5. Robust thalamic nuclei segmentation from T1-weighted MRI

Abstract

Accurate segmentation of thalamic nuclei, crucial for understanding their role in healthy cognition and in pathologies, is challenging to achieve on standard T1-weighted (T1w) magnetic resonance imaging (MRI) due to poor image contrast. White-matter-nulled (WMn) MRI sequences improve intrathalamic contrast but are not part of clinical protocols or extant databases. Here, we introduce Histogram-based polynomial synthesis (HIPS), a fast preprocessing step that synthesizes WMn-like image contrast from standard T1w MRI using a polynomial approximation. HIPS was incorporated into our Thalamus Optimized Multi-Atlas Segmentation (THOMAS) pipeline, developed and optimized for WMn MRI. HIPS-THOMAS was compared to a convolutional neural network (CNN)-based segmentation method and THOMAS modified for T1w images (T1w-THOMAS). The robustness and accuracy of the three methods were tested across different image contrasts, scanner manufacturers, and field strength. HIPS-synthesized images improved intra-thalamic contrast and thalamic boundaries, and their segmentations yielded significantly better mean Dice, lower percentage of volume error, and lower standard deviations compared to both the CNN method and T1w-THOMAS. Finally, using THOMAS, HIPS-synthesized images were as effective as WMn images for identifying thalamic nuclei atrophy in alcohol use disorders subjects relative to healthy controls, with a higher area under the ROC curve compared to T1w-THOMAS (0.79 vs 0.73)., Comment: 5 Figures, 3 Tables, 1 Supplemental Figure, Submitted to Scientific Reports Apr 12 2023

Abstract

Recognition memory is the ability to recognize previously encountered objects. Even this relatively simple, yet extremely fast, ability requires the coordinated activity of large‐scale brain networks. However, little is known about the sub‐second dynamics of these networks. The majority of current studies into large‐scale network dynamics is primarily based on imaging techniques suffering from either poor temporal or spatial resolution. We investigated the dynamics of large‐scale functional brain networks underlying recognition memory at the millisecond scale. Specifically, we analyzed dynamic effective connectivity from intracranial electroencephalography while epileptic subjects (n = 18) performed a fast visual recognition memory task. Our data‐driven investigation using Granger causality and the analysis of communities with the Louvain algorithm spotlighted a dynamic interplay of two large‐scale networks associated with successful recognition. The first network involved the right visual ventral stream and bilateral frontal regions. It was characterized by early, predominantly bottom‐up information flow peaking at 115 ms. It was followed by the involvement of another network with predominantly top‐down connectivity peaking at 220 ms, mainly in the left anterior hemisphere. The transition between these two networks was associated with changes in network topology, evolving from a more segregated to a more integrated state. These results highlight that distinct large‐scale brain networks involved in visual recognition memory unfold early and quickly, within the first 300 ms after stimulus onset. 39. From Perception to Recognition Memory: Time Course and Lateralization of Neural Substrates of Word and Abstract Picture Processing

Abstract

Through study of clinical cases with brain lesions as well as neuroimaging studies of cognitive processing of words and pictures, it has been established that material-specific hemispheric specialization exists. It remains however unclear whether such specialization holds true for all processes involved in complex tasks, such as recognition memory. To investigate neural signatures of transition from perception to recognition, according to type of material (words or abstract pictures), high-resolution scalp ERPs were recorded in adult humans engaged either in categorization or in memory recognition tasks within the same experimental setup. Several steps in the process from perception to recognition were identified. Source localization showed that the early stage of perception processing (N170) takes place in the fusiform gyrus and is lateralized according to the nature of stimuli (left side for words and right side for pictures). Late stages of processing (N400/P600) corresponding to recognition are material independent and involve anterior medial-temporal and ventral prefrontal structures bilaterally. A crucial transitional process between perception (N170) and recognition (N400/P600) is reflected by the N270, an often overlooked component, which occurs in anterior rhinal cortices and shows material-specific hemispheric lateralization.

41. Brain Responses Differ to Faces of Mothers and Fathers

Abstract

We encounter many faces each day but relatively few are personally familiar. Once faces are familiar, they evoke semantic and social information known about the person. Neuroimaging studies demonstrate differential brain activity to familiar and non-familiar faces; however, brain responses related to personally familiar faces have been more rarely studied. We examined brain activity with fMRI in adults in response to faces of their mothers and fathers compared to faces of celebrities and strangers. Overall, faces of mothers elicited more activity in core and extended brain regions associated with face processing, compared to fathers, celebrity or stranger faces. Fathers' faces elicited activity in the caudate, a deep brain structure associated with feelings of love. These new findings of differential brain responses elicited by faces of mothers and fathers are consistent with psychological research on attachment, evident even during adulthood.
