18. Miniaturised control of acidity in multiplexed microreactors

Abstract

The control of acidity influences the structural assembly of biopolymers that are essential for a wide range of applications. Its miniaturization can increase the speed and the possibilities of combinatorial throughput for their manipulation, similarly to the way that the miniaturization of transistors allows the high throughput of logical operations in microelectronics. Here we present a device containing multiplexed micro-reactors, each one enabling independent electrochemical control of the acidity in ~ 2.5 nL volumes, with a large acidity range in aqueous solutions from pH 3 to 7 and an accuracy of at least 0.4 pH units. The attained pH within each microreactor (with footprints of ~ 0.3 mm2 for each spot) was kept constant for long retention times (~10 minutes) and over repeated cycles >100. The acidity is driven by redox proton exchange reactions, which can be driven at different rates that influence the efficiency of the device in order to achieve more charge exchange (larger acidity range) or better reversibility. By the performance in the acidity control the miniaturisation and the possibility to multiplex paves the way for the control of combinatorial chemistry through pH and acidity controlled reactions.

37. Race specific and non-specific resistance to Magnaporthe oryzaeand QTL mapping in wild introgression lines using the standard differential system

Abstract

Blast disease caused by Magnaporthe oryzaeis one of the major challenges to rice crop production worldwide. Wild and related species are potential source of novel alleles or genes for crop improvement especially for biotic stress resistance. This study was designed to characterize blast resistance in a set of wild introgression lines developed in the background of Indian mega rice variety Swarna. The introgression lines (INLs) under this study showed spectra of resistance across the different blast isolates inoculated. Monogenic differential varieties with known specific Pigenes and the susceptible checks were used in phenotyping. This study confirmed the usefulness of the differential system consisting of monogenic varieties and differential blast isolates for the systematic characterization of resistance in novel germplasm and to understand the genetic architecture of blast resistance. Twelve race specific quantitative trait loci (QTL) for partial resistance to disease were detected in these wild introgression lines from O.nivarausing composite interval mapping. Of these, 10 were showing resistance to only one isolate each, while QTL detected in chromosome 3 showed resistance against isolates JPF514 and PHL16. While most of the QTLs mapped to previously reported defense related genes whereas two QTLs qBL2.2and qBL5.1between RM106—RM5460 and RM5140—RM289 with PVE% of 12.28 and 12.48 respectively are novel with no known blast resistance related genes reported within the locus. This study helps to provide developing the basis for future investigations on race specific or broad-spectrum resistance in rice and related crop species.

Abstract

In recent years, there has been a lot of interest in nanocellulose (NC) and nanocellulose-reinforced composite materials research and development. This is due to its unique capabilities and features, including its ability to be renewable and sustainable, as well as its high mechanical strength, lightweight, and low cost. In this study, NC was extracted from Cajanus Cajan pea pods using an acid hydrolysis technique. Scanning Electron Microscopy (SEM) was used to characterise the isolated nanocellulose. NCs were incorporated in carboxylated acrylonitrile rubber (XNBR) by an eco-friendly latex stage mixing process combined with sonication to prepare XNBR/NC nanocomposites. The presence of H-bonds between XNBR and NC resulted in homogeneous NC dispersion inside the XNBR matrix as well as better interfacial adhesion. This resulted in a significant improvement in the tensile properties of XNBR/NC composites when compared to neat rubber. Tensile strength has been improved by 54% by incorporation of only 5 phr NC. High crosslinking between nitrile rubber chains and cellulose nanoparticles is also indicated by a decrease in solvent uptake. The influence of nanocellulose content on morphology and mechanical properties was also examined. Abstract

NLR34449 is a popular, blast resistant, high-yielding variety, with highly desired fine-grain type and hence commands a premium price in the market. The nature and genetics of resistance to blast disease in this variety was not reported previously. Molecular marker-based screening using gene linked markers for ten blast resistance genes showed amplification of the resistance allele only with respect to the marker RM224, which is specific for Pi1gene.In order to confirm the role of Pi1gene with respect to blast disease resistance in Nellore Mahsuri, a F2mapping population derived from the cross Nellore Mahsuri/Improved Samba Mahsuri (blast susceptible variety) was studied. The F2plants segregated in ratio of 3 resistant: 1 susceptible indicating monogenic, dominant nature of inheritance with respect to blast resistance. Thus, the current study demonstrated that blast resistance in NLR34449 is contributed by Pi1.

Abstract

The development of nutrient-use efficient rice lines is a priority amidst the changing climate and depleting resources viz., water, land, and labor for achieving sustainability in rice cultivation. Along with the traditional transplanted irrigated system of cultivation, the dry direct-seeded aerobic system is gaining ground nationwide. The root-related traits play a crucial role in nutrient acquisition, adaptation and need to be concentrated along with the yield-attributing traits. We phenotyped an association panel of 118 rice lines for seedling vigour index (SVI) traits at 14 and 21 days after sowing (DAS), root-related traits at panicle initiation (PI) stage in polythene bags under controlled aerobic condition, yield and yield-related traits under the irrigated condition at ICARIIRR, Hyderabad, Telangana; irrigated and aerobic conditions at ARS, Dhadesugur, Raichur, Karnataka. The panel was genotyped using simple sequence repeats (SSR) markers and genome-wide association studies were conducted for identifying marker-trait associations (MTAs). Significant correlations were recorded for root length, root dry weight with SVI, root volume at the PI stage, number of productive tillers per plant, spikelet fertility, the total number of grains per panicle with grain yield per plant under irrigated conditions, and the total number of grains per panicle with grain yield per plant under aerobic condition. The panel was divided into three sub-groups (K = 3) and correlated with the principal component analysis. The maximum number of MTAs were found on chromosomes 2, 3, and 12 with considerable phenotypic variability. Consistent MTAs were recorded for SVI traits at 14 and 21 DAS (RM25310, RM80, RM22961, RM1385), yield traits under irrigated conditions (RM2584, RM5179, RM410, RM20698, RM14753) across years at ICAR-IIRR, grain yield per plant (RM22961, RM1146) under the aerobic condition, grain yield per plant at irrigated ICAR-IIRR and SVI (RM5501), root traits at PI stage (RM2584, RM80, RM410, RM1146, RM18472). Functionally relevant genes near the MTAs through in-silico expression analysis in root and panicle tissues viz., HBF2 bZIP transcription factor, WD40 repeat-like domain, OsPILS6a auxin efflux carrier, WRKY108, OsSCP42, OsMADS80, nodulin-like domain-containing protein, amino acid transporter using various rice expression databases were identified. The identified MTAs and rice lines having high SVI traits (Langphou, TI-128, Mouli, TI-124, JBB-631-1), high yield under aerobic (Phouren, NPK-43, JBB-684, Ratnamudi, TI-112), irrigated conditions (KR-209, KR-262, Phouren, Keibi-Phou, TI-17), robust root traits like root length (MoirangPhou-Angouba, Wangoo-Phou, JBB-661, Dissi, NPK-45), root volume (Ratnachudi, KJ-221, Mow, Heimang-Phou, PUP-229) can be further employed in breeding programs for the targeted environments aimed at improving seedling vigour, yield-related traits under irrigated condition, aerobic condition as adaptability to water-saving technology.
