Blood vessel walls are made of organized fibrous tissue with intrinsic birefringence. Even in its very early stages, hypertension can change the structure of a blood vessel wall. We showed that this structural change can be quantitatively measured non-invasively in the human retina using polarization-sensitive optical coherence tomography (PS-OCT). Organizational loss in vessel wall tissue was quantified in hypertensive patients and compared to data obtained from age-matched healthy subjects. The wall tissue in patients with hypertension was shown to be thicker, and exhibited lower birefringence, presumably due to a loss of tissue organization. The blood vessel wall birefringence index (BBI) combines vessel wall birefringence and thickness into one number and is introduced here to readily distinguish between blood vessel walls of hypertensive and healthy subjects. Based on receiver operating characteristic (ROC) curves, BBI showed 99% sensitivity and 100% specificity when discriminating normotensive (N = 11) and hypertensive (N = 11) subjects. Accurately determining the thickness of the blood vessel wall is not possible without polarization-sensitivity. Moreover, just blood vessel wall thickness or wall birefringence were not sufficient to achieve this high classification performance. Retinal vessel wall measurements with PS-OCT cannot be affected by a patient being anxious, by hormones or other blood values, since the measurement is intrinsic to the optical and mechanical properties of the vessel wall. Furthermore, this relatively low-cost system combines a very short imaging and analysis time with high sensitivity and specificity, making it highly suitable for low-cost screening.

Two designs with a multiplexed superluminescent diode for ultra-high-resolution spectral-domain polarization-sensitive optical coherence tomography (UHR-PS-OCT) are introduced. In the first design, a Wollaston prism separates orthogonal polarization states next to each other on one linescan camera; the other design uses a beam displacer to separate orthogonal states onto two lines of a linescan camera with multiple rows of detectors. The coherence lengths measured with the two systems were 3.6 µm and 2.9 µm ( n = 1.38 ), respectively. Measurements were collected from the fovea of a healthy subject, a healthy subject with astigmatism, and a patient with central serous retinopathy (CSR). A single volumetric scan provides double pass retardance induced by the retinal nerve fiber layer birefringence (RNFL) and its birefringence, the cumulative double pass retardance induced by the Henle fiber layer, and the retardance that is induced by the retinal pigment epithelium–Bruch’s membrane complex. The high axial resolution in UHR-PS-OCT is particularly helpful for the measurements of thin retinal tissue, such as the RNFL in the fovea, where birefringence values of around 1°/µm were found. Tilting of the retina due to a CSR or by off centering the imaging beam in the pupil causes an artificial increase in the double pass retardance induced by the RNFL and Henle fiber layer.

Clinical visualization and quantification of the amount and distribution of airway smooth muscle (ASM) in the lungs of individuals with asthma has major implications for our understanding of airway wall remodeling as well as treatments targeted at the ASM. This paper theoretically investigates the feasibility of quantifying airway wall thickness (focusing on the ASM) throughout the lung in vivo by means of bronchoscopic polarization-sensitive optical coherence tomography (PS-OCT). Using extensive human biobank data from subjects with and without asthma in conjunction with a mathematical model of airway compliance, we define constraints that airways of various sizes pose to any endoscopic imaging technique and how this is impacted by physiologically relevant processes such as constriction, inflation and deflation. We identify critical PS-OCT system parameters and pinpoint parts of the airway tree that are conducive to successful quantification of ASM. We further quantify the impact of breathing and ASM contraction on the measurement error and recommend strategies for standardization and normalization.

Hydration of contact lenses is an important parameter as it governs the oxygen transport to the cornea. Dehydration of contact lenses during lens wear causes discomfort due to dry eye and alterations in lens fitting characteristics. Terahertz (THz) imaging and optical coherence tomography (OCT) together can provide unique information about the hydration profile of the contact lens and its structure. Here, we present results that combine the information measured from OCT and THz systems on contact lenses which aid in determining the change in water content and thickness over time. Overlaid THz and OCT images that align well are shown, making it possible to automatically scale and co-register the two data sets. Hence, we show that a combined OCT/THz system can deliver distinctive knowledge about the capacity of contact lenses to maintain their comfort and optical characteristics over time.

Selective retinal therapy (SRT) employs a micro-second short-pulse lasers to induce localized destruction of the targeted retinal structures with a pulse duration and power aimed at minimal damage to other healthy retinal cells. SRT has demonstrated a great promise in the treatment of retinal diseases, but pulse energy thresholds for effective SRT procedures should be determined precisely and in real time, as the thresholds could vary with disease status and patients. In this study, we present the use of a multi-port fiber-based interferometer (MFI) for highly sensitive real-time SRT monitoring. We exploit distinct phase differences among the fiber ports in the MFI to quantitatively measure localized fluctuations of complex-valued information during the SRT procedure. We evaluate several metrics that can be computed from the full complex-valued information and demonstrate that the complex contour integration is highly sensitive and most correlative to pulse energies, acoustic outputs, and cell deaths. The validity of our method was demonstrated on excised porcine retinas, with a sensitivity and specificity of 0.92 and 0.88, respectively, as compared with the results from a cell viability assay.

A new method based on polarization-sensitive optical coherence tomography (PS-OCT) is introduced to determine the polarization properties of human retinal vessel walls, in vivo. Measurements were obtained near the optic nerve head of three healthy human subjects. The double pass phase retardation per unit depth (DPPR/UD), which is proportional to the birefringence, is higher in artery walls, presumably because of the presence of muscle tissue. Measurements in surrounding retinal nerve fiber layer tissue yielded lower DPPR/UD values, suggesting that the retinal vessel wall tissue near the optic nerve is not covered by retinal nerve fiber layer tissue (0.43°/µm vs. 0.77°/µm, respectively). Measurements were obtained from multiple artery-vein pairs, to quantify the different polarization properties. Measurements were taken along a section of the vessel wall, with changes in DPPR/UD up to 15%, while the vessel wall thickness remained relatively constant. A stationary scan pattern was applied to determine the influence of involuntary eye motion on the measurement, which was significant. Measurements were also analyzed by two examiners, with high inter-observer agreement. The measurement repeatability was determined with measurements that were acquired during multiple visits. An improvement in accuracy can be achieved with an ultra-broad-bandwidth PS-OCT system since it will provide more data points in-depth, which reduces the influence of discretization and helps to facilitate better fitting of the birefringence data.

Thickness and birefringence properties of the vein and artery walls of healthy subjects were quantified following retinal imaging with polarization-sensitive optical coherence tomography and the integrity and structure of vessel walls were assessed.

Blood flow is a key parameter in disease assessment. A time-series optical coherence tomography angiography method is implemented to measure the axial blood flow velocity and direction in a phantom and human tissue, in vivo.

Terahertz (THz) imaging and optical coherence tomography (OCT) provide similar information over the same scales. Here, we present an iterative algorithm that combines the information from OCT and THz within an object to determine both the depth of the reflecting layers at different locations. Also, we can extract unknown refractive index of the object for both the OCT wavelengths and THz frequencies. We validate this algorithm using a phantom test object with embedded layers and show that the depths and refractive index values obtained from the algorithm agreed with the measured values to within 3%. We further demonstrate for the first time that OCT and THz images can be co-registered and aligned using unsupervised image registration. Hence, we show that a combined OCT/THz system can provide unique information beyond the capability of the separate modalities alone, with possible applications in the medical and industrial sectors.

In this Letter, we describe a single-pixel polarization-sensitive imaging technique, capable of generating the birefringence map of a thin specimen by using single-pixel detectors. Spatially modulated light is circularly polarized to illuminate the specimen. The transmitted light through the specimen is then focused via a lens and measured by position-sensitive detectors in two orthogonal polarization channels. The measurement of the irradiance and centroid position of the optical focus and subsequent computations enable the production of polarization-dependent wavefront maps, which can then be utilized to reconstruct sample birefringence information. We demonstrate the feasibility of our method by measuring distribution of optic-axis orientation and phase retardation of various birefringent samples.

Polarization-sensitive optical coherence tomography (PS-OCT) derived birefringence values effectively identify skeletal muscle structural disruption due to muscular dystrophy and exercise-related muscle damage in animal models in ex vivo tissue. The purpose of this investigation was to determine if a PS-OCT needle probe inserted into the leg of a human subject could accurately identify various anatomical structures with implications for use as a diagnostic tool for the determination of skeletal muscle pathology. A healthy middle-aged subject participated in this study. A custom-built PS-OCT system was interfaced with a side-viewing fiber-optic needle probe inserted into the subject’s vastus lateralis muscle via a motorized stage for 3D data acquisition via rotation and stepwise pullback. The deepest recorded PS-OCT images correspond to a depth of 6 mm beneath the dermis with structural images showing uniform, striated muscle tissue. Multiple highly birefringent band-like structures with definite orientation representing connective tissue of the superficial aponeurosis appeared as the depth of the needle decreased. Superficial to these structures the dominating appearance was that of adipose tissue and low birefringent but homogeneous scattering tissue. The data indicate that a PS-OCT needle probe can be inserted into live human skeletal muscle for the identification of relevant anatomical structures that could be utilized to diagnose significant skeletal muscle pathology.

As data acquisition for retinal imaging with optical coherence tomography (OCT) becomes faster, efficient collection of photons becomes more important to maintain image quality. One approach is to use a larger aperture at the eye's pupil to collect more photons that have been reflected from the retina. A 2.8-mm beam diameter system with only seven reflecting surfaces was developed for low-loss retinal imaging. The larger beam size requires defocus and astigmatism correction, which was done in a closed loop adaptive optics method using a Shack-Hartmann wavefront sensor and a deformable mirror (DM) with 140 actuators and a ±2.75 μm stroke. This DM facilitates defocus correction ranging from approximately -3 D to +3 D. Comparing the new system with a standard 1.2-mm system on a model eye, a signal-to-noise gain of 4.5 dB and a 2.3 times smaller speckle size were measured. Measurements on the retinas of five subjects showed even better results, with increases in dynamic range up to 13 dB. Note that the new sample arm only occupies 30 cm × 60 cm, which makes it highly suitable for imaging in a clinical environment. While the left side was imaged with a standard 1.2-mm OCT system, the right side was imaged with the 2.8-mm system. Both images were collected with the same integration time and incident power, after correction of aberrations. Using the dynamic range within the images, which is determined by comparing the highest pixel value to the noise floor, a difference in dynamic range of 10.8 dB was measured between the two systems.

We present a new optical coherence tomography (OCT) angiography method for imaging tissue microvasculature in vivo based on the characteristic frequency-domain flow signature in a short time series of a single voxel. The angiography signal is generated by Fourier transforming the OCT signal time series from a given voxel in multiple acquisitions and computing the average magnitude of non-zero (high-pass) frequency components. Larger temporal variations of the OCT signal caused by blood flow result in higher values of the average magnitude in the frequency domain compared to those from static tissue. Weighting of the signal by the inverse of the zero-frequency component (i.e., the sum of the OCT signal time series) improves vessel contrast in flow regions of low OCT signal. The method is demonstrated on a fabricated flow phantom and on human skin in vivo and, at only 5 time points per voxel, shows enhanced vessel contrast in comparison to conventional correlation mapping/speckle decorrelation and speckle variance methods.

A less-complex 2.8-mm beam diameter spectral domain optical coherence tomography system with an adaptive optics module presented. In this system a Shack-Hartmann wavefront sensor used for aberration sensing and the Deformable mirror used for aberration correction. We demonstrated the diffraction-limited resolution performance of this system on model retina. On the model, measured speckle size with present system is 2.2 times smaller than a 1.2-mm beam diameter OCT system. Further, on the model eye SNR gain of 6.7 dB was quantified with the present system over a 1.2-mm beam diameter OCT system. Relatively small size of 25 cm by 50 cm, less complexity, large field of view of the present system as compared to the conventional AO-OCT systems, would make it suitable for ophthalmic clinical applications.

Morphological changes in the outer retina such as drusen are established biomarkers to diagnose age-related macular degeneration. However, earlier diagnosis might be possible by taking advantage of more subtle changes that accompany tissues that bear polarization-altering properties. To test this hypothesis, we developed a method based on polarizationsensitive optical coherence tomography with which volumetric data sets of the macula were obtained from 10 young (54 years) subjects. All young subjects and 5 of the older subjects had retardance values induced by the retinal pigment epithelium and Bruch’s membrane (RPE-BM) complex that were just above the noise floor measurement (5°−13° at 840 nm). We present an adaptive-optics optical coherence tomography (AO-OCT) system with 3.4-mm beam diameter. A deformable mirror is used for the correction of two radial Zernike orders (defocus, vertical and oblique astigmatism). The aberrations are corrected sequentially with a Shack-Hartmann wave-front sensor and the deformable mirror. This system fills a gap between a standard clinical 1.2-mm beam diameter OCT system and a 6-mm beam diameter AO-OCT system. We also present 8° by 8° en face OCT images from a patient with macular degeneration. This system has a 25 cm by 50 cm footprint, which makes it considerably smaller to conventional 6-mm beam diameter AO-OCT system. Because of its larger field of view and smaller size, it is likely to be useful in the ophthalmic clinics for high-resolution imaging of the human eye retina.

A spectrometer design with a multiple line line-scan camera and beam displacer is presented for ultra-high resolution optical coherence tomography measurements of the human retina at 840 nm. The beam displacer offsets the two orthogonal polarization states on the same line-scan camera, which reduces k-space mapping complexity, as data in both polarization channels can be mapped with the same procedure. Its coherence length is 2.8 μm in tissue (n = 1.38). Birefringence values of 1°/μm and higher were found in a circle with a radius of 2.5° eccentricity centered on the fovea, and in the raphe, pointing at a higher packing density of microtubules and a lower concentration of glia. Birefringence measurements may be more helpful in the modeling of individual structure-function maps than thickness measurements, as they are not affected by glial content.

The organization of fibrillar tissue on the micrometer scale carries direct implications for health and disease but remains difficult to assess in vivo. Polarization-sensitive optical coherence tomography measures birefringence, which relates to the microscopic arrangement of fibrillar tissue components. Here, we demonstrate a critical improvement in leveraging this contrast mechanism by employing the improved spatial resolution of focus-extended optical coherence microscopy (1.4 µm axially in air and 1.6 µm laterally, over more than 70 µm depth of field). Vectorial birefringence imaging of sheep cornea ex vivo reveals its lamellar organization into thin sections with distinct local optic axis orientations, paving the way to resolving similar features in vivo.

A general mathematical model based on Mueller-matrix calculation is presented to describe the optical behavior of a dual-crystal electro-optic modulator. The two crystals inside the modulator are oriented at ± 45° with respect to the horizontal, thereby cancelling natural birefringence and temperature-induced birefringence. We describe the behavior of the modulator as a function of the ellipticity of the crystals, the rotation angles of the crystals and the applied voltage. By fitting the measured data with a Mueller-matrix model that uses values for the ellipticity and orientation angles of the crystals, the simulated data and the experimental measurements could be matched. This Mueller-matrix includes physical properties of the thermally compensated electro optic modulator, and the matrix can be used in simulations where these device-specific properties are important, for instance in the modeling of a polarization-sensitive optical coherence tomography system.

Optical coherence tomography (OCT) images of the retina are inevitably affected by the finite width of the coherence function and noise. To make low-reflective layers visible, the raw OCT signal is log transformed; to reduce the effect of noise the images can be low-pass filtered. We determined the effects of these operations on layer thickness assessment, as a function of signal-to-noise ratio (SNR), by performing measurements in a phantom eye and modeling. The log transform appeared to be the key factor in a SNR-dependent overestimation of peak widths and a less predictive bias in the widths of low-reflective layers. Early detection of axonal tissue loss in retinal nerve fiber layer (RNFL) is critical for effective treatment and management of diseases such as glaucoma. This study aims to evaluate the capability of ultrahigh-resolution optical coherence tomography with adaptive optics (UHR-AO-OCT) for imaging the RNFL axonal bundles (RNFBs) with 3×3×3 μm3 resolution in the eye. We used a research-grade UHR-AO-OCT system to acquire 3°×3° volumes in four normal subjects and one subject with an arcuate retinal nerve fiber layer defect (n=5; 29–62yrs). Cross section (B-scans) and en face (C-scan) slices extracted from the volumes were used to assess visibility and size distribution of individual RNFBs. In one subject, we reimaged the same RNFBs twice over a seven month interval and compared bundle width and thickness between the two imaging sessions. Lastly we compared images of an arcuate RNFL defect acquired with UHR-AO-OCT and commercial OCT (Heidelberg Spectralis). Individual RNFBs were distinguishable in all subjects at 3° retinal eccentricity in both cross-sectional and en face views (width: 30–50μm, thickness: 10–15μm). At 6° retinal eccentricity, RNFBs were distinguishable in three of the five subjects in both views (width: 30–45μm, thickness: 20–40μm). Polarization-sensitive optical coherence tomography can be used to measure the birefringence of biological tissue such as the human retina. Previous measurements with a time-domain polarization-sensitive optical coherence tomography system revealed that the birefringence of the human retinal nerve fiber layer is not constant, but varies as a function of location around the optic nerve head. Here we present a spectral-domain polarization-sensitive optical coherence tomography system that uses a spectrometer configuration with a single line scan camera and a Wollaston prism in the detection arm. Since only one camera has to be synchronized with other components in the system, the design is simplified considerably. This system is 60 times faster than a time-domain polarization-sensitive optical coherence tomography system. Data was acquired using concentric circular scans around the optic nerve head of a young healthy volunteer and the acquisition time for 12 circular scans was reduced from 72 s to 1.2 s. The acquired data sets demonstrate variations in retinal thickness and double pass phase retardation per unit depth that were similar to data from the same volunteer taken with a time-domain polarization-sensitive system. The double pass phase retardation per unit depth of the retinal nerve fiber layer varied between 0.18 and 0.40 degrees/mum, equivalent to a birefringence of 2.2 * 10(-4) and 4.8 * 10(-4) respectively, measured at 840 nm.

Optical coherence tomography (OCT) is a non-invasive imaging technique, which has previously demonstrated potential for use in dermatology. The purpose of this study is to demonstrate how improvements in image quality, speed, and functionality enable qualitative and quantitative information to be obtained from in vivo human skin. We developed a portable fiber-optic based OCT imaging device that requires only 1 second to simultaneously provide high-resolution images of skin structure, collagen birefringence, and blood flow. Images of normal human skin were acquired in vivo, and features compared with clinical and histologic observations. The layered structure and appendages of skin were apparent in conventional OCT images, and correlated well with corresponding histology. Polarization-sensitive OCT images simultaneously revealed birefringent regions within the dermis corresponding to the location of collagen fibers, as confirmed with polarized light microscopy. Properties of collagen-rich tissues including tendon and scar tissues were quantified. Location of blood flow was also displayed alongside structural and polarization-sensitive images. Significant improvements in OCT technology have been made since its early application in dermatology. In particular, combining the previously described structural and Doppler imaging functions with polarization-sensitive imaging increases the utility of the technique for rapid, non-invasive investigations in the skin.

PURPOSE. Thinning of the retinal nerve fiber layer and changes in retinal nerve fiber layer (RNFL) birefringence may both precede clinically detectable glaucomatous vision loss. Early detection of RNFL changes may enable treatment to prevent permanent loss of vision. Polarization-sensitive optical coherence tomography (PS-OCT) can provide objective information on RNFL thickness and birefringence. METHODS. PS-OCT scans around the optic nerve head (ONH) of two healthy young volunteers were made using 10 concentric circles of increasing radius. Both the mean RNFL thickness and mean retinal nerve fiber birefringence for each of 48 sectors on a circle were determined with data analysis. RESULTS. Both the RNFL thickness and birefringence varied as a function of sector around the ONH. The RNFL became thinner with increasing distance from the ONH. In contrast, the birefringence did not vary significantly as a function of radius. CONCLUSIONS. Birefringence of healthy RNFL is constant as a function of scan radius but varies as a function of position around the ONH, with higher thickness values occurring superior and inferior to the ONH. Measured double-pass phase retardation per unit depth around the ONH ranged between 0.10 and 0.35 deg/m, equivalent to birefringences of 1.2 10 4 and 4.1 10 4 respectively, measured at a wavelength of 840 nm. Consequently, when a spatially constant birefringence around the ONH is assumed, the conversion of scanning laser polarimetry (SLP) phase-retardation measurements to RNFL thickness may yield incorrect values. The data do not invalidate the clinical value of a phase-retardation measurement, but affect the conversion of phase retardation to RNFL thickness. An ultra-high-speed spectral domain optical Doppler tomography (SD-ODT) system is used to acquire images of blood flow in a human retina in vivo, at 29,000 depth profiles (A-lines) per second and with data acquisition over 99% of the measurement time. The phase stability of the system is examined and image processing algorithms are presented that allow accurate determination of bi-directional Doppler shifts. Movies are presented of human retinal flow acquired at 29 frames per second with 1000 A-lines per frame over a time period of 3.28 seconds, showing accurate determination of vessel boundaries and time-dependent bi-directional flow dynamics in artery-vein pairs. The ultra-high-speed SD-ODT system allows visualization of the pulsatile nature of retinal blood flow, detects blood flow within the choroid and retinal capillaries, and provides information on the cardiac cycle. In summary, accurate video rate imaging of retinal blood flow dynamics is demonstrated at ocular exposure levels below 600 microW.

The transfer function for optical wavefront aberrations in single-mode fiber based optical coherence tomography is determined. The loss in measured OCT signal due to optical wavefront aberrations is quantified using Fresnel propagation and the calculation of overlap integrals. A distinction is made between a model for a mirror and a scattering medium model. The model predictions are validated with measurements on a mirror and a scattering medium obtained with an adaptive optics optical coherence tomography setup. Furthermore, a one-step defocus correction, based on a single A-scan measurement, is derived from the model and verified. Finally, the pseudo-convex structure of the optical coherence tomography transfer function is validated with the convergence of a hill climbing algorithm. The implications of this model for wavefront sensorless aberration correction are discussed.

A growing body of evidence suggests that phototransduction can be studied in the human eye in vivo by imaging of fast intrinsic optical signals (IOS). There is consensus concerning the limiting influence of motion-associated imaging noise on the reproducibility of IOS-measurements, especially in those employing spectral-domain optical coherence tomography (SD-OCT). However, no study to date has conducted a comprehensive analysis of this noise in the context of IOS-imaging. In this study, we discuss biophysical correlates of IOS, and we address motion-associated imaging noise by providing correctional post-processing methods. In order to avoid cross-talk of adjacent IOS of opposite signal polarity, cellular resolution and stability of imaging to the level of individual cones is likely needed. The optical Stiles-Crawford effect can be a source of significant IOS-imaging noise if alignment with the peak of the Stiles-Crawford function cannot be maintained. Therefore, complete head stabilization by implementation of a bite-bar may be critical to maintain a constant pupil entry position of the OCT beam. Due to depth-dependent sensitivity fall-off, heartbeat and breathing associated axial movements can cause tissue reflectivity to vary by 29% over time, although known methods can be implemented to null these effects. Substantial variations in reflectivity can be caused by variable illumination due to changes in the beam pupil entry position and angle, which can be reduced by an adaptive algorithm based on slope-fitting of optical attenuation in the choriocapillary lamina.

An optical coherence tomography (OCT) system with a 2.8-mm beam diameter is presented. Sensorless defocus correction can be performed with a Badal optometer and astigmatism correction with a liquid crystal device. OCT B-scans were used in an image-based optimization algorithm for aberration correction. Defocus can be corrected from ? 4.3 ?? D to + 4.3 ?? D and vertical and oblique astigmatism from ? 2.5 ?? D to + 2.5 ?? D . A contrast gain of 6.9 times was measured after aberration correction. In comparison with a 1.3-mm beam diameter OCT system, this concept achieved a 3.7-dB gain in dynamic range on a model retina. Both systems were used to image the retina of a human subject. As the correction of the liquid crystal device can take more than 60 s, the subject’s spectacle prescription was adopted instead. This resulted in a 2.5 times smaller speckle size compared with the standard OCT system. The liquid crystal device for astigmatism correction does not need a high-voltage amplifier and can be operated at 5 V. The correction device is small ( 9 ?? mm × 30 ?? mm × 38 ?? mm ) and can easily be implemented in existing designs for OCT.

A Several key structures and molecules in the human retina are known to exhibit birefringence, and therefore can be probed with polarimetric imaging. By using scanned illumination, the contrast is increased, revealing otherwise undetectable pathology.

We present a generalized analysis of fiber-based polarization-sensitive optical coherence tomography with an emphasis on determination of sample optic axis orientation. The polarization properties of a fiber-based system can cause an overall rotation in a Poincare sphere representation such that the plane of possible measured sample optic axes for linear birefringence and diattenuation no longer lies in the QU-plane. The optic axis orientation can be recovered as an angle on this rotated plane, subject to an offset and overall indeterminacy in sign such that only the magnitude, but not the direction, of a change in orientation can be determined. We discuss the accuracy of optic axis determination due to a fundamental limit on the accuracy with which a polarization state can be determined as a function of signal-to-noise ratio.

We developed an optical coherence tomography (OCT) prototype with a sample arm that uses a 3.4 mm beam, which is considerably larger than the 1.2 to 1.5 mm beam that is used in commercialized OCT systems. The system is equipped with adaptive optics (AO), and to distinguish it from traditional AO-OCT systems with a larger 6 mm beam we have coined this concept AO-assisted OCT. Compared to commercialized OCT systems, the 3.4 mm aperture combined with AO improves light collection efficiency and imaging lateral resolution. In this paper, the performance of the AOa-OCT system was compared to a standard OCT system and demonstrated for imaging of age-related macular degeneration (AMD). Measurements were performed on the retinas of three human volunteers with healthy eyes and on one eye of a patient diagnosed with AMD. The AO-assisted OCT system imaged retinal structures of healthy human eyes and a patient eye affected by AMD with higher lateral resolution and a 9° by 9° field of view. This combination of a large isoplanatic patch and high lateral resolution can be expected to fill a gap between standard OCT with a 1.2 mm beam and conventional AO-OCT with a 6 mm beam and a 1.5° by 1.5° isoplanatic patch.

We present the use of sub-micron resolution optical coherence tomography (OCT) in quality inspection for printed electronics. The device used in the study is based on a supercontinuum light source, Michelson interferometer and high-speed spectrometer. The spectrometer in the presented spectral-domain optical coherence tomography setup (SD-OCT) is centered at 600 nm and covers a 400 nm wide spectral region ranging from 400 nm to 800 nm. Spectra were acquired at a continuous rate of 140,000 per second. The full width at half maximum of the point spread function obtained from a Parylene C sample was 0:98 m. In addition to Parylene C layers, the applicability of sub-micron SD-OCT in printed electronics was studied using PET and epoxy covered solar cell, a printed RFID antenna and a screen-printed battery electrode. A commercial SD-OCT system was used for reference measurements.

o our knowledge, this is the first demonstration of in vivo depth-resolved birefringence measurements of the human retinal nerve fiber layer (RNFL) by use of polarization-sensitive optical coherence tomography (PS-OCT). Because glaucoma causes nerve fiber layer damage, which may cause loss of retinal birefringence, PS-OCT is a potentially useful technique for the early detection of glaucoma. We built a fiber-based PS-OCT setup that produces quasi-real-time images of the human retina in vivo. Preliminary measurements of a healthy volunteer showed that the double-pass phase retardation per unit depth of the RNFL near the optic nerve head is 39 +/- 6degrees/100 mum. Wide field of view (FOV) retinal imaging with high resolution has been demonstrated for quantitative analysis of retinal microstructures. An adaptive optics scanning laser ophthalmoscope (AO-SLO) that was built in our laboratory was improved by a customized scanning protocol for scanning wide region. A post-processing program was developed for generating wide FOV retinal images. The high resolution retinal image with 1.7 degree by 3.0 degree FOV were obtained.

The effect of using collimator lenses with different focal lengths on the performance of a spectral-domain adaptive optics optical coherence tomography (AO-OCT) system has been studied. In vivo OCT scans of a healthy human retina were taken separately with different collimator lenses. Although shorter focal length lenses provide a smaller beam diameter at the pupil of the eye, and therefore a larger diffraction-limited spot size, on the return path the shorter focal length collimators demonstrate a better performance focusing the sinc-function-like intensity distribution returning from the eye on the fiber tip. The results might have applications in the OCT imaging of challenging cases.

Ultrahigh speed line scan detectors based on CMOS technology have been recently demonstrated in ultrahigh resolution spectral-domain optical coherence tomography (UHR-SD-OCT) for retinal imaging. While successful, fundamental tradeoffs exist been image acquisition time, image sampling density, and sensitivity, all of which impact the extent of motion artifacts, visualization of fine spatial detail, and detection of faint reflections. Here we investigate these tradeoffs for imaging retinal nerve fiber bundles (RNFBs) using UHR-SD-OCT with adaptive optics (AO). Volume scans of 3°x3° and 1.5°x1.5° were acquired at retinal locations of 3° nasal and 6° superior to the fovea on a healthy subject. Dynamic AO compensation across a 6 mm pupil provided near-diffraction-limited performance. The acquisition rates were 22.5k lines/s and 125k lines/s with A-lines spaced at 0.9 μm and 1.8 μm and B-scans at 1.8 μm and 9 μm. Focus was optimized for visualizing the retinal nerve fiber bundles (RNFBs). En face projection and crosssectional views of the RNFBs were extracted from the volumes and compared to images acquired with established conventional CCD-based line-scan camera. The projection view was found highly sensitive to eye motion artifacts, yet could only be partially compensated with coarser sampling, since fine sampling was necessary to observe the microscopic features in the RNFBs. For the cross-sectional view, speckle noise rather than eye motion artifacts limited bundle clarity. The highest B-scan density (1.8 μm spacing) coupled with B-scan averaging proved the best combination. Regardless of view, the higher line rate provided better RNFB clarity.

We evaluate a novel non-invasive optical technique for observing fast physiological processes, in particular phototransduction, in single photoreceptor cells in the living human eye. The method takes advantage of the interference of multiple reflections within the outer segments (OS) of cones. This self-interference phenomenon is highly sensitive to phase changes such as those caused by variations in refractive index and scatter within the photoreceptor cell. A high-speed (192 Hz) flood-illumination retina camera equipped with adaptive optics (AO) is used to observe individual photoreceptors, and to monitor changes in their reflectance in response to visible stimuli (“scintillation”). AO and high frame rates are necessary for resolving individual cones and their fast temporal dynamics, respectively. Scintillation initiates within 5 to 10 ms after the onset of the stimulus flash, lasts 300 to 400 ms, is observed at visible and near-infrared (NIR) wavelengths, and is highly sensitive to the coherence length of the imaging light source. To our knowledge this is the first demonstration of in vivo optical imaging of the fast physiological processes that accompany phototransduction in individual photoreceptors.

An ultra-high-speed spectral-domain optical coherence tomography system (SD-OCT) was developed for imaging the human retina and optic nerve in vivo at a sustained depth profile (A-line) acquisition speed of 29 kHz. The axial resolution was 6 microm in tissue and the system had shot-noise-limited performance with a maximum sensitivity of 98.4 dB. 3-dimensional data sets were collected in 11 and 13 seconds for the macula and optic nerve head respectively and are presented to demonstrate the potential clinical applications of SD-OCT in ophthalmology. Additionally, a 3-D volume of the optic nerve head was constructed from the acquired data and the retinal vascular network was visualized.

Ultra-high-resolution (UHR) optical coherence tomography (OCT) imaging technique is using adaptive optics, to perform better better treatment and monitoring of retinal diseases. Researchers have developed a new generation of adaptive optics (AO) UHR OCT (AO-UHR-OCT) instrument that combines an ultrabroadband spectral light source and an achromatizing element, to increase measured axial resolution to around 3.5 μm. The measured axial resolution is increased without reducing lateral resolution and image quality. The new instrument uses an AO system in the spectral/Fourier domain OCT sample arm, to facilitate correction of a patient's ocular aberrations. The detection channel of the OCT spectrometer acquires spectral fringes of light, back-scattered from the retina, along with light from the reference arm.

The combination of adaptive optics (AO) and optical coherence tomography (OCT) has been successfully applied to in vivo retinal imaging, motivated by the potential of unprecedented 3D resolution. The eye, however, not only suffers from monochromatic aberrations, which are corrected by AO, but also substantial chromatic aberrations, which are not. To correct chromatic aberrations, achromatizing lenses have been recently employed in ultrahigh resolution AO-OCT instruments. These lenses, like their earlier predecessors for vision improvement, effectively correct the eye's longitudinal chromatic aberration (LCA), but make no attempt at the complementary transverse chromatic aberration (TCA). This raises an important concern as to the degrading impact of TCA on the 3D resolution of AO-OCT and OCT retina cameras. To address this, we undertook a theoretical analysis of TCA for high-resolution retinal imaging. The theoretical analysis included the two primary contributors of TCA for retinal imaging: (1) errors in the lateral positioning of the eye and (2) off-axis imaging. The analysis predicted the extent to which TCA impacts retinal imaging and the conditions under which it can be held at acceptable levels for AO-OCT and OCT. Several near-infrared bands were chosen that correspond to common OCT light sources.

We have developed an improved adaptive optics - optical coherence tomography (AO-OCT) system and evaluated its performance for in vivo imaging of normal and pathologic retina. The instrument provides unprecedented image quality at the retina with isotropic 3D resolution of 3.5 x 3.5 x 3.5 microm(3). Critical to the instrument's resolution is a customized achromatizing lens that corrects for the eye's longitudinal chromatic aberration and an ultra broadband light source (Delta lambda=112 nm lambda(0)= approximately 836 nm). The eye's transverse chromatic aberrations is modeled and predicted to be sufficiently small for the imaging conditions considered. The achromatizing lens was strategically placed at the light input of the AO-OCT sample arm. This location simplifies use of the achromatizing lens and allows straightforward implementation into existing OCT systems. Lateral resolution was achieved with an AO system that cascades two wavefront correctors, a large stroke bimorph deformable mirror (DM) and a micro-electromechanical system (MEMS) DM with a high number of actuators. This combination yielded diffraction-limited imaging in the eyes examined. An added benefit of the broadband light source is the reduction of speckle size in the axial dimension. Additionally, speckle contrast was reduced by averaging multiple B-scans of the same proximal patch of retina. The combination of improved micron-scale 3D resolution, and reduced speckle size and contrast were found to significantly improve visibility of microscopic structures in the retina.

The directional component of the retinal reflection, i.e., the optical Stiles-Crawford effect (SCE), is well established to result from the waveguiding property of photoreceptors. Considerable uncertainty, however, remains as to which retinal reflections are waveguided and thus contribute. To this end we have developed a retina camera based on spectral-domain optical coherence tomography (SD-OCT) that axially resolves (approximately 5 microm) these reflections and permits a direct investigation of the SCE origin at near infrared wavelengths. Reflections from the photoreceptor inner/outer segments junction (IS/OS) and near the posterior tip of the outer segments (PTOS) were found highly sensitive to beam entry position in the pupil with a considerable decrease in brightness occurring with an increase in aperture eccentricity. Reflections from the retinal pigment epithelium (RPE) were largely insensitive. The average directionality (rho(oct) value) at 2 degree eccentricity across the four subjects for the IS/OS, PTOS, and RPE were 0.120, 0.270, and 0.016 mm(-2), respectively. The directionality for the IS/OS approached typical psychophysical SCE measurements, while that for the PTOS approached conventional optical SCE measurements. Precise measurement of the optical SCE was found to require significant A-scan averaging.
