Abstract

Despite decades of intensive search for compounds that modulate the activity of particular protein targets, a large proportion of the human kinome remains as yet undrugged. Effective approaches are therefore required to map the massive space of unexplored compound–kinase interactions for novel and potent activities. Here, we carry out a crowdsourced benchmarking of predictive algorithms for kinase inhibitor potencies across multiple kinase families tested on unpublished bioactivity data. We find the top-performing predictions are based on various models, including kernel learning, gradient boosting and deep learning, and their ensemble leads to a predictive accuracy exceeding that of single-dose kinase activity assays. We design experiments based on the model predictions and identify unexpected activities even for under-studied kinases, thereby accelerating experimental mapping efforts. The open-source prediction algorithms together with the bioactivities between 95 compounds and 295 kinases provide a resource for benchmarking prediction algorithms and for extending the druggable kinome.

Abstract

Stereotypies are common in captive animals, but it remains unclear if they are pathological by-products of captive conditions or if they have an adaptive function. Here, we address this question using crib-biting, a common type of stereotypic behaviour in domestic horses, thought to result from stressful or frustrating environments. Since chronic stress is known to affect learning abilities via dopaminergic modulation in the basal ganglia, we predicted that stereotypic horses would underperform in learning tasks compared to healthy control animals, in line with the pathological by-product hypothesis. We exposed 19 crib-biters and 18 control horses in six spatial tasks, and collected behavioural and physiological data (heart rate, heart rate variability and salivary cortisol) to assess learning performance and stress levels. We found that, during the learning phase, 10 of 19 crib-biters showed stereotypic behaviour. Surprisingly, crib-biters that displayed the stereotypic behaviour (Group A) had lower salivary cortisol values compared to crib-biters that did not display the stereotypic behaviour (Group B) or control horses. (Group C), after the first part of the experiment. In addition, a smaller proportion of horses in Group A displayed behaviours indicating frustration during one of the test compared to Group B. Moreover, we found no difference between the three groups in their learning performance nor in their heart rate or heart rate variability measures. Our results therefore suggest that crib-biting has an adaptive function and can help horses to reduce stress and frustration during learning tasks, which then enables them to show normal learning performance

Abstract

Visual images in news photographs guide individuals’ understandings of people, places and events, especially when news audiences are unable to personally experience those represented images. When 41 Time newsmagazine covers from the first five years of the U.S.-led war on Iraq are considered through a framing analysis, four frames surface: The Sanitized War, Against the Powers-That-Be; The American Soldier in a Time of War; and The "Other" of the War, or "Us versus Them." These findings highlight the power of media messages to frame identity ideologies and stress the importance of complementing quantitative studies with qualitative approaches. ----------------------------------------------------- Las imágenes periodísticas orientan la comprensión sobre individuos, lugares y eventos, especialmente cuando las audiencias no pueden experimentar personalmente aquello representado en la prensa. Al analizar 41 portadas de la revista Time de los primeros cinco años de la Guerra de Irak liderada por los Estados Unidos, se identifican cuatro encuadres: La guerra desinfectada, Contra el poder, El soldado estadounidense en tiempos de guerra y El otro en la guerra (Ellos contra nosotros). Estos hallazgos subrayan el poder de los mensajes mediales para enmarcar ideologías de identidad y destacan la importancia de complementar estudios cuantitativos con enfoques cualitativos.

20. Framing ideology: How Time magazine represents nationalism and identities through visual reporting

Abstract

Visual images in news photographs guide individuals’ understandings of people, places and events, especially when news audiences are unable to personally experience those represented images. When 41 Time newsmagazine covers from the first five years of the U.S.-led war on Iraq are considered through a framing analysis, four frames surface: The Sanitized War, Against the Powers-That-Be; The American Soldier in a Time of War; and The "Other" of the War, or "Us versus Them." These findings highlight the power of media messages to frame identity ideologies and stress the importance of complementing quantitative studies with qualitative approaches. ----------------------------------------------------- Las imágenes periodísticas orientan la comprensión sobre individuos, lugares y eventos, especialmente cuando las audiencias no pueden experimentar personalmente aquello representado en la prensa. Al analizar 41 portadas de la revista Time de los primeros cinco años de la Guerra de Irak liderada por los Estados Unidos, se identifican cuatro encuadres: La guerra desinfectada, Contra el poder, El soldado estadounidense en tiempos de guerra y El otro en la guerra (Ellos contra nosotros). Estos hallazgos subrayan el poder de los mensajes mediales para enmarcar ideologías de identidad y destacan la importancia de complementar estudios cuantitativos con enfoques cualitativos.

Abstract

Background: Physical exercise is an important component of respiratory rehabilitation because it reverses skeletal muscle dysfunction, a clinically important manifestation of COPD associated with reduced health- related quality of life (HRQL) and survival. However, there is controversy regarding the components of the optimal exercise protocol. A study was undertaken to systematically evaluate and summarise randomised controlled trials (RCTs) comparing different exercise protocols for COPD patients. Methods: Six electronic databases, congress proceedings and bibliographies of included studies were searched without imposing language restrictions. Two reviewers independently screened all records and extracted data on study samples, interventions and methodological characteristics of included studies. Results: The methodological quality of the 15 included RCTs was low to moderate. Strength exercise led to larger improvements of HRQL than endurance exercise (weighted mean difference for Chronic Respiratory Questionnaire 0.27, 95% CI 0.02 to 0.52). Interval exercise seems to be of similar effectiveness as continuous exercise, but there are few data on clinically relevant outcomes. One small RCT which included patients with mild COPD compared the effect of high and low intensity exercise (at 80% and 40% of the maximum exercise capacity, respectively) and found larger physiological training effects from high intensity exercise. 4 Conclusions: Strength exercise should be routinely incorporated in respiratory rehabilitation. There is )5 insufficient evidence to recommend high intensity exercise for COPD patients and investigators should conduct larger high quality trials to evaluate exercise intensities in patients with moderate to severe COPD. 36. Risk factors associated with behavioural disorders of crib-biting, weaving and box-walking in Swiss horses.

Abstract

Summary Reasons for performing study: Studies on the prevalence of behavioural disorders in horses and on associated risk factors have revealed inconsistent results. There are many studies on the neuropharmacological, surgical ormechanical therapy of stereotypies, but little is known about their causation. Objectives: To explore risk factors associated with the occurrence of behavioural disorders in horses. Methods: A sample of horse owners, selected randomly and representative for Switzerland, was contacted in a postal survey. Answers were provided for 622 stables (response rate 35.2%). Individual data of 2341 horses were examined with path analysis (multivariable linear and logistic regression), and adjustment made for possible confounding effects due to age and breed. Results: Out of 60 possible risk factors, 11 were associated with the outcome at the univariable level (null-hypothesis path model) and 3 factors remained after the backward logistic regression procedure. Mature Warmbloods and Thoroughbreds, assessed by the owners to be reactive, fed 4 times a day and without daily pasture, had increased odds of displaying crib-biting, weaving and box-walking. Furthermore, indirect associations of 5 factors with the outcome were identified. Conclusions: The final logistic regression model of risk factors leads to the hypotheses that causal prevention of stereotypic behaviours should be based upon housing and management conditions which allow tactile contact with other horses (e.g. mutual grooming), daily free movement (paddock or pasture), as well as the provision of high amounts of roughage but of little or no concentrates. Potential clinical relevance: It is one of the aims of population medicine to prevent the development of behavioural disorders. Further research is needed to test the concluding hypotheses in experimental studies or to verify them in the context of similar observational studies. [ABSTRACT FROM AUTHOR]

Abstract

This introduction to the femme fatale, as a cinematic and critical phenomenon, will explore the ways in which feminist film criticism has read and re‐read this figure, and the ways in which both the figure itself and the critical readings it has engendered, have articulated ideas of gender, sexuality, and power. The cinematic femme fatale is associated primarily with the film noir of the 1940s–1950s, in which context she is commonly read as articulating male anxieties about the position of women following World War II, and as symptomatic of deeper anxieties around female sexuality and agency. At the same time critics have recognized in her a potential for resistance and an expression of female empowerment notwithstanding the often tragic consequences of her ambitions. In the decades that have followed, the figure of the femme fatale has proved an object of endless fascination for filmmakers, audiences, and critics alike. Abstract

Abstract Background Tumor cell proliferation is a predictor of survival in cutaneous melanoma. The aim of the present study was to evaluate the prognostic impact of mitotic count, Ki-67 expression and novel proliferation markers phosphohistone H3 (PHH3), minichromosome maintenance protein 4 (MCM4) and mitosin, and to compare the results with histopathological variables. Methods 202 consecutive cases of nodular cutaneous melanoma were initially included. Mitotic count (mitosis per mm2) was assessed on H&E sections, and Ki-67 expression was estimated by immunohistochemistry on standard sections. PHH3, MCM4 and mitosin were examined by staining of tissue microarrays (TMA) sections. Results Increased mitotic count and elevated Ki-67 expression were strongly associated with increased tumor thickness, presence of ulceration and tumor necrosis. Furthermore, high mitotic count and elevated Ki-67 expression were also associated with Clark's level of invasion and presence of vascular invasion. High expression of PHH3 and MCM4 was correlated with high mitotic count, elevated Ki-67 expression and tumor ulceration, and increased PHH3 frequencies were associated with tumor thickness and presence of tumor necrosis. Univariate analyses showed a worse outcome in cases with elevated Ki-67 expression and high mitotic count, whereas PHH3, MCM4 and mitosin were not significant. Tumor cell proliferation by Ki-67 had significant prognostic impact by multivariate analysis. Conclusions Ki-67 was a stronger and more robust prognostic indicator than mitotic count in this series of nodular melanoma. PHH3, MCM4 and mitosin did not predict patient survival.

Abstract

Abstract Background Tumor necrosis and apoptotic activity are considered important in cancer progression, but these features have not been much studied in melanomas. Our hypothesis was that rapid growth in cutaneous melanomas of the vertical growth phase might lead to tissue hypoxia, alterations in apoptotic activity and tumor necrosis. We proposed that these tumor characteristics might be associated with changes in expression of cell adhesion proteins leading to increased invasive capacity and reduced patient survival. Methods A well characterized series of nodular melanoma (originally 202 cases) and other benign and malignant melanocytic tumors (109 cases) were examined for the presence of necrosis, apoptotic activity (TUNEL assay), immunohistochemical expression of hypoxia markers (HIF-1 α, CAIX, TNF-α, Apaf-1) and cell adhesion proteins (αvβ3 integrin, CD44/HCAM and osteopontin). We hypothesized that tumor hypoxia and necrosis might be associated with increased invasiveness in melanoma through alterations of tumor cell adhesion proteins. Results Necrosis was present in 29% of nodular melanomas and was associated with increased tumor thickness, tumor ulceration, vascular invasion, higher tumor proliferation and apoptotic index, increased expression of αvβ3 integrin and poor patient outcome by multivariate analysis. Tumor cell apoptosis did also correlate with reduced patient survival. Expression of TNF-α and Apaf-1 was significantly associated with tumor thickness, and osteopontin expression correlated with increased tumor cell proliferation (Ki-67). Conclusion Tumor necrosis and apoptotic activity are important features of melanoma progression and prognosis, at least partly through alterations in cell adhesion molecules such as increased αvβ3 integrin expression, revealing potentially important targets for new therapeutic approaches to be further explored.
