This study evaluates the influence of inadequate transport conditions on the microbiological quality of chilled chicken meat packaged in plain and modified atmosphere packaging (MAP). The experiments simulated the temperature increase during sample transport to 8, 11, 14, 17, 20, and 25°C with exposure times of 1, 2, 3, and 4 h. Aerobic plate count (APC), psychrotrophic microorganisms count (PMC), β-D-glucuronidase-positive Escherichia coli, and Salmonella spp. were evaluated immediately after the exposure to the elevated temperature (0 h), 3 h, and 24 h after the return to the temperature of ≤4°C. The upper acceptable limits for APC and PMC were set for each combination of investigated chicken meat and packaging type, taking also the initial bacterial condition into account. Chilled chicken breast samples in plain packaging exceeded the APC limits in 16 cases and PMC limits in 20 cases when exposed to temperatures of >4°C, while only 2 MAP samples exceeded APC limits and 8 samples PMC limits, respectively. In chicken legs, 8 samples in plain packaging exceeded the APC limits and 15 the PMC limits, while 12 samples in MAP exceeded the APC limits and 19 the PMC limits. In 402 samples (31.9%) in which the presence of E. coli was detected, its amount ranged from 1.70 to 3.65 log CFU.g−1. It was more commonly detected in chicken legs (255 of 630; 40.5%) than chicken breasts (147 of 630; 23.3%) but was not related to exposure temperature, exposure time, or time until examination. The presence of Salmonella spp. was not detected in any of the samples. Data acquired in the presented study will be used in the development of software helping the national supervisory authorities in the Czech Republic to evaluate whether inadequate transport of samples to analytical laboratories could have affected the microbiological profile of the sample.

With ever-developing analytical capabilities, the protection of the dairy chain from contamination by residues of veterinary drugs is improving. Legislative requirements are an inherent part of this process. Of antibiotics in dairy farming, representatives of the beta-lactams, tetracyclines, sulfonamides, (fluoro)quinolones, aminoglycosides, or polypeptide antibiotics are the most widely used. Due to the typically low levels of antibiotic residues in milk, mass spectrometry is the most commonly used detection technique. However, the interference of the sample matrix is one of its main limiting factors, and therefore, it needs to be eliminated. In the first step, the lipid fraction is removed and proteins are precipitated, followed by solid-phase or liquid–liquid extraction. The current trends include the reduction in the consumption of organic solvents (to reduce occupational hazards and burden to the environment) and automation, eliminating the influence of human error and optimizing the workflow. These trends lead to the development of new microextraction and automated techniques as well as the use of new sorbents and/or (green) solvents of natural origin. To capture the latest developments in the field and the relatively recent aforementioned trends, this review focuses on papers investigating antimicrobial residues in milk that were published between 2015 and 2024, providing an overview of emerging techniques applicable to antibiotic residue detection in milk samples.

The presence of residues of antimicrobial substances in milk has been an important hygienic and technological parameter of raw milk quality since the 1960s. The presented review focuses on screening methods (microbiological inhibition methods and rapid specific tests) that are used in the control of antimicrobial residues in milk in the context of their historical development up to the present. We briefly explain the principles of the methods and discuss their pros and cons. The aim was to provide both the historical perspective on this topic and provide useful information on screening methods that are currently routinely used for the detection of residues of antimicrobials at farms, in the dairy industry, and in milk quality control laboratories.

The main objective of this work was to determine the significant physicochemical and bioactive parameters of honeys originating from Iraqi Kurdistan. For this purpose, honey samples (n = 16) were taken directly from Kurdish beekeepers from different places of Dohuk and Erbil Governorate in 2018 and 2022. The following parameters were analyzed: moisture content, electrical conductivity, diastase activity, hydroxymethylfurfural content, pH, free acidity, content of saccharides, total phenolic content, total flavonoid content, color, color intensity and DPPH, FRAP. Another objective of this work was to compare the properties of Kurdistan honeys with the requirements of European Union law and with the honeys of Central Europe. A total of 48 honey samples were included in the comparison. It was detected that the honeys produced in Iraqi Kurdistan met the strict quality criteria set by European law, which is important information for local beekeepers and experts in relation to honey production and an opportunity to set local limits of honey quality. Despite the different climatic and topographical conditions, the available flora, and the different species of bees, the properties of Iraqi Kurdistan honeys were similar to the properties of honeys from the Czech and Slovak Republics.

Enterococci are a group of microorganisms that have a controversial position from some scientific points of view. The species of the greatest clinical importance are E. faecalis and E. faecium, which are common agents of nosocomial infections. However, enterococci also have important applications in the dairy industry, as they are used as non-starter lactic acid bacteria (NSLAB) in a variety of cheeses, especially artisanal cheeses. The aim of this study was to determine the presence of representatives from the Enterococcus genus using PCR and MALDI-TOF MS methods on samples of raw milk, processing environment swabs, and cheese from four different artisanal dairy plants in Slovakia. Among the 136 isolates of enterococci, 9 species of genus Enterococci (E. faecalis, E. faecium, E. durans, E. devriesi, E. hirae, E. italicus, E. casseliflavus, E. malodoratus, and E. gallinarum) were identified and were tested for their antimicrobial resistance (AMR) to 8 antibiotics (amoxicillin, penicillin, ampicillin, erythromycin, levofloxacin, vancomycin, rifampicin, and tetracycline); most of them were resistant to rifampicin (35.3%), ampicillin (22.8%), and tetracycline (19.9%). A PCR analysis of vanA (4.41%) and tetM (14.71%) revealed that antimicrobial resistance genes were present in not only phenotypic resistant isolates of enterococci but also susceptible isolates. The investigation of antimicrobial resistance in enterococci during the cheesemaking process can be a source of valuable information for public health in the concept of “One Health”.

Multiple factors can facilitate invasion success, with the absence of natural enemies, such as predators and parasites, recognised as conferring a significant advantage on invasive over native species. Pumpkinseed Lepomis gibbosus (Centrarchidae) represents one of the most successful freshwater fish invaders in Europe. Previous research has highlighted genetic differences between pumpkinseed populations in Türkiye and those in other European regions, attributed to rapid adaptation to new environmental conditions. This study aimed to investigate whether these highly adapted pumpkinseed populations in Türkiye benefit from a potential release from parasites, as proposed by the enemy-release hypothesis. Genetic characterisation of pumpkinseed populations from both European and Asian parts of Türkiye revealed that they share the same cytochrome c oxidase I haplotype as European populations. Microsatellite analysis indicated low genetic diversity, with STRUCTURE analysis confirming the clustering of all Turkish populations, suggesting a common source. Consistent with the low genetic diversity indicative of a small founding population, we observed a limited number of co-introduced parasite species, including the myxozoan Myxobolus dechtiari, the monogenean Onchocleidus dispar, and the digenean Posthodiplostomum centrarchi. Parasite infection by local parasites acquired in Türkiye was rare. Parasite diversity, species richness, and equitability were low, with only nine parasite taxa identified in all four pumpkinseed populations. The most diverse parasite community was found in Değirmenköy Reservoir, located in the European part of Türkiye, where seven parasite taxa were identified. While our study did not uncover genetically distinct pumpkinseed populations in Türkiye, the fish demonstrated resilience against most local parasite species, potentially providing them with an advantage over native species, aligning with the enemy-release hypothesis.

The aim of this study was to evaluate the impact of sumac on the oxidation status, sensory properties, and physicochemical and microbiological parameters of common carp (Cyprinus carpio) fish during chilled storage. Fillets of carp were treated with ground sumac at percentages of 0% (T0), 0.5% (T1), 1.5% (T2) and 2.5% (T3) and analysed after 24 and 72 h of chilled storage. The fat content, oxidation parameters (thiobarbituric acid reactive substances, TBARS), antioxidant capacity, total volatile basic nitrogen (TVB-N), free fatty acids (FFA), sensory properties, colour indicators (lightness L*, redness a*, yellowness b*), water activity, pH value, aerobic plate count (APC), psychrotrophic microorganism count (PMC), and coliform bacteria count were determined. The TBARS values of T1, T2, and T3 decreased significantly (p < 0.05) after 24 h of chilled storage. The antioxidant capacity increased in a dose-dependent and time-dependent manner. As regards organoleptic properties, sumac was able to reduce the natural fishy odour, increase the sour odour and flavour, and mask the fish odour and flavour significantly. A significant effect of sumac on the redness (a*) values of muscles was observed, which increased with elevation of the sumac ratio. A decrease in fillet pH and lower APC and PMC were observed. No effect was found on water activity or the growth of coliform bacteria. The study found that sumac plays a role in the oxidation status, pH value and organoleptic properties of fish fillets, which could be beneficial for the fish and fishery products industry.

Abstract Background Gestational diabetes mellitus (GDM) represents a risk factor for both mother and her offspring in a short-term (perinatal morbidity) and long-term horizon (postpartum diabetes or foetal programming). Several studies focused at peri/postnatal outcomes of GDM mother´s offspring, however relatively few (and none in Czech population) were designed as prospective. The aim of the study was to ascertain eventual anthropometric and developmental abnormalities and/or morbidity in offspring of GDM mothers compare to controls in a 5-year follow-up using a parent-reported parameters related to psychomotor development and common paediatric morbidities including a sub-study of offspring of GDM mothers experiencing adverse perinatal outcomes. Methods A 5 year follow up study of offspring of GDM mothers (n = 26) vs those with a normal pregnancy (n = 63). An electronic questionnaire was used to obtain the parameters (such as growth, psychomotor development, vaccination, morbidity history etc.) available to parents from the parent-held infant health record. Data on pregnancy and delivery were available from the previous study. Results Offspring of GDM mothers had delayed psychomotor development in early childhood, but in 5 years of age they seemed to gradually achieve results of a control group. Children with macrosomia had a higher percentile of weight-for-height and were significantly more frequently ill than those with a normal birth weight. Offspring of obese mothers had worse verbal language skills in early childhood and a higher percentile of weight-for-height. Conclusion Maternal gestational diabetes and obesity can be considered an important determinant of postnatal offspring development and health status, which further advocates for broader implementation of preventive strategies.

Objectives: To report the clinical outcomes of the Victo™ (Promedon, Cordoba, Argentina) adjustable artificial urinary sphincter (AUS) implantation in a cohort of patients with severe urinary incontinence (UI) after prostate surgery. Patients and Methods: This study enrolled patients with UI following prostate surgery who underwent a Victo implantation between May 2018 and December 2023. Patients were prospectively evaluated at baseline, and at 3 and 12 months after device activation, and thereafter annually. The 24‐h pad‐weight test (24hPWT) was used to assess severity of UI, while the Patient Global Impression of Improvement (PGI‐I) and patient satisfaction according to a Likert scale were used to measure patient‐reported outcomes. Results: A total of 96 patients with a median (interquartile range [IQR]) age of 68 (65–72) years were included in the final analysis. The median (IQR) follow‐up was 3 (1–4) years. In all, 10 patients completed the 5‐year follow‐up. After the treatment, we observed a significant reduction in 24hPWT by the median of 83% (P < 0.001) at 3 months and by a median of 79% (P < 0.001) at 3 years. According to the PGI‐I, a total of 87%, 92%, 87%, 81%, 83%, and 50% (five of 10) of patients rated their condition/incontinence as 'very much improved', 'much improved' or 'little improved' at 3 months, 1‐, 2‐, 3‐, 4‐, and 5‐year follow‐up visits, respectively. The proportion of patients, who were 'very satisfied' or 'satisfied' with the treatment outcome was 79%, 80%, 75%, 69%, 80%, and 60% (six of 10) at 3 months, 1‐, 2‐, 3‐, 4‐, and 5‐years, respectively. There were a total of 13 (14%) device failures during the follow‐up period. Conclusion: In conclusion, our data suggest that Victo AUS significantly reduces the severity of UI after prostate surgery and provides a reasonably high patient‐reported satisfaction with treatment outcomes at mid‐term follow‐up. Synodontis Cuvier, 1816 is a species-rich group of African catfishes. Prior to this analysis, 13 species of Synodontis were recognized from Lake Tanganyika and its tributaries, composing the only extant lacustrine radiation of the genus, with a unique origin of obligate brood parasitism among all fishes. Species differentiation and phylogenetic relationships of this group remained largely unresolved. Herein, the taxonomy of Synodontis in the Lake Tanganyika basin is revised, including redescriptions of all species and an updated identification key. Genetic (mitochondrial and nuclear genomic markers) and morphological data suggest a lower species diversity than previously recognized, supporting the distinction of 10 Synodontis species in the lake basin. Based on our findings, we recognize Synodontis grandiops , Synodontis ilebrevis , and Synodontis lucipinnis as junior synonyms of Synodontis multipunctatus , Synodontis polli , and Synodontis petricola , respectively. No recent material of Synodontis dhonti and Synodontis tanganyicae (with Synodontis lacustricolus as a synonym) was available. Morphometric/meristic data suggest their close relationship to Synodontis granulosus. Sequencing of historical specimens inferred S. tanganyicae as sister to S. granulosus but with a 1.7% divergence based on mitochondrial data. In contrast to previous studies, phylogenomic data support the lacustrine assemblage of Synodontis in Lake Tanganyika as monophyletic. [ABSTRACT FROM AUTHOR]

The aim of this article was to evaluate the amount of salt dosed in various proportions into minced pork and beef. Determination of salt was conducted by the atomic absorption spectrometry (AAS) method and, for comparison, by the near infrared spectroscopy (NIR) method. The meat (beef neck and pork topside) was ground through a plate with a hole size of 8 mm. Salt doses were from 0 to 2.0%, at intervals of 0.2%. Each sort of meat was divided into two parts of 350 g: 1) with initial grinding (coarse grinding), and 2) with subsequent grinding through a plate with a hole size of 3 mm (fine grinding). The salt content in fresh pork meat (0% salt) ranged between 0.13 and 0.16%, whereas the content in beef ranged from 0.14 to 0.19% (as analysed by the AAS method). The proportion of added salt had a statistically significant effect on the salt content in meat. No significant differences were found between coarse and fine grinding and between pork and beef meat. The predicted natural salt content in meat was 0.15 and 0.19% for pork, and 0.20 and 0.21% for beef. The proportion of analysed salt was then determined as 0.99-1.06 times the dosed salt. The authors do not recommend the use of the NIR method, which provides reliable results for the main chemical components of meat, i.e. protein and fat, for the determination of the proportions of salt in meat. In recent years, pumpkinseed Lepomis gibbosus (Actinopterygii: Centrarchidae) have spread intensively to many parts of Europe, including Ukraine. In this study, we (1) assess intra-population genetic variability in five widely-spaced Ukrainian pumpkinseed populations and compare their population genetic structure with other European populations and (2) provide a comprehensive survey of pumpkinseed parasites across the region. Discriminant analysis of principal components and FST analyses based on microsatellites indicated that all five populations formed discrete clusters. Within Europe, Ukrainian populations were most closely associated with populations from the River Danube, suggesting that the Danube is the main source of all Ukrainian pumpkinseed populations. The parasite fauna comprised 15 taxa, most of which (92%) were native to North America (monogenea Onchocleidus similis and O. dispar; myxozoan Myxobolus dechtiari). Parasites acquired in the species' new range occurred accidentally, with only Trichodina ciliates found relatively frequently and the eye flukes Diplostomum pseudospathaceum and Tylodelphys clavata occurring at higher prevalence. Absence of specific monogeneans in pumpkinseed from estuaries (Khadzhibey and Sukhyi Lymans) indicates low tolerance of Onchocleidus species to salinity.

Abstract Background African annual killifishes (Nothobranchius spp.) are adapted to seasonally desiccating habitats (ephemeral pools), surviving dry periods as dormant eggs. Given their peculiar life history, geographic aspects of their diversity uniquely combine patterns typical for freshwater taxa (river basin structure and elevation gradient) and terrestrial animals (rivers acting as major dispersal barriers). However, our current knowledge on fine-scale inter-specific and intra-specific genetic diversity of African annual fish is limited to a single, particularly dry region of their distribution (subtropical Mozambique). Using a widespread annual killifish from coastal Tanzania and Kenya, we tested whether the same pattern of genetic divergence pertains to a wet equatorial region in the centre of Nothobranchius distribution. Results In populations of Nothobranchius melanospilus species group across its range, we genotyped a part of mitochondrial cytochrome oxidase subunit 1 (COI) gene (83 individuals from 22 populations) and 10 nuclear microsatellite markers (251 individuals from 16 populations). We found five lineages with a clear phylogeographic structure but frequent secondary contact. Mitochondrial lineages were largely congruent with main population genetic clusters identified on microsatellite markers. In the upper Wami basin, populations are isolated as a putative Nothobranchius prognathus, but include also a population from a periphery of the middle Ruvu basin. Other four lineages (including putative Nothobranchius kwalensis) coexisted in secondary contact zones, but possessed clear spatial pattern. Main river channels did not form apparent barriers to dispersal. The most widespread lineage had strong signal of recent population expansion. Conclusions We conclude that dispersal of a Nothobranchius species from a wet part of the genus distribution (tropical lowland) is not constrained by main river channels and closely related lineages frequently coexist in secondary contact zones. We also demonstrate contemporary connection between the Ruvu and Rufiji river basins. Our data do not provide genetic support for existence of recently described cryptic species from N. melanospilus complex, but cannot resolve this issue.
