Abstract

Rapid digestion of protein pp60(sup c-src) tyrosine kinase (TK) enables mapping of the time course for autophosphorylation of src TK on three tyrosine sites and provides direct evidence for an intramolecular pY530-SH2 complex. The digestion process involves perfusive-immobilized enzyme cartridges in combination with electrospray ionization mass spectrometry, and the results support earlier models on phosphorylation. The pY530-SH2 interaction modulates signal transduction through a five-fold down-regulation of src TK activity along with inhibition of protein-protein interactions.

Abstract

The forward and backward reactions of pp60(super c-src) tyrosine kinase enzyme have a bi-bi rapid equilibrium random mechanism. The addition of substrates in the reactions is random and sequential. The high-affinity binding of the enzyme involves interactions between the peptide amino acid and the enzyme. Due to the low binding synergy, the affinity of an inhibitor bound to one substrate is unaffected by another substrate bound to the enzyme. The reactions stop when MgATP and (FGE)(sub 3)-pY-(GEF)(sub 2)GD or MgADP and (FGE)(sub 3)Y(GEF)(sub 2)GD bind to the enzyme.

Abstract

Autophosphorylation of pp60(super c-src) tyrosine kinase (srcTK) is an intermolecular process. The electrospray ionization mass spectrum of srcTK reveals that MgATP is effective in increasing the phosphorylation process. The enzyme srcTK is similar to receptor tyrosine kinases in its ability to dimerize and indicates that the dimerization which precedes activation does not necessitate binding to a membrane or a hydrophobic N-terminal region.

19. Model-Driven Analytics of Energy Meter Data in Smart Homes

Abstract

The purpose of this study is to develop a deeper understanding of the health issues facing adolescent females (ages 18-21) in rural, northeastern Afghanistan. Incorporating participant observations, in-depth interviews, and narrative inquiries, this study seeks to illustrate adolescent females’ perspectives on health issues. To achieve this goal, ten adolescent females were interviewed in rural, northeastern Afghanistan during 2010. The participants were between 18- and 21-years old. The one-on-one interviews were conducted in a multiple-response format and were structured around three research questions: How does a young female’s understanding of health issues shape her identity in northeastern Afghanistan? In what ways do the narrative stories of Afghan females link to issues such as education, health, and family dynamics? In what ways are the narrative stories of Afghan females linked to their cultural beliefs about health? The participants were asked to discuss their perceptions of life, health, body image, illness, and related topics. The participants struggled to articulate answers to these questions, but their personal narratives and body language vividly illustrated the issues they struggled to express. Their narrative responses are reduced in this study to produce a dynamic perspective on adolescent females’ perceptions of health issues in northeastern Afghanistan. During the course of this study, it became apparent that: Family influences shaped the participants’ social world. They relied upon their families for daily communication, information, and moral and emotional support. Despite this social dependence, the participants demonstrated a marked reluctance to discuss health issues with their mothers or with other older females in their households. The participants rarely connected health with its traditional meaning (i.e. physical or mental well-being). For them, health was related to a good life—free from stress and care—and education. There is still much more to learn about adolescent health in rural Afghanistan; yet an understanding of these cultural constructs of family, health, and education is necessary to pursue further inquiries. This study’s findings provide the groundwork for future research and discussion—and, ultimately, a deeper understanding of adolescent females’ perceptions of health in rural Afghanistan.

Abstract

To examine the possible role of basic fibroblast growth factor (FGF) in regulating the effects of TNFa, we tested the effect of FGF on TNFa-mediated PGE2 production and TNFa receptor expression in human fibroblasts. We found that, while FGF alone had no effect on PGE2 production, it enhanced the amount of PGE2 produced in response to TNFa between 3 and 11-fold. FGF stimulated TNFa-induced PGE2 production independent of potential TNFa-mediated IL-1 production, as neither anti-IL-1 mAbs nor IL-1 receptor antagonist protein (IRAP) inhibited TNFa induced-PGE2 production or the stimulatory effect of FGF. A one minute exposure of cells to FGF prior to removal was sufficient to significantly enhance TNFa-induced PGE2 production; the maximal FGF effect was reached after a 6 h preincubation. We also found that FGF significantly enhanced TNFa receptor expression. Untreated fibroblasts expressed ˜3,900 receptors/cell, while cells treated with FGF for 6h expressed ˜9,500 receptors/cell, a 2.4-fold increase in receptor number; there was no apparent change in affinity for TNFa (Kd 3.8×10-11 M). The FGF-mediated increase in TNFa receptor expression and TNFa-mediated PGE2 production could be abolished by FGF mAbs, indicating a specific FGF effect. These results show that FGF increases TNFa receptor expression and suggest that this may account, at least in part, for the ability of FGF to enhance TNFa-mediated PGE2 production in human fibroblasts.

50. Exploration of the Sequence Specificity of pp60c-srcTyrosine Kinase

Abstract

The minimum length required for phosphorylation of a peptide by pp60c-srctyrosine kinase (srcTK) was delineated in this work. Budde (M. D. Anderson University of Texas, personal communication) suggested that the peptide (FGE)3Y(GEF)2GD (peptide I) was a “good” srcTK substrate. Peptide I yielded a 251-fold higher kcat/Kmthan RRLIEDAEYAARRG, a peptide substrate based upon the autophosphorylation site of srcTK. This was due to a 38-fold lower Kmand a 6.6-fold increase in kcat. N-terminal truncation of up to 8 residues in a series of peptides yielded only a 3-fold decrease in activity. Removal of the final N-terminal residue resulted in a 10-fold loss in substrate activity, primarily as a result of an increase in the Km. C-terminal truncations ending in the amide yielded no significant loss in activity until the Y+3 residue was removed, which resulted in a 73-fold decrease in kcat/Kmrelative to peptide I. The latter was due primarily to an increase in Km. The results from peptides truncated on both termini suggest that subsite recognition N- and C-terminal relative to the site of phosphorylation can be examined independently. In addition, the observation that only 5 residues are required for significant substrate activity suggests that small molecule inhibitors based upon interactions with the phosphoacceptor site may be developed.
