Gelatin-Alginate (GA) is frequently employed as a carrier because it is non-toxic, biodegradable, and has a three-dimensional network structure. Meanwhile, cerium oxide nanoparticles (nCeO2) demonstrated high antioxidant enzyme simulation activity. Therefore, in order to develop a porous hydrogel scaffold for the application of bone tissue engineering, an appropriate-type GA-nCeO2 hydrogel scaffold was developed and evaluated.Methods: GA-nCeO2 hydrogel scaffold was prepared by the lyophilized method and characterized. The surface morphology and cell adhesion of the scaffold were observed by the scanning electron microscope. CCK8 and live-dead staining methods were used to evaluate its biological safety and cell proliferation. Then the osteogenic differentiation in early and late stages was discussed. The expression of osteogenic genes was also detected by RT-PCR. Finally, a bone defect model was made in SD rats, and bone formation in vivo was detected.Results: The results showed that GA-nCeO2 hydrogel scaffold exhibited a typical three-dimensional porous structure with a mean pore ratio of 70.61 ± 1.94%. The GA-nCeO2 hydrogel was successfully endowed with simulated enzyme activity including superoxide dismutase (SOD) and catalase (CAT) after the addition of nCeO2. Osteoblasts demonstrated superior cell proliferation and adhesion on composite scaffolds, and both mineralization test and gene expression demonstrated the strong osteogenic potential of GA-nCeO2 hydrogel. The outcomes of hematoxylin and eosin (H&E) staining and Masson trichrome staining in the femoral defect model of SD rats further supported the scaffold’s favorable biocompatibility and bone-promoting capacity.Conclusion: Due to its favorable safety, degradability, and bone formation property, GA-nCeO2 hydrogel was anticipated to be used as a potential bone defect healing material.Graphical Abstract: Keywords: bone regeneration, cerium oxide nanoparticles, hydrogel, bone defect

Drug–drug interaction (DDI) may occur between them because their metabolic pathways overlap and can be inhibited by voriconazole, including cytochrome P450 (CYP) isozymes 3A4 and 2C9.Objective: In the present study, we aimed to investigate the DDI between ruxolitinib and voriconazole in patients with hematological malignancies.Methods: A total of 12 patients with hematologic malignancies were enrolled in this single-arm, single-center, Phase I/II, fixed sequence self-control study. All subjects received 5 mg ruxolitinib alone, followed by the co-administration of ruxolitinib and voriconazole. The plasma concentrations of the two drugs were determined by two well-validated high-performance liquid chromatography-tandem mass spectrometry methods. Phoenix WinNonlin software was used to compare the differences in maximum plasma concentration (Cmax), time to Cmax (Tmax), terminal elimination half-life (T1/2), and apparent plasma clearance (CL/F), as well as area under the curve from time zero to last (AUClast) and AUC from time zero to infinity (AUCinf) between the two periods.Results: After pre-treatment with voriconazole, no significant change existed in Tmax, while Cmax, T1/2, AUClast, and AUCinf of ruxolitinib were significantly increased by 50.4%, 81.3%, 110.1%, and 118.3%, respectively, and CL/F was significantly decreased to 43.6% compared with patients receiving ruxolitinib alone.Conclusion: Our findings confirmed a moderate inhibitory DDI between ruxolitinib and voriconazole as voriconazole decreased the elimination and increased the exposure of ruxolitinib in patients with hematologic malignancies. We recommended a dose reduction regimen when voriconazole and ruxolitinib were coadministered. Drug monitoring might help determine the ruxolitinib treatment concentration for aGVHD patients, improve efficacy, and reduce toxicity.Keywords: pharmacokinetics, ruxolitinib, voriconazole, drug–drug interaction, graft-versus-host disease

The lncRNA HCP5 has been reported to affect the progression of tumor in several types of cancer. Here, in this research, we focus on the role and function of lncRNA HCP5 in human colorectal cancer.Materials and Methods: Tissue samples from colorectal cancer patients were used for detecting the expression of HCP5 by qRT-PCR. Proliferation, migration, invasion and apoptotic cells were assessed by CCK-8, colony formation, transwell assays and flow cytometry in SW480 and HCT-116 cells. The interactions between miR-299-3p and HCP5 or PFN1 were analyzed and confirmed by online database and luciferase reporter assays. The changes in PFN1 and AKT proteins were measured by Western blot. In vivo experiment was used to confirm the role of HCP5 in CRC.Results: The expression of HCP5 had a higher level in colorectal cancer samples and cells by qRT-PCR, comparing with the normal colorectal tissues and human normal colon epithelial cell. It was revealed that knockdown of HCP5 inhibited viabilities, migration and invasion, while inducing apoptosis in SW480 and HCT-116 cells. Then, HCP5 negatively regulated the expressions of miR-299-3p, which negatively regulated the expressions of PFN1 by targeting PFN1. Furthermore, miR-299-3p inhibitor could alleviate the inhibiting effect by si-HCP5 on cell process of SW480 and HCT-116 cells. In addition, the lncHCP5/miR-299-3p/PFN1 axis could affect the progression of CRC through activating the AKT signaling. Last, we confirmed that knockdown of HCP5 inhibited the progression of CRC with an in vivo experiment.Conclusion: The experiments and analyses support our hypothesis that knockdown of lncRNA HCP5 suppresses the progression of colorectal cancer by miR-299-3p/PFN1/AKT axis.Keywords: lncRNA HCP5, miR-299-3p, PFN1, AKT, human colorectal cancer

With the development of semiconductor technology, the size of transistors continues to shrink. In complex radiation environments in aerospace and other fields, small-sized circuits are more prone to soft error (SE). Currently, single-node upset (SNU), double-node upset (DNU) and triple-node upset (TNU) caused by SE are relatively common. TNU’s solution is not yet fully mature. A novel and low-cost TNU self-recoverable latch (named NLCTNURL) was designed which is resistant to harsh radiation effects. When analyzing circuit resiliency, a double-exponential current source is used to simulate the flipping behavior of a node’s stored value when an error occurs. Simulation results show that the latch has full TNU self-recovery. A comparative analysis was conducted on seven latches related to TNU. Besides, a comprehensive index combining delay, power, area and self-recovery—DPAN index was proposed, and all eight types of latches from the perspectives of delay, power, area, and DPAN index were analyzed and compared. The simulation results show that compared with the latches LCTNURL and TNURL which can also achieve TNU self-recoverable, NLCTNURL is reduced by 68.23% and 57.46% respectively from the perspective of delay. From the perspective of power, NLCTNURL is reduced by 72.84% and 74.19%, respectively. From the area perspective, NLCTNURL is reduced by about 28.57% and 53.13%, respectively. From the DPAN index perspective, NLCTNURL is reduced by about 93.12% and 97.31%. The simulation results show that the delay and power stability of the circuit are very high no matter in different temperatures or operating voltages.

There has been very limited study on the stability of a Shack-Hartmann wavefront sensor (SHWS) since its emergence in the early 1970s. In this paper, through experimental study of the system stability of a digital SHWS, a special lenslet array with long focal range is designed and implemented with a spatial light modulator to improve the system performance. Diffractive lenses with long focal length range can provide pseudo-nondiffracting beams and a long range of focusing plane. The performance and effect of the modified SHWS with this lenslet array are investigated, and the experimental results show that the system stability and measurement repeatability are not sensitive to the sensing distance and stay at an acceptable level. The Shack-Hartmann wavefront sensor (SHWS) recently has been extensively researched for optical surface metrology due to its extendable dynamic range compared with interferometry technique. In this paper, we proposed to use a digital SHWS to measure toroidal surfaces, which are widely used in many optical systems due to their different symmetries and curvatures in the X and Y directions. For what is believed to be the first time, an asymmetrical optical lenslet array implemented by a spatial light modulator was presented to tackle the measurement challenge. This unconventional design approach has a great advantage to provide different optical powers in the X and Y directions so that focusing spots can be formed and captured on the detector plane for accurate centroid finding and precise wavefront evaluation for 3D shape reconstruction of the toroidal surface. A digital SHWS system with this extraordinary microlens array was built to verify the design concept, and the experimental results were presented and analyzed.
