Quantifying the spatio-temporal distribution of arthropods in tropical rainforests represents a first step towards scrutinizing the global distribution of biodiversity on Earth. To date most studies have focused on narrow taxonomic groups or lack a design that allows partitioning of the components of diversity. Here, we consider an exceptionally large dataset (113,952 individuals representing 5,858 species), obtained from the San Lorenzo forest in Panama, where the phylogenetic breadth of arthropod taxa was surveyed using 14 protocols targeting the soil, litter, understory, lower and upper canopy habitats, replicated across seasons in 2003 and 2004. This dataset is used to explore the relative influence of horizontal, vertical and seasonal drivers of arthropod distribution in this forest. We considered arthropod abundance, observed and estimated species richness, additive decomposition of species richness, multiplicative partitioning of species diversity, variation in species composition, species turnover and guild structure as components of diversity. At the scale of our study (2km of distance, 40m in height and 400 days), the effects related to the vertical and seasonal dimensions were most important. Most adult arthropods were collected from the soil/litter or the upper canopy and species richness was highest in the canopy. We compared the distribution of arthropods and trees within our study system. Effects related to the seasonal dimension were stronger for arthropods than for trees. We conclude that: (1) models of beta diversity developed for tropical trees are unlikely to be applicable to tropical arthropods; (2) it is imperative that estimates of global biodiversity derived from mass collecting of arthropods in tropical rainforests embrace the strong vertical and seasonal partitioning observed here; and (3) given the high species turnover observed between seasons, global climate change may have severe consequences for rainforest arthropods.

In den Jahren 2004 und 2005 wurden vierzehn Hartholzauenbestände entlang der mitteleuropäischen und rumänischen Donau untersucht. Ziel war es, die Lebensgemeinschaften holzbesiedelnder (xylobionter) und blattfressender (phyllophager) Käfer der Eiche auf den Einfluss natürlicher und anthropogen verursachter Umweltfaktoren hin zu untersuchen. Von Interesse waren insbesondere das Überflutungsregime und eine mit Ausdeichung einhergehende Bewirtschaftung und die dadurch bedingte Ausprägung von Strukturparameter wie Kronentotholz, Altholzvorräte, Bestandsstruktur und Pflanzenartenzusammensetzung. Die Untersuchung wurde in bewirtschafteten und unbewirtschafteten, in überfluteten und nicht überfluteten sowie in geschlossenen und offenen Habitaten durchgeführt. Methodisch fanden zur Erfassung der Käfer hauptsächlich die Baumkronenbenebelung mit Insektizid und der Einsatz von Flugfensterfallen über die Vegetationsperiode hinweg Anwendung. Durch diesen vieldimensionalen Aufbau der Untersuchungen konnten eine Reihe verschiedener Fragestellungen durch die Analyse der Gemeinschaftsstrukturen und Abundanzen xylobionter und phyllophager Käfer vergleichend behandelt werden. Übergeordnet stellte sich die Frage, ob Umweltfaktoren und Schlüsselstrukturen identifizierbar sind, welche die Unterschiede in den Artengemeinschaften zwischen den verschiedenen Untersuchungsflächen, aber auch zwischen Mitteleuropa und Rumänien sowie zwischen Xylobionten und Phyllophagen erklären können. Weitere Aspekte waren das Auftreten gefährdeter Käferarten sowie die phänologische Entwicklung über die Vegetationsperiode hinweg. Nicht zuletzt sollten xylobionte und phyllophage Käferarten identifiziert werden, welche als Indikatorarten für die jeweiligen Habitatausprägungen der Hartholzaue dienen können, sowie ein Beitrag zur Erforschung des Lebensraumes Aue und der Baumkronen in Mitteleuropa und Rumänien geleistet werden. Insgesamt konnten knapp 39 000 Käferindividuen zu 964 Arten bestimmt werden. Davon waren 351 Arten (13 000 Individuen) xylobiont und 213 (15 400 Individuen) phyllophag. Die Auswertungen zeigten, dass sich im Rahmen dieser Arbeit und auf Basis der aufgenommenen Umweltfaktoren die xylobionten Lebensgemeinschaften wesentlich besser eigneten als die phyllophagen, landschaftsökologische und naturschutzfachliche Fragestellungen zum Einfluss verschiedener Umweltfaktoren im Hartholzauwald zu beantworten. Letztere reagierten stark auf das bestandspezifische Pflanzenspektrum und mit diesem auf biogeographische und klimatische Einflüsse. Lediglich der direkte Zusammenhang oligophager Käferarten mit auwaldtypischen Gehölzen wie Weide und Pappel führte zu einer entsprechend deutlichen Differenzierung überfluteter und ausgedeichter Bestände. Die xylobionten Käfer zeigten dagegen klare Reaktionen auf Strukturparameter und Umweltfaktoren. Während die genebelten Gemeinschaften einen deutlichen Bezug zu den Strukturen des untersuchten Baumes besaßen, wurden die flugaktiven Xylobionten stärker von den Strukturen des gesamten Habitates beeinflusst. Besiedler von altem Totholz reagierten dabei insbesondere auf das Totholzvolumen, während Frischholzbesiedler eine starke Beziehung zur Totholzoberfläche im Kronenraum zeigten. Naturnahe und regelmäßig überflutete bzw. überstaute Wälder wiesen aufgrund hoher Mengen an altem Totholz sowie durch eine Habitattradition in Form von alter Baumsubstanz sehr hohe Abundanzen altholzbesiedelnder Käfer sowie sehr hohe Anteile gefährdeter und seltener Arten auf. Es konnten hier zudem einige Urwaldreliktarten sowie Neu- und Wiederfunde für Bayern erhalten werden. Eine Ausdeichung und Bewirtschaftung führte stets zu veränderten Artenzusammensetzungen und Abundanzen der xylobionten Käfer. Bewirkt wurde dies durch das Aufkommen überflutungsempfindlicher standortfremder Baumarten und veränderter Totholzqualität durch eine geringere Bestandsfeuchte. In forstlich geprägten Wäldern verweist die relativ niedrige Zahl gefährdeter Arten zudem auf einen Bruch der Habitattradition durch Entfernung alter Baumsubstanz. Zwar ausgedeichte, aber nachhaltig als Mittelwälder bewirtschaftete offene Beständen zeigten den teilweisen Erhalt einer an großdimensionierte Altholzstrukturen gebundenen Fauna, welcher durch Relikteichen bewirkt wurde. Zusätzlich konnte sich eine wärmeliebende, oft frischholzbesiedelnde Artengemeinschaft von hoher Wertigkeit etablieren. Für den Erhalt einer auentypischen Fauna sowohl der xylobionten als auch der phyllophagen Käfer ist eine hohe Bestandfeuchte durch zumindest regelmäßige Überstauung notwendig, welche zugleich eine Bewirtschaftung einschränkt und eine Habitat- und Strukturtradition ermöglicht. Die strukturschaffende Kraft von Hochwasserereignissen sowie eine hohe Heterogenität in Kleinsthabitaten und mikroklimatischen Bedingungen bleiben von besonderer Bedeutung für die Artenvielfalt naturnaher Hartholzauwälder. In the years 2004 and 2005, 14 hardwood floodplain stocks along the central european and romanian danube river have been examined. The goal was to investigate the influence of natural and anthropogenous environmental factors on the communities of xylobiont and phyllophagous beetles of the oak. Of particular interest was the flooding regime and the with embarkement accompanying management, as well as the development of structure parameters like crown dead wood, old wood supplies, inventory structure, and plant type composition. The investigation was made in managed and unmanaged, in flooded and unflooded, as well as in closed and open habitats. Mainly, canopy fogging with insecticide and flight intercept traps over the vegetation period were methodically applied for the collection of the beetles. By this multi-dimensional structure of the investigations, a number of different questions could be treated comparatively by the analysis of the community structures and the abundancies of xylobiont and phyllophagous beetles. Superordinately the question arose whether environmental factors and key structures are identifiable, which can explain the differences in the biological communities between the different investigation areas, and in addition, between Central Europe and Romania as well as between Xylobionts and Phyllophages. Further aspects were the occurrence of endangered species as well as the phaenological development over the vegetation period. Last of all, xylobiont and phyllophagous species of beetle should be identified, which can serve as indicators for their respective habitats of the hardwood floodplain. Additionally, this study should be a contribution for the investigation of the habitat floodplain and canopy in Central Europe and Romania. Altogether nearly 39,000 beetle individuals could be identified for 964 species. Of these, 351 species (13,000 individuals) were xylobiont and 213 (15,400 individuals) phyllophagous. The evaluations pointed out that, in the context of this work and on basis of the captured environmental factors, the xylobiont partnerships were substantially better suitable than the phyllophagous to answer questions of landscape ecology and nature protection regarding the influence of different environmental factors in the hardwood floodplain. The latters react strongly to the stock-specific plant spectrum and with this to biogeographical and climatic influences. Only the direct connection of oligophagous species with floodplain-typical wood such as willow and pappel leads to an accordingly clear differentiation of flooded and embarked stocks. In addition, a higher biodiversity of oak-settling phyllophagous beetles appears on natural as well as dry and open surfaces than in forestal-shaped and denser forests. In comparison, the xylobiont beetles showed clear reactions to structure parameters and environmental factors. While the fogged communities possessed a clear relation to the structures of the examined tree, the flight-active xylobionts were affected more strongly by the structures of the entire habitat. Settlers of old dead wood reacted thereby in particular to the dead wood volume, while fresh wood settlers showed a strong relationship with the dead wood surface in the canopy area. Natural and regularly flooded and/or overdammed forests exhibited very high abundancies of old wood-settling beetles and very high portions of endangered and rare species due to high quantities of old dead wood as well as by a habitat tradition in the form of old tree substance. Furthermore, several urwald relic species as well as new finds and refinds for Bavaria could be received. Embarkement and cultivation always led to changed species compositions and abundancies of xylobiont beetles. This was caused by the emergence of flooding-sensitive, location-strange tree species and changed dead wood quality by a lower humidity. In forestal shaped forests the relatively low number of endangered species refers to a break of the habitat tradition by the removing of old tree substance. Embarked, but sustainable managed as coppice with standard open stocks showed the partial preservation of a fauna bound to large dimensioned old wood structures, which was caused by relic oaks. Additionally, a thermophilic, often freshwood-settling biological community of high significance could be established. For the preservation of a floodplain-typical fauna for both the xylobiont and the phyllophagous beetles, a high humidity by at least regular phreatic water is necessary, which at the same time limits cultivation and makes a habitat and structure tradition possible. The structure-creating strength of flood events as well as a high heterogeneity in micro habitats and micro-climatic conditions remain of special importance for the diversity of species of natural hard wood floodplains.

Background To understand mycobacterial pathogenesis analysis of gene expression by quantification of RNA levels becomes increasingly important. However, current preparation methods yield mycobacterial RNA that is contaminated with chromosomal DNA. Results After sonication of RNA samples from Mycobacterium smegmatis genomic DNA is efficiently removed by DNaseI in contrast to untreated samples. Conclusions This procedure eliminates one of the most prevalent error sources in quantification of RNA levels in mycobacteria.

À ce jour, par manque de réplications spatiales, les travaux réalisés sous diverses longitudes ne permettent pas de dire si, dans les forêts tropicales, la plus grande biodiversité en arthropodes se trouve au niveau du sol ou de la canopée. Le projet Ibisca (Inventaire de la biodiversité des insectes du sol et de la canopée), qui a comme cadre la forêt protégée de San Lorenzo au Panama, s'inscrit dans cette problématique avec un double objectif: (1) étudier les relations existant entre la bêta-biodiversité et la stratification verticale des arthropodes et (2) étudier les variations saisonnières de cette biodiversité. Pour les récoltes au niveau de la canopée, des moyens d'accès variés, qu'ils soient directs (grue gérée par le Smithsonian, réseaux de cordes, "radeau des cimes", "bulle des cimes" et "ikos") ou indirects (fumigation d'insecticide) ont été mis en oeuvre, pour la première fois de façon concomitante, lors de la première phase d'Ibisca en septembre - octobre 2003 (saison des pluies) sur neuf sites différents (400 m2). Au cours de ce projet, 15 méthodes différentes d'échantillonnage ont été utilisées. Les taxa - cibles, environ une quarantaine, ont été choisies pour leur représentativité numérique et en fonction de leur appartenance à des phylogénies et des guildes écologiques variées. Lors de l'année 2004, à trois reprises, des réplications temporelles partielles ont été ou seront menées sur au moins trois des neuf sites (ceux accessibles par la grue): en pleine saison sèche (février - mars: réplication partielle, c'est-à-dire ne faisant intervenir qu'une partie des techniques d'échantillonnage), en tout début de saison des pluies (mai: réplication totale) et en pleine saison des pluies (octobre: réplication partielle). L'interprétation des données (stratification verticale et bêta-diversité) bénéficiera d'informations recueillies au cours d'études connexes (inventaire exhaustif de végétation à DBH > 10 cm, mesures d'ouverture de la canopée, d'incidence lumineuse, du taux d'herbivorie, etc.). L'ensemble des données recueillies est intégré dans une base de données collective, accessible aux chercheurs impliqués dans le projet via Internet.
