3. Protective Capacity of Helichrysum italicum Infusion Against Intestinal Barrier Disruption and Translocation of Salmonella Infantis.

Abstract

Background: Helichrysum italicum is a Mediterranean plant with well-known anti-inflammatory activity, but our previous whole transcriptome analysis has found that H. italicum infusion (HII) can also affect cytoskeletal rearrangement and tight junctions. The goal of the present study was to determine if HII improves the intestinal barrier (IB) dysfunction and by what mechanism. Methods: Caco-2 cells on Transwell inserts were used as a model of IB permeability. Heat-killed (HKB) or live Salmonella Infantis bacteria were used to induce IB integrity disruption upon three different testing conditions: pre-, co-, and post-treatment with 0.2 v/v% HII. Transepithelial electrical resistance values were used as an indicator of monolayer integrity before and after all treatments, and RT-PCR was used to assess the expression of tight junction proteins (TJPs) and inflammatory cytokines known to regulate intestinal permeability. Results: We found that all three treatments with HII improved the HKB-induced integrity disruption and decreased the down-regulation of TJP1, OCLN, and CLDN1, with the greatest effect observed in the pre-treated cells. Treatment with HII also decreased the up-regulation of CLDN2, TNF-α, IL-1β, and IL-6. In addition, pre-treatment of Caco-2 cells with HII prevented translocation of S. Infantis but did not prevent adhesion and invasion. Conclusion: This study showed that HII can improve inflammation-disrupted IB function by indirect modulation of mRNA expression of TJPs, especially in a preventive manner. [ABSTRACT FROM AUTHOR]

15. Experiences with rabies eradication programs

Abstract

Oral vaccination as a method of rabies eradication in the field was first started in Switzerland in 1978 and after 1984 several other EU countries followed this practice. Due to oral vaccination some European countries are now rabies-free in terrestrial animals. In Slovenia, after the first experimental oral vaccination and study of vaccination models from 1988 -1992, the spring-autumn campaigns have been carried out since 1995. The model of oral vaccination of wildlife requires 16-20 baits per km2 in the vaccination area. The baits were distributed by plane. They were dispersed from a height of 300-500 m. The aeroplanes' paths were 1000 metres apart. In the vaccination campaigns two vaccines were used. Lysvulpen®, produced by the Bioveta company at the Czech Republic, was laid down in the southwestern part of the country, and Fuchsoral®, produced by the German company Impfstoffwerk Dessau-Tornau, was placed in the eastern part of Slovenia. A rapid decline of rabies was evidenced from 1995 to 1999, when the oral vaccination program in the whole territory using the aircraft baits distributing system was practiced. In 1999, only 6 rabies cases were laboratory-confirmed, whereas in 1995, 1089 rabies cases were documented. Of the 14 rabies cases detected in 1998, 12 were found as an island in a circle with a radius of 30 km in the centre of the vaccinated area. In 2000 and 2001, rabies incidence increased again, so it was decided to change the baits distribution system in the year 2001. The vaccination by crossing flights in certain areas was introduced. In the next year (2002), after changing the vaccination strategy, positive cases rapidly dropped and only 15 cases in 2002, and 8 cases were found in 2003, near the non-vaccinated border with Croatia.

24. Physical activity regulates the intestinal microbiota composition: Telesna aktivnost uravnava sestavo črevesne mikrobiote

Abstract

Gut microbiota is the name given today to the bacterial population living in our intestine. It provides nutrients, metabolites and affects the immune system. Recent ani-mals and human studies suggest that regular physical activity increases the presence of beneficial microbial species of gut microbiota and improves the health status of the host. When gut bacteria diversity reduces, there are systemic consequences leading to gastrointestinal, physiological and psychological distress. This review describes the communication pathway of the microbiota-gut-brain axes and other possible mecha-nisms by which physical activity causes changes in microbiota composition. Further-more, it provides the latest evidence of the beneficial role of exercise, which in turn can affect health and various disease processes. The results of research studies in this area are increasingly becoming a focus of scientific attention. Črevesna mikrobiota je bakterijska združba v črevesju. Zagotavlja hranila, meta-bolite in vpliva na imunski sistem. Nedavne študije na živalih in ljudeh nakazujejo, da redna telesna aktivnost povečuje prisotnost koristnih bakterijskih vrst, ki so del čreve-sne mikrobiote in na ta način izboljšuje zdravstveno stanje gostitelja. Upad raznolikosti črevesnih bakterij vodi v sistemske posledice, ki povzročajo gastrointestinalne, fiziolo-ške in psihološke težave. Pregledni članek opisuje komunikacijsko pot, os mikrobiota--črevesje-možgani ter druge možne mehanizme, s katerimi telesna aktivnost povzroča spremembe v sestavi mikrobiote. Poleg tega ponuja najnovejše dokaze o koristni vlogi telesne aktivnosti, kar lahko vpliva na zdravje in različne bolezenske procese. Rezultati raziskav na tem področju so vse bolj v središču pozornosti znanstvenikov.

Abstract

Polymerase chain reaction (PCR) has enabled enormous progress in the field of molecular biology in the last twenty-five years. It became popular due to its high sensitivity and specificity and was introduced to numerous scientific fields. Today many different variants of PCR exist, with real-time PCR being the most common. This method is being successfully used at our institution for a variety of different applications, among which gene expression analyzes and development of protocols for clinical diagnostics currently dominate. But the extreme flexibility and customizability of real-time PCR, coupled with our team’s expertise, ensures efficient application of this technique to various new research projects.

Abstract

Molecular methods provide new alternatives for characterization of viruses and for the diagnosis of animal diseases. A wide range of PCR assays have been developed for routine detection of viral nucleic acid in different clinical specimens. Sequences obtained by direct sequencing of PCR products are used to deduce phylogenetic relationships among virus isolates in order to trace virus evolution and routes of dissemination. Direct sequence analysis of PCR products is now regulary used for the identification of genetic variants of variouse viruses.
