The role of mosquito vectors in spreading disastrous diseases to living organisms, especially to humans is inevitable and undeniable. The impacts of the available chemical and synthetic insecticides on non-specific organisms as well as on nature are being the reason behind the search for target-specific, biocompatible and eco-friendly alternatives. The Madhuca longifolia seed extract and cry proteins from Bacillus thuringiensis-based nanocomposites (Cp-Ml-ZnO NCs) were produced to conquer the above-mentioned issues. The Cp-Ml-ZnO NCs (100 μg/mL) expressed better scavenging potentiality on 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) radicals than Ml seed extract and Ml-ZnO NPs. Listeria monocytogenes have the ability to form biofilms, which aid in the contamination of food and the evasion of antimicrobials. Consumption of L. monocytogenes laden food can promote mild to severe infection in humans and cause serious health issues. Therefore, biofilm development by L. monocytogenes is considered to be a major concern for both healthcare and food safety. This study attempted to target chorismate synthase, an essential protein predicted to be involved in the biofilm pathway. Nigella sativa is renowned for its applications in folk medicine; hence, bioactive ingredients reported were used for molecular docking studies. In the absence of a three-dimensional structure of chorismate synthase from L. monocytogenes, a homology model was generated using the Modeller program. A model with the highest DOPE score was chosen and validated. The reliable model was subjected to docking studies with 30 ligands from N. sativa. From this approach, α-longipinene was unveiled as the best hit. Further in vitro studies demonstrated the antibiofilm potential of α-longipinene against L. monocytogenes. Overall, the study reveals lead molecules from N. sativa as promising antibiofilm agents against L. monocytogenes. Hence, extended investigation with lead molecules will provide sustainable strategies to prevent biofilm-mediated problems due to L. monocytogenes.

Silver (Ag) and zinc oxide (ZnO) are considered to be harmful nanoparticles (NPs) to the aquatic organisms as their intake causes toxic impacts to wildlife, through direct ingestion or by the transference along trophic levels. Over usage and ultimate disposal of metallic particles from the industries subsequently lead to pollution of the aquatic environment. Exposure of NPs in aquatic ecosystem alters biological and physicochemical parameters of the water and aquatic organisms and determines their potential ecotoxicological impacts. Prolonged exposure of aquatic organisms to these NPs results in differential bioaccumulation and distribution into internal organs like liver, kidney, gills, brain, and muscle tissue. The contact of NPs to aquatic organisms induces various types of toxic traits including cytotoxicity, genotoxicity, and epigeneticity. Taking this in consideration, this present review focuses on the comparative toxic impact of ZnO and Ag NPs towards both vertebrates and invertebrates in aquatic ecosystems.

The prevalence of infectious diseases in the aquaculture industry and a limited number of safe and effective oral vaccines has imposed a challenge not only for fish immunity but also a threat to human health. The availability of fish oral vaccines has expanded recently, but little is known about how well they work and how they affect the immune system. The unsatisfactory efficacy of existing oral vaccinations is partly attributable to the antigen degradation in the adverse gastrointestinal environment of fishes, the highly tolerogenic gut environment, and inferior vaccine formulation. To overcome such challenges in designing: an easier, cost-efficient, and effective vaccination method, several encapsulation methods are being adopted to safeguard antigens from the intestinal atmosphere for their immunogenic functions. Oral vaccination is easily degraded by gastric acids and enzymes before reaching the immunological site; however, this issue can be solved by encapsulating antigens in poly-biodegradable nanoparticles, transgenic designed bacteria, plant systems, and live feeds. To enhance the immunological impact, each antigen delivery method operates at a different level. During the nineteenth and twentieth century, production and usage of plastic material have increased and led to a serious problem for their non-degradable property, persistence in the environment. Natural factors act on plastic material fragmented into microplastics. Entering of microplastics causes threats to all ecosystems and their organisms. In this urgent need, this review summarizes (1) sources and transport of microplastics into aquatic ecosystems, (2) ecological impacts of microplastics in aquatic biota and their environment, (3) bacterial remediation of environmental microplastics and (4) mechanism involved bacterial remediation. The terrestrial biosphere serves as a major hub for microplastic wastes and it has been settled in the aquatic ecosystem through the improper disposal methods of human beings. The aquatic biota attains severe effects such as reduced food consumption, fecundity and triggered inflammation; moreover, microplastics ingestion leads to changes in physical, chemical, and gene levels of aquatic organisms. Hence, it is an urgent need to prevent further entry of microplastics (from origin) and eliminate the persisted microplastics in the environment. Recently, researchers are trying to use bacterial strains for microplastics degradation as an eco-friendly and more safe method because during degradation the daughter metabolites carbon dioxide, methane, and water will be produced. Microplastics are a major concern pollutant in earth. Unfortunately, ingestion of microplastics in each trophic organisms via the food chain. In this situation, eradicating the environmental microplastics through an eco-friendly approach using bacteria is a effective remediation technique. Furthermore, research needed using different formulations including bacteria and their based components, consortium of bacteria have enhanced the efficacy of degradation.

Lectins are hemagglutinating glycoproteins which act as pathogen recognition receptors that can bind to the pathogen-associated molecular patterns. Shrimp lectin (Md-Lec) is a lectin that is reported to have antimicrobial property. In this study, the CuS nanoparticles (CuS NPs) which were previously reported to have wide range of antimicrobial activity were conjugated with Md-Lec. Thus, their minimum inhibitory concentration (MIC) against aquatic pathogens was reduced four times than that of their individual activity. Moreover, antibacterial and antibiofilm assay was performed with Md-Lec, pCuS NPs, and Md-Lec conjugated with pCuS NPs (Md-Lec-pCuS NPs) against Aeromonas hydrophila at 25, 50, and 100 μg/mL. The hydrophobicity index and EPS (extracellular polysaccharide) production of A. hydrophila were decreased after treating with Md-Lec-pCuS NPs. The non-specific immune parameters—lysozyme assay, peroxidase assay, and respiratory burst activity—were elevated considerably in the infected fishes that are treated with Md-Lec-pCuS NPs at their MIC. Further, immune gene (COX2, MyD88, and LEC) expression analysis at different time intervals was also done in the infected Etroplus suratensis that are treated with Md-Lec-pCuS NPs at 3.13 μM concentration. The results reveal that the immune gene expression was upregulated at the 6th hour of infection in the infected fish that are treated with Md-Lec-pCuS NPs (3.13 μM) and thus it can be used as a therapeutic agent which can act as an immunostimulant for the production of disease-free fishes in aquaculture industry.

Fabrication and use of nanoparticles have progressively enlarged within the last decade. Herein the silver nanoparticles (AgNps) were synthesized via the extract from Marsilea quadrifolia (Mq) as a decreasing and steadying mediator. The Mq-AgNPs demonstrated superior toxicity on Zika virus vector, Aedes aegypti with the LC50 value of 10.69 µg·mL−1. The Mq-AgNPs were established securely to non-target organisms Artemia nauplii and Ceriodaphnia cornuta, and no structural or anatomical alterations confirm its security to the surroundings. The antibacterial activity exposed that Mq-AgNPs showed superior growth inhibition effect against the tested bacteria. Furthermore, the Mq-AgNPs showed strong antioxidant activities when compared to Mq leaf extract. Overall, our results highlight that Mq-AgNPs are hopeful and biodegradable devices against mosquito vectors of therapeutic significance, with moderate toxicity against non-target aquatic animals.

Metal oxide nanoparticles (NPs) have gained attention in biomedicine due to their broad spectrum of applications, such as targeted drug delivery, their use as antibacterial agents or in cancer treatments. In particular, zinc oxide nanoparticles (ZnO NPs) have become one of the most popular metal oxide NPs in biomedical applications. The present study describes for the first time the biogenic synthesis of rod shaped ZnO NPs using C. annuum L. var. grossum (L.) Sendt) extract as a capping agent. The purified C. annum-based zinc oxide nanorods (Ca-ZnO NRs) were characterized by different physico-chemical techniques. UV–vis spectroscopy depicted the absorbance peak at 372 nm. The size of nanorods ranges between 70 and 80 nm as revealed by FE-TEM micrographs and the mean size was 72 nm. Ca-ZnO NRs exhibited antibacterial activity in a dose-dependent manner and a greater activity was observed against S. enterica. Ca-ZnO NRs have shown antioxidant activity (91%) at 100 μg ml−1 when compared to the standard (ascorbic acid) (98%). A significant inhibition of bovine serum albumin (BSA) protein denaturation evidenced their anti-inflammatory property. Further, Ca-ZnO NRs showed hemolysis of red blood cells (RBC) (~ 3%) (up to 3 μg ml−1) which is much lesser than permissible limit.

It is a well explored facts that need of biosynthesized nanomaterials has emerged for its several advantages in biomedical applications and the search for potential biosynthesized-nanomaterials is still ongoing. In this study an attempt was made to meet the requirement of potential biosynthesized-silver nanoparticles using the aqueous seed extract of N. sativa (Bc-AgNPs). X-ray powder diffraction (XRD) confirms the biosynthesis of silver nanoparticle (supports the observation of UV–Vis spectroscopy) with face centred cubic structure. Fourier-transform infrared spectroscopy (FTIR) confirms the capping of the active ingredients of seed extract and further the average size of 34 nm was confirmed by high-resolution transmission electron microscopy (HR-TEM) and energy dispersive X-ray analysis (EDX) confirms the purity of Bc-AgNPs. Due to the capping of the active ingredients of seed extract, the Bc-AgNPs have shown significant antibacterial activity against Gram negative bacteria and at the lower concentration of 100 µg*ml−1 it has 89% scavenging activity against free radicals. A strong anti-inflammatory (membrane stabilization) activity towards RBCs (98% at 100 µg*ml−1) was recorded when compared to the positive control (aceclofenac). In addition to the previous activities, a greater inhibition of carbohydrate hydrolyzing enzymes such as α-amylase, α-glucosidase and DPP-IV highlighted the antidiabetic potential of these green synthesized AgNPs. Overall, the efficient bioactivities of Bc-AgNPs advocates its future potential in biomedical products.

In the present study, phytochemical analysis, synthesis and characterization of zinc oxide nanoparticles (NG-ZnO NPs) from neem gum as a capping agent. GC–MS analysis confirms the phytochemical oxadiazol-2yl-benzothiazole-(21.5%) on the synthesized ZnO nanoparticles. Various sophisticated analytical instrumentations characterized the morphological and crystalline behaviour of NG-ZnO NPs. The analytical results confirm the spherical crystals of NG-ZnO NPs in size range of 60–80 nm size. The NG-ZnO NPs shown significantly efficient antibacterial and antibiofilm activity against P. aeruginosa at 100 µg/mL. Further, in vitro study on human liver cancer cell line (Hep G2) indicated significantly inhibited the cell proliferation at 100 µg/mL in 24 h. Also, NG-ZnO NPs have also shown potent mosquito larvicidal activity on C. quinquefasciatus at 100 µg/mL. Based on findings, it is recommended that neem gum capped ZnO NPs is potential nano biomedical agents to be used in new drug formulations.

Bio-molecule assisted synthesis of nanoparticles using protein templates and the self-assembly of the bio-molecules together with small peptides, denatured protein, and DNA is a new advancement. Specific interactions with nanoparticles ultimately dictate the size and crystallinity of the nanomaterials and the formation of nanoparticles using protein for structural integrity. Here, we are using protein-based synthesis method for preparing highly crystalline ZnONPs. Crustin(Cr)-capped ZnONPs were synthesized and confirmed by Ultraviolet-vis spectroscopy (UV–Vis), Fourier-transform infrared spectroscopy (FTIR), Raman, X-ray diffraction (XRD), High-resolution transmission electron microscopy (HR-TEM), and Zeta potential analysis. UV–Vis spectrum revealed the preliminary confirmation of the Cr-ZnONPs. By using XRD patterns, confirmed the crystalline structure of Cr-ZnONPs. In addition, TEM analysis revealed that the particle size measurements are within the 50 nm range. Further, Cr-ZnONPs tested against different Gram-negative and Gram-positive bacteria at 50 µg/mL. The great potential of the antibacterial and biofilm control ability of Cr-ZnONPs at 50 µg/mL was observed. Moreover, the control of thrush infective fungal Candida albicans cells and eco-toxicological analysis of Cr-ZnONPs showed that no increase in toxicity up to 100 µg/L was observed in Ceriodaphnia cornuta. Overall, Cr-ZnONPs can be used for potential photocatalytic, antibacterial, antifungal and antibiofilm applications.

Nanotechnology is currently gaining immense attention to combat food borne bacteria, and biofilm. Diabetes is a common metabolic disease affecting majority of people. A better therapy relies on phytomediated nanoparticle synthesis. In this study, W. somnifera leaf extract-assisted ZnO NPs (Ws-ZnO NPs) was synthesized and characterized. From HR-TEM analysis, it has been found that the hexagonal wurtzite particle is 15.6 nm in size and − 12.14 mV of zeta potential. A greater antibacterial effect of Ws-ZnO NPs was noticed against E. faecalis and S. aureus at 100 µg mL−1. Also, the biofilm of E. faecalis and S. aureus was greatly inhibited at 100 µg mL−1 compared to E. coli and P. aeruginosa. The activity of α-amylase and α-glucosidase enzyme was inhibited at 100 µg mL−1 demonstrating its antidiabetic potential. The larval and pupal development was delayed at 25 µg mL−1 of Ws-ZnO NPs. A complete mortality (100%) was recorded at 25 µg mL−1. Ws-ZnO NPs showed least LC50 value (9.65 µg mL−1) compared to the uncoated ZnO NPs (38.8 µg mL−1) and leaf extract (13.06 µg mL−1). Therefore, it is concluded that Ws-ZnO NPs are promising to be used as effective antimicrobials, antidiabetic and insecticides to combat storage pests.
