Directed enzyme prodrug therapy (DEPT) strategies show promise in mitigating chemotherapy side effects during cancer treatment. Among these, the use of immobilized enzymes on solid matrices as prodrug activating agents (IDEPT) presents a compelling delivery strategy, offering enhanced tumor targeting and reduced toxicity. Herein, we report a novel IDEPT strategy by employing a His-tagged Leishmania mexicana type I 2′-deoxyribosyltransferase (His-LmPDT) covalently attached to glutaraldehyde-activated magnetic iron oxide nanoparticles (MIONPs). Among the resulting derivatives, PDT-MIONP3 displayed the most favorable catalyst load/retained activity ratio, prompting its selection for further investigation. Substrate specificity studies demonstrated that PDT-MIONP3 effectively hydrolyzed a diverse array of 6-oxo and/or 6-amino purine 2′-deoxynucleosides, including 2-fluoro-2′-deoxyadenosine (dFAdo) and 6-methylpurine-2′-deoxyribose (d6MetPRib), both well-known prodrugs commonly used in DEPT. The biophysical characterization of both MIONPs and PDT-MIONPs was conducted by TEM, DLS, and single particle ICPMS techniques, showing an ideal nanosized range and a zeta potential value of −47.9 mV and −78.2 mV for MIONPs and PDT-MIONPs, respectively. The intracellular uptake of MIONPs and PDT-MIONPs was also determined by TEM and single particle ICPMS on HeLa cancer cell lines and NIH3T3 normal cell lines, showing a higher intracellular uptake in tumor cells. Finally, the selectivity of the PDT-MIONP/dFAdo IDEPT system was tested on HeLa cells (24 h, 10 µM dFAdo), resulting in a significant reduction in tumoral cell survival (11% of viability). Based on the experimental results, PDT-MIONP/dFAdo presents a novel and alternative IDEPT strategy, providing a promising avenue for cancer treatment. [ABSTRACT FROM AUTHOR]

Physicochemical and rheological analyses are carried out on ferrogels based on Agar gum in combination with octenyl succinic anhydride (OSA) starch in the presence of magnetic nanoparticles (MNPs) coated with a stabilizing agent (PNiPAAm). The objective of this work is to prepare nanocapsules of MNPs coated with OSA starch present in a rigid network of Agar to obtain a nanocomposite gel (ferrogel). It is found that the effect of MNPs on the properties of ferrogels has a direct relationship with PNiPAAm/OSA starch interactions. The results obtained show that the ferrogels have stable thermal properties and a high melting point (99.7 °C) for high concentrations of MNPs (0.1%). The rheological properties show that this concentration (0.1%) has a negative effect on the crosslinking of Agar, therefore a drop in viscosity. In addition, it is shown that each pore comprises three populations of MNPs (primary: 3–4 nm, secondary: 6–9 nm, and clusters: ≥12 nm). Moreover, it is found that MNPs are coated with OSA starch. In addition, the prepared materials possess at low concentration of MNPs an interesting stiffness and uniform surface morphology with a minimum of clusters leading to stable systems that can be used as biomaterials in nanomedecine. 1,10‐Phenanthroline d‐ or f‐metal complexes can be utilised in biomedical applications such as imaging or therapeutics. Herein, we designed bimetallic d‐block metal‐phenanthroline f‐block metal‐1,4,7,10‐tetraazacyclododecane‐1,4,7,1,0‐tetraacetic acid (DOTA) conjugates as theranostic agents to simultaneously achieve both of these applications. Luminescence studies show the 1,10‐phenanthroline‐Eu(III)‐DOTA complexes displayed an off/on/off pH‐dependent switch, demonstrating their potential as pH‐responsive lanthanide luminescence probes. Relaxometry studies showed that the 1,10‐phenanthroline‐Gd(III)‐DOTA complexes present a r1 of 5.15±0.05 mM−1 s−1 and could thus be used as magnetic resonance (MR) contrast agents. Complexation of Pt(II) by the 1,10‐phenanthroline moiety resulted in quenching of the Eu(III) luminescence, but an enhancement of the Gd(III) relaxivity (r1=7.53±0.69 mM−1 s−1). Cell viability studies of the d–f hybrids in a cancer cell line showed the potential of these complexes as anticancer agents, as the IC50 for the Pt(II)/Gd(III) complex (IC50=24.9 μM) was lower than that of cisplatin (IC50=31.6 μM). As such, Pt(II)‐1,10‐phenanthroline‐Gd(III)‐DOTA complexes are promising theranostic agents for cancer therapy. [ABSTRACT FROM AUTHOR]

To investigate magnetostructural relationships in colloidal magnetite (Fe3O4) nanoparticles (NPs) at high temperature (300-900 K), we measured the temperature dependence of magnetization (M) of oleate-capped magnetite NPs ca. 20 nm in size. Magnetometry revealed an unusual irreversible high-temperature dependence of M for these NPs, with dip and loop features observed during heating-cooling cycles. Detailed characterizations of as-synthesized and annealed Fe3O4 NPs as well as reference ligand-free Fe3O4 NPs indicate that both types of features in M(T) are related to thermal decomposition of the capping ligands. The ligand decomposition upon the initial heating induces a reduction of Fe3+ to Fe2+ and the associated dip in M, leading to more structurally and compositionally uniform magnetite NPs. Having lost the protective ligands, the NPs continually sinter during subsequent heating cycles, resulting in divergent M curves featuring loops. The increase in M with sintering proceeds not only through elimination of a magnetically dead layer on the particle surface, as a result of a decrease in specific surface area with increasing size, but also through an uncommonly invoked effect resulting from a significant change in Fe3+/Fe2+ ratio with heat treatment. The interpretation of irreversible features in M(T) indicates that reversible M(T) behavior, conversely, can be expected only for ligand-free, structurally and compositionally uniform magnetite NPs, suggesting a general applicability of high-temperature M(T) measurements as an analytical method for probing the structure and composition of magnetic nanomaterials.

This work aimed to develop new ferromagnetic hydrogels based on xanthan gum (XG) and its crosslinked derivative (CXG). The CXG derivative was obtained by chemical reticulation of XG using epichlorohydrin. The formulated XG-Fe3O4 and CXG-Fe3O4 hydrogels were then characterized in terms of structure, morphology, thermal, and rheological properties. The crosslinking of native XG was demonstrated by FTIR and thermal testings. It was noted that the swelling kinetics were affected by crosslinking and an increase in the swelling capacity was observed for the ferromagnetic hydrogels. Morphological images showed a better matrix consistency and improved stability. Additionally, the thermogravimetric analysis indicated a difference in weight loss of the hydrogels in the presence and absence of nanoparticles. The rheological characteristics indicated that both crosslinking and inclusion of Fe3O4 led to the formation of a more rigid structure, which was expressed by the high viscosity values. These novel magnetic hydrogels are interesting candidates for targeted drug delivery as intelligent biopolymeric materials. [ABSTRACT FROM AUTHOR]
