Strain gradient continuum damage modelling has been applied to quasistatic brittle fracture within an approach based on a maximum energy-release rate principle. The model was implemented numerically, making use of the FEniCS open-source library. The considered model introduces non-locality by taking into account the strain gradient in the deformation energy. This allows for stable computations of crack propagation in differently notched samples. The model can take wedges into account, so that fracture onset can occur at wedges. Owing to the absence of a damage gradient term in the dissipated energy, the normal part of the damage gradient is not constrained on boundaries. Thus, non-orthogonal and non-parallel intersections between cracks and boundaries can be observed. [ABSTRACT FROM AUTHOR]

Osteosarcoma (OS) is a rare form of primary bone cancer, impacting approximately 3.4 × 10 6 individuals worldwide each year, primarily afflicting children. Given the limitations of existing cancer therapies, the emergence of nanotheranostic platforms has generated considerable research interest in recent decades. These platforms seamlessly integrate therapeutic potential of drug compounds with the diagnostic capabilities of imaging probes within a single construct. This innovation has opened avenues for enhanced drug delivery to targeted sites while concurrently enabling real-time monitoring of the vehicle's trajectory. In this study, we developed a nanotheranostic system employing the layer-by-layer (LbL) technique on a core containing doxorubicin (DOXO) and in-house synthesized carbon quantum dots. By utilizing chitosan and chondroitin sulfate as polyelectrolytes, we constructed a multilayered coating to encapsulate DOXO and docetaxel, achieving a coordinated co-delivery of both drugs. The LbL-functionalized nanoparticles exhibited an approximate size of 150 nm, manifesting a predominantly uniform and spherical morphology, with an encapsulation efficiency of 48% for both drugs. The presence of seven layers in these systems facilitated controlled drug release over time, as evidenced by in vitro release tests. Finally, the impact of the LbL-functionalized nanoparticles was evaluated on U2OS and Saos-2 osteosarcoma cells. The synergistic effect of the two drugs was found to be crucial in inducing cell death, particularly in Saos-2 cells treated with nanoparticles at concentrations higher than 10  μ g/ml. In this paper, we exploit some results in the theory of irreversible phenomena to address the study of quasi-static brittle fracture propagation in a two-dimensional isotropic continuum. The elastic strain energy density of the body has been assumed to be geometrically nonlinear and to depend on the strain gradient. Such generalized continua often arise in the description of microstructured media. These materials possess an intrinsic length scale, which determines the size of internal boundary layers. In particular, the non-locality conferred by this internal length scale avoids the concentration of deformations, which is usually observed when dealing with local models and which leads to mesh dependency. A scalar Lagrangian damage field, ranging from zero to one, is introduced to describe the internal state of structural degradation of the material. Standard Lamé and second-gradient elastic coefficients are all assumed to decrease as damage increases and to be locally zero if the value attained by damage is one. This last situation is associated with crack formation and/or propagation. Numerical solutions of the model are provided in the case of an obliquely notched rectangular specimen subjected to monotonous tensile and shear loading tests, and brittle fracture propagation is discussed., Competing Interests: We have no competing interests.

31P nuclear magnetic resonance spectroscopy has been applied to quantitate phospholipids in human blood plasma and in separate lipoprotein fractions. The addition of suitable detergents to samples produces an excellent chemical shift dispersion and allows the identification and integration of the peaks of the most important phospholipids. Results are in agreement with those obtained with enzymatic colorimetric and TLC methods: our method is characterized by good accuracy and reproducibility.

Surgical specimens from 22 patients with infiltrating ductal carcinoma of the breast were analyzed by high-resolution 1H NMR spectroscopy at 80 and 300 MHz. Results were compared with those obtained from 9 benign (proliferative dysplasia without atypia) breast samples. NMR spectra at 80 MHz showed well-resolved water and lipid peaks so it was possible to calculate specific T1 and T2 relaxation times. The longitudinal T1 relaxation process of the water was found to be properly represented by a monoexponential function, whereas the transverse T2 decay was well accounted for by a biexponential model. Relaxation time measurements of the water were found to be a valuable tool in breast tissue characterization, in particular (a) mean T1 values from malignant and control populations are well distinguished (P less than 0.0008); (b) mean T2 values of both fast and slow components from the two populations are statistically different (P less than 0.0001) and no overlap exists in the distributions of the second component. Three-hundred-megahertz spectra allowed a detailed analysis of the lipidic component of the sample: malignant tumors were found to present in the region 1.3-1.2 ppm a species characterized by a very slow transverse relaxation time (T2 = 200-1000 ms).
