in posizioni 5 e 6 dell'anello benzisotiazolonico una serie di sostituenti dotati opposte proprietà elettroniche. La reattività dei composti di nuova sintesi è stata testata in vitro contro glutatione con la messa a punto di metodi analitici in HPLC-MS/ MS al fine di identificare tutte le specie chimiche generate nella reazione e raccogliere informazioni essenziali sul potenziale ossidativo di ogni benzisotiazolone. Il ruolo delle cisteine regolatorie sull’attività di MGL, la loro sensibilità nei riguardi degli stati di ossido-riduzione e la loro capacità di rispondere a cambiamenti del potenziale redox sono stati esplorati in una serie di saggi farmacologici utilizzando MGL umano ricombinante. In questa serie di esperimenti, hMGL è stato preincubato per un certo periodo di tempo in tampone con l’aggiunta di diversi agenti ossidanti o riducenti, e successivamente, nel primo caso, utilizzando un probe (dimedone) per identificare un possibile prodotto di ossidazione dei residui di cisteina, l’acido sulfenico. Successivamente, analizzando il digerito triptico di hMGL tramite spettrometria di massa MALDI-TOF/TOF, dopo reazione con perossido di idrogeno e dimedone, è stato inequivocabilmente identificato il residuo di cisteina che era stato ossidato ad acido sulfenico e che quindi era stato in grado di reagire covalentemente con il dimedone. Infine, durante un periodo di tre mesi svolto nel laboratorio del Prof. Kasper Rand, presso l’università di Copenaghen, sono stati studiati i cambiamenti conformazionali indotti su hMGL a seguito dell’interazione con un inibitore a struttura benzisotiazolonica, utilizzando la spettrometria di massa di scambio idrogeno/deuterio (H/DX-MS).
