4. Charting Unexplored Dwarf Galaxy Territory With RR Lyrae

Abstract

Observational bias against finding Milky Way (MW) dwarf galaxies at low Galactic latitudes (b < 20 deg) and at low surface brightnesses (fainter than 29 mag arcsec^-2, in the V-band) currently limits our understanding of the faintest limits of the galaxy luminosity function. This paper is a proof-of-concept that groups of two or more RR Lyrae stars reveal MW dwarf galaxies at d > 50 kpc in these unmined regions of parameter space, with only modest contamination from interloper groups when large halo structures are excluded. For example, a friends-of-friends (FOF) search with a linking length of 500 pc could reveal dwarf galaxies more luminous than M_V = -3.2 mag and with surface brightnesses as faint as 31 mag arcsec^-2 (or even fainter, depending on RR Lyrae specific frequency). Although existing public RR Lyrae catalogs are highly incomplete at d > 50 kpc and/or include <1% of the MW halo's volume, a FOF search reveals two known dwarfs (Bootes I and Sextans) and two dwarf candidate groups possibly worthy of follow-up. PanSTARRS 1 (PS1) may catalog RR Lyrae to 100 kpc which would include ~15% of predicted MW dwarf galaxies. Groups of PS1 RR Lyrae should therefore reveal very low surface brightness and low Galactic latitude dwarfs within its footprint, if they exist. With sensitivity to RR Lyrae to d >600 kpc, LSST is the only planned survey that will be both wide-field and deep enough to use RR Lyrae to definitively measure the Milky Way's dwarf galaxy census to extremely low surface brightnesses, and through the Galactic plane., Comment: Accepted by AJ

Abstract

We present an extensive search for RR Lyrae stars in and around the ultra-faint Milky Way companions Segue 2 and Segue 3. The former (M_V = -2.5, Belokurov et al. 2009) appears to be an extremely faint dwarf galaxy companion of the Milky Way. The latter (M_V = 0.0, Fadely et al. 2011) is among the faintest star clusters known. We use B and V band time-series imaging obtained at the WIYN 0.9 meter telescope at Kitt Peak National Observatory to search for RR Lyrae in these objects. In our Segue 2 observations, we present a previously unknown fundamental mode (RRab) RR Lyrae star with a period of P_ab = 0.748 days. With this measurement, we revisit the inverse correlation between and <[Fe/H]> established in the literature for Milky Way dwarf galaxies and their RR Lyrae. In this context, the long period of Segue 2's RRab star as well as the known significant spread in metallicity in this dwarf galaxy are consistent with the observed trend in and <[Fe/H]>. We derive the first robust distance to Segue 2, using both its RRab star and spectroscopically confirmed blue horizontal branch stars. Using [Fe/H] = -2.16 and -2.44 dex, we find d_RRL = 36.6 +2.5/-2.4 and 37.7 +/- 2.7 kpc; assuming [Fe/H] = -2.257 dex, we find d_BHB = 34.4 +/- 2.6 kpc. Abstract

On Mars, near the equator, much of the terrain in Gale Crater consists of bedrock outcrops separated by relatively smooth, uniform regolith surfaces. In scattered sites, however, distinct patterns—in the form and texture of the ground surface—contrast sharply with the typical terrain and with eolian bedforms. This paper focuses on these diverse, intriguing ground patterns. They include ∼1 to >10 m-long linear disruptions of uniform regolith surfaces, alignments, and other arrangements of similar-sized rock fragments and shallow, ∼0.1 m-wide sandy troughs 1–10 m in length. Similar features were recognized early in the Mars Science Laboratory (MSL) mission, but they received only limited attention until Curiosity, the MSL rover, encountered striking examples in the Glen Torridon region. Herein, the ground patterns are illustrated with rover images. Potential mechanisms are briefly discussed in the context of the bedrock composition and atmospheric conditions documented by Curiosity. The evidence suggests that the patterns are active forms of spontaneous granular organization. It leads to the hypothesis that the patterns arise and develop from miniscule, inferred cyclic expansion and contraction of the bedrock and regolith, likely driven by oscillating transfers of energy and moisture between the atmosphere and the terrain. The hypothesis has significant implications for studies of contemporary processes on Mars on both sides of the atmosphere-lithosphere interface. The ground patterns, as well as ripples and dunes formed by the wind, constitute remarkable extra-terrestrial examples of granular self-organization, complex phenomena well known in diverse systems on Earth.

Abstract

Despite the importance of sand and dust to Mars geomorphology, weather, and exploration, the processes that move sand and that raise dust to maintain Mars' ubiquitous dust haze and to produce dust storms have not been well quantified in situ, with missions lacking either the necessary sensors or a sufficiently active aeolian environment. Perseverance rover's novel environmental sensors and Jezero crater's dusty environment remedy this. In Perseverance's first 216 sols, four convective vortices raised dust locally, while, on average, four passed the rover daily, over 25% of which were significantly dusty ("dust devils"). More rarely, dust lifting by nonvortex wind gusts was produced by daytime convection cells advected over the crater by strong regional daytime upslope winds, which also control aeolian surface features. One such event covered 10 times more area than the largest dust devil, suggesting that dust devils and wind gusts could raise equal amounts of dust under nonstorm conditions.

Abstract

Inositol 1,4,5-trisphosphate receptors (IP3Rs) initiate a diverse array of physiological responses by carefully orchestrating intracellular calcium (Ca2+) signals in response to various external cues. Notably, IP3R channel activity is determined by several obligatory factors, including IP3, Ca2+, and ATP. The critical basic amino acid residues in the N-terminal IP3-binding core (IBC) region that facilitate IP3 binding are well characterized. In contrast, the residues conferring regulation by Ca2+ have yet to be ascertained. Using comparative structural analysis of Ca2+-binding sites identified in two main families of intracellular Ca2+-release channels, ryanodine receptors (RyRs) and IP3Rs, we identified putative acidic residues coordinating Ca2+ in the cytosolic calcium sensor region in IP3Rs. We determined the consequences of substituting putative Ca2+ binding, acidic residues in IP3R family members. We show that the agonist-induced Ca2+ release, single-channel open probability (P0), and Ca2+ sensitivities are markedly altered when the negative charge on the conserved acidic side chain residues is neutralized. Remarkably, neutralizing the negatively charged side chain on two of the residues individually in the putative Ca2+-binding pocket shifted the Ca2+ required to activate IP3R to higher concentrations, indicating that these residues likely are a component of the Ca2+ activation site in IP3R. Taken together, our findings indicate that Ca2+ binding to a well-conserved activation site is a common underlying mechanism resulting in increased channel activity shared by IP3Rs and RyRs.

Abstract

Voltage-dependent calcium channels (Car) open in response to changes in membrane potential, but their activity is modulated by [Ca.sup.2+] binding to calmodulin (CAM). Structural studies of this family of channels have focused on CaM bound to the IQ motif; however, the minimal differences between structures cannot adequately describe CaM's role in the regulation of these channels. We report a unique crystal structure of a 77-residue fragment of the [Ca.sub.v]1.2 [[alpha].sub.1] subunit carboxyl terminus, which includes a tandem of the pre-IQ and IQ domains, in complex with [Ca.sup.2+].CaM in 2 distinct binding modes. The structure of the [Ca.sub.v]1.2 fragment is an unusual dimer of 2 coiled-coiled pre-IQ regions bridged by 2 [Ca.sup.2+].CaMs interacting with the pre-IQ regions and a canonical [Ca.sub.v]1-IQ-[Ca.sup.2+]-CaM complex. Native [Ca.sub.v]1.2 channels are shown to be a mixture of monomers/dimers and a point mutation in the pre-IQ region predicted to abolish the coiled-coil structure significantly reduces [Ca.sup.2+]-dependent inactivation of heterologously expressed [Ca.sub.v]1.2 channels. structure | function | voltage-gated calcium channel

Abstract

The pro- and antiapoptotic proteins belonging to the B-cell lymphoma-2 (Bcl-2) family exert a critical control over cell-death processes by enabling or counteracting mitochondrial outer membrane permeabilization. Beyond this mitochondrial function, several Bcl-2 family members have emerged as critical modulators of intracellular Ca2+ homeostasis and dynamics, showing proapoptotic and antiapoptotic functions. Bcl-2 family proteins specifically target several intracellular Ca2+-transport systems, including organellar Ca2+ channels: inositol 1,4,5-trisphosphate receptors (IP3Rs) and ryanodine receptors (RyRs), Ca2+-release channels mediating Ca2+ flux from the endoplasmic reticulum, as well as voltage-dependent anion channels (VDACs), which mediate Ca2+ flux across the mitochondrial outer membrane into the mitochondria. Although the formation of protein complexes between Bcl-2 proteins and these channels has been extensively studied, a major advance during recent years has been elucidating the complex interaction of Bcl-2 proteins with IP3Rs. Distinct interaction sites for different Bcl-2 family members were identified in the primary structure of IP3Rs. The unique molecular profiles of these Bcl-2 proteins may account for their distinct functional outcomes when bound to IP3Rs. 40. The observed winter circulation at InSight’s landing site and its impact on understanding the year-round circulation and aeolian activity in Elysium Planitia and Gale Crater

Abstract

InSight landed < 550km from the Mars Science Laboratory rover. Both missions carried a suite of meteorological instruments, but MSL’s wind sensor was damaged on landing and failed after ~two Mars years [1]. This made it difficult to characterize the diurnal and seasonal cycles of winds in Gale Crater. Fortunately, InSight’s wind sensor is returning good wind data for the landing season (Ls~300°, northern winter). A complementary presentation provides a detailed look at the observed diurnal cycle of InSight winds and its variability [2].\ud \ud Here, we use atmospheric models, primarily the MarsWRF multi-scale model, run at ~km to 5km scale resolution over the MSL/InSight region, to provide predictions based on our current understanding of the underlying physics. We then improve the models by modifying them to better match the circulation at InSight in northern winter, and use these results to better understand the circulation in Gale Crater. Finally, we extend our simulations over a full Mars year to provide insight into the orientation, morphology, and migration of aeolian features in the vicinity of InSight and MSL.\ud \ud In this region, the large-scale circulation is largely controlled by the seasonal Hadley circulation, with near-surface winds blowing from ~south to north in northern summer and ~north to south in winter. Both landing sites also sit on or close to the hemispheric dichotomy boundary, resulting in daytime upslope flows (from ~north to south) and nighttime downslope flows (from ~south to north); these tend to enhance the Hadley circulation during the daytime in northern winter and at night in summer.\ud \ud For InSight’s location, most models predict winds from between the ~north and west virtually all sol in northern winter [e.g. 3], although there is more model-to-model variation in wind speed. In Gale Crater, however, the situation is complicated by the presence of very large topographic gradients. For example, models suggest that in northern winter the ~north-to-south Hadley circulation winds are enhanced at night by strong downslope winds on the northern rim slopes, resulting in very strong winds at MSL’s location [e.g. 4].\ud \ud Our goal is to develop a model of the seasonal cycle of winds over the entire region which simultaneously matches the InSight and partial MSL wind datasets and the observed aeolian features. Unfortunately, while evidence points toward most aeolian activity near MSL occurring in northern winter at night [5], MSL’s wind sensor was unable to measure nighttime winds well [1]. By measuring regional winds over the full diurnal cycle in northern winter, InSight wind data allow us to (i) directly measure the winds approaching Gale Crater from the ~north, (ii) constrain and improve MarsWRF and other models, and hence (iii) better understand the true nature of the winds and aeolian processes in Gale Crater and over the entire region.\ud \ud [1] Newman, C. Abstract

Ryanodine receptors (RyRs) are tetrameric ligand-gated Ca(2+) release channels that are responsible for the increase of cytosolic Ca(2+) concentration leading to muscle contraction. Our current understanding of RyR channel gating and regulation is greatly limited due to the lack of a high-resolution structure of the channel protein. The enormous size and unwieldy shape of Ca(2+) release channels make X-ray or NMR methods difficult to apply for high-resolution structural analysis of the full-length functional channel. Single-particle electron cryo-microscopy (cryo-EM) is one of the only effective techniques for the study of such a large integral membrane protein and its molecular interactions. Despite recent developments in cryo-EM technologies and break-through single-particle cryo-EM studies of ion channels, cryospecimen preparation, particularly the presence of detergent in the buffer, remains the main impediment to obtaining atomic-resolution structures of ion channels and a multitude of other integral membrane protein complexes. In this review we will discuss properties of several detergents that have been successfully utilized in cryo-EM studies of ion channels and the emergence of the detergent alternative amphipol to stabilize ion channels for structure-function characterization. Future structural studies of challenging specimen like ion channels are likely to be facilitated by cryo-EM amenable detergents or alternative surfactants.
