Abstract

Abstract Background Chagas disease is a health threat for many people, mostly those living in Latin America. One of the most important problems in treatment is the limitation of existing drugs. Prodigiosin, produced by Serratia marcescens (Rhodnius prolixus endosymbiont), belongs to the red-pigmented bacterial prodiginine family, which displays numerous biological activities, including antibacterial, antifungal, antiprotozoal, antimalarial, immunosuppressive, and anticancer properties. Here we describe its effects on Trypanosoma cruzi mitochondria belonging to Tc I and Tc II. Results Parasites exposed to prodigiosin altered the mitochondrial function and oxidative phosphorylation could not have a normal course, probably by inhibition of complex III. Prodigiosin did not produce cytotoxic effects in lymphocytes and Vero cells and has better effects than benznidazole. Our data suggest that the action of prodigiosin on the parasites is mediated by mitochondrial structural and functional disruptions that could lead the parasites to an apoptotic-like cell death process. Conclusions Here, we propose a potentially useful trypanocidal agent derived from knowledge of an important aspect of the natural life cycle of the parasite: the vector-parasite interaction. Our results indicate that prodigiosin could be a good candidate for the treatment of Chagas disease.

Abstract

La enseñanza del inglés en el sector oficial colombiano ha sido un reto en ciertos contextos por décadas, debido a una variedad de razones tales como limitadas oportunidades para el desarrollo profesional de los profesores, poca infraestructura y falta de recursos. En relación con esta última, en el pasado, pocos materiales estaban disponibles para el uso o referencia de los (las) docentes en su labor de apoyo del desarrollo la Competencia Comunicativa en inglés de sus estudiantes. Hoy día, las entidades gubernamentales han tomado algunas acciones que apuntan al desarrollo de las competencias en inglés de los estudiantes, de tal manera que ellos sean capaces de utilizar sus habilidades para comunicarse en una lengua extranjera. En consecuencia, el material propuesto a docentes e instituciones para guiar la enseñanza del inglés motivo este estudio de investigación acción, ejecutado con el objetivo de explorar el desarrollo de la Competencia Comunicativa en lectura y escritura a través de la implementación de secuencias didácticas basadas en el Currículo Sugerido de inglés para Colombia expedido por el Ministerio de Educación Nacional. Los participantes de esta investigación fueron veinte seis estudiantes de décimo grado de una institución educativa pública localizada en el área rural de Sahagún, Córdoba. Los instrumentos utilizados para recolectar la información fueron: exámenes, entrevistas, encuestas y tareas escritas que los estudiantes tuvieron que completar. Los resultados principales revelaron que los estudiantes tuvieron un leve avance en su Competencia Comunicativa en lectura y escritura.

Abstract

Se presenta el estudio de la actividad antibacteriana de la especie vegetal Diplostephium tolimense Cuatrec. (Asteraceae), colectada en el páramo del nevado del Tolima, la cual cuenta con pocos estudios químicos y farmacológicos, al igual que muchas otras especies de este ecosistema. A partir de un extracto etanólico crudo y mediante partición biodirigida se obtiene la fracción más simple de mayor actividad frente a Staphylococcus aureus (ATCC 25923). Paralelamente se realizan análisis fitoquímicos a las porciones de mayor bioactividad obtenidas durante todo el proceso. Se observa que la acción antibacteriana se incrementa al aumentar la simplicidad química del extracto y que terpenos y flavonoides parecen estar relacionados con la acción revelada. Se trata del primer reporte, hasta la fecha conocido, de actividad antibacteriana in vitro para D. tolimense. A study of antibacterial activity of the Diplostephium tolimense Cuatrec (Asteraceae) vegetal species, which was collected in the paramo of Tolima, is presented. This species, as many others of this ecosystem, has had few chemical and pharmacological studies. The simplest fraction with the largest activity against Staphylococcus aureus (ATCC 25923), is obtained starting from a raw ethanol extract and by means of bioassay guided partition. Parallelly, phytochemical analyses to the portions of greater bioactivity, obtained during the whole process, are carried out. It is observed that antibacterial action increases as the chemical simplicity increases and that the presence of terpenes and flavonoids seems to be related with this action. This is the first report, so far well-known, of in vitro antibacterial activity for D. tolimense.

26. Effect of an Oxadiazoline and a Lignan on Mycolic Acid Biosynthesis and Ultrastructural Changes of Mycobacterium tuberculosis.

Abstract

Tuberculosis (TB) is an important disease that causes thousands of deaths around the world. Resistance against antitubercular available drugs has been reported; so, research on new effective antimycobacterial molecules is needed. Antimycobacterial activity of three lignans and two synthetic hydrazones was assessed against Mycobacterium tuberculosis H37Rv by antimycobacterial microdilution assay (TEMA). An oxadiazoline (AC451) and a lignan (ethoxycubebin) were the most active compounds (MIC 6.09 and 62.4 μM, resp.). Several changes in mycolic acid profile of treated bacteria were detected with both compounds by mass spectrometry analysis. Additionally, the level of reduction of mycolic acids in ethoxycubebin treatment was correlated to disruption in bacterial morphology.
