Cell–material interactions are crucial for many biomedical applications, including medical implants, tissue engineering, and biosensors. For implants, while the adhesion of eukaryotic host cells is desirable, bacterial adhesion often leads to infections. Surface free energy (SFE) is an important parameter that controls short- and long-term eukaryotic and prokaryotic cell adhesion. Understanding its effect at a fundamental level is essential for designing materials that minimize bacterial adhesion. Most cell adhesion studies for implants have focused on correlating surface wettability with mammalian cell adhesion and are restricted to short-term time scales. In this work, we used quartz crystal microbalance with dissipation monitoring (QCM-D) and electrical impedance analysis to characterize the adhesion and detachment of S. cerevisiae and E. coli, serving as model eukaryotic and prokaryotic cells within extended time scales. Measurements were performed on surfaces displaying different surface energies (Au, SiO2, and silanized SiO2). Our results demonstrate that tuning the surface free energy of materials is a useful strategy for selectively promoting eukaryotic cell adhesion and preventing bacterial adhesion. Specifically, we show that under flow and steady-state conditions and within time scales up to ∼10 h, a high SFE, especially its polar component, enhances S. cerevisiae adhesion and hinders E. coli adhesion. In the long term, however, both cells tend to detach, but less detachment occurs on surfaces with a high dispersive SFE contribution. The conclusions on S. cerevisiae are also valid for a second eukaryotic cell type, being the human embryonic kidney (HEK) cells on which we performed the same analysis for comparison. Previous studies have shown that selective synthetic cell receptors can be produced by cell imprinting on polymer layers. However, knowledge on the fundamental detection mechanisms remains limited. In this article, while using yeast cells (Saccharomyces cerevisiae) as model cells, the factors influencing cellular recognition by surface-imprinted polymers (SIPs) are studied by means of spectroscopic and microscopy techniques and a transducer platform based on interfacial thermal transport, the so-called heat-transfer method (HTM). These analyses indicate that cell imprinting creates selective binding sites on the surface of the SIP layer in the form of binding cavities that match the cells in shape and size. Also, we show that phospholipid moieties are incorporated into the SIP cavities during imprinting, while membrane proteins do not seem to be transferred. More importantly, we demonstrate that the incorporated phospholipids significantly enhance cell adhesion to the SIP, and thus play a significant role in the cell-SIP binding mechanism. Furthermore, the hydrophobicity of the SIP layer was found to be considerably higher when compared with a non-imprinted polymer layer (NIP), an effect that could not be attributed to the presence of cavities on the surface of the SIP layer. This work focuses on the development of a label-free biomimetic sensor for the specific and selective detection of bacteria. The platform relies on the rebinding of bacteria to synthetic cell receptors, made by surface imprinting of polyurethane-coated aluminum chips. The heat-transfer resistance (Rth) of these so-called surface imprinted polymers (SIPs) was analyzed in time using the heat-transfer method (HTM). Rebinding of target bacteria to the synthetic receptor led to a measurable increase in thermal resistance at the solid–liquid interface. Escherichia coli and Staphylococcus aureus were used as model organisms for several proof-of-principle experiments, demonstrating the potential of the proposed platform for point-of-care bacterial testing. The results of these experiments indicate that the sensor is able to selectively detect bacterial rebinding to the SIP surface, distinguishing between dead and living E. coli cells on one hand and between Gram-positive and Gram-negative bacteria on the other hand (E. coli and S. aureus). In addition, the sensor was capable of quantifying the number of bacteria in a given sample, enabling detection at relatively low concentrations (104 CFU mL–1 range). As a first proof-of-application, the sensor was exposed to a mixed bacterial solution containing only a small amount (1%) of the target bacteria. The sample was able to detect this trace amount by using a simple gradual enrichment strategy.

We investigated the mass sensitivity of Love mode SAW biosensors by experimental and theoretical techniques. A first experimental approach is based on the analysis of the dispersion curve of the sensor, which helps to determine by derivation a value of the mass sensitivity. In order to obtain the dispersion curve, we have performed a chemical wet etching procedure, which enables the continuous monitoring of the transfer function during the etching of the entire guiding layer. A second experimental approach is based on the addition and removal of layers in known quantities. The mass sensitivity is estimated in different cases: etching of a thin gold layer, copper electrodeposition, and surface adsorption of an ionic surfactant. The results obtained by these techniques are compared to a theoretical model. In the theoretical model, the layered structure of the acoustic sensor is described in terms of mechanical transmission lines and the model is used to determine the sensitivity of the acoustic device to density and viscosity variations of thin films and bulk materials. The attenuation and phase shifts are quantified and analyzed to identify the type of interaction between the adsorbed biomolecules layer and the sensor. We conclude that the Love mode SAW biosensor must be considered and optimized as a viscosity sensor and an independent layer thickness measurement is needed to apply adequately the theoretical model.

One significant challenge facing biosensor development is packaging. For surface acoustic wave based biosensors, packaging influences the general sensing performance. The acoustic wave is generated and received thanks to interdigital transducers and the separation between the transducers defines the sensing area. Liquids used in biosensing experiments lead to an attenuation of the acoustic signal while in contact with the transducers. We have developed a liquid cell based on photodefinable epoxy SU-8 that prevents the presence of liquid on the transducers, has a small disturbance effect on the propagation of the acoustic wave, does not interfere with the biochemical sensing event, and leads to an integrated sensor system with reproducible properties. The liquid cell is achieved in two steps. In a first step, the SU-8 is precisely patterned around the transducers to define 120 mu m thick walls. In a second step and after the dicing of the sensors, a glass capping is placed manually and glued on top of the SU-8 walls. This design approach is an improvement compared to the more classical solution consisting of a pre-molded cell that must be pressed against the device in order to avoid leaks, with negative consequences on the reproducibility of the experimental results. We demonstrate the effectiveness of our approach by protein adsorption monitoring. The packaging materials do not interfere with the biomolecules and have a high chemical resistance. Biosensors offer the opportunity to sense the biological world providing valuable information for medical diagnostics, analytical chemistry, environmental monitoring and fundamental research. Convergence of engineered (bio)chemical surfaces with micro- and nano- systems promises tremendous advances and potential cost reductions in biotechnology. This paper introduces some key challenges facing biosensor development, focussing on opportunities that arise from microsystem platforms utilising novel materials and processes. Examples from our work are presented illustrating the implementation of acoustic wave sensors and novel FET-type sensors.
