A produção de hortaliças representa um setor importante do agronegócio brasileiro. Apesar de ser um mercado altamente diversificado em termos de produtos, cerca de doze espécies de olerícolas representam mais de 80% do valor total gerado. A cadeia produtiva de hortaliças possui vários entraves ao aumento da produção em razão de adversidades climáticas, custo dos insumos e incidência de pragas e doenças. Diversos problemas fitossanitários ainda desafiam e limitam a produtividade de diversas culturas, merecendo destaque patógenos que sobrevivem no solo por longos períodos, que possuem ampla gama de hospedeiras, incluindo muitas plantas invasoras, e que são difíceis de controlar. Espécies dos gêneros Pythium e Phytopythium estão entre os patógenos mais comumente distribuídos no solo, sendo que a dificuldade de controle dessas espécies em hortaliças baseia-se na oferta reduzida de defensivos, elevado números de espécies patogênicas e o surgimento de populações resistentes aos ingredientes ativos utilizados. Dentre os principais fungicidas utilizados no controle de oomicetos merece destaque o mefenoxam, o qual é amplamente utilizado no mundo para diversas culturas. Apesar de ser considerado efetivo, populações de Pythium e Phytopythium resistentes ao mefenoxam já têm sido observadas, comprometendo assim o controle em campo. Dessa forma, o objetivo desse trabalho foi identificar isolados de Pythium e Phytopythium obtidos de plantas olerícolas e invasoras provenientes de diversas regiões produtoras de hortaliças do Brasil, avaliar a sensibilidade ao fungicida mefenoxam in vitro e determinar a gama de hospedeiras entre diversas espécies olerícolas. Para a identificação inicial dos isolados, o DNA genômico foi extraído para o sequenciamento das regiões ITS do rDNA e citocromo c oxidase II (Cox II). Em seguida, avaliou-se a sensibilidade in vitro ao fungicida mefenoxam nas concentrações de 5, 10 e 100 g.mL-1 e a gama de hospedeiras dos isolados em 17 cultivares pertencentes a 14 culturas olerícolas diferentes. Por fim, foi realizado um levantamento de espécies de Pythium e Phytopythium associadas a plantas invasoras, com sintomas de tombamento ou podridão de raiz, em campos de produção de hortaliças no Brasil. Estas foram identificadas da mesma forma que os isolados obtidos de hortaliças. A comparação das sequências da região ITS de 127 isolados provenientes de 25 culturas olerícolas, resultou na identificação de 10 espécies de Pythium que atacam diferentes culturas, a saber: P. acanthophoron, P. aphanidermatum, P. deliense, P. myriotylum, P. oopapillum, P. spinosum, P. splendens, P. sylvaticum, P. tracheiphilum e P. ultimum var. ultimum, e três espécies de Phytopythium, Phy. chamaehyphon, Phy. cucurbitacearum e Phy. vexans. Entre os 65 isolados testados para a sensibilidade in vitro a mefenoxam, apenas dois isolados de P. ultimum var. ultimum (Pyt 481 e Pyt 553), possuem reação intermediária, com crescimento micelial maior que 50% em relação à testemunha na concentração de 10 g.mL-1 . Esses mesmos isolados tiveram crescimento reduzido na concentração de 100 g.mL-1 . Na avaliação das possíveis hospedeiras, as espécies com ampla gama de hospedeiras entre as cultivares avaliadas, foram: P. myriotylum, P. deliense, P. aphanidermatum e P. ultimum var. ultimum. O teste de patogenicidade em 17 cultivares comprova que P. myriotylum, P. deliense e P. aphanidermatum foram capazes de causar doença em dez, nove e oito cultivares, respectivamente. Contrariamente, Pythium sylvaticum infecta somente o pimentao e P. tracheiphilum (Pyt 635) apenas alface. Phytopythium cucurbitacearum não causou doença nas culturas avaliadas. Phytopythium chamaehyphon foi reisolado de pimenta e rúcula, enquanto Phy. vexans causou podridão de cenoura, coentro e rúcula. As principais espécies de Pythium recuperadas de plantas invasoras foram P. aphanidermatum (doze isolados) e P. ultimum var. ultimum (oito isolados) de um total de 34 isolados. As espécies P. myriotylum (dois isolados), P. sylvaticum, Phy. chamaehyphon e Phy. oedochilum (um isolado de cada) também foram identificadas. Os resultados desse trabalho comprovam a diversidade de espécies de Pythium e Phytopythium associadas a hortaliças e plantas invasoras no Brasil, sendo P. aphanidermatum a espécie com maior prevalência e ampla distribuição entre as regiões produtoras. A utilização de mefenoxam é uma alternativa viável para o controle de espécies de Pythium e Phytopythium em áreas produtoras de hortaliças. Esses resultados ressaltam a importância da identificação precisa de oomicetos, determinação da gama de hospedeiros e sensibilidade a fungicidas dos isolados para recomendação de estratégias de manejo em cultivos de hortaliças. Vegetable crops represents an important sector of Brazilian agribusiness. Despite being a highly diversified market in terms of products, about twelve species of vegetables represent more than 80% of the total value generated. The vegetable production chain has several obstacles to increasing production due to climatic adversities, the cost of inputs and the incidence of pests and diseases. Several phytosanitary problems still challenge and limit the productivity of several crops, with emphasis on soilborne plantpathogens that survive in the area for long periods, have a wide host range, including many weeds, and are difficult to control. Species of Pythium and Phytopythium genus are among the pathogens most commonly distributed in the soil, and the difficulty in controlling these species in vegetables is based on the reduced supply of pesticides, high numbers of pathogenic species and the emergence of resistant populations to the active ingredients used. Among the main fungicides used in the control of oomycetes, mefenoxam deserves mention, which is widely used in the world for several cultures. Despite being considered effective, resistants Pythium and Phytopythium populations to mefenoxam have already been observed, compromising the control in the field. Thus, the aim of this work was to idenfity Pythium and Phytopythium isolates obtained from vegetable and weed from different vegetable producing regions of Brazil, to evaluate the sensitivity to the fungicide mefenoxam in vitro and to determine the host range among different vegetable species. For the initial identification of the isolates, genomic DNA was extracted for sequencing of ITS region of the rDNA and cytochrome oxidase II (Cox II). Then, evaluated the in vitro sensitivity to the fungicide mefenoxam at 5, 10 and 100 g.mL-1 concentrations and the host range of some isolates in 17 cultivars belonging of 14 vegetable crops. Finally, a survey was carried out of Pythium and Phytopythium species associated with weeds with symptoms of damping-off or root rot obtained from vegetable production fields in Brazil. These were identified in the same way as the isolates obtained from vegetables. The sequences comparation of ITS region of 127 isolates from 25 vegetable crops, resulted in identification of 10 species of Pythium that attack different cultures, including: P. acanthophoron, P. aphanidermatum, P. deliense, P. myriotylum , P. oopapillum, P. spinosum, P. splendens, P. sylvaticum, P. tracheiphilum and P. ultimum var. ultimum, and three species of Phytopythium, Phy. chamaehyphon, Phy. cucurbitacearum and Phy. vexans. Among the 65 isolates tested for in vitro sensitivity to mefenoxam, only two P. ultimum var. ultimum isolates (Pyt 481 and Pyt 553), showed intermediate reaction, with mycelial growth higher than 50% in relation to the control in the 10 g.mL-1 concentration. These same isolates have reduced colony diameter in the 100 g.mL-1 concentration. The other isolates were sensitive to mefenoxam in the culture medium. In the evaluation of possible hosts, the species with widest range of hosts among the cultivars evaluated were: P. myriotylum (Pyt 604), P. deliense (Pyt 589), P. aphanidermatum (Pyt 586 and Pyt 600) and P. ultimum var. ultimum (Pyt 502). The patogenicity test in 17 cultivars prove that P. myriotylum, P. deliense and P. aphanidermatum caused disease in tem, nine and eight cultivars, respectively. Contrary, Pythium sylvaticum (Pyt 692) infected only peppers and P. tracheiphilum (Pyt 635) only lettuce. Phytopythium cucurbitacearum (Pyt 650) did not cause disease in any evaluated crop. Phytopythium chamaehyphon was reisolated from pepper and arugula, and Phy. vexans (Pyt 535) caused root rot in carrots, coriander and arugula. The main Pythium species recovered from weeds were P. aphanidermatum (twelve isolates) and P. ultimum var. ultimum (eight isolates) of a total of 34 isolates. Pythium myriotylum (two isolates), P. sylvaticum, Phy. chamaehyphon and Phy. oedochilum (one isolate from each) were also identified. The results of this work demonstrate that there is high diversity of Pythium and Phytopythium species associated with vegetables and weeds in Brazil, with P. aphanidermatum being the species with the highest prevalence and wide distribution among the producing regions. The use of mefenoxam is a viable alternative for Pythium spp. and Phytopythium spp. control in vegetable-producing areas. These results emphasize the importance of accurate identification of oomycetes, determination of the host range and sensitivity to fungicides of the isolates to recommend management strategies in vegetable crops.
