Ashwagandha (Withania somnifera) is one of the recognized plant species that considered of most traditional natural supplements. Tissue culture is an efficient method as fast and affordable in plant propagation. Few studies have discussed the genetic impact of such method on ashwagandha plant. The aim of this research was to identify the genetic stability of micropropagated plantlets and to assess the impact of in vitro-propagation on somaclonal variability in ashwagandha using start codon-targeted (SCoT), sequence-related amplified polymorphism (SRAP) and DNA-barcoding assays. SCoT marker assay produced a total number of 132 bands with an average of 11 bands per primer, where scorable PCR fragments were generated from all primers. The phylogenetic tree constructed using SCoT binary data, revealed genetic variability among studied plant samples. SRAP primer combinations showed a total of 78 bands by an average of 11.1 bands / combination, in which all combinations produced scored PCR fragments. Over SRAP assay, one specific band was obtained that was present in different ashwagandha micropropagated plant samples compared to the control (mother plant). This PCR fragments were obtained using me1F/em1R primer combination (287 bp). The phylogenetic tree constructed using SRAP data was successful to differentiate between micro-propagated plants and the control. The DNA- barcoding analysis using chloroplast gene RNA polymerasel (rpoCl) gene was used to detect the soma- clonal variation between control and one micro-propagated plant of ashwagandha. The phylogenetic tree constructed using DNA-barcoding sequences was successful to differentiate between the two samples, where control and micropropagated plantlets were grouped in two different groups. This study suggests the valuableness of using SRAP and DNA-barcoding in detecting soma-clonal variation among micropropagated plantlest of ashwagandha.

Water stress is a critical abiotic stress for plant reduction in arid and semiarid zones and, has been discovered to be detrimental to the development of seedlings as well as the growth and physiological characteristics of many crops such as cotton. The objectives of our study were to determine the combining ability and genetic components for five quantitative traits [(leaf area (LA), leaf dry weight (LDW), plant height (PH), fiber length (2.5 percent SL), and lint cotton yield/plant (LCY/P)] under water shortage stress, a half diallel cross between six cotton genotypes representing a wide range of cotton characteristics was evaluated in RCBD with four replications. The genotype mean squares were significant for all traits studied, demonstrating significant variation among genotypes for all characters under water shortage stress. LCY/P had the highest phenotypic and genotypic correlation co-efficient with PH, LDW, and LA shortage. The highest direct effect on lint cotton yield was exhibited by leaf area (3.905), and the highest indirect effects of all traits were through LA, with the exception of 2.5 percent SL, which was through LDW. The highest dissimilarity (Euclidean Distance) between parental genotypes was between G.87 and G.94, followed by G.87 and Menoufi. G.94 was also a well-adapted genotype, and the combinations G.87 x G.94 and G.87 x Menoufi may outperform their parents. The combining ability analysis revealed highly significant differences between parental GCA effects and F1 crosses SCA effects. The variation of GCA and SCA demonstrated the assurance of additive and non- additive gene action in the inheritance of all traits studied. In terms of general combining ability (GCA) effects, parental genotype G.94 demonstrated the highest significant and positive GCA effects for all traits studied, with the exception of 2.5 percent SL, where G.87 revealed the highest significant and positive GCA effects. The effects of specific combining ability (SCA) revealed that the cross (G.87 x2G.94) revealed stable, positive, and significant SCA for all of the studied traits.

Phosphorous (P) plays the prominent role to promote the plants storage functions and structural roles, as it is recognized as a vital component of ADP, ATP, Cell wall as well as a part of DNA. Soils acts as the sink to supply P to plants because soil pH and its physical condition are the main factor which regulate the solubility and availability P element. Phosphorus is not deficient in Pakistani soils but its availability to plants is the serious matter of concern. A pot experiment was conducted to evaluate P dynamics in two different soil series of Pakistan (Bahawalpur and Lyallpur) using Maize as test crop. The treatments applied were T0: Control (without any fertilizer), T1: Recommended DAP @648 mg pot−1, T2: Half dose DAP @324 mg pot−1, T3: Recommended rate of TSP @900 mg pot−1, T4: Half dose TSP @450 mg pot−1. Soil analysis showed that Bahawalpur soil has sandy clay loam texture with 33% clay and Lyallpur series has sandy loam texture with 15.5% clay; furthermore, these soil contain 4.6 and 2.12% CaCO3 respectively. Results showed an increase in P concentration in roots (23 mg kg−1) with the application of half dose of TSP in Lyallpur series and lowest in Bahawalpur series (14.6 mg kg−1) at recommended dose of DAP. Concentration of P in shoots responded the same; increase at half dose of TSP (16.7 mg kg−1) and lowest at full dose of DAP in Bahawalpur series as (15.58 mg kg−1). Adsorbed P (17 mg kg−1) was recorded highest in Bahawalpur soil with more clay amount in pot with DAP application but lower in Lyallpur soil series (14 mg kg−1) with the application of applied TSP. The PUE was recorded highest in Lyallpur series with the application of half dose of TSP and it was 61% more than control and was Highest in Bahawalpur series was with the application of recommended dose of DAP is 72% more than control treatment. On estimation; results showed that applied sources made an increase in P availability than control, but TSP gave better P uptake than DAP unless of rates applied. Soil of Lyallpur series showed better uptake of P and response to applied fertilizers than Bahawalpur series which showed more adsorption of P by high clay and CaCO3 amount. Conclusively, the study suggested that soil series play a crucial role in choosing fertilizer source for field application.

Grapevine (Vitis vinifera L.) fruit crops are a significant source of antioxidants, fibre, and nutrients; all are vital for a healthy diet and play a key role in the economy of several advanced and developing countries. It is of great importance to generate true-to-type plant products using in vitro propagation system. Thus, somaclonal variations can multiply very rapidly which leads to loss of the main features of parent rootstocks. In this research, a mixture of three Polymerase Chain Reaction (PCR)-based molecular marker methods – (conserved DNA derived polymorphism) CDDP, (Inter-simple sequence repeat) ISSR, and DNA barcoding – have been used to verify micro propagated grapevine genetic stability. Both ISSR and CDDP primer combinations produced scorable PCR fragments. The total number of bands was 98 and 109, with an average of 9.8 and 10.9 bands/primer in ISSR and CDDP assays, respectively. On the other hand, about 20 polymorphic bands were collected by CDDP primers, of CDDP-3 and CDDP-11produced 1 and 5 bands, with a polymorphism percentage of 11% and 33%, respectively. About 5 different unique PCR bands were detected in the mother plant (control) and were not observed in micro propagated plantlets (MP) samples of grapevine plant or vice versa. The phylogenetic trees were constructed using ISSR and CDDP assays diverged the control from MP samples at 1.3% and 7%, respectively. The phylogenetic tree constructed using (chloroplast gene RNA polymerase1) rpoC1 gene, multiple sequence alignment revealed that rpoC1 gene sequencing detected small genetic differences between control and MP samples of the grapevine and clustered grapevine control and MP samples to a single cluster with other Vitis species. This experiment reveals the potentiality of using CDDP, ISSR, and DNA barcoding in detecting the somaclonal variation of grapevine varieties subjected to tissue culture as a tool for plant conservation and breeding programs.

Graphical abstract, This study was carried out in a demonstrated field in El-Sharkia Governorate, Egypt, during the winter of season 2020 to evaluate the leverage of four post-emergence herbicides i.e., tribenuron-methyl, clodinafop- propargyl, pyroxsulam and pinoxaden compared to control on total protein and amino acid contents in three wheat cultivars (Shandwel 1, Giza 171, and Sakha 95). Generally, the use of foliar herbicides led to a significant decrease in essential, non-essential amino acids and protein contents. However, tribenuran-methyl herbicide signifcantly increased the levels of proline, glycine, arginine, and histidine, but cystine and threonine not affected as compared to control. On the other hand, foliar herbicide application was significantly increased physiological , biochemical parameters and yield of Shandweel cultivar as compared to the other varieties. The physiological and biochemical models of dual-herbicide-tolerant wheat cultivars add to our understanding of the crop. In recent agricultural systems, herbicide tolerant plants are important for long-term weed management. Therefore, the study recommended the safely usage of Tribenuran-methyl as foliar herbicide in weed managment.

The utilization of vermi-technology to reduce allelopathic effects is a cost-effective, efficient, and environmentally appropriate alternative to traditional chemical and mechanical methods. The current investigation was an effort to obtain vermicompost from C. erectus and its binary combination with soil and farmyard manure (FYM) using E. foetida. The pH, EC, organic carbon, macro and micro-nutrients, CO2 emission, the average growth rate of the worms, number of worms, number of cocoons, and weight gained by earthworms were analyzed by standard methods. The present study also investigated the effect of produced vermicompost on the growth and yield of mung beans (Vigna radiata L). The maximum reduction in soil pH was observed (6.47) in vermicompost of C. erectus leaves, among other treatments. The highest N (1.86%), P (0.15%), and K (0.41%) contents were found in the vermicompost of C. erectus leaves + FYM. Similarly, the maximum plant height (36.00 cm) was achieved in vermicompost of C. erectus leaves + FYM compared to other treatments. The highest SPAD value was observed (56.37) when the soil was treated with vermicompost (C. erectus leaves + FYM) @ 5 t ha?1, followed by the treatment where vermicompost (C. erectus leaves + soil) @ 8 t ha?1 was applied. The soil amendment of vermicompost (C. erectus leaves + FYM) @ 5 t ha?1 showed competitive results (in terms of the growth and yield parameters of mung beans) compared to other types of vermicompost and C. erectus biomass. Avoidable or inappropriate nitrogen (N) fertilizer rates harmfully affect the yield production and ecological value. Therefore, the aims of this study were to optimize the rate and timings of N fertilizer to maximize yield components and photosynthetic parameter of soybean. This field experiment consists of five fertilizer N rates: 0, 75, 150, 225 and 300 kg N ha−1 arranged in main plots and four N fertilization timings: V5 (trifoliate leaf), R2 (full flowering stage) and R4 (full poding stage), and R6 (full seeding stage) growth stages organized as subplots. Results revealed that 225 kg N ha−1 significantly enhanced grain yield components, total chlorophyll (Chl), photosynthetic rate (PN), and total dry biomass and N accumulation by 20%, 16%, 28%, 7% and 12% at R4 stage of soybean. However, stomatal conductance (gs), leaf area index (LAI), intercellular CO2 concentration (Ci) and transpiration rate (E) were increased by 12%, 88%, 10%, 18% at R6 stage under 225 kg N ha−1. Grain yield was significantly associated with photosynthetic characteristics of soybean. In conclusion, the amount of nitrogen 225 kg ha−1 at R4 and R6 stages effectively promoted the yield components and photosynthetic characteristics of soybean.

Background and objective Roses are the world's best-known garden plants, established as ornamental plants cultivated for their blooms. Taif rose (Rosa damascena trigintipetala) refers to the Damascus Rose species and is regarded one of Taif Governorate's most significant financial goods, which produces an extremely fragrant commercially precious essential oil. The objective of current study was to assess the genetic stability of micropropagated Taif rose and to assess the usefulness of Conserved DNA Derived Polymorphism (CDDP) and DNA-barcoding genes such as; rpoC1 (chloroplast gene RNA polymerase1) in the detection of somaclonal variation. Materials and methods Ten combinations of CDDP PCR primers were employed and the rpoC1 gene region was sequenced for mother plant (control) and micropropagated plantlets of Taif rose plant. Results Based on CDDP data, phylogenetic divergence indicated that the distinct specimens of Taif rose micro-propagated plantlets and control were genetically differentiated by a difference of 1% of genetic dissimilarity. Phylogenetic tree which developed using rpoC1 DNA showed that rpoC1 DNA sequencing discovered a genetic difference between the control and micro-propagated plantlets of Taif rose. Conclusion Furthermore, CDDP and DNA barcoding using rpoC1 gene have demonstrated their usefulness in investigating the genetic history of Rosa species and their ability to explore genetic mutation.

The WRKY transcription factors (TFs) network is composed of WRKY TFs’ subset, which performs a critical role in immunity regulation of plants. However, functions of WRKY TFs’ network remain unclear, particularly in non-model plants such as pepper (Capsicum annuum L.). This study functionally characterized CaWRKY30—a member of group III Pepper WRKY protein—for immunity of pepper against Ralstonia solanacearum infection. The CaWRKY30 was detected in nucleus, and its transcriptional expression levels were significantly upregulated by R. solanacearum inoculation (RSI), and foliar application ethylene (ET), abscisic acid (ABA), and salicylic acid (SA). Virus induced gene silencing (VIGS) of CaWRKY30 amplified pepper’s vulnerability to RSI. Additionally, the silencing of CaWRKY30 by VIGS compromised HR-like cell death triggered by RSI and downregulated defense-associated marker genes, like CaPR1, CaNPR1, CaDEF1, CaABR1, CaHIR1, and CaWRKY40. Conversely, transient over-expression of CaWRKY30 in pepper leaves instigated HR-like cell death and upregulated defense-related maker genes. Furthermore, transient over-expression of CaWRKY30 upregulated transcriptional levels of CaWRKY6, CaWRKY22, CaWRKY27, and CaWRKY40. On the other hand, transient over-expression of CaWRKY6, CaWRKY22, CaWRKY27, and CaWRKY40 upregulated transcriptional expression levels of CaWRKY30. The results recommend that newly characterized CaWRKY30 positively regulates pepper’s immunity against Ralstonia attack, which is governed by synergistically mediated signaling by phytohormones like ET, ABA, and SA, and transcriptionally assimilating into WRKY TFs networks, consisting of CaWRKY6, CaWRKY22, CaWRKY27, and CaWRKY40. Collectively, our data will facilitate to explicate the underlying mechanism of crosstalk between pepper’s immunity and response to RSI.

The current experiment was adopted during the summer 2018, fall 2018/2019 and summer 2019 respectively at the Experimental Farm of Baloza station, Desert Research Center. North Sinai Governorate, Egypt to study the effect of different doses of irradiation (0, 20, 30 and 40 Gy), three irrigation levels (100, 80 and 60% field capacity on growth, yield and its quality of some potato cultivars (Spunta, Cara, Caruso and Hermes). Treated Spunta cultivar pre planting with 20 (Gy) and irrigated with 80% field capacity was the best treatment for increasing number of aerial stem/plants, leaf area, total chlorophyll in leaves, average tuber weight, and total yield/fed. Hermes cultivar with 20 (Gy) and irrigation level of 80% was the best for increasing dry matter content in tuber in both mutagenic generations.

Weed infestations significantly reduce the growth and yield of field crops. Herbicides are mostly used for weed management due to their quick results. However, resistant biotypes to available herbicides are rapidly increasing around the word. This situation calls for the development of alternative weed management strategies. Crop rotation and allelopathic water extracts are regarded as the most important alternative weed management strategies. Therefore, this two-year study assessed the impact of different annual crop rotations by weed management strategies’ interactions on weed infestation and productivity of wheat crop. Wheat was planted in five rotations, i.e., (i) fallow-wheat, (ii) rice-wheat, (iii) cotton-wheat, (iv) mungbean-wheat and (v) sorghum-wheat. The weed management strategies included in the study were, (i) false seedbed, (ii) application of 12 L ha−1 allelopathic plant water extracts (1:1:1:1 ratio of sorghum, sunflower, mulberry and eucalyptus), (iii) herbicide application, (iv) weed-free (weed control) and (v) weedy-check (no weed control). Herbicide application was the most effective treatment in lowering weed densities and biomass during both years followed by false seedbed, while allelopathic crop water extracts were least effective. The lowest weed infestation was noted in sorghum-wheat rotation followed by cotton-wheat and mungbean-wheat, while fallow-wheat had the highest weed infestation. Weedy-check treatment caused significant reduction in wheat growth and yield, whereas the highest grain yield was recorded from weed-free and herbicide application treatments. Grain yield of wheat planted after sorghum was suppressed, however, yield improved when wheat was planted after mungbean. Planting wheat after mungbean in a weed-free environment, achieved through chemical and/or mechanical means, is the best strategy to obtain higher wheat yields.

Maize (Zea mays L.) is one of the important cereal crops along with wheat and rice worldwide. The purpose of this study was to use classical genetic approaches to assess the resistance of various maize parents and hybrids to the northern corn leaf blight (NCLB) disease in two different locations in Egypt. Eight parents, 28 F1, and 2 check hybrids were evaluated. The analysis of variance showed high significant variations between maize parents and their hybrids for the studied parameters and NCLB disease, besides there are significant variations between both locations. Results of maize parents showed that Sids 63, Giza 602, and Giza 628 cultivars exhibited the highest values and were resistant to NCLB in both locations comparing with Nubaria 39 and Gemmiza 18 that were susceptible to NCLB disease. Concerning the maize hybrids, analysis of variance and mean squares of growth characters in both locations indicated high significant variations between the maize hybrids including the check hybrids. When combined between the two locations for current parameters against NCLB, the data pointed that the Sakha location values for maize hybrids were much closed to the combining data in parents and the hybrids detected high resistance to this disease comparing with Nubaria location. All tested maize lines (38 lines), including parents and hybrids were classified as follows, two lines were rated as 1 (highly resistant), three were rated as 2 (resistant), sixteen were rated as 3 (moderate resistant), eight were rated 4 (moderately susceptible) and nine were rated 5 (susceptible). The data explaining that the crossing between high resistant maize cultivars produced high levels of resistance to NCLB disease. Therefore, our results verified that classical breeding could efficiently increase the resistance levels of maize germplasm against NCLB disease by developing new cultivars with superior performance in terms of grain yield, disease resistance and grain quality.

This study aimed to evaluate the nematicidal activity of two marine algae (Colpomenia sinuosa and Corallina mediterranea) extracts and their synthesized silver nanoparticles against the root-knot nematode (Meloidogyne incognita) infecting tomato plant. Scanning Electron Microscope (SEM) showed that the obtained nanoparticles were aggregated in anisotropic Ag particles. Transmission Electron Microscope (TEM), results showed the particles size was less than 40 nm. Whenever, FT-IR analysis spectrum presented sharp absorbance between 440 and 4000 cm-1 for the obtained nanoparticles, with 13 distinct peaks ranged from 3915 − 474.Both of methylene chloride extract and its synthesized green silver nanoparticles were applied against the M. incognita. The results indicated that the synthesized silver nanoparticles of C. sinuosa exhibited the highest nematicidal activity. Besides, they reduced number of nematode galls, number of egg-masses per root and eggs/egg mass, as well as growth parameters of the treated plants with nanoparticles were enhanced comparing with the other treatments. While the methylene chloride extract of C. sinuosa exhibited higher activity than that of C. mediterranea, and the most effective eleuent of this solvent was Hexane: methylene chloride: ethyl acetate (1: 0.5: 0.5, v/v/v). The 3rd fraction of this eluent was the most effective one when it was applied on M. incognita, resulting in 87.5 % mortality after 12 h and 100 % after 24 and 72 h of exposure. The analysis of this fraction revealed the presence of seven bioactive constituents. Conclusively, the synthesized silver nanoparticles of C. sinuosa could be considered as alternative chemical nematicides.

Sunflower (Helianthus annuus L.), a member of the Asteraceae, is one of the major oilseed crops around the world. Charcoal rot caused by Macrophomina phaseolina (Tassi) Goid is the most damaging disease of sunflowers globally. Fungicides are mostly used to control charcoal rot, however, these cause environmental pollution and pose adverse effects on the ecosystem. Therefore, ecofriendly management options are inevitable for the management of charcoal rot disease. Plant mineral nutrition, the use of plant growth-promoting rhizobacteria and biochar have recently been manipulated for the management of different plant diseases. However, the interactive effects of all these treatments have rarely been tested on charcoal rot suppression in sunflowers. This study assessed the influence of sulfur (0 and 2.25 mg/kg) combined with farmyard manure biochar (2%), NPK (20:20:20 mg/kg) and three different plant growth-promoting rhizobacteria (PGPR) strains on the charcoal rot suppression growth, yield, biochemistry and physiology of sunflower. The PGPR strains included in the study were Bacillus sp. strain MR-1/2 (regarded as PGPR1), Achromobacter sp. strain FB-14 (regarded as PGPR2) and Planomicrobium sp. strain MSSA-10 (regarded as PGPR3). The charcoal rot infestation was induced by inoculating the soil with M. phaseolina, and the impacts of the different treatments were studied on the disease infestation, growth, yield, biochemistry and physiology of sunflowers under 0 and 2.25-mg/kg S application. The results revealed that farmyard manure biochar and Planomicrobium sp. strain MSSA-10 in combination with 2.25-mg/kg S proved effective for the management of charcoal rot disease through regulating the antioxidant enzymes’ activities and strengthening the immune system of sunflower plants. The studied health markers (total chlorophyll content and carotenoids) and stress markers (total protein content, catalase and peroxidase) were significantly altered by the applied treatments under 0 and 2.25-mg/kg S applications. The findings of the experiment indicated that both farmyard manure biochar and Planomicrobium sp. strain MSSA-10, combined with 2.25-mg/kg S, could be used to enhance the crop yield and manage charcoal rot disease in sunflowers. Farmyard manure biochar and Planomicrobium sp. strain MSSA-10 are an easy-to-apply, cost-effective, ecofriendly and sustainable option for the management of charcoal rot disease in sunflowers.

Climate change is causing soil salinization, resulting in huge crop losses throughout the world. Multiple physiological and biochemical pathways determine the ability of plants to tolerate salt stress. Chili (Capsicum annum L.) is a salt-susceptible crop; therefore, its growth and yield is negatively impacted by salinity. Irreversible damage at cell level and photo inhibition due to high production of reactive oxygen species (ROS) and less CO2 availability caused by water stress is directly linked with salinity. A pot experiment was conducted to determine the impact of five NaCl salinity levels, i.e., 0,1.5, 3.0, 5.0 and 7.0 dS m-1 on growth, biochemical attributes and yield of two chili genotypes (‘Plahi’ and ‘A-120’). Salinity stress significantly reduced fresh and dry weight, relative water contents, water use efficiency, leaf osmotic potential, glycine betaine (GB) contents, photosynthetic rate (A), transpiration rate (E), stomatal conductance (Ci), and chlorophyll contents of tested genotypes. Salinity stress significantly enhanced malondialdehyde (MDA) contents and activities of the enzymatic antioxidants such as superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD). In addition, increasing salinity levels significantly reduced the tissue phosphorus and potassium concentrations, while enhanced the tissue sodium and chloride concentrations. Genotype ‘Plahi’ had better growth and biochemical attributes compared to ‘A-120’. Therefore, ‘Plahi’ is recommended for saline areas to improve chili production.

Background and Objective: This study focused on evaluation of SBPase promoter in driving transgenes under drought, high light intensity and salinity as major abiotic stresses limiting plant growth and production in various areas of the world. Wheat production has remained constant despite the rapidly growing population around the world. \ud \ud Materials and Methods: This had made it necessary to develop other varieties with higher yield in order to satisfy the future demand. When elite wheat varieties’ photosynthesis is genetically manipulated, it gives a high possibility for yield increase. Lack of a properly defined molecular tool-box promoter for driving gene expression for manipulation of photosynthesis, hinders the progression of this field. Gus histochemical staining and quantitative PCR were used to examine the function of Brachypodium distachyon sedoheptulose-1, 7-bisphosphatase (SBPase) promoter to drive the expression of GUS in the stably transformed wheat plants under various stresses conditions. \ud \ud Results: Results revealed that B. distachyon SBPase promoter fully drive the GUS expression in the stable wheat transformants (cv. Cadenza) in the leaf tissues under normal and high light conditions. It also drove GUS gene expression under drought and salinity stress conditions.
