This chapter focuses on the basic principles of X-ray fluorescence (XRF) and micro-XRF imaging applicable to cultural heritage research, presenting basic figures of merit and application examples based on the use of advanced laboratory and synchrotron radiation sources. Related methods of analysis making use of absorption edge phenomena by X-ray absorption spectroscopy, including X-ray absorption near-edge structure and extended X-ray absorption fine structure spectroscopy will also be discussed.

Microbeam X-ray fluorescence (XRF) spectroscopy is applied, next to an art-historical analysis, to determine the time and place of origin of Chinese blue-and-white porcelain. In the performed experiments, two groups of kraak porcelain samples (A and B) have been analyzed with XRF spectroscopy to obtain quantitative data, (trace) elemental distributions and fingerprints within different regions of interest. The outcome was processed with Principal Component Analysis (PCA), and with the obtained results it was possible to conclude that the two groups show similar elemental fingerprints and were manufactured in the same time period with a comparable use of raw materials. Additionally, X-ray absorption spectroscopy (XAS) measurements have been performed on the pigment layer, confirming the use of Cobalt Blue as an underglaze pigment.

A novel 3D elemental and morphological analysis approach is presented combining X-ray computed tomography (μCT), X-ray fluorescence (XRF) tomography, and confocal XRF analysis in a single laboratory instrument (Herakles). Each end station of Herakles (μCT, XRF-CT, and confocal XRF) represents the state-of-the-art of currently available laboratory techniques. The integration of these techniques enables linking the (quantitative) spatial distribution of chemical elements within the investigated materials to their three-dimensional (3D) internal morphology/structure down to 1-10 μm resolution level, which has not been achieved so-far using laboratory X-ray techniques. The concept of Herakles relies strongly on its high precision (around 100 nm) air-bearing motor system that connects the different end-stations, allowing combined measurements based on the above X-ray techniques while retaining the coordinate system. In-house developed control and analysis software further ensures a smooth integration of the techniques. Case studies on a Cu test pattern, a Daphnia magna model organism and a perlite biocatalyst support material demonstrate the attainable resolution, elemental sensitivity of the instrument, and the strength of combining these three complementary methodologies.

When investigating unique geological samples, such as cometary, asteroidal, or interstellar material from sample return missions or inclusions in deep Earth diamonds, a nondestructive method is preferred. The presented nondestructive highly sensitive wavelength-dispersive X-ray fluorescence spectroscopy (WD-XRF) technique is designed to measure the L-lines of REE between 4.5 and 7 keV with a sensitivity down to the ppm level. REE fluorescence L-lines are often only separated by a few eV from neighboring XRF-lines and cannot be resolved by an energy dispersive approach especially in the presence of transition metal K-lines. In our spectrometer the characteristic X-rays emitted by the sample are dispersed by a fixed Ge(111) analyzer crystal over the active area of an energy dispersive pn-charge-coupled-device (pnCCD) detector, enabling high energy resolution detection of X-rays differentiated by their corresponding Bragg angles. The use of an energy-dispersive 2D detector enables the simultaneous acquiring of XRF-lines while eliminating any ambiguities due to potential contribution from higher order diffraction effects or other diffraction planes and thereby increases the sensitivity by reducing the (scatter) background. This detection method shows an energy resolution of 12 eV for the Ti–Kα fluorescence line and has a sensitivity down to 0.50 ppm for REE L-lines. The method was optimized specifically for the nondestructive analysis of inclusions in deep Earth diamonds, yielding in situ quantitative information about up-to-now inaccessible elemental (REE) composition patterns together with the more abundant transition metals like Ti, Cr, Mn, and Fe. A large number of painted pre-Islamic ceramics from the Kur River Basin (Fars province, Iran) was collected on surveys and excavations conducted by the late Prof. L. Vanden Berghe (Ghent University, Belgium) during the 1950s. Handheld X-ray fluorescence spectrometry was undertaken on a selection of these ceramics to evaluate the applicability of the technique to study the fabric of these ceramics and the pigments used for decoration. The aim is to obtain elemental data, from which information about raw materials, manufacturing techniques and exchange systems can be extracted. A total of 26 sherds was analyzed dating from the Neolithic to the Late Bronze Age and belonging to different sites and styles. Five compositional groups could be identified in the dataset. These elemental groups differ from the groups obtained by the macroscopic study and are related to the production process and/or the provenance of the primary materials.

At the French synchrotron facility SOLEIL, a new X-ray imaging facility PUMA (Photons Utilisés pour les Matériaux Anciens) has been made available to scientific communities studying materials from cultural heritage. This new instrument aims to achieve 2D and 3D imaging with microscopic resolution, applying different analytical techniques including X-ray fluorescence spectroscopy (XRF), X-ray absorption spectroscopy (XAS), X-ray diffraction and phase-contrast imaging. In order to discover its capabilities a detailed analytical characterization of this beamline as an analytical and imaging tool is deemed necessary. It is crucial to understand the structural origins of macroscopic properties in silicate glasses for their high-tech applications. An example of such an application is chemically strengthened boroaluminosilicate glasses that are exposed to an ion exchange process during which alkali ions (e.g. Na+) are replaced by larger (e.g. K+) ions. Despite the empirically thorough understanding of this exchange process, much less is known about the fundamental physics of the process. Since Fe atoms are a suitable probe for monitoring chemical stress-induced changes in the local structure in the chemically strengthened glasses, a set of chemically strengthened boroaluminosilicate glasses containing 1 mol% Fe2O3 are here studied using depth-resolved confocal X-ray absorption near-edge structure (XANES) and extended X-ray absorption fine structure (EXAFS) spectroscopy. Information on the Fe oxidation state, coordination number, and bond distance as a function of the sample depth and glass composition is obtained. These new insights on chemical stress-induced changes will aid in the further development and improvement of such damage-resistant glasses.

A new, commercially available, mobile system combining X-ray diffraction and X-ray fluorescence has been evaluated which enables both elemental analysis and phase identification simultaneously. The instrument makes use of a copper or molybdenum based miniature X-ray tube and a silicon-Pin diode energy-dispersive detector to count the photons originating from the samples. The X-ray tube and detector are both mounted on an X-ray diffraction protractor in a Bragg-Brentano theta:theta geometry. The mobile instrument is one of the lightest and most compact instruments of its kind (3.5 kg) and it is thus very useful for in situ purposes such as the direct (non-destructive) analysis of cultural heritage objects which need to be analyzed on site without any displacement. The supplied software allows both the operation of the instrument for data collection and in-depth data analysis using the International Centre for Diffraction Data database. This paper focuses on the characterization of the instrument, combined with a case study on pigment identification and an illustrative example for the analysis of lead alloyed printing letters. The results show that this commercially available light-weight instrument is able to identify the main crystalline phases non-destructively, present in a variety of samples, with a high degree of flexibility regarding sample size and position. Tile stoves arise in the Low Countries from the 14th century onwards. Although they have been well documented in northern and central Europe, they have remained an almost forgotten aspect of the material culture of the historic county of Flanders (Belgium). In recent years the amount of stove tiles found in archaeological excavations has increased considerably, raising questions concerning their production and origin. Micro X-ray fluorescence spectroscopy was used to determine the elemental composition of these tiles to see if they could be divided into groups and what these groups could tell us about the distribution of workshops, resources and finished products.

A novel plug-and-play setup based on polycapillary X-ray optics enables three-dimensional (3D) confocal X-ray fluorescence (XRF) and X-ray absorption spectroscopy down to 8 × 8 × 11 μm3 (17 keV) at the European Synchrotron Radiation Facility Collaborative Research Group Dutch–Belgian Beamline, BM26A. A complete description and analytical characterization is presented, together with two recently performed experimental cases. In Deep Earth diamond Sao Luiz-Frankfurt am Main 16, an olivine-rich inclusion was mapped with full 3D XRF elemental imaging. The preliminary tests on Iron Gall ink contained in an historical document, a letter from the court of King Philip II of Spain, reveal both the delicate nature of Iron Gall ink and the lack of Fe–Ni chemical bonding.

The study presents the application of combined micro-Spatially Offset Raman Spectroscopy (micro-SORS) and confocal X-ray fluorescence (confocal XRF) to the non-destructive investigation of micrometer scale stratified painted systems. Micro-SORS can be applied in situations where a high turbidity of layers prevents the acquisition of conventional Raman signals from depth and in the presence of Raman scattering compounds, facilitating highly chemically specific (molecular) characterization of the layers. In contrast, confocal XRF provides elemental information on a well-defined volume at a given depth. In this study, selected specimens have been analysed with both methods; molecular and elemental data have been compared and integrated for a comprehensive characterization of the stratigraphy. This dual approach represents a new analytical modality in the area of Cultural Heritage and is also beneficial in areas where non-destructive interrogation of thin layers and films is required.

A new three-dimensional (3D) micro X-ray fluorescence (μXRF) methodology based on a novel 2D energy dispersive CCD detector has been developed and evaluated at the P06 beamline of the Petra-III storage ring (DESY) in Hamburg, Germany. This method is based on the illumination of the investigated sample cross-section by a horizontally focused beam (vertical sheet beam) while fluorescent X-rays are detected perpendicularly to the sheet beam by a 2D energy dispersive (ED) CCD detector allowing the collection of 2D cross-sectional elemental images of a certain depth within the sample, limited only by signal self-absorption effects. 3D elemental information is obtained by a linear scan of the sample in the horizontal direction across the vertically oriented sheet beam and combining the detected cross-sectional images into a 3D elemental distribution data set. Results of the 3D μXRF analysis of mineral inclusions in natural deep Earth diamonds are presented to illustrate this new methodology.

Here, we report analyses by synchrotron X-ray fluorescence microscopy of the elemental composition of eight candidate impact features extracted from the Stardust Interstellar Dust Collector (SIDC). Six of the features were unambiguous tracks, and two were crater-like features. Five of the tracks are so-called “midnight” tracks—that is, they had trajectories consistent with an origin either in the interstellar dust stream or as secondaries from impacts on the Sample Return Capsule (SRC). In a companion paper reporting synchrotron X-ray diffraction analyses of ISPE candidates, we show that two of these particles contain natural crystalline materials: the terminal particle of track 30 contains olivine and spinel, and the terminal particle of track 34 contains olivine. Here, we show that the terminal particle of track 30, Orion, shows elemental abundances, normalized to Fe, that are close to CI values, and a complex, fine-grained structure. The terminal particle of track 34, Hylabrook, shows abundances that deviate strongly from CI, but shows little fine structure and is nearly homogenous. The terminal particles of other midnight tracks, 29 and 37, had heavy element abundances below detection threshold. A third, track 28, showed a composition inconsistent with an extraterrestrial origin, but also inconsistent with known spacecraft materials. A sixth track, with a trajectory consistent with secondary ejecta from an impact on one of the spacecraft solar panels, contains abundant Ce and Zn. This is consistent with the known composition of the glass covering the solar panel. Neither crater-like feature is likely to be associated with extraterrestrial materials. We also analyzed blank aerogel samples to characterize background and variability between aerogel tiles. We found significant differences in contamination levels and compositions, emphasizing the need for local background subtraction for accurate quantification.

X-ray absorption near-edge structure (XANES) spectroscopy is a well-known nondestructive technique that allows for chemical state and local structure determination. Spatially resolved oxidation state imaging is possible performing full-field transmission mode XANES experiments, providing chemical state information on the illuminated sample area, but these experiments are limited to thin, concentrated samples. Here we present the use of a unique energy dispersive (ED) pnCCD detector, the SLcam, for full-field fluorescence mode XANES experiments, thereby significantly relaxing the constraints on sample thickness. Using this new detection methodology, spatially resolved chemical state information on millimeter-sized sample areas can be obtained with microscopic resolution in moderate measuring times (less than 15 h), obtaining a XANES profile for each of nearly 70,000 points in a single measurement without the need of scanning the sample through the beam. Besides a description of the use of this detector for micro-XANES applications, we also present the proof of concept for fluorescence mode micro-XANES using a Fe(0)/Fe2O3 model sample and a Nitisol soil sample, which was measured to obtain iron chemical state distribution information.

“Mad Meg”, a figure of Flemish folklore, is the subject of a famous oil-on-panel painting by the Flemish renaissance artist Pieter Bruegel the Elder, exhibited in the Museum Mayer van den Bergh (Antwerp, Belgium). This article reports on the in situ chemical characterization of this masterpiece by using currently available state-of-the-art portable analytical instruments. The applied non-destructive analytical approach involved the use of a) handheld X-ray fluorescence instrumentation for retrieving elemental information and b) portable X-ray fluorescence/X-ray diffraction instrumentation and laser-based Raman spectrometers for obtaining structural/molecular information. Next to material characterization of the used pigments and of the different preparation layers of the painting, also the verification of two important historical iconographic hypotheses is performed concerning the economic way of painting by Brueghel, and whether or not he used blue smalt pigment for painting the boat that appears towards the top of the painting. The pigments identified are smalt pigment (65% SiO2 + 15% K2O + 10% CoO + 5% Al2O3) for the blue color present in all blue areas of the painting, probably copper resinate for the green colors, vermillion (HgS) as red pigment and lead white is used to form different colors. The comparison of blue pigments used on different areas of the painting gives no differences in the elemental fingerprint which confirms the existing hypothesis concerning the economic painting method by Bruegel.

Because of its major contributions to the world economy and cultural heritage, Chinese porcelain and its technology can be considered as very important. In this research, the applicability of Raman spectroscopy and X-Ray Fluorescence (XRF) spectroscopy to study the pigments of the ruan-cai colour palette of Chinese porcelain dating from the 18th century was evaluated. Next to the investigations based on stylistic analysis, the use of other analysis techniques is important to obtain complementary information. Raman spectroscopy and XRF spectroscopy are two non-invasive, analytical techniques proven to be useful for the determination of the nature and composition of pigments used to decorate ceramics. As a result, analytical analysis allows us to retrieve more information about the production of Chinese porcelain. Based on visual inspection of the decoration, the objects could be divided into two groups and to evaluate the use of the combined analytical approach, all groups and subgroups were sampled. Before Raman and XRF analyses, the samples were cut, embedded and polished, in order to be able to measure the transversal section of the samples. In this way, each layer of the porcelain samples could be analysed separately. The results show that the blue colour is cobalt-aluminium oxide, in red zones haematite (α-Fe2O3) is present and the green area contains a mixture of CuO in a lead-rich basis and a solid solution of malachite (Cu2CO3(OH)2). The opaque yellow and white colours were identified as lead tin yellow type II (PbSn1−xSixO3, where x ∼ 0.25) and a lead arsenate, respectively.

The capabilities and limitations of nanosecond laser ablation ICP-mass spectrometry for bulk and spatially resolved (elemental mapping) analysis of iron meteorites were assessed. The quantitative data obtained were compared to those obtained i. via multi-element solution ICP-MS (after digestion) relying on external calibration and, ii. high-accuracy determination of selected platinum group elements using solution ICP-MS after target element isolation using anion exchange chromatography and deploying isotope dilution. Results generated by the different methods described show good agreement. Significant matrix effects were observed to affect the results of the ns-LA-ICP-MS analysis of iron meteorites, making quantification via a matrix-matched standard a prerequisite. Careless use of intensity distribution maps without proper correction for laser-solid coupling efficiency can lead to incorrect interpretation of the element maps. ns-LA-ICP-MS was shown to be suitable for fast and quasi-nondestructive analysis of iron meteorites, not only homogeneous ataxites and hexahedrites, but also more heterogeneous ones, when considering ablated areas of a sufficient size. In the context of elemental mapping using LA-ICP-MS, Pearson's product–moment correlation analysis was demonstrated to be a powerful tool that can provide valuable information on the fractionation of the elements in the parent bodies of meteorites.

The development of a novel and straightforward procedure for the preparation of matrix-matched calibration standards for the quantitative imaging of iron (Fe) in biological tissues by laser ablation (LA)-ICP-MS with on-tissue internal standard addition is described. This simple approach enabled on-tissue addition of Rh as internal standard to samples (with heterogeneous Fe distribution) and calibrants (with homogeneous Fe distribution). This is achieved without altering the original Fe distribution of the sample. Calibration standards were prepared by full horizontal immersion of slides with mounted homogenised sheep brain tissue section into the corresponding solution containing 0.5, 0.75, 1, 5, 10 and 20 mg kg−1 Fe (each also containing 250 µg kg−1 Rh as IS) in pure methanol for 30 minutes (6 immersions, each for 5 minutes). Subsequent air-drying (bench drying at room temperature) for approximately 5 minutes was undertaken in between consecutive immersions, to prevent long-term exposure of the tissue to lipid degradation. Tissue-matched standards were characterised in-house for Fe composition, homogeneity and stability (at storage temperatures of −80 °C, −20 °C, 4 °C and 25 °C for up to 2 months) in order to investigate their suitability as calibrants for quantitative LA-ICP-MS. The homogeneity data suggested that the materials are homogeneous in terms of Fe and Rh distribution with RSDs (n = 30) of 8.3% and 4.7%, respectively. The Fe measurement precision was improved by approximately a factor of 2 when normalising 56Fe intensities to 103Rh intensities; the RSD (n = 30) for 56Fe/103Rh was 3.6%. The produced calibration standards were found to be stable when stored at room temperature for approximately 50 days, suggesting that they can be reused for multiple batches. Using LA coupled to double-focusing sector field ICP-MS in medium resolution mode (m/Δm = 4000), linear calibration over a range of 107 to 1519 mg kg−1 Fe (R2 = 0.99) was achieved with a limit of detection of 1.84 mg kg−1 Fe. Assessment of the accuracy of the method for the quantitative imaging of Fe in tissues was undertaken by comparison of the LA-ICP-MS data with that obtained by micro-XRF; the average Fe concentrations in selected tissue regions obtained by using XRF fell within the window defined by the LA-ICP-MS values and their associated standard deviations.

Larvae of Mytilus spp. are among the most Cu sensitive marine species. In this study we assessed the combined effect of salinity and dissolved organic carbon (DOC) on Cu accumulation on mussel larvae. Larvae were exposed for 48 h to three Cu concentrations in each of nine salinity/DOC treatments. Synchrotron radiation X-ray fluorescence was used to determine the Cu concentration in 36 individual larvae with a spatial resolution of 10 × 10 μm. Cu body burden concentrations varied between 1.1 and 27.6 μg/g DW larvae across all treatments and Cu was homogeneously distributed at this spatial resolution level. Our results indicate decreasing Cu accumulation with increasing DOC concentrations which can be explained by an increase in Cu complexation. In contrast, salinity had a nonlinear effect on Cu. This cannot be explained by copper speciation or competition processes and suggests a salinity-induced alteration in physiology.

International audience; The Stardust Interstellar Preliminary Examination team analyzed thirteen Al foils from the NASA Stardust interstellar collector tray in order to locate candidate interstellar dust (ISD) grain impacts. Scanning electron microscope (SEM) images reveal that the foils possess abundant impact crater and crater-like features. Elemental analyses of the crater features, with Auger electron spectroscopy, SEM-based energy dispersive X-ray (EDX) spectroscopy, and scanning transmission electron microscope-based EDX spectroscopy, demonstrate that the majority are either the result of impacting debris fragments from the spacecraft solar panels, or intrinsic defects in the foil. The elemental analyses also reveal that four craters contain residues of a definite extraterrestrial origin, either as interplanetary dust particles or ISD particles. These four craters are designated level 2 interstellar candidates, based on the crater shapes indicative of hypervelocity impacts and the residue compositions inconsistent with spacecraft debris.

The unique potential of nanoscale elemental imaging of major/minor and trace-level elemental distributions within thin biological tissue sections of the ecotoxicological model organism Daphnia magna is demonstrated by synchrotron radiation nano-X-ray fluorescence (nano-XRF). The applied highly specialized sample preparation method, coupled with the high spatial resolution (∼180 nm) and high X-ray photon flux (6 × 10(11) photons/s) available at the European Synchrotron Radiation Facility (ESRF) ID22NI beamline proved to be critical for the high-quality visualization of (trace-)metal distributions on the submicron level within the target structures of interest. These include the branchial sacs on the thoracic appendages (epipodites) of D. magna, which are osmoregulatory regions where ion exchange occurs. For the main element of interest (Zn), detection limits of 0.7 ppm (3 ag) was reached in fast-scanning mode using an acquisition time of 0.3 s/pixel. As demonstrated, synchrotron radiation nano-XRF revealed the elemental distributions of Ca, Fe, and Zn within this osmoregulatory region on the submicron scale, aiding the exploration of possible detoxification mechanisms of Zn within D. magna at the subtissue level.

The development of highly brilliant synchrotron facilities all around the world is opening the way to new research in biological sciences including speciation studies of trace elements in plants. In this paper, for the first time, iron (Fe) speciation in xylem sap has been assessed by X-ray absorption near-edge structure (XANES) spectroscopy at the highly brilliant synchrotron PETRA III, beamline P06. Both standard organic Fe-complexes and xylem sap samples of Fe-deficient tomato plants were analyzed. The high photon flux provided by this X-ray synchrotron source allows on one side to obtain good XANES spectra in a reasonable amount of time (approx. 15 min for 200 eV scan) at low Fe concentrations (sub parts-per-million), while on the other hand may cause radiation damage to the sample, despite the sample being cooled by a stream of liquid nitrogen vapor. Standard Fe-complexes such as Fe(III)-succinate, Fe(III)-α-ketoglutarate, and Fe(III)-nicotianamine are somehow degraded when irradiated with synchrotron X-rays and Fe(III) can undergo photoreduction. Degradation of the organic molecules was assessed by HPLC-UV/Vis analyses on the same samples investigated by X-ray absorption spectroscopy (XAS). Fe speciation in xylem sap samples revealed Fe(III) to be complexed by citrate and acetate. Nevertheless, artifacts created by radiation damage cannot be excluded. The use of highly brilliant synchrotrons as X-ray sources for XAS analyses can dramatically increase the sensitivity of the technique for trace elements thus allowing their speciation in xylem sap. However, great attention must be paid to radiation damage, which can lead to biased results.

Conventionally, this information can be obtained in a spatially resolved manner by scanning a sample through a focused X-ray beam. Recently, full-field methods have been developed to obtain direct 2D chemical state information by imaging a large sample area. These methods are usually in transmission mode, thus restricting the use to thin and transmitting samples. Here, a fluorescence method is displayed using an energy-dispersive pnCCD detector, the SLcam, characterized by measurement times far superior to what is generally applicable. Additionally, this method operates in confocal mode, thus providing direct 3D spatially resolved chemical state information from a selected subvolume of a sample, without the need of rotating a sample. The method is applied to two samples: a gold-supported magnesia catalyst (Au/MgO) and a natural diamond containing Fe-rich inclusions. Total reflection X-ray fluorescence (TXRF) analysis is an excellent tool to determine major, minor and trace elements in minuscule amounts of samples, making this technique very suitable for pigment analysis. Collecting minuscule amounts of pigment material from precious works of art by means of a cotton swab is a well-accepted sampling method, but poses specific challenges when TXRF is to be used for the characterization of the unknown material.

Ovarian cancer is amongst the most common types of cancer in women, with a relatively low overall cure rate of approximately 30%. This is therefore an important incentive to urge for further research in order to maximize the chances of survival for these patients. Intraperitoneal chemotherapy with Cisplatin is an effective treatement for ovarian cancer; however, many questions still remain concerning the ideal treatment protocol and tumor resistance towards the drug, which should be resolved for optimal application of this therapy. For the first time in-vivo grown tumors treated with both hyper- and normothermic intraperitoneal chemotherapy have been studied using nano-XRF spectroscopy to examine the platinum (Pt) distribution within the analyzed tissues. These measurements prove Pt resides predominantly outsides the cancer cells in the stroma of the tissue. These findings indicate the resistance mechanism of the cancer cells prevents Cisplatin from diffusing through their cell membranes. This is an important addition to the existing knowledge on the resistance mechanism providing insights which might help to overcome this effect. In our aim to find the optimal treatment protocol, no significant differences were found between the two examined procedures. A more extensive data set will be needed to draw definite conclusions.

A new method for the quantification of X-ray fluorescence (XRF) was derived based on the fundamental parameter method (FPM). The FPM equations were adapted to accommodate the special case of confocal nano-XRF, i.e. X-ray nano-beam excitation coupled with confocal detection, taking into account the special characteristics of the detector channel polycapillary. A thorough error estimation algorithm based on the Monte Carlo method was applied, producing a detailed analysis of the uncertainties of the quantification results. The new FPM algorithm was applied on confocal nano-XRF data obtained from cometary dust returned by NASA's Stardust mission, recorded at beamline ID13 of the European Synchrotron Radiation Facility.

Recent studies have suggested that exposure of the freshwater invertebrate Daphnia magna to dietary Zn may selectively affect reproduction without an associated increase of whole body bioaccumulation of Zn. The aim of the current research was therefore to investigate the hypothesis that dietary Zn toxicity is the result of selective accumulation in tissues that are directly involved in reproduction. Since under field conditions simultaneous exposure to both waterborne and dietary Zn is likely to occur, it was also tested if accumulation and toxicity under combined waterborne and dietary Zn exposure is the result of interactive effects. To this purpose, D. magna was exposed during a 16-day reproduction assay to Zn following a 5 × 2 factorial design, comprising five waterborne concentrations (12, 65, 137, 207, and 281 μg Zn/L) and two dietary Zn levels (49.6 and 495.9 μg Zn/g dry wt.). Tissue-specific Zn distribution was quantified by synchrotron radiation based confocal X-ray fluorescence (XRF). It was observed that the occurrence of reproductive inhibition due to increasing waterborne Zn exposure (from 65 μg/L to 281 μg/L) was accompanied by a relative increase of the Zn burdens which was similar in all tissues considered (i.e., the carapax, eggs, thoracic appendages with gills and the cluster comprising gut epithelium, storage cells and ovaries). In contrast, the impairment of reproduction during dietary Zn exposure was accompanied by a clearly discernible Zn accumulation in the eggs only (at 65 μg/L of waterborne Zn). During simultaneous exposure, bioaccumulation and toxicity were the result of interaction, which implies that the tissue-specific bioaccumulation and toxicity following dietary Zn exposure are dependent on the Zn concentration in the water. Our findings emphasize that (i) effects of dietary Zn exposure should preferably not be investigated in isolation from waterborne Zn exposure, and that (ii) XRF enabled us to provide possible links between tissue-specific bioaccumulation and reproductive effects of Zn.

The Breviary of Arnold of Egmond is one of the most wealthily illuminated fifteenth century manuscripts in the Northern Netherlands. The manuscript originally contained a number of full-page miniatures, which were all removed at an unknown date before 1902. The three remaining miniatures studied here, are today part of different collections, but they were brought together for an exhibition. Although several historical and art historical details of this breviary have extensively been studied, no examination of the materials used was undertaken before. Analytical techniques, such as mobile Raman spectroscopy, can be used to characterise and identify these materials in a non-invasive way. This paper presents the results of the in situ Raman analysis of three full-page miniatures of the Breviary of Arnold of Egmond. During this study, different pigments could be identified, such as lead white (2PbCO(3)·Pb(OH)(2)), lead-tin yellow type I (Pb(2)SnO(4)), ultramarine (Na(8-10)Al(6)Si(6)O(24)S(2-4)), massicot (PbO), vermilion (HgS) and red lead (Pb(3)O(4)). Next to identification of the pigments, visual analysis was used to detect differences and similarities between the stylistic elements of the three analysed folios.

A selection of illuminations of the 12th century manuscript Liber Floridus was analysed with Raman spectroscopy (in situ and laboratory measurements), X-ray fluorescence spectroscopy, UV-fluorescence photography and infrared reflectography (IRR). The aim of this study is to determine the pigments used, in order to search for anachronisms. Using a combination of Raman spectroscopy (molecular information) and X-ray fluorescence spectroscopy (elemental information) following pigments could be identified: ultramarine (Na(8-10)Al(6)Si(6)O(24)S(2-4)), azurite (2CuCO(3)·Cu(OH)(2)), caput mortuum (Fe(2)O(3)), vermilion (HgS), orpiment (As(2)S(3)) and lead white (2PbCO(3)·Pb(OH)(2)). Moreover, two synthetic red pigments, PR4 and PR176, and a degradation product, gypsum (CaSO(4)·2H(2)O), were present in the manuscript. To establish the origin of the modern materials UV-fluorescence photography was used. Infrared reflectography (IRR) was applied to visualise the underdrawing of the investigated folios.

The enamel of 20 Email Champlevé objects dating between the 12th and 19th centuries was investigated by means of microscopic and portable X-ray fluorescence analysis (μ-XRF and PXRF). Seven of these objects were microsampled and the fragments were analyzed with electron probe microanalysis (EPMA) and vacuum μ-XRF to obtain quantitative data about the composition of the glass used to produce these enameled objects. As a result of the evolution of the raw materials employed to produce the base glass, three different compositional groups could be discriminated. The first group consisted of soda-lime-silica glass with a sodium source of mineral origin (with low K content) that was opacified by addition of calcium antimonate crystals. This type of glass was only used in objects made in the 12th century. Email Champlevé objects from the beginning of the 13th century onward were enameled with soda-lime-silica glass with a sodium source of vegetal origin. This type of glass, which has a higher potassium content, was opacified with SnO2 crystals. The glass used for 19th century Email Champlevé artifacts was produced with synthetic and purified components resulting in a different chemical composition compared to the other groups. Although the four analytical techniques employed in this study have their own specific characteristics, they were all found to be suitable for classifying the objects into the different chronological categories.

A stream of 1-20 μm sized mineral inclusions having the negative crystal shape of its host within an "ultra-deep" diamond from Rio Soriso (Juina area, Mato Grosso State, Brazil) has been studied with confocal μ-X-ray absorption near edge structure (μXANES) at the Fe K and Mn K edges. This technique allows the three-dimensional nondestructive speciation of the Fe and Mn containing minerals within the inclusion cloud. The observed Fe-rich inclusions were identified to be ferropericlase (Fe,Mg)O, hematite and a mixture of these two minerals. Confocal μ-X-ray fluorescence (μXRF) further showed that Ca-rich inclusions were present as well, which are spatially separated from or in close contact with the Fe-rich inclusions. The inclusions are aligned along a plane, which most likely represents a primary growth zone. In the close vicinity of the inclusions, carbon coated planar features are visible. The three-dimensional distribution indicates a likely fluid overprint along an open crack. Our results imply that an imposed negative diamond shape of an inclusion alone does not exclude epigenetic formation or intense late stage overprint.

Micro X-ray fluorescence analysis (μ-XRF) is one of the newest branches of XRF, which has developed very rapidly since 1990. In this review, we first discuss a number of papers that describe advances in instrumentation and (calibration) methods that can contribute towards more reliable quantitative analysis using μ-XRF. This first part also includes recent papers that aim to characterize the heterogeneity of standards useful for calibrating μ-XRF set-ups. We then present a short overview of recently published papers in which the (semi-)quantitative nature of μ-XRF is exploited, including studies in which μ-XRF is compared to other microanalytical methods providing compositional information or is used in combination with other methods in order to characterize materials more completely.

We detected additional CAI-like material in STARDUST mission samples of comet 81P/Wild 2. Two highly refractory cometary dust fragments were identified in the impact track 110 [C2012, 0, 110, 0, 0] by applying high resolution synchrotron induced confocal and conventional XRF analysis (HR SR-XRF). The use of a polycapillary lens in front of the detector for confocal spectroscopy dramatically improves the fidelity of particle measurements by removing contribution from the surrounding aerogel. The high spatial resolution (300 × 300 nm2; 300 × 1000 nm2) obtained allowed the detailed non-destructive in situ (trapped in aerogel) study of impacted grains at the sub-μm level. For the two largest particles of the track, the terminal particle and a second particle along the impact track, Ca concentration is up to 30 times higher than CI and Ti is enriched by a factor of 2 compared to CI. High resolution (HR) SR-XRF mapping also reveals that the highest concentrations of Ca, Ti, Fe (and Ni) measured within each grain belongs to different areas of the respective maps which indicate that the particles are composed of several chemically diverse mineral phases. This is in agreement with the finding of a complex phase assemblage of highly refractory minerals in the first ever detected Stardust mission CAI grain “Inti” of Track 25. Principle component analysis (PCA) is a powerful tool for extracting the dominant mineral components and was applied to the two grains indicating that regions in the terminal particle and the second particle are consistent with anorthite or grossite and gehlenite, monticellite or Dmitryivanovite (CaAl2O4), respectively. Our new findings demonstrate that the HR SR-XRF with confocal geometry and PCA analysis is capable of identifying CAI-like fragments without the need to extract particles from the aerogel matrix which is a time-consuming, complex and destructive process. Furthermore, the detection of new CAI-like fragments in the coma dust of comet 81P/Wild 2 strengthens the observation that strong mixing effects and, therefore, mass transport before or during comet formation must have occurred at least up to the region where Kuiper Belt comets formed (∼30 AU).

Using a significantly improved method for X-ray fluorescence (XRF) tomography with submicrometer spatial resolution, the fully three-dimensional (3D) distribution of main and trace elements from calcium to selenium was determined at a spatial resolution level of 200 nm within a unique (∼2 μm) comet coma particle brought to Earth by the National Aeronautical and Space Administration (NASA)’s Stardust mission. The measurements were based on synchrotron radiation XRF nanotomography that has been applied to the terminal particle located in an aerogel host, which was used to capture comet and interstellar grain particles in space. Using a symmetric dual-detector arrangement for the presented nano-XRF tomography measurements, a rotation angle of 180° was sufficient for the tomographic reconstruction. Dedicated correction algorithms were developed to correct for submicrometer-level distortions within the tomographic dataset arising from instabilities of the sample positioning system during data collection.

The aim of the present paper is to characterize a cross-linked horse heart cytochrome c (HHC) film on cysteamine-modified gold electrodes. The HHC film was deposited using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC) as a coupling agent. Attenuated total reflection infrared (ATR-IR) spectroscopic analyses were performed to characterize the newly formed surface on a qualitative and conformational level. The film thickness was measured using a noncontact optical surface profiler, while quantitative data and information on the heterogeneity of the film were obtained by means of synchrotron radiation X-ray micro fluorescence (SR micro-XRF). Results indicate that, in addition to electrochemical studies, spectroscopic analysis methods are essential to gain insight in the effect of immobilization strategies on protein conformations. New reference materials consisting of cobalt thin films on gold were prepared by sputter deposition. The thickness and homogeneity of the films were characterized using synchrotron radiation micro-XRF. The samples can be used as reference materials to quantify cobalt phthalocyanine and cobalt porphyrin modified gold electrodes which have been analyzed with synchrotron radiation micro-XRF.

A new laboratory scale X-ray fluorescence (XRF) imaging instrument, based on an X-ray microfocus tube equipped with a monocapillary optic, has been developed to perform XRF computed tomography experiments with both higher spatial resolution (20 μm) and a better energy resolution (130 eV @Mn-K(α)) than has been achieved up-to-now. This instrument opens a new range of possible applications for XRF-CT. Next to the analytical characterization of the setup by using well-defined model/reference samples, demonstrating its capabilities for tomographic imaging, the XRF-CT microprobe has been used to image the interior of an ecotoxicological model organism, Americamysis bahia. This had been exposed to elevated metal (Cu and Ni) concentrations. The technique allowed the visualization of the accumulation sites of copper, clearly indicating the affected organs, i.e. either the gastric system or the hepatopancreas. As another illustrative application, the scanner has been employed to investigate goethite spherules from the Cretaceous-Paleogene boundary, revealing the internal elemental distribution of these valuable distal ejecta layer particles.
