Objetivo: Descrever a perspectiva dos profissionais acerca do grupo de familiares destinado aos acompanhantes de pacientes da oncologia pediátrica. Método: Relato de experiência de profissionais da equipe assistencial sobre o grupo de acolhimento aos familiares da unidade de oncologia pediátrica em hospital de referência do sul do Brasil. A atividade é realizada semanalmente e coordenada por duas enfermeiras assistenciais, juntamente com a psicóloga da unidade. Resultados: Os encontros semanais são caracterizados por um espaço de acolhimento, em que ocorre a valorização do cuidador através de um momento de convivência social. É utilizado como um espaço de troca de experiência entre familiares a respeito da doença do seu filho, proporcionando o desenvolvimento de uma rede de apoio, assim como o fortalecimento de vínculos com a equipe assistencial. Por ser um espaço seguro e de educação em saúde, os profissionais ali presentes acabam desmistificando algumas temáticas, como também somando informações pertinentes sobre rotinas da unidade, cuidados a serem seguidos durante o tratamento. Além disso, são trabalhadas demandas psicológicas dos acompanhantes, as quais surgem durante o processo de tratamento da doença. Desta forma, novos questionamentos emergem, e as temáticas são adaptadas para serem abordadas de acordo com as necessidades expostas pelos familiares, trazendo uma devolutiva de novos assuntos em próximos grupos. Conclusão: Por meio deste momento semanal foi possível observar o fortalecimento do vínculo com a equipe assistencial. As famílias relatam sentir-se mais inseridas nos contextos do cuidado, gerando autonomia no que tange os conhecimentos acerca de temáticas que submergem durante o período da internação hospitalar. Além disso, proporciona um melhor entendimento do tratamento e seus impactos na vida cotidiana da família, promovendo um momento de protagonismo e de valorização do acompanhante.

The large deuterium/tritium target is the key component to achieve high yield and high current neutron source, which is the precondition for the wide application of deuterium/tritium neutron source. At present, there are mature deuterium/tritium titanium targets in scientific or engineering applications abroad, and the neutron yield generally reaches the level of 1013/s. However, the technology of large-area target film in China is immature. The scientific and industrial application of large-area target film is rarely reported. The main problem is that the deuterium/tritium target can not reach the high yield, and the large area of the target film prepared is uneven, the binding force is poor and easy to fall off. In this paper, in order to solve the above mentioned problems and optimize the performance of the target film, magnetron sputtering coating and multi-arc ion plating were combined to prepare large-area titanium films with copper or molybdenum substrate and diameter greater than φ500 mm. At the same time, the feasibility of erbium film, scandium film and zirconium film preparation was explored by preparing large area deuterium-titanium or tritium-titanium targets with high binding force and high deuterium-tritium-titanium ratio. Deuterium and tritium targets with deuterium/tritium titanium ratio greater than 1.85 are produced by using self-developed deuterium/tritium target production system after degassing, purification, high temperature deuterium absorption, tail gas recovery and other processes. In order to better utilize the functions of each film layer, scanning electron microscopy was used to observe the cross-section of the film layer in the experiment. The results show that the clear interface is observed among the base layer, transition layer, and functional layer. The prepared titanium film was tested for neutron yield using a deuterium ion accelerator, and the effects of deuterium ion beam current and anode voltage on neutron yield were analyzed. The experiment shows that with the increase of deuterium beam current, neutron yield shows a linear increase in the range of 700 μA. The effect of anode acceleration voltage on neutron yield increases exponentially in a quadratic relationship. The neutron yield of the developed deuterium target is more than 8.0×108/s, and the neutron yield of the tritium target is estimated according to the reaction section of the deuterium target and the tritium target. Under the same conditions, the neutron yield of the tritium target is more than 1.0×1011/s. The test results show that the developed φ500 mm large area deuterium/tritium target can achieve the high-yield neutron output of the high-current neutron source, reaching the highest neutron flux of 1013/s in the world.

Confucianism is a cultural school guided by Confucianism, which is of great significance to the development of Chinese philosophy. This paper first introduces the steps of the outlier data mining algorithm of weighted deep forest, describes the construction method of weighted deep forest, divides the data into several different dimensions, and enters the random forest training in turn. Then outlier detection is introduced, dividing the data into classes with different densities and calculating their local isolation factors and outlier data. Then the algorithm’s performance is tested, and finally, the influence of Confucianism on Chinese philosophy and the main position of Confucian philosophy in the history of Chinese philosophy is analyzed. In terms of algorithm performance, the number T of different decision trees has an impact on AUC. When the number T of subtrees is 50, the AUC value starts to converge, and when the number T is 100 and 200, the AUC value of the algorithm is close to 1. In terms of the influence of Confucianism on Chinese philosophy, 75% of the people think that Confucianism Confucianism is precocious and has enriched the connotation and promoted the evolution of Chinese philosophy. Confucianism is an essential part of the Chinese philosophical calendar, advancing Chinese philosophy and moral culture.

ABSTRACT: Three different medicinal plants that consisted of the formulated mixture (CAVAC-1901) have been traditionally used for distinct medicinal purposes in different areas. Angelica dahurica has been used as an important ingredient of a prescription, Gumiganghwal-tang, for the common cold and influenza. Curcuma longa has been utilized for the treatment of asthma, and jaundice. Pinus densiflora (Korean red pine) has been used to improve memory and brain function for the treatment of vascular. Industrial livestock, which are characterized by dense breeding, are vulnerable to influenza infection, causing severe economic loss and social problems. However, there are no viable alternatives due to the risk of the occurrence of variants. Therefore, the aim of this study was to discover anti-influenza combinations of different medicinal plants with the concept of a multicomponent and multitarget (MCMT) strategy in traditional Chinese medicine (TCM). As part of a continuous project, 3 medicinal plants whose inhibitory activity against influenza A was previously reported at the compound level, and the inhibition of cytopathic effects (CPEs) by these formulated mixtures was evaluated against influenza A virus H1N1. A selected combination with an optimal ratio exhibiting synergistic activity was assessed for its antiviral activity in chickens against the highly pathogenic avian influenza (HPAI) H5N6. The selected combination (CAVAC-1901) showed potent inhibitory effects on the expression of neuraminidase and nucleoprotein, by RT-qPCR, Western blot, and immunofluorescence assays. The antiviral activity was more evident in chickens infected with H5N6. The sample-treated group (50 mg/kg/d) decreased mortality and virus titers in various organs. Our results indirectly suggest synergistic inhibitory activity of the combination of 3 different medicinal plants with different modes of action. Taken together, an optimally formulated mixture (CAVAC-1901) could serve as an effective alternative to current measures to minimize damage caused by HPAIs.

IntroductionIbuprofen is one of the most common non-steroidal anti-inflammatory drugs used to close patent ductus arteriosus (PDA) in preterm infants. PDA is associated with bronchopulmonary dysplasia (BPD), while PDA closure by ibuprofen did not reduce the incidence of BPD or death. Previous studies have indicated an anti-angiogenesis effect of ibuprofen. This study investigated the change of angiogenic factors after ibuprofen treatment in preterm infants.MethodsPreterm infants with hemodynamically significant PDA (hsPDA) were included. After confirmed hsPDA by color doppler ultrasonography within 1 week after birth, infants received oral ibuprofen for three continuous days. Paired plasma before and after the ibuprofen treatment was collected and measured by ELISA to determine the concentrations of platelet-derived growth factor-BB (PDGF-BB) and vascular endothelial growth factor A (VEGF-A), and hypoxia-inducible factor-2α (HIF-2α).Results17 paired plasma from infants with hsPDA were collected. The concentration of PDGF-BB and VEGF-A significantly decreased after ibuprofen treatment (1,908 vs. 442 pg/mL for PDGF-BB, 379 vs. 174 pg/mL for VEGF-A). HIF-2α level showed a tendency to decrease after ibuprofen treatment, although the reduction was not statistically significant (p = 0.077).ConclusionThis study demonstrated decreased vascular growth factors after ibuprofen exposure in hsPDA infants.

The intervertebral disc’s ability to resist load and facilitate motion arises largely from osmotic swelling pressures that develop within the tissue. Changes in the disc’s osmotic environment, diurnally and with disease, have been suggested to regulate cellular activity, yet knowledge of in vivo osmotic environments is limited. Therefore, the first objective of this study was to demonstrate proof-of-concept for a method to measure intra-tissue swelling pressure and osmolality, modeling micro-osmometer fluid flux using Darcy’s law. The second objective was to compare flux-based measurements of the swelling pressure within nucleus pulposus (NP) tissue against ionic swelling pressures predicted by Gibbs-Donnan theory. Pressures (0.03- 0.57 MPa) were applied to NP tissue (n = 25) using equilibrium dialysis, and intra-tissue swelling pressures were measured using flux. Ionic swelling pressures were determined from inductively coupled plasma optical emission spectrometry measurements of intra-tissue sodium using Gibbs-Donnan calculations of fixed charge density and intra-tissue chloride. Concordance of 0.93 was observed between applied pressures and flux- based measurements of swelling pressure. Equilibrium bounds for effective tissue osmolalities engendered by a simulated diurnal loading cycle (0.2-0.6 MPa) were 376 and 522 mOsm/kg H2O. Significant differences between flux and Gibbs-Donnan measures of swelling pressure indicated that total tissue water normalization and non-ionic contributions to swelling pressure were significant, which suggested that standard constitutive models may underestimate intra-tissue swelling pressure. Overall, this micro-osmometer technique may facilitate future validations for constitutive models and measurements of variation in the diurnal osmotic cycle, which may inform studies to identify diurnal- and disease-associated changes in mechanotransduction.

Cooling technology is widely applied in modern turbines to protect the turbine blades, and extracting high-pressure cooling air from a compressor exerts a remarkable influence on the gas-turbine performance. However, the three-dimensional optimal design of a turbine in modern industrial practice is usually carried out by pursuing high component efficiency without considering possible changes in coolant requirement; hence, it may not exactly lead to improvement in the gas-turbine cycle efficiency. In this study, the turbine stator was twisted and leaned to achieve higher comprehensive efficiency, which is the cycle-based efficiency definition for a cooled turbine that considers both turbine aerodynamic performance and coolant requirement. First, the influence of twist and compound lean on turbine aerodynamic performance, considering stator-hub leakage, was investigated. Then, a method to predict the coolant requirement for turbines with different stator designs was applied, to evaluate coolant-requirement change at the design condition. The optimized turbines were finally compared to demonstrate the necessity of considering the coolant-requirement change in the optimal design. This indicated that proper twisting to open the throat area in the stator hub and compound lean to the pressure surface side could help improve the cooled-turbine comprehensive efficiency.

Voltage-gated sodium channels are targets for a range of pharmaceutical drugs developed for the treatment of neurological diseases. Cannabidiol (CBD), the non-psychoactive compound isolated from cannabis plants, was recently approved for treatment of two types of epilepsy associated with sodium channel mutations. This study used high-resolution X-ray crystallography to demonstrate the detailed nature of the interactions between CBD and the NavMs voltage-gated sodium channel, and electrophysiology to show the functional effects of binding CBD to these channels. CBD binds at a novel site at the interface of the fenestrations and the central hydrophobic cavity of the channel. Binding at this site blocks the transmembrane-spanning sodium ion translocation pathway, providing a molecular mechanism for channel inhibition. Modelling studies suggest why the closely-related psychoactive compound tetrahydrocannabinol may not have the same effects on these channels. Finally, comparisons are made with the TRPV2 channel, also recently proposed as a target site for CBD. In summary, this study provides novel insight into a possible mechanism for CBD interactions with sodium channels.

Abstract Background The causes of birth defects (BDs) are complex and include genetic and environmental factors and/or their interactions. More research is needed to describe the epidemiology of BDs within specific regions of China. This study focused on differences in the prevalence of BDs based on ethnicity in a large city in Guangxi Province, China. Methods Surveillance data of infants born in 114 registered hospitals in Liuzhou between 2011 and 2015 were analyzed to determine the epidemiology of BDs across five major ethnic groups. We calculated the prevalence of BDs and relative risk of BDs by ethnicity. Results There were 260,722 perinatal infants of which 6581 had BDs, with the average prevalence of 25.24 per 1000 perinatal infants (PIs). Prevalence data showed an obvious uptrend over the past 5 years. Han had the highest prevalence of total BDs (28.98‰), followed by Zhuang (25.19‰), Yao (18.50‰), Miao (15.78‰) and Dong (14.24‰). Relative to the Han; Zhuang, Miao, Yao, and Dong had a lower risk of musculoskeletal and urogenital malformations; Miao and Yao had a lower risk of cardiovascular malformation; and Dong had a lower risk of cardiovascular and craniofacial malformation. Several maternal risk factors were found to be associated with BDs (e.g., maternal and gestational age, number of antenatal care visits). Conclusion This study provided a comprehensive description of ethnic differences in the risk of BDs in Liuzhou City, China. Observed ethnic differences in the risk of BDs may be related to genetic susceptibilities, environment, cultural customs, or to potential combinations of these factors.

Abstract Background Although the majority of Candida infections occur in the developing world, candidemia epidemiology is poorly understood in these countries. The aim of this study was to investigate the epidemiology of non-Candida albicans (non-C. albicans) candidemia among neonates at Liuzhou Maternity and Child Healthcare Hospital in China. Methods A retrospective review of all positive blood culture about Candida species in neonatal intensive care unit was conducted between January 2012 and November 2015. Information about demographics, risk factors and outcome of candidemia were collected. Univariate and multivariate logistic regression models were used to identify the risk factors associated with the development of non-C.albicans candidemia. Results The prevalence of candidemia in infants was 1.4%. Non-C.albicans was responsible for 56.5% of neonatal candidemia. The predisposing factors for development of non-C.albicans candidemia among infants included mechanical ventilation [odds ratio (OR), 95% confidence interval (95%CI) = 3.13, 1.07–9.14; P = 0.037] and use of assisted reproductive technology (OR, 95%CI = 4.52, 1.39–14.77; P = 0.012). The overall mortality rate of candidemia was 8.7% and non-C.albicans attributed to 83.3% of all mortalities. Conclusions Non-C.albicans species are the major cause of candidemia in local neonatal group. The study highlights the urgent needs to evaluate the possibility of development of non-C.albicans candidemia in neonates exposed to these risk factors and much emphasis must be laid on the early implementation of medical intervention to reduce the incidences of candidemia in neonates.

Femtosecond degenerate four-wave mixing (fs DFWM) experiments are applied to study the rotational coherences of perdeutero-benzene (C6D6) and pyridine (C5H5N). These systems are investigated at 20–40 K in a collision-free seeded supersonic jet and the spectra are compared to those obtained in a gas cell (298 K). The differences between the non-rigid, symmetric oblate C6D6 and the slightly asymmetric oblate C5H5N are discussed. From the simulation and fitting, rotational constants are obtained in good agreement with literature data. For the first time, a fs DFWM spectrum (including asymmetry transients) of a non-rigid, asymmetric rotor molecule (C5H5N) was successfully fitted. [Copyright &y& Elsevier]

Abstract Background The prevalence and clinical characteristics of neonatal candidemia are poorly understood in western China. The aim of our study was to evaluate the epidemiological features of neonatal candidemia in the Liuzhou Maternity and Child Healthcare Hospital. Methods A retrospective case-control study was conducted between January 2012 and November 2015. Electronic databases were reviewed and data on Candida species were isolated from blood cultures and candidemia incidence, risk factors, and mortality were extracted. Univariate and multivariate logistic regression analysis were performed to identify risk factors associated with candidemia. Results During the 4-year period, candidemia was identified in 69 newborns, for an incidence rate of 13.6 per 1000 admissions. Prolonged antibiotic therapy duration [odds ratio (OR), 95% confidence incidence (95% CI) = 1.06, 1.01–1.10], total parenteral nutrition [OR, 95% CI = 6.03, 2.10–17.30] and neurodevelopmental impairment (OR, 95% CI = 7.34, 1.18–45.80) were all associated with increased odds of candidemia development in infants (P value was 0.010, 0.001, 0.033, respectively). The overall mortality rate was 7.2% in the candidemia group. Conclusions Prolonged duration of antibiotic therapy, presence of total parenteral nutrition and neurodevelopmental impairment were the major risk factors associated with neonatal candidemia. This study highlights the importance of the early detection, diagnosis and treatment of neonatal candidemia.

The mechanical behaviour and cellular metabolism of intervertebral discs (IVDs) and articular cartilage are strongly influenced by their proteoglycan content and associated osmotic properties. This osmotic environment is a biophysical signal that changes with disease and may contribute to the elevated matrix breakdown and altered biologic response to loading observed in IVD degeneration and osteoarthritis. This study tested the hypothesis that changes in osmo-sensation by the transient receptor potential vallinoid-4 (TRPV4) ion channel occur with disease and contribute to the inflammatory environment found during degeneration. Immunohistochemistry on bovine IVDs from an inflammatory organ culture model were used to investigate if TRPV4 is expressed in the IVD and how expression changes with degeneration. Western blot, live-cell calcium imaging, and qRT-PCR were used to investigate whether osmolarity changes or tumour necrosis factor α (TNFα) regulate TRPV4 expression, and how altered TRPV4 expression influences calcium signalling and pro-inflammatory cytokine expression. TRPV4 expression correlated with TNFα expression, and was increased when cultured in reduced medium osmolarity and unaltered with TNFα-stimulation. Increased TRPV4 expression increased the calcium flux following TRPV4 activation and increased interleukin-1β (IL-1β) and IL-6 gene expression in IVD cells. TRPV4 expression was qualitatively elevated in regions of aggrecan depletion in degenerated human IVDs. Collectively, results suggest that reduced tissue osmolarity, likely following proteoglycan degradation, can increase TRPV4 signalling and enhance pro-inflammatory cytokine production, suggesting changes in TRPV4 mediated osmo-sensation may contribute to the progressive matrix breakdown in disease.

Pinus densiflora was screened in an ongoing project to discover anti-influenza candidates from natural products. An extensive phytochemical investigation provided 26 compounds, including two new megastigmane glycosides (1 and 2), 21 diterpenoids (3–23), and three flavonoids (24–26). The chemical structures were elucidated by a series of chemical reactions, including modified Mosher’s analysis and various spectroscopic measurements such as LC/MS and 1D- and 2D-NMR. The anti-influenza A activities of all isolates were screened by cytopathic effect (CPE) inhibition assays and neuraminidase (NA) inhibition assays. Ten candidates were selected, and detailed mechanistic studies were performed by various assays, such as Western blot, immunofluorescence, real-time PCR and flow cytometry. Compound 5 exerted its antiviral activity not by direct neutralizing virion surface proteins, such as HA, but by inhibiting the expression of viral mRNA. In contrast, compound 24 showed NA inhibitory activity in a noncompetitive manner with little effect on viral mRNA expression. Interestingly, both compounds 5 and 24 were shown to inhibit nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression in a dose-dependent manner. Taken together, these results provide not only the chemical profiling of P. densiflora but also anti-influenza A candidates.

Annulus fibrosus (AF) defects from annular tears, herniation, and discectomy procedures are associated with painful conditions and accelerated intervertebral disc (IVD) degeneration. Currently, no effective treatments exist to repair AF damage, restore IVD biomechanics and promote tissue regeneration. An injectable fibrin-genipin adhesive hydrogel (Fib-Gen) was evaluated for its performance repairing large AF defects in a bovine caudal IVD model using ex vivo organ culture and biomechanical testing of motion segments, and for its in vivo longevity and biocompatibility in a rat model by subcutaneous implantation. Fib-Gen sealed AF defects, prevented IVD height loss, and remained well-integrated with native AF tissue following approximately 14,000 cycles of compression in 6-day organ culture experiments. Fib-Gen repair also retained high viability of native AF cells near the repair site, reduced nitric oxide released to the media, and showed evidence of AF cell migration into the gel. Biomechanically, Fib-Gen fully restored compressive stiffness to intact levels validating organ culture findings. However, only partial restoration of tensile and torsional stiffness was obtained, suggesting opportunities to enhance this formulation. Subcutaneous implantation results, when compared with the literature, suggested Fib-Gen exhibited similar biocompatibility behaviour to fibrin alone but degraded much more slowly. We conclude that injectable Fib-Gen successfully sealed large AF defects, promoted functional restoration with improved motion segment biomechanics, and served as a biocompatible adhesive biomaterial that had greatly enhanced in vivo longevity compared to fibrin. Fib-Gen offers promise for AF repairs that may prevent painful conditions and accelerated degeneration of the IVD, and warrants further material development and evaluation.

Ecklonia cava is edible seaweed that is found in Asian countries, such as Japan and Korea; and, its major components include fucoidan and phlorotannins. Phlorotannins that are isolated from E. cava are well-known to have an antioxidant effect and strong antiviral activity against porcine epidemic diarrhea virus (PEDV), which has a high mortality rate in piglets. In this study, the bioactive components were determined based on two different approaches: (i) bio-guided isolation using the antiviral activity against the H1N1 viral strain, which is a representative influenza virus that originates from swine and (ii) high-resolution mass spectrometry-based dereplication, including relative mass defects (RMDs) and HPLC-qTOFMS fragmentation analysis. The EC70 fraction showed the strongest antiviral activity and contained thirteen phlorotannins, which were predicted by dereplication. Ten compounds were directly isolated from E. cava extract and then identified. Moreover, the dereplication method allowed for the discovery of two new phlorotannins. The structures of these two isolated compounds were elucidated using NMR techniques and HPLC-qTOFMS fragmentation analysis. In addition, molecular modelling was applied to determine the absolute configurations of the two new compounds. The antiviral activities of seven major phlorotannins in active fraction were evaluated against two influenza A viral strains (H1N1 and H9N2). Six of the compounds showed moderate to strong effects on both of the viruses and phlorofucofuroeckol A (12), which showed an EC50 value of 13.48 ± 1.93 μM, is a potential active antiviral component of E. cava.

Three H9N2 avian influenza viruses were isolated from the Dongting Lake wetland, among which one was from fresh egret feces, the other two were from chicken cloacal swabs in poultry markets. Phylogenetic analyses suggested that eight genes of the egret-derived H9N2 virus might come from Korean-like or American-like lineages. The two poultry-derived H9N2 viruses were reassortants between the CK/BJ/94-like and G1-like viruses. Except the PB1 genes (90.6%), the nucleotide sequence of other internal genes of the two viruses exhibited high homology (>95%). In addition, they also exhibited high homology (96-98.3%) with some genes of the H7N9 virus that caused an epidemic in China in 2013. Nucleotide sequence of the poultry-derived and egret-derived H9N2 viruses shared low homology. Infection studies showed that the egret-derived H9N2 virus was non-pathogenic to both mice and chickens, and the virus was unable to infect chickens even through 8 passages continuously in the lung. On the other hand, the chickens infected by poultry-derived viruses showed obvious clinical symptoms and even died; the infected mice showed no noticeable clinical symptoms and weight loss, but viruses could be detected in their lungs. In conclusion, for the egret-derived H9N2 virus, it would take a long adaptation process to achieve cross-species transmission in poultry and mammals. H9N2 viruses isolated at different times from the same host species in the same geographical region presented different evolutionary status, and virus isolated from different hosts in the same geographical region exhibited genetic diversity. Therefore, it is important to continue the H9N2 virus surveillance for understanding their evolutionary trends so as to provide guidance for disease control and prevention.

We have studied the generation of volatile hydrocarbons by fatty acid-modified L1210 leukemia cells in tissue culture as a measure of lipid peroxidation. There was considerable generation of ethane, and this was dependent on cell number and Fe2+ concentration; it was eliminated by antioxidants and augmented by ascorbic acid. The assay was sensitive and reproducible; ethane was detected when as little as 0.03% of the cellular n-3 (omega-3) fatty acids were peroxidized. To gain further understanding we used a lipid modification model that allows study of cells enriched with fatty acids of different degrees of unsaturation. The quantity of ethane generated was greatest by cells modified with fatty acids of the n-3 family, and there was a high direct correlation of percentage of n-3 fatty acids contained in cellular lipids with peroxidation as measured by ethane generation. Ethane generation was more sensitive in detecting peroxidation than loss of polyunsaturated fatty acids. We conclude that lipid-supplemented leukemic cells produce ethane, and that the rate of generation is a sensitive, quantitative, and highly useful measure of lipid peroxidation when small amounts of iron are present.

In this paper, we report the use of femtosecond time-resolved degenerate four-wave mixing rotationally resolved spectroscopy to obtain very accurate structural information on the symmetric top cyclohexane. Apart from highlighting the versatility of this method in determining accurate structures of large and complex molecules without dipole moment, the present study also details the comparison of the experimentally determined rotational constant B[subo] with that obtained from high-level ab initio calculations. The theoretical calculations, which were carried out at both the second-order Møller-Plesset (MP2) and coupled-cluster with single, double, and perturbative triple substitutions [CCSD(T)] levels of theory, also take into account vibrational averaging effects. Rapid eye movement (REM) sleep has been hypothesized to promote emotional resilience, but any neuronal circuits mediating this have not been identified. We find that in mice, somatostatin (Som) neurons in the entopeduncular nucleus (EP Som )/internal globus pallidus are predominantly active during REM sleep. This unique REM activity is both necessary and sufficient for maintaining normal REM sleep. Inhibiting or exciting EP Som neurons reduced or increased REM sleep duration, respectively. Activation of the sole downstream target of EP Som neurons, Vglut2 cells in the lateral habenula (LHb), increased sleep via the ventral tegmental area (VTA). A simple chemogenetic scheme to periodically inhibit the LHb over 4 days selectively removed a significant amount of cumulative REM sleep. Chronic, but not acute, REM reduction correlated with mice becoming anxious and more sensitive to aversive stimuli. An important factor for investigating climate change in the Sanjiangyuan is the evolution of the spatio-temporal pattern of lakes in this region. The present study used the Google Earth Engine (GEE) platform to extract lakes from 2000 to 2020. The present approach created a lake distribution dataset yearly and analyzed spatial and temporal patterns over 20 years. The analysis of lakes focused on the reaction of the Sanjiangyuan Lakes area to changes in climate, glaciers, and permafrost. The findings indicated that the Sanjiangyuan region contains 143 lakes, the majority of which are predominantly small, measuring 1-10 km 2 . The small lakes account for 60.14 % of the total and are primarily located in the source regions of the Yangtze River and Yellow River. The findings demonstrated that the Sanjiangyuan lakes experienced a significant expansion over the past two decades, particularly from 2011 to 2020. These lakes are divided into expanded, atrophic, and stable categories. Expanded lakes showed significant inter-annual trends in expansion, while atrophic lakes showed smaller fluctuations. The area of stable lakes experienced a consistent decline after 2010, despite a consistent expansion tendency from 2001 to 2010. Moreover, the results indicated that alterations in the size of glaciers and ice reserves in the Sanjiangyuan region have had the greatest influence on the fluctuation in lake area.
