Bio-based polymers are a promising material with which to tackle the use of disposable and non-degradable plastics in agriculture, such as mulching films. However, their poor mechanical properties and the high cost of biomaterials have hindered their widespread application. Hence, in this study, we improved polysaccharide-based films and enriched them with plant nutrients to make them suitable for mulching and fertilizing. Films were produced combining sodium carboxymethyl cellulose (CMC), chitosan (CS), and sodium alginate (SA) at different weight ratios with glycerol and CaCl2 as a plasticizer and crosslinker, respectively, and enriched with ammonium phosphate monobasic (NH4H2PO4). A polysaccharide weight ratio of 1:1 generated a film with a more crosslinked structure and a lower expanded network than that featuring the 17:3 ratio, whereas CaCl2 increased the films' water resistance, thermal stability, and strength characteristics, slowing the release rates of NH4+ and PO43−. Background: Soils under organic farming systems exhibit better quality and higher biological activity than conventional systems. Manure addition, especially coupled with reduced or no tillage, significantly enhances microbial biomass and activity by improving soil physical properties and providing carbon (C) and nitrogen (N) sources. While several studies have examined the effects of transitioning from conventional to organic farming on soil chemistry and biochemistry, limited research has explored the influence of land use variations on soil fertility within long‐term organic farming systems. Aims: Therefore, the aim of this study was to assess how three different land uses—pasture, vegetable crops, and orchard—affected soil fertility under a long‐term organic farming system. Methods: Soil samples were collected from the 0 to 15 cm layer of plots used for pasture, vegetable crops and orchard, being the latter cover cropped with legumes, and analyzed to determine chemical and biochemical soil parameters. Results: Contrary to expectations, high land use intensity (vegetable crops and orchard soils) resulted in increased soil organic C and total N, compared to low intensity (pasture). Such an increase was ascribed to farmyard manure addition that counteracted the negative impact of tillage. Consequently, microbial biomass C and activity also increased. The greatest availability of organic substrates favored bacteria, particularly gram‐positive strains, shaping the microbial community. However, despite changes of microbial biomass and of the main microbial groups, microbial activity was only slightly affected, suggesting high functional redundancy of microorganisms in long‐term organic farming soil. Conclusions: Results suggested that if land use intensification provides for organic supply, its negative impact on soil fertility may be mitigated. [ABSTRACT FROM AUTHOR]

The classical chloroform fumigation-incubation (CFI) and fumigation-extraction (CFE) methods are nowadays among the most used for determining soil microbial biomass, although the chloroform lysing of microbial cells is not always complete. Here, we have tested a physical method, used for sterilizing foods but never in soil, based on N2 or CO2 high pressurization (N2HP or CO2HP, respectively) to cause microbial cell lysis. The N2HP and CO2HP were tested on two soils differing for their organic matter content, one agricultural (AGR) and one forest (FOR), and firstly were compared with the CFI. The CO2 extra-flush from both soils during 10-d incubation by N2HP was lower than that by CFI method, whereas that by CO2HP was greater. Then, the lysis by CO2HP was compared with that by the CFE method by varying CO2 pressure and duration. The CO2HP, at proper conditions, was more efficient than CFE method to cause the lysis of soil microbial cells. Moreover, both CO2 pressure value and duration were important in increasing the extractable organic C compared to the CFE. The most successful combination of high CO2 pressure and duration was 4.13 MPa and 32 h. However, we cannot exclude that CO2HP might have caused the release of soil organic C not ascribable to living organic matter. Further studies using 13C and/or 15N-labeled microbial cells should assess the release of abiotic organic C. [ABSTRACT FROM AUTHOR]

A microcosm was assembled to physically separate soil from roots and was used to study both the impact of living roots on the soil-plant system during early stages of growth and plant responses to abiotic stress. Oat (Avena sativa L.) seedlings were grown in the microcosm unit for 44 days. Twenty-three days after planting, 0.154 mg CdSO4/g dry soil was added. Plants grown in Cd-treated microcosms showed considerable inhibition of shoot growth rates, and leaf chlorophyll content. Soil microbial biomass C and respiration increased with plant age, and most of the measured biochemical indicators decreased with increasing distance from the soil root interface, thus demonstrating the rhizosphere effect, likely due to the quick assimilation of rhizodeposits by rhizosphere microflora. On the other hand, short-term Cd contamination sometimes had an inhibitory effect on soil respiration, qC[O.sub.2], ATP content, and phosphatase activity, while stimulating microbial biomass, mainly at the rhizosphere level. The decrease in rhizosphere microbial activity observed after Cd application to soil may be due to a synergic effect of the metal directly on microbial cells and indirectly on plants, which reduced shoot growth rate and chlorophyll content, resulting in decreased availability of root exudates.
