Abstract

Italian ryegrass is a grass weed commonly found in winter wheat fields that are competitive with winter wheat for moisture and nutrients. Ryegrass can cause substantial reductions in yield and grain quality if not properly controlled with the use of herbicides. To control the cost and environmental impact we detect weeds in drone and satellite imagery. Satellite imagery is too coarse to be used for precision spraying, but can aid in planning drone flights and treatments. Drone images on the other hand have sufficiently good resolution for precision spraying. However, ryegrass is hard to distinguish from the crop and annotation requires expert knowledge. We used the Python segmentation models library to test more than 600 different neural network architectures for weed segmentation in drone images and we map accuracy versus the cost of the model prediction for these. Our best system applies herbicides to over 99% of the weeds while only spraying an area 30% larger than the annotated weed area. Abstract

Ageing is characterized by a progressive loss of physiological integrity, leading to impaired function and increased vulnerability to death1. Despite rapid advances over recent years, many of the molecular and cellular processes that underlie the progressive loss of healthy physiology are poorly understood2. To gain a better insight into these processes, here we generate a single-cell transcriptomic atlas across the lifespan of Mus musculus that includes data from 23 tissues and organs. We found cell-specific changes occurring across multiple cell types and organs, as well as age-related changes in the cellular composition of different organs. Using single-cell transcriptomic data, we assessed cell-type-specific manifestations of different hallmarks of ageing-such as senescence3, genomic instability4 and changes in the immune system2. This transcriptomic atlas-which we denote Tabula Muris Senis, or 'Mouse Ageing Cell Atlas'-provides molecular information about how the most important hallmarks of ageing are reflected in a broad range of tissues and cell types.

Abstract

Here we present a compendium of single-cell transcriptomic data from the model organism Mus musculus that comprises more than 100,000 cells from 20 organs and tissues. These data represent a new resource for cell biology, reveal gene expression in poorly characterized cell populations and enable the direct and controlled comparison of gene expression in cell types that are shared between tissues, such as T lymphocytes and endothelial cells from different anatomical locations. Two distinct technical approaches were used for most organs: one approach, microfluidic droplet-based 3'-end counting, enabled the survey of thousands of cells at relatively low coverage, whereas the other, full-length transcript analysis based on fluorescence-activated cell sorting, enabled the characterization of cell types with high sensitivity and coverage. The cumulative data provide the foundation for an atlas of transcriptomic cell biology.

7. Navigating on Mobile Shopping Applications: Examining the Interplay among Online Customer Experience, Customer Brand Relationships and Customer Citizenship Behavior.

Abstract

Online customer experience is now a crucial business component. The present study examines the association between Online Customer Experience (OCE), Customer Brand Relationships (CBR) and Customer Citizenship Behavior (CCB) in the context of mobile shopping applications (MSAs) through a survey of 410 Indian consumers of MSAs, following judgment sampling. Subsequently, CB-SEM was used. OCE's pragmatic and affective dimension significantly impacts CBR outcomes of satisfaction, loyalty and repurchase intention toward MSAs. CBR outcomes exert different impacts on CCB components, loyalty being the most crucial predictor of these components. Abstract

The ability to slow or reverse biological ageing would have major implications for mitigating disease risk and maintaining vitality1. Although an increasing number of interventions show promise for rejuvenation2, their effectiveness on disparate cell types across the body and the molecular pathways susceptible to rejuvenation remain largely unexplored. Here we performed single-cell RNA sequencing on 20 organs to reveal cell-type-specific responses to young and aged blood in heterochronic parabiosis. Adipose mesenchymal stromal cells, haematopoietic stem cells and hepatocytes are among those cell types that are especially responsive. On the pathway level, young blood invokes new gene sets in addition to reversing established ageing patterns, with the global rescue of genes encoding electron transport chain subunits pinpointing a prominent role of mitochondrial function in parabiosis-mediated rejuvenation. We observed an almost universal loss of gene expression with age that is largely mimicked by parabiosis: aged blood reduces global gene expression, and young blood restores it in select cell types. Together, these data lay the groundwork for a systemic understanding of the interplay between blood-borne factors and cellular integrity.

15. Model-based Filtering of Resting-State Blood-Oxygen-Level-Dependent Signal

Abstract

Resting-state functional magnetic resonance imaging (rs-fMRI) is one of the promising non-invasive technology that helps in the detection of neurodegenerative and neurological disorders, localisation of the different areas of the brain and understanding the connectivity between them. It involves the acquisition of time series of MR images while the brain is at "resting-state" and serves as a biomarker for various neurological conditions. The acquired resting-state blood-oxygen-level-dependent (BOLD) data, however, can be corrupted with various artefacts (head movements, physiological movements like breathing etc.). Several preprocessing pipelines have been developed to counter the effect of these known artefacts. However, there might be some artefacts that could not have been attenuated from the signal and might lead to incorrect assessment. Therefore, the thesis proposes model-based filtering of the minimally preprocessed (free from known artefacts) resting-state BOLD signal. It was observed that these signals have long memory dependencies. Hence, the usage of autoregressive fractional integrative process filters is proposed for this purpose. Furthermore, the utility of the approach is demonstrated by removing the effect of white noise from a synthetic signal with statistical properties similar to the resting-state BOLD signal. Afterwards, the proposed method is implemented on the minimally preprocessed resting-state BOLD data of 98 subjects from the Human Connectome Project. The results suggest that the proposed filter, in contrast to the low-pass filter, attenuates the higher frequencies but do not eliminate them. Additionally, four different evaluation measures (power spectrum, functional connectivity using Pearson's correlation and coherence, and eigenmode analysis) were considered. Abstract

Ageing is the single greatest cause of disease and death worldwide, and understanding the associated processes could vastly improve quality of life. Although major categories of ageing damage have been identified—such as altered intercellular communication, loss of proteostasis and eroded mitochondrial function1—these deleterious processes interact with extraordinary complexity within and between organs, and a comprehensive, whole-organism analysis of ageing dynamics has been lacking. Here we performed bulk RNA sequencing of 17 organs and plasma proteomics at 10 ages across the lifespan of Mus musculus, and integrated these findings with data from the accompanying Tabula Muris Senis2—or ‘Mouse Ageing Cell Atlas’—which follows on from the original Tabula Muris3. We reveal linear and nonlinear shifts in gene expression during ageing, with the associated genes clustered in consistent trajectory groups with coherent biological functions—including extracellular matrix regulation, unfolded protein binding, mitochondrial function, and inflammatory and immune response. Notably, these gene sets show similar expression across tissues, differing only in the amplitude and the age of onset of expression. Widespread activation of immune cells is especially pronounced, and is first detectable in white adipose depots during middle age. Single-cell RNA sequencing confirms the accumulation of T cells and B cells in adipose tissue—including plasma cells that express immunoglobulin J—which also accrue concurrently across diverse organs. Finally, we show how gene expression shifts in distinct tissues are highly correlated with corresponding protein levels in plasma, thus potentially contributing to the ageing of the systemic circulation. Together, these data demonstrate a similar yet asynchronous inter- and intra-organ progression of ageing, providing a foundation from which to track systemic sources of declining health at old age. Bulk RNA sequencing of organs and plasma proteomics at different ages across the mouse lifespan is integrated with data from the Tabula Muris Senis, a transcriptomic atlas of ageing mouse tissues, to describe organ-specific changes in gene expression during ageing.

Abstract

Ageing is characterized by a progressive loss of physiological integrity, leading to impaired function and increased vulnerability to death1. Despite rapid advances over recent years, many of the molecular and cellular processes that underlie the progressive loss of healthy physiology are poorly understood2. To gain a better insight into these processes, here we generate a single-cell transcriptomic atlas across the lifespan of Mus musculus that includes data from 23 tissues and organs. We found cell-specific changes occurring across multiple cell types and organs, as well as age-related changes in the cellular composition of different organs. Using single-cell transcriptomic data, we assessed cell-type-specific manifestations of different hallmarks of ageing—such as senescence3, genomic instability4 and changes in the immune system2. This transcriptomic atlas—which we denote Tabula Muris Senis, or 'Mouse Ageing Cell Atlas'—provides molecular information about how the most important hallmarks of ageing are reflected in a broad range of tissues and cell types.
