3. Process development for recovery and purification of extracellular vesicles for biological products

Abstract

Extracellular vesicles (EVs) have rapidly attracted research and commercial interest during the past decade. Potential applications of EVs are diverse and include drug delivery, biomarkers and therapeutics. Despite this growing interest, manufacturing processes for EV production are still in the development phase. The recovery and purification of EVs remain a major hurdle in the progression of fundamental research and the commercial application of EV-based products. Many methods have been described to isolate and purify EVs, each having distinct advantages and challenges. Ultracentrifugation has historically been the most frequently cited EV isolation method, however it is highly operator-dependent, time-consuming and challenging to scale. To this end, the work presented in this thesis explores a scalable and reproducible form of pseudo-affinity chromatography which uses the ligand heparin. Initially, an in-house EV production process was established based on protocols transferred from the project sponsor, ReNeuron. This multi-step process was used to produce concentrated and pre-purified EV material derived from cultures of adherent CTX0E03 neural stem cells or suspension human embryonic kidney cells. This EV material was used as the chromatography column load material in the following three chapters. Initially, heparin affinity chromatography (HAC) using a linear elution gradient was developed for EV purification. This work provides evidence to support an affinity between EVs and heparin, which is currently limited to two conflicting literature articles. This protocol resulted in an EV recovery of at least 68.7%, based on particle counts, which was a 72.6% increase over that using a standard size-exclusion chromatography set-up. Subsequently, a HAC protocol using a step elution gradient was developed for EV fractionation. EV fractionation is a relatively novel development in the EV field that aims to separate EV subpopulations with potentially more desirable properties. This protocol fractionated EVs into two discrete eluted populations that exhibited significantly different tetraspanin immunophenotypes, variable expression of the exosome-associated protein TSG101 and differences in purity. 98.8% of residual protein and 99.0% of DNA were removed using this protocol. Finally, custom chromatography resins were created through the coupling of heparin and related sugars to agarose to gain insights into the structural features of heparin that may be important for EV binding. Specifically, the use of the heparin analogue, tinzaparin, which has a shorter and more consistent chain length, resulted in improved EV binding over the use of standard unfractionated heparin. Overall, HAC shows promise as a method to capture and purify EVs and the work presented in this thesis advances understanding in several important areas from providing evidence to support EV-heparin binding and gaining insights into structural features that facilitate this interaction, to proposing two applications for HAC, EV purification and EV fractionation.

11. Sub-Millisecond Visible-Light Gating of a Zinc Oxide Nanowire

Abstract

Semiconductor nanowires are the building blocks of many nanoscale electrical and neuromorphic circuits. Here, we demonstrate a simple arrangement wherein an ethanol-adsorbed ZnO single nanowire, deposited between gold electrodes using dielectrophoresis exhibits significant change in resistance when activated by visible light. Here we have observed that the transition timescale between two stable ohmic states, one in the dark and the other in the illuminated regime in a single nanowire can occur in the submillisecond order, which is 7 orders of magnitude lower than our previously reported switching timescale in bulk ZnO thin films. A mathematical model of the light activated resistance switching mechanism is proposed based on the adsorption-desorption kinetics of oxygen molecules at the surface of the nanowires., Comment: 16 pages, 5 figures

Abstract

As technology continues to evolve, the demand for renewable and sustainable energy continues to grow. As the use of renewable energies, specifically photovoltaics, is continually being adopted and incorporate into everyday life, it is evident that a need for an increase in the amount of energy that is derived from these processes is necessary. In the exploration of dye sensitized solar cells (DSSC) a recent and increased involvement in the application of carbon based nanomaterials and the effects of their unique electronic properties is being investigated. Additionally, as the development of DSSC continues to break way, different methods of dye selection and photoanode scaffolding are being researched to ultimately increase the Power Conversion Efficiency (PCE) of these cells.

17. Fractal-Based Electrolytic Capacitor Electrodes: Scaling Behavior with Respect to Fractal Order and Complexity

Abstract

In past decades, the application of fractals to electrode design for enhanced signaling and electrochemical performance was a popular subject and enabled the growth of consumer micro-electronics. Supercapacitors, which are energy storage devices with many promising characteristics, have largely grown alongside of such developments in electronics, but little work has been done to use fractal electrodes in supercapacitors. In this work, plane-filling and fractal patterns were used in designing laser scribed graphene supercapacitor electrodes, allowing the scaling laws of capacitance with respect to fractal order and complexity to be examined for the first time. An interesting exponential relationship between capacitance and fractal order for the more open structured fractals was observed, the exponent of which was proportional to the Hausdorff dimension. Additional non-linear relationships between capacitance and order were observed for other structures which was correlated with inter-plate repulsion and differences in path length. These findings provide the first step in maximizing the efficiency of fractal-based electrolytic devices by exploring the non-intuitive trends in capacitance with respect to fractal order and complexity.

Abstract

In this paper we describe a simple and inexpensive method to generate plasma using a kitchen microwave. The microwave-generated plasma is characterized by spectroscopic analysis and compared with the absorption spectra of a gas discharge tube. A Paschen-like curve is observed leading to a hypothesis of the microwave plasma generation mechanism in air. We have also demonstrated that this microwave generated air plasma can be used in a multitude of applications such as: a) surface modification of a substrate to change its wettability; b) surface modification to change electrical/optical properties of a substrate; and c) enhancement of adhesive forces for improved bonding of polymeric microfluidic molds, such as bonding polydimethylsiloxane (PDMS) chips to glass covers. The Wahls diet is a modified Paleolithic diet that emphasizes dark green leafy vegetables, colorful fruits, high-quality animal proteins, and omega-3 polyunsaturated fatty acids, while limiting grains, legumes, dairy products, sugar, and processed foods containing proinflammatory omega-6 fatty acids. The Wahls diet may reduce inflammation, oxidative stress, and mitochondrial dysfunction and has plausible mechanisms for slowing amyotrophic lateral sclerosis (ALS) progression. However, research on its dietary components in the ALS animal models has yielded conflicting results. Though multiple cohort studies suggest high carotenoids, omega-3 fatty acids and fruit intake are associated with reduced ALS risks, neither the diet nor its components has been demonstrated to slow down ALS progression in case studies or clinical trials. On the contrary, the Wahls diet, a restrictive, low-carbohydrate and low glycemic index diet, caused an average weight loss of 7.2% BMI in multiple sclerosis clinical trials, which is a significant concern for people living with amyotrophic lateral sclerosis (PALS) as weight loss is associated with faster ALS progression and shorter survival. Abstract

Lion's Mane (Hericium erinaceus) has historically been used as traditional medicine in Asia and Europe for its potential benefits in fighting infection and cancer. It has gained interest in the neurodegenerative disease field because of its mechanisms of action; these include anti-inflammation, neuroprotection, and promoting neurite growth demonstrated in various cell and animal models. A very small, double-blind, placebo-controlled trial in patients with mild cognitive impairment showed a temporary improvement in cognitive function; this finding has yet to be replicated. However, there have been no studies in ALS cell or animal models or in humans with ALS. Lion's Mane appears safe and inexpensive when consumed in powder or capsule, but one anaphylactic case was reported after a patient consumed fresh Lion's Mane mushroom. Currently, we do not have enough information to support the use of Lion's Mane for treating ALS. We support further research in ALS disease models and clinical trials to study its efficacy.
