Construction of the genome of Hericium coralloides, a species of edible mushroom, and identification of the genes involved in terpenoid biosynthesis can determine the biology and genetics of terpenoids. The present study describes the assembly of a high-quality chromosome-scale genome of H. coralloides using Pacbio HiFi sequencing and Hi-C technology. This genome consisted of 13 chromosomes, a total size of 43.6 Mb, contigs of N50 3.6 Mb, GC content at 54%, and BUSCOs integrity of 96.9%. Genes associated with terpenoid biosynthesis were predicted by KEGG enrichment analysis and homologous alignment. The Her011461 and Her008335 genes, encoding proteins in the terpenoid backbone synthesis pathway, were found to encode geranylgeranyl pyrophosphate and farnesyl diphosphate synthases, key enzymes in the biosynthesis of geranylgeranyl diphosphate, a precursor of several diterpenoids. Her011463 was found to be involved in regulating diterpene cyclase. The Her005433, Her006724, Her010605, and Her010608 genes were found to encode sesquiterpene synthesis. Most of these genes were more highly expressed in dikaryotic mycelia than in the primordium and fruiting bodies, indicating that terpenoids may be more abundant in dikaryotic mycelia. To our knowledge, this study is the first to assemble the H. coralloides genome at the chromosome scale and to identify the genes involved in terpenoid biosynthesis.

A typhoon or hurricane is one of the most destructive high-impact weather events. In this study, the genesis and development processes of Typhoon Nangka (2015), which occurred over the Western Pacific in 2015, were investigated based on the comprehensive observation data from three satellites, i.e., the Himawari-8 satellite, the CloudSat satellite and the Global Precipitation Measurement mission satellite (GPM), focusing on the characteristics of typhoon structure, precipitation and cloud. The results (Results) show that during the developing stage of Typhoon Nangka, the cloud system was relatively complex and changed significantly, with large raindrops dominating the precipitation around the eyewall in the first quadrant, and the convection in the eyewall and outer rainband burst upward to 17 km. In addition, three features were obvious: stratiform precipitation was dominant in the inner rainband, both the precipitation type (stratiform or convective) and intensity were distributed unevenly in the outer rainband, and large water content was located in the warm layer of clouds. Moreover, the collision growth and breakup of water droplets tended to be stable. The precipitation in the typhoon eyewall, inner rainband and outer rainband was significantly different; stratiform precipitation mainly occurred in the inner rainband, while convective precipitation mainly appeared in the eyewall and outer rainband. The cloud system was distributed asymmetrically, and the upper-layer and lower-layer clouds were closely related, dominated by single-layer clouds. There were deep convective clouds in the eyewall, and cirrus clouds with the broadest range across the eyewall. The coverage range of cirrus clouds was close to the radius of the typhoon. There were stratocumulus, altostratus and cumulus in the low levels.

Flammulina filiformis is a popular mushroom which has been regarded as a potential model fungus for mycelium growth, fruiting body development, and stress response studies. Based on a genome-wide search, four genes encoding heterotrimeric G protein α subunits were identified in F. filiformis. The data of conserved domain analysis showed that these genes contain only one subgroup I of Gα subunit (Gαi), similar to many other fungi. To explore the function of Gαi, FfGa1 over-expression (OE) and RNA interference (RNAi) strains were generated using the Agrobacterium tumefaciens-mediated transformation (ATMT) approach. RNAi transformant strains showed remarkably reduced growth on PDA medium and added sensitivity to cell wall-enforcing agents with maximum growth inhibition, but showed better growth in response to hypertonic stress-causing agents, while OE strains exhibited more resistance to thermal stress and mycoparasite Trichoderma as compared to the wild-type and RNAi strains. Taken together, our results indicated that FfGa1 positively regulates hyphal extension, and is crucial for the maintenance of cell wall integrity and protection against biotic and abiotic (hypertonic and thermal) stress.

The characteristics and causes of a drop in temperature during a cold wave process in the early winter of 2020/2021 were analyzed. The results show that the air temperature at 700 -600 hPa over China was firstly and mostly influenced by the cold wave process, and then the cold air gradually extended to the lower layer, causing the most severe cooling in North China and its nearby areas. During the cold wave, the longitude of the upper-level jet over the Chinese mainland was larger; the Ural blocking high and the East Asian trough were stronger, so that the geopotential height gradient between the two was also significantly larger; the meridional air flow was abnormally strong, which was conducive to the southward transport of cold air from the middle and high latitudes. Results of the diagnostic analysis further show that the outbreak of the cold wave and the negative temperature tendency anomaly in the key area were mainly caused by the meridional temperature horizontal advection anomaly, while the temperature rise accompanied by abnormal air subsidence compensated for the abnormal decrease in temperature, which was conducive to the gradual rise of temperature in the key area.
