Biodegradace komerčního PVA za přítomnosti směsné čistírenské mikrobiální kultury adaptované na PVA probíhala s lagovou fází max. 20 hod, úplný rozklad nastal za cca 100 hod. Doba rozkladu PVA obsaženého ve směsných fólií obsahujících výše jmenované komponenty v různém procentuálním zastoupení byla cca 100 hod s lagovou fází cca 20 hod. Pokles rychlostní konstanty podle DPVA byl způsoben přítomností snadno rozložitelného substrátu (rozkládal se přednostně). Z výsledků jednoznačně vyplývá, že použitá "plniva" nemají v daném procentuálním zastoupení negativní vliv na lagovou fázi, dobu a rychlost rozkladu. Modifikované přírodní polymery mohou být plnivem vhodným pro výrobu směsných filmů na bázi PVA. Biologický rozklad ligninu 2 byl podle očekávání mnohem pomalejší; 45 % ligninu (podle DCO2) bylo rozloženo za 400 hod testu. Při rozkladu ligninu 2 docházelo k produkci huminových látek. Biodegradation of plastic films based on polyvinyl alcohol and protein hydrolysate under aerobic condition was assessed by means of respirometric test (bod measurements) and/or Zahn-Wellens test. The tests were provided in water envirofnment after inoculation by activated sludge drawn-up from waste water treatment plant. 13 mixed film blends were tested it was found that biodegradation of these blends was practicaly non-independečntable on their compoisiton.

Water solubility of polyvinyl alcohol (PVA) is related to degree of hydrolysis, molecular weight and modification during blending in the presence of other processing additives. In the present paper the effect of collagen hydrolysate (CH) and glycerol on PVA water sensitivity has been investigated. CH content affects water penetration into the prepared blown films, affecting therefore their solubility. An increasing content of CH in PVA based blends shortens the time to the first disruption of the film after immersing in water, restraining the negative effect of glycerol on solubility. Pure PVA film presented limited biodegradation at low temperature (5 oC). The CH addition in the blend significantly increases biodegradation rate at that temperature. PVA/CH blends properties are of practical relevance for applications as hospital laundry bags and containers of water-soluble substances, such as chemical agents for treatment of waste and potable water, fertilizers, washing agents, sanitary products, etc.

Research was conducted into biodegradability of mixeds polymer films based on poly(vinyl alcohol), protein hydrolyzate (collagen hydrolyzate from wastes after chrome tanning) and sglycerol in an aqueous aerobic environment. Evaluation of biodegradation was based on carbon dioxide produced in the gas phase. Pure PVAL was degraded by a current mixed culture for water-treatment (unadapted) only after an approx. 10-day lag phase, during breakdown of mixed film the protein component and glycerol were broken down first and PVAL degradation occurred in the second stage. Biodegradation could be well described by 1st-order formal chemical kinetics. Repeated degradation by an adapted culture proceeded in a single stage with considerably shorter lag phase (< 30 h) at a simultaneously approx. 1.5-fold greater breakdown rate (rate constants). During degradation of substrates containing PVAL, microbiological tests proved an approx. 100-fold increase in numbers of PVAL-degrading bacteria. Added protein hydrolyzate + glycerol in PVAL contributed to increasing biodegradability more than followed from proportional representation of individual components.

Hybrid blends based on poly(vinyl alcohol) (PVA) and collagen hydrolysate (CH), an abundant, added value waste product of the leather industry, have been processed by melt blow extrusion to environmentally degradable films. Optimisation of the blend compositions was performed in respect of mechanical properties of the films. The experimental design method (DOE) was used for the understanding of the structure-property relationships in the hybrid blends using glycerol as a plasticiser. Parameters were selected from torwue measurements, melt flow index, thermogravimetric analysis, as well as tensile strength and elongation at break of the plasticised blends. The use of the DOE method offers the possibility of identifying a range of blend compositions suitable to yield products with valuable mechanical and thermal properties. Biodegradation experiments performed under anaerobic conditions evidenced a positive effect of collagen hydrolysate on the mineralization rate of PVA/CH blends.

Předkládaná práce popisuje termické a mechanické chování změkčených bio-plastů a bio-kompozitů se zaměřením na použití v 3D tisku. Zkoumán byl vliv chemické struktury změkčovadla na bázi derivátů kyseliny citronové na termické a mechanické vlastnosti vzorků změkčené polymerní směsi poly-3-hydroxybutyrátu a kyseliny polymléčné. Vliv změkčovadla na polymerní matrici a jejich vzájemná kompatibilita byla posuzována rychlostí zamísení změkčovadla do taveniny, respektive rychlostí migrace změkčovadla ven z materiálu za zvýšené teploty. Pomocí modulované diferenciální skenovací kalorimetrie byl sledován vliv struktury změkčovadla na teplotu skelného přechodu a krystalizační kinetiku změkčeného materiálu. Sledováno bylo také chování materiálu při 3D tisku. Pomocí tahové zkoušky byly stanoveny mechanické vlastnosti tištěných těles, především jejich tažnost. Nejvyšší efekt změkčení byl pozorován za použití změkčovadla tributylcitrátu, kdy bylo dosaženo poklesu teploty skelného přechodu o a zvýšení tažnosti o 150 % oproti neměkčené referenci. Takto změkčená polymerní směs vykazovala i dostatečné 3D tiskové vlastnosti a byla využita jako matrice pro kompozity v další části práce. Kompozity byly plněny kaolinem, vápencem, halloysitem, sráženou silikou, mastkem, hydroxidem hořečnatým a sekaným lněným vláknem. U kompozitů byla sledována distribuce částic pomocí skenovací elektronové mikroskopie v závislosti na použití povrchové úpravy plniva. Pomocí viskozimetrie a diferenciální skenovací kalorimetrie byl u kompozitů sledován vliv plniva na reologické chování, krystalizační kinetiku a termickou stabilitu kompozitů. Jejich mechanické vlastnosti a teplota průhybu při zatížení byly sledovány na tělesech připravených 3D tiskem. Kaolin vykazoval v kompozitním materiálu homogenní distribuci částic a zanedbatelný vliv na termickou stabilitu a nukleaci matrice. Kaolinem plněný kompozit navíc vykazoval oproti neplněné referenci o 18 % menší warping při tisku, proto byl kaolin vyhodnocen ja, This dissertation work deals with the thermic and the mechanical behavior of plasticized bio-plastics and bio-composites for the 3D printing applications. The influence of plasticizer chemical structure on thermic and mechanical properties of plasticized polymeric blends from the poly-3-hydroxybutyrate and the poly lactic acid was investigated. Used plasticizers are based on derivative of citric acid. The influence of plasticizers on polymeric matrix and their compatibility was estimated by gear torque rate of melt mixer, respectively rate of plasticizer migration from the material during higher temperature. The plasticizer structure influence on the glass transition temperature and on the kinetics of crystallization of plasticized material was investigated by modulated differential scanning calorimetry. The behavior of material during 3D printing was also observed. Mechanical properties of printed samples, especially their elongation at break, were determined by tensile tests. The largest softening effect was observed using tributylcitrate plasticizer, where the glass temperature decreased by 35 °C and elongation at break increased by 150% compared to non-plasticized reference material. This plasticized polymeric blend showed also sufficient 3D printing properties and was used as the matrix for composites in the next part of this work. Composites were filled by kaolin, limestone, halloysit, fumed silica, talc, magnesium hydroxide and chopped flax fibers. Particle distribution in composites in dependence of used surface treatment of filler was observed by scanning electron microscopy. The influence of composite filler on rheological properties, crystallization kinetics and thermal stability of composites, was observed by viscometry and differential scanning calorimetry. Dizertační práce se zabývala syntézou a studiem kompozitních materiálů pro potenciální lékařské využití. Teoretická část je zaměřena na biomateriály, zejména na kompozity složené z polyvinylalkoholu a hydroxyapatitu(PVA/HA). Byly připraveny kompozitní membrány složené z polyvinylalkoholu s různým hmotnostním zastoupením hydroxyapatitu - 0%, 10%, 20%, 30%, 40% a 50%. Hydroxyapatit (HA) byl připraven srážecí metodou z hydrogenfosforečnanu amonného a tetrahydrátu dusičnanu vápenatého ve vodném alkalickém prostředí. Vzniklá suspenze se smísila s roztokem polyvinylalkoholu, který byl připraven rozpuštěním ve vodě o teplotě 85° C. Jednotlivé směsi byly odlity do formy a sušeny po dobu 7 dní při teplotě 30 ° C, vzniklé 0,5 mm tenké membrány byly analyzovány ATR-FTIR spektroskopií k identifikaci funkčních skupin v kompozitu, dále byla provedena XRD analýza. Zkouška tahem a TGA měření byly realizovány k určení vlivu HA na mechanické vlastnosti, respektive změnu tepelné odolnosti kompozitů ve srovnání s čistým PVA. Byla provedena zkouška bioaktivity v simulovaném krevním roztoku (SBF) po dobu 2h, 7 a 28 dnů. SEM byla použita k charakterizaci povrchové mikrostruktury biocompositních membrán před a po ponoření do SBF. Na povrchu testovaných membrán vznikla vrstva apatitu, která je charakteristická pro bioaktivní materiály. Bylo zjištěno, že s rostoucím množstvím HA částic docházelo ke vzniku aglomerátů v kompozitu, které vznikly mimo jiné jako důsledek růstu krystalů HA během sušení membrán. Bioaktivita rostla s delším působením SBF na vzorky., The work is focused on the synthesis and the study of biocomposite materials for potential medical application. The theoretical part is a literature overview focused on different types of biomaterials especially on poly(vinyl alcohol) - hydroxyapatite (PVA/HA) composites. A set of polyvinyl alcohol (PVA) membranes with various weight percent - 0%, 10%, 20%, 30%, 40% and 50% of hydroxyapatite (HA) were prepared. Hydroxyapatite was prepared by precipitation procedure using starting materials of diammonium hydrogen phosphate and calcium nitrate tetrahydrate in water alkaline environment and then mixed with solution of poly(vinyl alcohol), which was prepared by dissolving it in water at 85°C. The different mixtures were casted in a mould and evaporated for 7 days at temperature 30 °C to obtain 0,5 mm thin membranes. The ATR-FTIR spectroscopy was used to identify the different functional groups in composite membranes, XRD testing was carried out to identify crystallized hydroxyapatite. The tensile testing and TGA measurement were realised to find the effect of HA amount on the mechanical properties and thermal stability of the membranes. The in vitro bioactivity tests in Simulated Blood Fluid (SBF) were performed for 2 hours, 7 and 28 days. SEM was used to characterise surface microstructure of biocomposite membranes before and after immersion in SBF. The surface of the tested membranes were analysed to investigate the formation of apatite, the characteristic of bioactivity. It was observed a formation of clusters within membranes with increasing amount of HA particles due to hydrogen bond and also the agglomeration and crystal growth of HA particles during drying of membranes. The bioactivity was found increasing with time immersion of biocomposite materials.

Tato práce je zaměřena na studium interakcí hyaluronanu s amfifilními molekulami s vysokou biokompatibilitou. Metodou fluorescenční sondy byly zkoumány interakce mezi kationtovým lipidem 1,2-dipalmitoyl-3-trimetylamonium-propan (DPTAP), mezi směsí tohoto kationtového lipidu se zwiterionickým fosfolipidem, 1,2-dipalmitoyl-sn-glycero-3-fosfocholinem (DPPC), a hyaluronanem, a to jak nativním, tak i s hydrofobně modifiko¬vaným. Bylo zjištěno, že u směsi DPPC a DPTAP dochází k tvorbě agregátů nezávisle na poměru lipidů ve směsi, a že s hyaluonanem interagují jak agregáty vzniklé z DPTAP, tak ze směsi DPPC a DPTAP při specifickém poměru koncentrací DPTAP a hyaluronanu. Dále byly studovány fyzikální vlastnosti membrán při různém poměru DPPC a DPTAP ve směsi a vliv cholesterolu na některé z těchto vlastností. V neposlední řadě byly studovány také systémy neionogenní tenzid-DPPC, konkrétně velikost vznikajících agregátů, termodynamika solubilizace a interakce s nativním hyaluronanem., This work is focused on the study of interaction between hyaluronan and high-biocompatible amphiphilic molecules. Using fluorescent probe method, screening of the interaction of cationic lipid 1,2-dipalmitoyl-3-trimethylammonium-propane (DPTAP), mixture of this cationic lipid with zwitterionic lipid, 1,2-dipalmitoyl-sn-glycero-3-phosphochloline (DPPC), with hyaluronan, both native and hydrophobically modified was carried out. Results showed the self-aggregation of DPPC and DPTAP independently on lipids ratio in the mixture and the interaction of DPTAP and DPPC/DPTAP aggregates with hyaluronan at specific ratio of DPTAP and hyaluronan concentration. Physical properties of formed membranes and the influence of cholesterol were also investigated at different DPPC and DPTAP concentration ratio. Last but not least, the non-ionic surfactant-DPPC systems were studied, namely, the size of the formed aggregates, the thermodynamics of solubilisation and the interaction with native hyaluronan.

Využity byly sloučeniny různých funkčních skupin; isokyanáty, karbodiimidy, alkoholy a epoxidy; a funkcionality; od dvou po polyfunkční. Cílem je charakterizovat kinetiku termické degradace tohoto polymeru během zpracování samotného a s přídavkem výše uvedených činidel v různých množstvích. Za tímto účelem bylo provedeno reologické měření sestávající ze čtyř po sobě jdoucích frekvenčních testů v rozsahu 0,1–50 Hz a ze získaných dat byl vyhodnocen parametr kvantifikující rychlost poklesu viskozity. Tento kinetický parametr umožnil porovnání účinků jednotlivých činidel. Dále byly připraveny reaktivní vzorky hnětením, při němž byl zaznamenán a diskutován kroutící moment, a také v roztocích, u kterých byla měřena viskozita. U takto připravených vzorků byly studovány tepelné vlastnosti, molekulová hmotnost a infračervená spektra. Z testovaných aditiv vykázaly nejlepší celkové výsledky v provedených experimentech polymerní karbodiimid Raschig 9000, hexamethylen diisokyanát, poly(glycidyl methakrylát) syntetizovaný pro účely této práce a poly(hexamethylen diisokyanát). Notably, the sample with a 100-fold molar overdose of Raschig had a 20–30% lower value of the viscosity loss factor than the reference in the frequency region 0.1–5 Hz. Results showed the self-aggregation of DPPC and DPTAP independently on lipids ratio in the mixture and the interaction of DPTAP and DPPC/DPTAP aggregates with hyaluronan at specific ratio of DPTAP and hyaluronan concentration. Physical properties of formed membranes and the influence of cholesterol were also investigated at different DPPC and DPTAP concentration ratio. Last but not least, the non-ionic surfactant-DPPC systems were studied, namely, the size of the formed aggregates, the thermodynamics of solubilisation and the interaction with native hyaluronan.

Dizertační práce se zabývala syntézou a studiem kompozitních materiálů pro potenciální lékařské využití. Teoretická část je zaměřena na biomateriály, zejména na kompozity složené z polyvinylalkoholu a hydroxyapatitu(PVA/HA). Byly připraveny kompozitní membrány složené z polyvinylalkoholu s různým hmotnostním zastoupením hydroxyapatitu - 0%, 10%, 20%, 30%, 40% a 50%. Hydroxyapatit (HA) byl připraven srážecí metodou z hydrogenfosforečnanu amonného a tetrahydrátu dusičnanu vápenatého ve vodném alkalickém prostředí. Vzniklá suspenze se smísila s roztokem polyvinylalkoholu, který byl připraven rozpuštěním ve vodě o teplotě 85° C. Jednotlivé směsi byly odlity do formy a sušeny po dobu 7 dní při teplotě 30 ° C, vzniklé 0,5 mm tenké membrány byly analyzovány ATR-FTIR spektroskopií k identifikaci funkčních skupin v kompozitu, dále byla provedena XRD analýza. Zkouška tahem a TGA měření byly realizovány k určení vlivu HA na mechanické vlastnosti, respektive změnu tepelné odolnosti kompozitů ve srovnání s čistým PVA. Byla provedena zkouška bioaktivity v simulovaném krevním roztoku (SBF) po dobu 2h, 7 a 28 dnů. SEM byla použita k charakterizaci povrchové mikrostruktury biocompositních membrán před a po ponoření do SBF. Na povrchu testovaných membrán vznikla vrstva apatitu, která je charakteristická pro bioaktivní materiály. Bylo zjištěno, že s rostoucím množstvím HA částic docházelo ke vzniku aglomerátů v kompozitu, které vznikly mimo jiné jako důsledek růstu krystalů HA během sušení membrán. Bioaktivita rostla s delším působením SBF na vzorky., The work is focused on the synthesis and the study of biocomposite materials for potential medical application. The theoretical part is a literature overview focused on different types of biomaterials especially on poly(vinyl alcohol) - hydroxyapatite (PVA/HA) composites. A set of polyvinyl alcohol (PVA) membranes with various weight percent - 0%, 10%, 20%, 30%, 40% and 50% of hydroxyapatite (HA) were prepared. Hydroxyapatite was prepared by precipitation procedure using starting materials of diammonium hydrogen phosphate and calcium nitrate tetrahydrate in water alkaline environment and then mixed with solution of poly(vinyl alcohol), which was prepared by dissolving it in water at 85°C. The different mixtures were casted in a mould and evaporated for 7 days at temperature 30 °C to obtain 0,5 mm thin membranes. The ATR-FTIR spectroscopy was used to identify the different functional groups in composite membranes, XRD testing was carried out to identify crystallized hydroxyapatite. The tensile testing and TGA measurement were realised to find the effect of HA amount on the mechanical properties and thermal stability of the membranes. The in vitro bioactivity tests in Simulated Blood Fluid (SBF) were performed for 2 hours, 7 and 28 days. SEM was used to characterise surface microstructure of biocomposite membranes before and after immersion in SBF. The surface of the tested membranes were analysed to investigate the formation of apatite, the characteristic of bioactivity. It was observed a formation of clusters within membranes with increasing amount of HA particles due to hydrogen bond and also the agglomeration and crystal growth of HA particles during drying of membranes. The bioactivity was found increasing with time immersion of biocomposite materials.

Předkládaná práce popisuje termické a mechanické chování změkčených bio-plastů a bio-kompozitů se zaměřením na použití v 3D tisku. Zkoumán byl vliv chemické struktury změkčovadla na bázi derivátů kyseliny citronové na termické a mechanické vlastnosti vzorků změkčené polymerní směsi poly-3-hydroxybutyrátu a kyseliny polymléčné. Vliv změkčovadla na polymerní matrici a jejich vzájemná kompatibilita byla posuzována rychlostí zamísení změkčovadla do taveniny, respektive rychlostí migrace změkčovadla ven z materiálu za zvýšené teploty. Pomocí modulované diferenciální skenovací kalorimetrie byl sledován vliv struktury změkčovadla na teplotu skelného přechodu a krystalizační kinetiku změkčeného materiálu. Sledováno bylo také chování materiálu při 3D tisku. Pomocí tahové zkoušky byly stanoveny mechanické vlastnosti tištěných těles, především jejich tažnost. Nejvyšší efekt změkčení byl pozorován za použití změkčovadla tributylcitrátu, kdy bylo dosaženo poklesu teploty skelného přechodu o a zvýšení tažnosti o 150 % oproti neměkčené referenci. Takto změkčená polymerní směs vykazovala i dostatečné 3D tiskové vlastnosti a byla využita jako matrice pro kompozity v další části práce. Kompozity byly plněny kaolinem, vápencem, halloysitem, sráženou silikou, mastkem, hydroxidem hořečnatým a sekaným lněným vláknem. U kompozitů byla sledována distribuce částic pomocí skenovací elektronové mikroskopie v závislosti na použití povrchové úpravy plniva. Pomocí viskozimetrie a diferenciální skenovací kalorimetrie byl u kompozitů sledován vliv plniva na reologické chování, krystalizační kinetiku a termickou stabilitu kompozitů. Jejich mechanické vlastnosti a teplota průhybu při zatížení byly sledovány na tělesech připravených 3D tiskem. Kaolin vykazoval v kompozitním materiálu homogenní distribuci částic a zanedbatelný vliv na termickou stabilitu a nukleaci matrice. Kaolinem plněný kompozit navíc vykazoval oproti neplněné referenci o 18 % menší warping při tisku, proto byl kaolin vyhodnocen ja, This dissertation work deals with the thermic and the mechanical behavior of plasticized bio-plastics and bio-composites for the 3D printing applications. The influence of plasticizer chemical structure on thermic and mechanical properties of plasticized polymeric blends from the poly-3-hydroxybutyrate and the poly lactic acid was investigated. Used plasticizers are based on derivative of citric acid. The influence of plasticizers on polymeric matrix and their compatibility was estimated by gear torque rate of melt mixer, respectively rate of plasticizer migration from the material during higher temperature. The plasticizer structure influence on the glass transition temperature and on the kinetics of crystallization of plasticized material was investigated by modulated differential scanning calorimetry. The behavior of material during 3D printing was also observed. Mechanical properties of printed samples, especially their elongation at break, were determined by tensile tests. The largest softening effect was observed using tributylcitrate plasticizer, where the glass temperature decreased by 35 °C and elongation at break increased by 150% compared to non-plasticized reference material. This plasticized polymeric blend showed also sufficient 3D printing properties and was used as the matrix for composites in the next part of this work. Composites were filled by kaolin, limestone, halloysit, fumed silica, talc, magnesium hydroxide and chopped flax fibers. Particle distribution in composites in dependence of used surface treatment of filler was observed by scanning electron microscopy. The influence of composite filler on rheological properties, crystallization kinetics and thermal stability of composites, was observed by viscometry and differential scanning calorimetry. Tato práce je zaměřena na studium interakcí hyaluronanu s amfifilními molekulami s vysokou biokompatibilitou. Metodou fluorescenční sondy byly zkoumány interakce mezi kationtovým lipidem 1,2-dipalmitoyl-3-trimetylamonium-propan (DPTAP), mezi směsí tohoto kationtového lipidu se zwiterionickým fosfolipidem, 1,2-dipalmitoyl-sn-glycero-3-fosfocholinem (DPPC), a hyaluronanem, a to jak nativním, tak i s hydrofobně modifiko¬vaným. Bylo zjištěno, že u směsi DPPC a DPTAP dochází k tvorbě agregátů nezávisle na poměru lipidů ve směsi, a že s hyaluonanem interagují jak agregáty vzniklé z DPTAP, tak ze směsi DPPC a DPTAP při specifickém poměru koncentrací DPTAP a hyaluronanu. Dále byly studovány fyzikální vlastnosti membrán při různém poměru DPPC a DPTAP ve směsi a vliv cholesterolu na některé z těchto vlastností. V neposlední řadě byly studovány také systémy neionogenní tenzid-DPPC, konkrétně velikost vznikajících agregátů, termodynamika solubilizace a interakce s nativním hyaluronanem., This work is focused on the study of interaction between hyaluronan and high-biocompatible amphiphilic molecules. Using fluorescent probe method, screening of the interaction of cationic lipid 1,2-dipalmitoyl-3-trimethylammonium-propane (DPTAP), mixture of this cationic lipid with zwitterionic lipid, 1,2-dipalmitoyl-sn-glycero-3-phosphochloline (DPPC), with hyaluronan, both native and hydrophobically modified was carried out.
