Short-crested breaking waves that result from directionally spread wave conditions dissipate energy and generate turbulence within the surf zone, altering sediment transport processes, wave runup, and forces on structures. Additionally, vertical vorticity generated near crest ends during breaking, which depends on the gradient in wave height along a crest, may enhance nearshore dispersion of pollutants, nutrients, and larvae. Although directionally spread irregular wave fields are ubiquitous on ocean and large lake coastlines, the dependence of short-crested breaking wave characteristics (including the along-crest length and number of crest ends) on offshore wave conditions is not well established. To assess this relationship, laboratory experiments with alongshore-uniform barred bathymetry were performed in a large-scale directional wave basin. A three-dimensional scanning lidar, trinocular camera stereo processing methods, and in situ measurements were used to study short-crested wave field breaking characteristics in the laboratory, yielding a dataset with dense spatio-temporal coverage relative to prior laboratory or field measurements. Wave height estimates are similar for remotely sensed and in situ observations, except in the outer surf zone where plunging breaking occurred. Directional wave properties estimated with an array of in situ or remotely sensed sea-surface elevation estimates are similar and yield smaller directional spreads than single-point colocated pressure and velocity based in situ estimates when waves are less directionally spread. Using a breaking crest identification procedure combining visible imagery and stereo sea-surface elevation, we find that the average along-crest length of breaking waves decreases and the average number of crest ends increases with increasing directional spread. Relative to observations, a parameterized relationship between directional spread and crest characteristics based on theory for non-breaking, refracting waves generally over-estimates breaking crest lengths and is similar to or underestimates the total number of crest ends observed in the surf zone. Phosducin-like protein (PhLP) is a widely expressed binding partner of the G protein βγ subunit complex (Gβγ) that has been recently shown to catalyze the formation of the Gβ7 dimer from its nascent polypeptides. Phosphorylation of PhLP at one or more of three consecutive serines (Ser-18, Ser-19, and Ser-20) is necessary for Gβγ dimer formation and is believed to be mediated by the protein kinase CK2. Moreover, several lines of evidence suggest that the cytosolic chaperonin complex (CCT) may work in concert with PhLP in the Gβγ-assembly process. The results reported here delineate a mechanism for Gβγ assembly in which a stable ternary complex is formed between PhLP, the nascent Gβ subunit, and CCT that does not include Gγ. PhLP phosphorylation permits the release of a PhLP·Gβ intermediate from CCT, allowing Gγ to associate with Gβ in this intermediate complex. Abstract: Functional and anatomical relationships among primary afferent fibers, blood vessels, and cancers are poorly understood. However, recent evidence suggests that physical and biochemical interactions between these peripheral components are important to both tumor biology and cancer-associated pain. To determine the role of these peripheral components in a mouse model of cancer pain, we quantified the change in nerve and blood vessel density within a fibrosarcoma tumor mass using stereological analysis of serial confocal optical sections of immunostained hind paw. To this end we introduced the Discoma coral-derived red fluorescent protein (DsRed2) into the NCTC 2472 fibrosarcoma line using the Sleeping Beauty transposon methodology, thus providing a unique opportunity to visualize tumor–nerve–vessel associations in context with behavioral assessment of tumor-associated hyperalgesia. Tumors from hyperalgesic mice are more densely innervated with calcitonin gene related peptide (CGRP)-immunoreactive nerve fibers and less densely vascularized than tumors from non-hyperalgesic mice. As hyperalgesia increased from Day 5 to 12 post-implantation, the density of protein gene product 9.5 (PGP9.5)-immunoreactive nerves and CD31-immunoreactive blood vessels in tumors decreased, whereas CGRP-immunoreactive nerve density remained unchanged. Importantly, intra-tumor injection of a CGRP1 receptor antagonist (CGRP 8–37) partially blocked the tumor-associated mechanical hyperalgesia, indicating that local production of CGRP may contribute to tumor-induced nociception through a receptor-mediated process. The results describe for the first time the interaction among sensory nerves, blood vessels and tumor cells in otherwise healthy tissue, and our assessment supports the hypothesis that direct tumor cell-axon communication may underlie, at least in part, the occurrence of cancer pain. Here we address the problem of coherent interference that arises in double-sum wavemakers of wave-resolving models. Identified as a key problem for experimental and numerical simulations since the late 1970s, this problem induces spurious persistent longshore variability and affects nearshore dynamics. To overcome this problem, we present the implementation of a single-sum wavemaker in the 3D wave-resolving model CROCO. The new wavemaker, which assigns only one pair of direction and frequency values to each component of the wave spectrum, definitively prevents coherent interference, unlike a conventional double-sum wavemaker that allows waves of different direction to share the same frequency. Each wave component must also strictly comply with the periodicity rules, to avoid any spurious boundary dynamics. We validate the single-sum wavemaker with experimental data collected in a wave basin with longshore-uniform bathymetry and compare results with the double-sum wavemaker simulations. We show that the new wavemaker produces transient rips devoid of any coherent interference effect and that, consequently, the model statistics closely match the experimental data. The new wavemaker therefore guarantees statistical integrity while reducing computational costs, a necessary step for realistic wave-resolving studies of nearshore dynamics. • Double-sum wavemakers produce coherent interference and persistent rip currents. • Single-sum wavemaker corrects this effect by eliminating coherent interferences. • The correction is validated for the first time with wave basin experiment. Smoking is usually initiated in adolescence, and is the leading preventable cause of death in the United States. Little is known, however, about the links between smoking and neurobiological function in adolescent smokers. This study aimed to probe prefrontal cortical function in late adolescent smokers, using a response inhibition task, and to assess possible relationships between inhibition-related brain activity, clinical features of smoking behavior, and exposure to cigarette smoking. Participants in this study were otherwise healthy late adolescent smokers (15-21 years of age; n=25), who reported daily smoking for at least the 6 months before testing, and age- and education-matched nonsmokers (16-21 years of age; n=25), who each reported smoking fewer than five cigarettes in their lifetimes. The subjects performed the Stop-signal Task, while undergoing functional magnetic resonance imaging. There were no significant group differences in prefrontal cortical activity during response inhibition, but the Heaviness of Smoking Index, a measure of smoking behavior and dependence, was negatively related to neural function in cortical regions of the smokers. These findings suggest that smoking can modulate prefrontal cortical function. Given the late development of the prefrontal cortex, which continues through adolescence, it is possible that smoking may influence the trajectory of brain development during this critical developmental period.
