The objective of the present study was to detect the genetic diversity of Anaplasma marginale strains in naturally infected calves from a rural property located in the northeastern region of the state of Pará, Eastern Amazon, which has a history of mortality due to anaplasmosis. Fourteen calves positive for A. marginale were selected using a semi-nested polymerase chain reaction for the target msp1α gene, with asymptomatic (n=3) and symptomatic (n=11) infections. After sequencing the samples, two genotypes were verified in the E and C regions and the structures in tandem repeats were determined. Nine different strains were found: eight related to the E genotype (α-β-β-Γ = one animal, asymptomatic; 16-F-17-F-F = two animals, symptomatic; α-β-F-F-F-F = one animal, asymptomatic; 31-62-62-61 = one animal, symptomatic; τ-10-3 = three animals, two symptomatic and one asymptomatic; α-β-β-β = one animal, symptomatic; τ-22 -13-18 = two animals, both symptomatic; β-β-β-BRA1-31 = two animals, both symptomatic), and one related to genotype C (23-24-25-31-27-27 = one animal, asymptomatic). Genotype E was predominant in 92.86% of the samples (13/14), followed by genotype C (7.14%). This study made it possible to detect the genetic diversity of A. marginale in calves from the selected dairy farm, in addition to identifying the BRA1 sequence in the animals of the present study, which was recently diagnosed in Minas Gerais, demonstrating the dispersion of A. marginale strains in herds from different Brazilian states. Genetic diversity of A. marginale was observed in both symptomatic and asymptomatic calves. There were no significant differences when clinical signs were compared to the genotype verified in the infected animals. The prevalence of pathogenicity was not observed. RESUMO: O objetivo do presente trabalho foi detectar a diversidade genética de cepas de Anaplasma marginale em bezerros naturalmente infectados oriundos de uma propriedade rural localizada na região nordeste do estado do Pará, Amazônia Oriental, a qual apresentava histórico de mortalidade devido à anaplasmose. Foram selecionados 14 bezerros positivos para A. marginale pela técnica de semi-nested PCR (nPCR) para o alvo no gene msp1α, com infecção assintomática (n=3) e sintomáticos (n=11). Após o sequenciamento das amostras foram verificados dois genótipos nas regiões E e C, e determinadas as estruturas em tandem repeats. Nove diferentes estirpes foram encontradas, sendo oito relacionadas ao genótipo E (α-β-β-Γ = um animal, assintomático; 16-F-17-F-F = dois animais, sintomáticos; α-β-F-F-F-F = um animal, assintomático; 31-62-62-61 = um animal, sintomático; τ-10-3 = três animais, dois sintomáticos e um assintomático; α-β-β-β = um animal, sintomático; τ-22-13-18 = dois animais, sintomáticos; β-β-β-BRA1-31 = dois animais, sintomáticos) e uma relacionada ao genótipo C (23-24-25-31-27-27 = um animal, assintomático). O genótipo E foi predominante em 92,86% das amostras (13/14), seguido pelo genótipo C (7,14%). O estudo possibilitou a detecção da diversidade genética de A. marginale em bezerros dessa propriedade leiteira, além de identificar a sequência BRA1 nos animais do presente estudo, a qual foi diagnosticada recentemente em Minas Gerais, o que demonstra a dispersão das estirpes de A. marginale nos rebanhos de diferentes estados brasileiros. A diversidade genética de A. marginale foi observada tanto em bezerros sintomáticos quanto em assintomáticos e não houve diferença significativa quando se comparou os sinais clínicos ao genótipo verificado no animal infectado, não observando a prevalência de patogenicidade de estirpes.

This study collected samples from 50 Holstein cows, most intensively bred, and from the Campos Gerais region, Paraná, with an average milk production of 30.21L/day. Samples of the liver, spleen and lymph nodes were collected to determine the levels of copper, cobalt, iron, molybdenum and zinc. Spleen and lymph nodes were subjected to histological analysis and evaluation of the degree of hemosiderosis. The average concentrations of copper (495.05ppm), molybdenum (4.19ppm), and zinc (274.49ppm) were higher than those established for the bovines. For cobalt 26% of the animals presented levels below the established level, which characterized cobalt deficiency. Only iron (299.12ppm) exhibited an adequate average level. Histopathologically hemosiderosis was observed mainly in the spleen (78%) and less frequently in the lymph nodes (20%). The observation of hemosiderin in the spleen and lymph nodes is not related to copper deficiency. Still it may be related to high levels of molybdenum, zinc, iron, or other undetermined causes. RESUMO: Para este estudo foram coletadas amostras de 50 vacas, da raça Holandesa, a maior parte criada intensivamente e oriunda da região dos Campos Gerais/PR, com média individual de produção de leite de 30,21L/dia. Foram coletadas amostras de fígado, baço e linfonodos para determinação dos níveis de cobre (Cu), cobalto (Co), ferro (Fe), molibdênio (Mo) e zinco (Zn). Baço e linfonodo foram submetidos à análise histológica e avaliação do grau de hemossiderose. A média das concentrações de Cu (495,05ppm), Mo (4,19ppm) e Zn (274,49ppm) encontrava-se acima dos níveis estabelecidos para bovinos. Para o Co, observou-se que 26% dos animais apresentaram níveis abaixo do estabelecido, o que caracterizou deficiência de Co. Apenas o Fe (299,12ppm) apresentou níveis médios adequados. Na histopatologia, hemossiderose foi observada, principalmente no baço (78%) e com menos frequência nos linfonodos (20%). A observação de hemossiderina no baço e linfonodos não está relacionada à deficiência de Cu, porém pode estar relacionada a elevados níveis de Mo, Zn e Fe ou a outras causas não determinadas.

Periodontitis is an inflammatory response in a susceptible host caused by complex microbiota, predominantly composed of Gram-negative anaerobic bacteria. Aiming to characterize the subgingival bacterial microbiota associated with ovine periodontitis, polymerase chain reaction (PCR) was performed in subgingival periodontal pocket samples of 14 sheep with severe periodontitis and in subgingival sulcus biofilm of 14 periodontally healthy sheep in search mainly of Gram-negative and Gram-positive microorganisms considered important periodontopathogens. The most prevalent bacteria in the sheep with periodontal lesions were Tannerella forsythia (78.6%), Treponema denticola (78.6%), Fusobacterium nucleatum (64.3%), and Porphyromonas gingivalis (50%), whereas in the healthy sheep, F. nucleatum (42.8%) was the most often detected bacterium. RESUMO: Foram avaliados os fatores epidemiológicos e as afecções podais de bovinos de corte no estado do Pará em 12 propriedades. Em todas foi realizado o estudo epidemiológico e em nove o estudo epidemiológico e o exame clínico dos animais. Nos centros de manejo foram observados fatores favoráveis a traumatismos nos dígitos dos bovinos, como piso das seringas calçado com pedras, troncos com exposição de ferragens e rampas dos embarcadores com pisos de pedras pontiagudas, além da falta de manutenção e erros estruturais. Em todas as fazendas observou-se um manejo inadequado dos animais. Em 91,7% das fazendas (11/12) havia piquetes com presença de troncos de árvores e 66,7% (8/12) apresentavam áreas de relevo inclinado com presença de pedras. Em 16,7% (2/12) das fazendas havia áreas de brejo com áreas alagadas. A sodomia foi relatada em todas as propriedades. Nenhuma fazenda adotava medidas profiláticas relacionadas às afecções podais. Foram examinados 498 bovinos entre fêmeas e machos. Os membros pélvicos foram mais acometidos, tanto nas fêmeas quanto nos machos. Foram diagnosticadas 629 lesões nas fêmeas, sendo as mais frequentes pododermatite séptica, deformações ungulares, pododermatite da sobreunha e dermatite digital. Nos machos diagnosticou-se 285 lesões e as mais frequentes foram pododermatite séptica, deformações ungulares e abrasão de talão. Conclui-se que erros de manejo associados às condições inadequadas das instalações foram fatores que contribuíram para a ocorrência de enfermidades podais em bovinos de corte nas fazendas estudadas; as características ambientais favoreceram o desenvolvimento das lesões; as associações de lesões foram mais prevalentes do que as simples, sendo diagnosticadas em animais em todas as fazendas estudadas; o diagnóstico precoce a campo não era realizado, o que contribuiu para aumentar a gravidade e a diversificação das afecções podais diagnosticadas. ABSTRACT: The epidemiological study was conducted in 12 farms with the history of foot disease occurrence. Clinical examination of the affected cattle for the diagnosis of foot disease was performed only on nine of these farms. Multiple risk factors were found for digit injuries, as syringes floors paved with rocks, working chutes with iron fittings exposure, loading ramps covered with sharp stones and the lack of maintenance and structural errors. On each farm there was an inappropriate handling of cattle. On 91.7% of them (11/12) existed pickets with trunks of trees and 66.7% (8/12) had areas of sloping relief with stones. Moreover, 16.7% (2/12) of the farms were in marsh areas and wetlands. Sodomy was reported for all farms. None of them adopted preventive measures related to foot problems. A total of 498 cattle, males and females, were examined. The hind limbs were the most affected in both, females and in males. In females 629 lesions were diagnosed. The septic pododermatitis was the most common, followed by claw deformities, pododermatitis in the paradigits and digital dermatitis. In males 285 injuries were diagnosed; the most common were septic pododermatitis, claw deformities and heel erosion. It was concluded that management errors associated with inadequate facilities were factors that contributed to the occurrence of foot diseases in beef cattle of the farms studied. The environmental characteristics favored the development of lesions; combined injuries were more prevalent than the simple ones diagnosed in cattle of all farms. Early diagnosis was not carried out in the field, what helped to increase the severity and the diversification of the foot problems.

The objective of this study was to describe the epidemiological, clinical and pathological aspects of an outbreak of periodontitis in sheep that occurred on a rural property in Benevides, in the state of Pará, Brazil. The first clinical signs of this disease in the herd - visible nodular swollen mandible - were observed about one month after the sheep began grazing on a recently renovated Panicum maximum cv. Massai pasture; the animals were also supplemented with elephant grass (Pennisetum purpureum). From the 545 adult Santa Ines, Dorper, Texel and crossbred sheep, 20 (3.7%) showed facial bulging, particularly of the mandible. These 20 sheep, which were generally over 36 months of age, had low body condition scores as well as dull and ruffled coats; many of them had loose or missing pre, upper or lower molars, showed the formation of abscesses and fistulas, exhibited pain upon palpation and had difficulty chewing. In the same herd, the prevalence of periodontal lesions at the premolars and molar teeth was evaluated via post-mortem examination of 39 young and 17 adult sheep. In 51.3% (20/39) of adolescents and in 100% of adults, periodontal lesions were found at least at one tooth of the dental arch, with unilateral or bilateral periodontal lesions at the maxillary and mandibular teeth. Histopathological analysis of 13 sheep revealed pyogranulomatous inflammation. The second and third maxillary premolars were the teeth that were most affected in young animals, whereas the third pre-molar and the molars, of the maxilla and mandible, were most often affected in adults. These lesions led to severe bone destruction, alterations in the dental arch, occlusion, tooth loss and periodontal abscesses. The epidemiological and clinic-pathological characteristics of this periodontitis outbreak, described here for the first time in sheep in Brazil, were similar to those for bovine periodontitis (“cara inchada”). RESUMO: O trabalho teve por objetivo descrever os aspectos epidemiológicos e clínico-patológicos de um surto de periodontite em ovinos, ocorrido em uma propriedade rural em Benevides, no estado do Pará, Brasil. Os primeiros sinais clínicos da presença da enfermidade no rebanho, visualizados como abaulamento nodular da mandíbula, foram observados aproximadamente um mês após a introdução dos animais em área de pastagem de Panicum maximum cv. Massai, que havia sido reformada recentemente, e suplementados com capim Elefante (Pennisetum purpureum). De 545 ovinos adultos, das raças Santa Inês, Dorper, Texel e seus mestiços, 20 (3,7%) apresentaram abaulamento facial, predominantemente mandibular. Os 20 animais, na sua maioria com idades acima dos 36 meses, apresentavam baixo escore corporal, pelos arrepiados e sem brilho, alguns com afrouxamento e perda dos dentes pré-molares e molares inferiores e superiores, formação de abscesso e fístula, demonstrando dor à palpação e dificuldade na mastigação. No mesmo rebanho, a prevalência de lesões periodontais nos dentes pré-molares e molares foi avaliada por meio do exame post-mortem em 39 ovinos jovens e 17 adultos. Assim, 51,3% (20/39) dos jovens e todos os adultos apresentavam lesões periodontais em pelo menos um dos dentes da arcada, com ocorrência uni ou bilateral nos maxilares e mandibulares. A análise histopatológica de 13 animais revelou processo inflamatório piogranulomatoso. Os dentes mais afetados nas arcadas dos animais jovens foram os segundo e terceiro pré-molares maxilares. Já nos adultos foram o terceiro pré-molar e os molares, tanto maxilares quanto mandibulares. As lesões causaram severa destruição óssea, alteração na arcada dentária, na oclusão, perda dental e ocorrência de abscessos periodontais. As características epidemiológicas e clínico-patológicas do surto de periodontite em ovinos, aqui descritos originalmente nesta espécie animal no País, assemelham-se às da periodontite bovina (“cara inchada”).

Resumo: Em uma propriedade localizada na região nordeste do Estado do Pará foram analisadas alternativas de suplementação mineral para bovinos de corte, da raça nelore, machos, com idades entre 18 e 20 meses e pesos de 299,5 a 308,5 kg. Foram avaliados o custo benefício, os valores dos minerais no fígado e nas costelas e o ganho de peso médio diário dos animais submetidos aos diferentes tratamentos. O estudo foi realizado durante seis meses (março a agosto de 2012), no período de ótima oferta de pastagem. Foram utilizados 60 bovinos, divididos em três grupos de 20 animais em delineamento experimental inteiramente casualizado. O Grupo 1 foi suplementado com uma mistura mineral seletiva (MMS) composta por 25kg de NaCl, 50kg de fosfato bicálcico,190g de sulfato de Cobre, 60g de sulfato de Cobalto e 15g de selenito de sódio; essa mistura continha, em um quilograma, 133g de Na, 123g de P, 631 mg de Cu, 167mg de Co e 9 mg de Se; essa mistura foi proposta com base no prévio diagnóstico clínico-nutricional do rebanho. O Grupo 2 foi suplementado apenas com NaCl (Grupo controle) e o Grupo 3 recebeu uma mistura mineral comercial (MMC), rotineiramente utilizada na fazenda, contendo, em um quilograma, 130g de P, 220g de Ca, 18g de Mg, 36g de S, 6.000mg de Zn, 1.500mg de Cu, 2.000mg de Mn, 200mg de Co, 90mg de I e 36mg de Se. O melhor custo benefício foi obtido com a mistura mineral seletiva. A pastagem não contém Co suficiente para atender as necessidades dos bovinos do grupo controle, mas foi capaz de suprir as necessidades de P, Cu, Zn e Se durante os cinco meses do experimento. Abstract: Three alternatives of mineral supplementation of 18 to 20-month-old male Nelore cattle with 299.5 to 308.5 kg of body weight were used at a farm in northeast of Pará state, Brazil. To evaluate which mineral mixture resulted in better cost-benefit, three groups of 20 head were supplemented for six months, from March to August of 2012. Group 1 was supplemented with a selected mineral mixture (SMM) composed of 25kg NaCl, 50kg dicalcium phosphate, 190g copper sulphate, 60g cobalt sulphate, and 15g sodium selenite, having 133g Na, 123g P, 631mg Cu, 167mg Co, and 9mg Se per kg; this mixture was made based in previous clinical-nutritional examination of herd. Group 2 (control), supplemented only with NaCl; and group 3 received a commercial mineral mixture (CMM), routinely used in this farm, with 130g P, 220g Ca, 18g Mg, 36g S, 6000mg Zn, 1500mg Cu, 2000mg Mn, 200mg Co, 90mg I, and 36mg Se per kg. Concentrations of phosphorus, copper, zinc and iron were determined in liver and 12th ribs. The best cost-benefit alternative was the SMM; the forage alone did not contain enough Co to meet the requirements of the control group, but provided enough P, Cu, Zn and Se during the five months of the experiment.

RESUMO: O objetivo deste trabalho foi verificar a presença de Brucella abortus e as lesões causadas por esse agente nos anexos fetais e nos fetos de búfalas. Para isso, 20 búfalas em diversos meses de gestação, sorologicamente positivas para brucelose, foram submetidas ao abate sanitário. A idade fetal foi determinada através de exames ultrassonográficos associados à mensuração dos fetos durante a necropsia. Do útero fechado desses animais foram coletadas amostras para histopatologia e qPCR. A partir do segundo mês de gestação foi possível detectar a presença de DNA de B. abortus em líquido amniótico, líquido alantoide e em útero e, a partir do quinto mês, na placenta, coração, baço, rim, pulmão, intestino, fígado e linfonodos dos fetos. Os principais achados anatomopatológicos foram placentite fibrinopurulenta necrótica e endometrite supurativa crônica. ABSTRACT: The objective of this study was to detect Brucella abortus and injuries caused by the bacteria in fetal membranes and fetuses. Twenty buffaloes serologically positive for brucellosis were used and subjected to stamping for collection of material from the closed uterus of several months gestation. Fetal age was determined by ultrasound examination and the size of fetuses was measured at necropsy. The samples were subjected to histopathology and qPCR. From the second month of pregnancy on it was possible to detect the presence of B. abortus DNA in amniotic fluid, allantoic liquid and uterus, and from the fifth month on in placenta, heart, spleen, kidney, lung, intestine, liver and lymph nodes of the fetuses. The main pathological findings were fibrinous suppurative necrotic placentitis, and chronic endometritis.
