The chemical industry explosion in the 20th century has led to increased environmental pollution, affecting fauna, flora, and waterways. These substances alter water's taste, color, and smell, making it unfit for consumption or toxic. Agricultural water networks face threats from pollution before and after treatment. Some chemical contaminants, like pesticides, are embedded in natural biogeochemical cycles. In this study, we developed a simple and low-cost procedure for the fabrication of needles coated with polydimethylsiloxane (PDMS) as an efficient sorbent for the microextraction of organic pollutant traces from water. The prepared needles were used as an alternative for commercial solid-phase micro-extraction (SPME) devices in analytical chemistry. The PDMS polymeric phase was characterized by Fourier-transform infrared spectroscopy (FT-IR), thermogravimetry (TGA), and scanning electron microscopy (SEM). The PDMS-coated needles were used for extraction of thirteen pesticides by direct-immersion solid-phase microextraction (DI-SPME) from contaminated waters, followed by determination with gas chromatography-mass spectrometry (GC-MS). The developed analytical method showed limits of detection (LODs) between 0.3 and 2.5 ng mL−1 and RSDs in the range of 0.8–12.2%. The homemade needles were applied for the extraction of pesticides in surface and ground aqueous samples collected from an agricultural area. A comparative study of a Schiff base reaction involving benzaldehyde and n-butylamine was carried out to improve the yield of the resulting imine. This reaction was carried out at different temperatures without and with the elimination of the water produced during the process by the pervaporation (PV) technique using a typical cylindrical cell. To reach this goal, different dense membranes made of crosslinked poly(vinyl alcohol) with different oxalic acid (crosslinker) contents were prepared by the solvent casting method. Different parameters influencing the performance of the membrane in the separation process including swellability, diffusivity, crosslinking density, and thermal properties were investigated. The total and partial cumulative transmembranar fluxes as well as the separation factor were studied and the separation process was monitored by HPLC analysis. The n-butyl-1-phenylmethanimine produced was characterized by FTIR and 1HNMR analyses. The results obtained were a clear improvement in the yield of the reaction. For example, the yield obtained from the Schiff base reaction occurring without assistance by PV varied from 58 to 84 wt% when the temperature changed from 5 to 45 °C. On the other hand, when the PV process was used to eliminate water from this reaction mixture, the yield went from 90.4 to 98.6% by weight in this same temperature order. The cumulative total and partial fluxes significantly decreased with time. On the other hand, the separation factor reached a maximum at about one hour at 5, 15, and 45 °C. At 25 °C, the maximum total flux was reached at about 2 h of the PV process. The best selectivity of the PVA-0.5 membrane with regard to water was obtained at 15 °C. It was also revealed from the results obtained that the cumulative total and partial flux decreased rapidly with time and the separation factor reached a maximum at one hour into the PV process, in which 1.51 × 104 was reached at 15 °C, 6.25 × 103 and 3.50 × 103 at one hour of the separation process, and 10.23 × 103 at 25 °C at 2 h of the water removal by PV. Arson is a dangerous crime; even a small amount of an ignitable liquid can cause considerable losses in property and even human life. According to the National Fire Protection Association, [...], The identification of ignitable liquids is very important and challenging aspect in arson crime investigations. The detection of gasoline and diesel fuel components using solid phase micro-extraction prior to gas chromatography-mass spectrometry for the forensic analysis of fire debris has been carried out. Previous works show that the absorption characteristics of the substrate are one of the most important factors in determining the evaporation rate of the accelerants. In order to determine the presence of the fuel residues, four of the most common substrate materials were tested in this work; wool, cotton, silk and polyester. The obtained results indicate that both gasoline and diesel fuel accelerants persisted longer on wool and silk than on the other selected substrates. Such information illustrates the influence of fuel persistence times after extinguishing and the best materials to be scanned for ignitable liquids at the fire scene. This paper describes the comprehensive fabrication of monolithic materials for use as stationary phases in capillary liquid chromatography. Several columns were synthesized in the confines of 320 µm i.d. fused-silica capillaries by single-step in situ copolymerization of benzyl methacrylate and ethylene dimethacrylate (EDMA). The polymerization procedure was optimized by varying the reaction time within the range of 0.5–20 h, and by changing the composition contents of the polymeric mixture. The EDMA content showed a predominant influence on the characteristics of the columns and hence, on their chromatographic properties. The optimum value of the thermal initiator corresponded to 5 mg/mL.Changes of the porous, hydrodynamic properties and morphology of the prepared columns were thoroughly investigated and characterized. Different solvents were used as the mobile phase to demonstrate that the resulting monoliths exhibited good permeability and mechanical stability, whereas swelling and shrinking behaviors were observed and discussed. The efficiency and performance toward different sets of analytes were obtained; mixtures of aromatic hydrocarbons and phenolic compounds were successfully separated and evaluated, and adding tetrahydrofuran to the mobile phase showed improvement in both resolution and peak shapes. We aimed in the current study, the identification of a marine bacterial amylase produced by Bacillus pacificus, which was associated with Turbinaria ornata. Cultural conditions were optimized for the highest amylase production on Tryptic soy broth media supplemented with starch 1% at initial pH 9, 55 °C for 24 h. The newly purified amylase was characterized for a possible biotechnological application. Data indicated that the obtained amylase with a molecular weight of 40 kD and the N-terminal sequence of the first 30 amino acids of amBp showed a high degree of homology with known alpha amylase, and was stable at 60 °C of pH 11. Among the tested substrate analogs, amBp was almost fully active on Alylose and Alylopectine (97%), but moderately hydrolyzed glycogen < sucrose < maltose < lactose. Therefore, the current amylase mainly generated maltohexaose from starch. Mg2+ and Zn2+ improved amylase activity up to 170%. While ethylenediamine tetraacetic acid (EDTA) similarly induced the greatest activity with purified amylase, PCMB had the least effect. Regarding all these characteristics, amylase from marine bacterial symbionts amBp has a new promising feature for probable therapeutic, industrial, and nutritional applications. [ABSTRACT FROM AUTHOR]
