The brain-specific tyrosine phosphatase, STEP (STriatal-Enriched protein tyrosine Phosphatase) is an important regulator of synaptic function. STEP normally opposes synaptic strengthening by increasing N-methyl D-aspartate glutamate receptor (NMDAR) internalization through dephosphorylation of GluN2B and inactivation of the kinases extracellular signal–regulated kinase 1/2 and Fyn. Here we show that STEP61is elevated in the cortex in the Nrg1+/−knockout mouse model of schizophrenia (SZ). Genetic reduction or pharmacological inhibition of STEP prevents the loss of NMDARs from synaptic membranes and reverses behavioral deficits in Nrg1+/−mice. STEP61protein is also increased in cortical lysates from the central nervous system-specific ErbB2/4 mouse model of SZ, as well as in human induced pluripotent stem cell (hiPSC)-derived forebrain neurons and Ngn2-induced excitatory neurons, from two independent SZ patient cohorts. In these selected SZ models, increased STEP61protein levels likely reflect reduced ubiquitination and degradation. These convergent findings from mouse and hiPSC SZ models provide evidence for STEP61dysfunction in SZ.

In 2009–2010 a Portuguese consortium was created to implement the methodologies proposed by the Dose Datamed II (DDM2) project, aiming to collect data from diagnostic X-ray and nuclear medicine (NM) procedures, in order to determine the most frequently prescribed exams and the associated ionizing radiation doses for the Portuguese population. The current study is the continuation of this work, although it focuses only on NM exams for the years 2011 and 2012.

High environmental temperatures during the hot months of the year reduce reproductive performance in cattle. Summer heat stress depression in fertility is a multifactorial problem; however, there is evidence that the bovine germinal vesicle and maturing oocyte, as well as the early embryo, are major targets of the deleterious effects of heat stress. Such adverse effects are less pronounced in heat-tolerant breeds (Bos indicus) than heat-sensitive breeds (Bos taurus). This genetic variation results from the greater thermoregulatory ability and cellular thermoresistance of heat-tolerant breeds. Heat-induced oocyte cellular damage occurs in both cytoplasmic and nuclear compartments. Heat shock has been shown to reduce oocyte nuclear maturation, induce apoptosis, compromise oocyte cytoskeleton, and impair oocyte mitochondrial function and developmental competence. However, the oocyte cytoplasm is more susceptible to heat shock than the nucleus. This effect is greater for Bos taurus than Bos indicus oocytes. The detrimental effects of heat shock are also critical during the first cleavage divisions when most of the embryonic genome is inactive; however, the bovine embryo becomes more resistant to increased temperature as it proceeds through development. Several studies demonstrated that Bos indicus embryos are more thermotolerant than Bos taurus embryos. Adaptive changes involved in acquisition of thermotolerance are likely derived from changes in gene expression and (or) activity of biochemical molecules that control cellular functions against stress. Recently, molecules such as IGF-I and caspase inhibitor z-DEVD-fmk have been shown to exert a thermoprotective role, rescuing heat-induced oocyte and embryo cellular damage and developmental competence. Therefore, cattle genotype and thermoprotective molecules can be considered as an alternative to modulate the effects of increased temperature in reproductive function.

Burning of vegetation can promote changes in soil physical properties and also create hydrophobic substances, which accumulate and result in the formation of water-repellent layers. A study was conducted between 2005 and 2011 on an Albaqualf, with natural grassland composed mainly of Andropogon lateralis, in southern Brazil, to investigate the induction of hydrophobicity by burning and changes in the soil hydro-physical characteristics. The treatments consisted of no burning, and burned 60, 36, 12, 6, 4, and 2 months before sampling. Burning management did not change the saturated hydraulic conductivity, whereas air permeability was significantly reduced in the surface layer, nearly reaching the original values 4 months after burning. Saturated hydraulic conductivity and air permeability correlated with total porosity and macroporosity. The surface layer was the most sensitive to fire effects, with an increased degree of hydrophobicity for up to 2 months after vegetation burning. Hydrophobicity and contact angle correlated positively with soil organic matter content.

Liquid secondary ion and electrospray mass spectrometry were used to study the complexation in‐source of 5,10,15‐tris(pentafluorophenyl)corrole with several divalent transition‐metal ions. The metallocorrole ions formed in‐source were identified by comparing their product ion mass spectra with the spectra of the same ions formed from metallocorroles obtained from classical procedures. Positive metallocorrole ion formation is accompanied by oxidation of the metal centre. Mechanisms were proposed for the oxidation processes, and data from negative‐ion spectra reinforced these mechanisms. Copyright © 2006 John Wiley & Sons, Ltd.

A 15-year-old female Holstein cow was presented with a history of anorexia and weight loss. Abnormal physical examination findings included poor body condition, dehydration, icterus, and photodermatitis on nonpigmented areas of the skin of the dorsum. At necropsy, the common bile duct was greatly enlarged, with thickened walls, and tightly adhered to the hepatic capsular surface and serosa of adjacent loops of the small intestine. Two sessile, yellowish, friable, well-circumscribed, cauliflower-like, bulging masses (14 and 8 cm in diameter) were attached to the inner surface of the common bile duct blocking its lumen. Microscopically, the tumor was composed of well-differentiated, columnar epithelial cells with oval or round, vesicular, basal nuclei arranged in papillary projections. Periodic acid-Schiff-positive material accumulated in the apical cytoplasm of the neoplastic cells. Immunostaining of the tumor cells was positive for cytokeratin (CK) of wide-spectrum screening and for CK 7. The diagnosis of papillary adenocarcinoma of the extrahepatic bile duct was based on the morphologic features of the neoplasm and evidence of local invasion. To the best of our knowledge, extrahepatic bile duct carcinomas have not been described in cattle.

The zinc complexes of diaryl bis(p‐nitrophenyl)porphyrins and β‐(1,3‐dinitroalkyl)tetraphenylporphyrins were studied by electrospray ionization (ESI) tandem mass spectrometry (MS/MS). All porphyrins showed the protonated molecule under ESI conditions. The protonated molecules were induced to fragment and the corresponding ESI tandem mass spectra were analysed. Porphyrins with two p‐nitrophenyl groups showed, as expected, characteristic fragmentations including either loss of one nitro group, as the major fragment of the tandem mass spectra, and loss of both nitro groups. In contrast, MS/MS of the β‐(1,3‐dinitroalkyl)porphyrins provided interesting and unexpected results such as the absence (or in insignificant abundance) of the ions formed by loss of one nitro group. However, these porphyrins show an abundant fragment due to combined loss of the two nitro groups. Also, the typical β‐cleavage of the alkyl chain is not observed per se, only when combined with loss of HNO2 or •NO2. Instead, α‐cleavage, with loss of the β‐pyrrolic substituent, is the most favourable process. Copyright © 2005 John Wiley & Sons, Ltd.

Fertility parameters of boar spermatozoa were evaluated in vitro, after freeze‐thawing the semen in three different extenders containing permeable and non‐permeable cryoprotectants: A (111‐OmM Tris, 31.4mM citric acid, 185.0mM glucose, 20 per cent egg yolk, 3 per cent glycerol and 100 iu/ml penicillin G); B (200mM Tris; 70.8mM citric acid, 55.5mM glucose, 20 per cent egg yolk, three per cent glycerol and 100 iu/ml penicillin G); C (200mM Tris, 70.8mM citric acid, 55.5mM fructose, 20 per cent egg yolk, 3 per cent glycerol and 100 iu/ml penicillin G). The freeze‐thawing techniques were the same for each extender. Eight ejaculates from four boars were obtained; the sperm‐rich fraction of each ejaculate was extended in each of the three media at a final concentration of 400 × 106sperm/ml, loaded into 0.5 ml straws and frozen at a rate of 30°C/minute to ‐ 196°C. The straws were thawed at 60°C for eight seconds. Sperm motility, acrosomal integrity and in vitro sperm penetration through the zona pellucida of gilt oocytes matured in vitro were evaluated. The motility of unfrozen spermatozoa was 93.1 per cent compared with 60.7 per cent, 48.2 per cent and 35 per cent for sperm frozen in extenders A, B and C respectively; these values were all significantly different (P<0.05). There was no significant decline in sperm motility after incubation for 30 minutes in extender A, but there were significant decreases in sperm motility after 30 minutes of incubation in B and C. The percentage acrosomal integrities were 97.2 per cent for the control and 45.5 per cent, 30.3 per cent and 16.8 per cent for the frozen‐thawed spermatozoa in extenders A, B and C respectively. The results of the in vitro penetration assay were 80.7 per cent when using control spermatozoa, and 42.2 per cent, 18.4 per cent and 3.3 per cent when using frozen‐thawed spermatozoa in extenders A, B and C respectively
