Background: The Bucephalidae is a large family of digenean trematodes but most previous analyses of its phylogenetic position have relied on a single mitochondrial gene or morphological features. Mitochondrial genomes (mitogenomes) remain unavailable for the entire family. To address this, we sequenced the complete mitogenome of Dollfustrema vaneyi and analyzed the phylogenetic relationships with other trematodes. Results: The circular genome of Dollfustrema vaneyi spanned 14,959 bp and contained 12 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and a major non-coding region. We used concatenated amino acid and nucleotide sequences of all 36 genes for phylogenetic analyses, conducted using MrBayes, IQ-TREE and PhyloBayes. We identified pronounced topological instability across different analyses. The addition of recently sequenced two mitogenomes for the Aspidogastrea subclass along with the use of a site-heterogeneous model stabilized the topology, particularly the positions of Azygiidae and Bucephalidae. The stabilized results indicated that Azygiidae was the closest lineage to Bucephalidae in the available dataset, and together, they clustered at the base of the Plagiorchiida. Conclusions: Our study provides the first comprehensive description and annotation of the mitochondrial genome for the Bucephalidae family. The results indicate a close phylogenetic relationship between Azygiidae and Bucephalidae, and reveal their basal placement within the order Plagiorchiida. Furthermore, the inclusion of Aspidogastrea mitogenomes and the site-heterogeneous model significantly improved the topological stability. The present study recorded the adult stages of two digenetic trematodes from the anterior part of the alimentary canal and pyloric caeca of two commercially important marine bony fishes belonging to the family Serranidae from the Red Sea in Egypt. They are opecoelid Cainocreadium epinepheli (Yamaguti, 1934) Durio and Manter, 1968 and Bucephalid Prosorhynchus serrani Durio and Manter, 1968 that infect areolate grouper, Epinephelus areolatus and yellow-edged lyretail, Variola louti, respectively. The incidence of infection is reported as 55.5% (5/9) for Cainocreadium epinepheli and 81.2 % (13/16) for Prosorhynchus serrani. The body of Cainocreadium epinepheli measured 2.003-3.667 (2.809) × 0.651-1.338 (0.986) (length × maximum width) while the body of Prosorhynchus serrani measured 1.664-2.210 (1.750) × 0.225-0.358 (0.287). Cainocreadium epinepheli is characterized by the presence of smooth tegument, median gonopore in the forebody, canalicular seminal receptacle, claviform cirrus sac, long excretory vesicle reaching forebody, pre-testicular intercaecal uterus, lobed ovary, non-filamentous eggs and has an extension of vitellaria into forebody. On the other hand, Prosorhynchus serrani devoids the oral and ventral suckers but has a single caecum, midbody-located mouth, conical rhynchus, pre-testicular ovary, and elongated seminal vesicle with curved proximal part of pars prostatica. The internal organs, body regions, and their ratios concerning total body length for both species were described, measured, and compared with some of the previously described species. Prosorhynchoides extenuatus n. sp. is described based on adult worms collected from the intestine of the chain pickerel, Esox niger Lesueur, 1818, from the Pascagoula River, Jackson County, Mississippi, U.S.A. Sequences of the ribosomal DNA from adult specimens (internal transcribed spacer region 2, and partial 28S ribosomal DNA gene) were identical to those of a cercaria developing in branched sporocysts within the Gulf wedge clam, Rangia cuneata (G. B. Sowerby I, 1832) and a metacercaria we collected from the threadfin shad, Dorosoma petenense (Günther, 1867) in the same river. The cercaria conforms to the description of bucephalid cercaria C Wardle, 1990, that was originally described from the same host in Texas, U.S.A. Novel sequence data are also provided for Prosorhynchoides potamoensisCurran & Overstreet, 2009, and Prosorhynchoides pusillus (Stafford, 1904) Margolis & Arthur, 1979. The 28S rDNA fragments from these species are aligned with bucephalid sequences available in GenBank and their relationships are analyzed using Bayesian inference analysis. The new species formed a branch within a clade of marine bucephalines, whereas P. potamoensis and P. pusillus branched within a clade of North American freshwater species currently classified in Bucephalinae and Paurorhynchinae. We surveyed 30 individuals of Tylosurus gavialoides (Castelnau) (Belonidae) collected from Moreton Bay, Queensland, Australia, and describe three new species of Prosorhynchoides Dollfus, 1929 from them. The new species are morphologically distinct from existing Prosorhynchoides spp. and 28S and ITS-2 ribosomal DNA data further supports our morphological findings. We also conduct the first mitochondrial DNA analysis of species of Prosorhynchoides . The new species from T. gavialoides form a strongly supported clade on the basis of the two ribosomal markers, further supporting the emerging hypothesis that bucephaline clades are strongly associated with host groups. We have not observed any of the new species reported here in over 3500 surveyed individuals of other piscivorous fish in Australia, suggesting that these species are host-specific at least to belonids, if not to only T. gavialoides . Our findings support previous reports that suggest that belonids are exceptional hosts for bucephalids. We predict that further sampling of the numerous other belonid species present in Australian waters, for which nothing is known of the bucephalid fauna, will uncover further bucephalid richness. The present study describes the ultrastructural organisation of the mature spermatozoon of the bucephalid digenean Prosorhynchoides arcuatus (Bucephalinae) collected from the Bluefish Pomatomus saltatrix (Teleostei: Pomatomidae) caught from the Gulf of Gabès in the Chebba (Tunisia) by means of transmission electron microscopy. The ultrastructural study reveals that the mature spermatozoon of P. arcuatus is a filiform cell which is tapered at both extremities. It exhibits the Bakhoum et al.’s type V of spermatozoon of digeneans characterised by the presence of two axonemes with the 9 + ‘1’ pattern of the Trepaxonemata, lateral expansion, external ornamentation of the plasma membrane located in the anterior part of the proximal region of the spermatozoon and associated with cortical microtubules, two bundles of parallel cortical microtubules with maximum number located in the anterior part of the spermatozoon, and one mitochondrion. Other characteristics are the presence of spine-like bodies, a posterior extremity containing only the nucleus, and the presence of a large amount of glycogen granules. Within the Digenea, the family Bucephalidae includes numerous species parasitizing mainly marine and freshwater fishes. This family includes five recognized subfamilies, and ultrastructural data on their sperm cells are very scarce. The existing data are restricted to the subfamily Bucephalinae. Thus, the present study is the first complete analysis of the sperm cell of a bucephalid belonging to the subfamily Prosorhynchinae. Herein, we describe the ultrastructure of the mature spermatozoon of Prosorhynchus aculeatus, a parasite of the conger eel Conger conger, assessed by means of transmission electron microscopy (TEM). The spermatozoon of P. aculeatus is a filiform cell that presents two axonemes of the 9+'1' pattern of trepaxonematan Platyhelminthes, parallel cortical microtubules, mitochondrion, nucleus, external ornamentation of the plasma membrane, spine-like bodies and a large amount of glycogen granules. According to the anterior location of the external ornamentation of the plasma membrane, P. aculeatus presents a Quilichini et al.'s type 1 spermatozoon. With respect to the posterior extremity, the sperm cell of P. aculeatus corresponds to the Quilichini et al.'s cryptogonimidean type. Our results are compared with those of the two previously studied bucephalids (Bucephalinae), Prosorhynchoides gracilescens and Pseudorhipidocotyle elopichthys. [ABSTRACT FROM AUTHOR]

The identity of a larval bucephalid infecting the eastern oyster, Crassostrea virginica (Gmelin, 1791), from a tidal river in Virginia is investigated using ribosomal deoxyribonucleic acid (rDNA). The internal transcribed spacer (ITS1, 5.8S, ITS2) region and a portion of the 28S rDNA were isolated from genomic DNA from sporocysts containing cercariae and compared with sequences from GenBank and from our previous collections of possibly related bucephalids. The studied larval bucephalid was 100% identical at the ITS1, 5.8S, and partial 28S rDNA sequences with Prosorhynchoides paralichthydis (Corkum, 1961) Curran and Overstreet, 2009; however, it differed from P. paralichthydis by 6 bases plus 3 deletions in the ITS2 region. This level of variation at the ITS2 region has been demonstrated among some Indo-Pacific species of Prosorhynchoides Dollfus, 1929, suggesting that the larval bucephalid represents an unidentified or innominate species of Prosorhynchoides that is closely related to P. paralichthydis., (© American Society of Parasitologists 2023.)

Prosorhynchoides fabulus n. sp. (Trematoda: Bucephalidae) is described from the intestine of white bass Morone chrysops (Moronidae) collected from the Neches River in the Big Thicket National Preserve in Texas, U.S.A. The new species is characterized by a relatively large cirrus sac extending anteriorly to the level of the ovary and cecum, a mouth nearly exactly midbody, obliquely arranged testes, a serpentine excretory bladder limited to the hindbody, and small size (<500 μm). The extent of the cirrus sac distinguishes P. fabulus n. sp. from most other species in the genus in North America. The new species displays similarities to Prosorhynchoides carvajali, Prosorhynchoides labiatus, and Prosorhynchoides megacirrus in the extent of the cirrus sac but differs from these species in combinations of the location of the vitellaria, position of the pharynx and mouth, extent of the excretory bladder, arrangement and position of the gonads, and body size. Two new species of bucephalid trematode (Platyhelminthes: Digenea) are described from the giant moray eel, Gymnothorax javanicus (Anguilliformes: Muraenidae), from off Lizard Island, Australia. We used a combined morphological and molecular-based approach targeting the internal transcribed spacer 2 (ITS2) of the ribosomal DNA (rDNA) and the D1–D3 region of the large subunit (28S) of rDNA to circumscribe the species. Dollfustrema durum n. sp. is distinguished from seven congeners in having 5–6 rows of enlarged body spines circling the anterior portion of the rhynchus. From the remaining 10 species, D. durum n. sp. differs in body length, and in having a caecum that terminates posteriorly to the confluent arc formed by the vitelline follicles, gonads predominantly anterior to the pharynx, testes in tandem, an anterior testis positioned posteriorly to the vitelline follicles, and the pre-vitelline field 23–40% of the body length. Heterobucephalopsis perardua n. sp. differs from Heterobucephalopsis gymnothoracis , the type- and only other reported species, in being two to three times smaller. Heterobucephalopsis , currently considered a genus inquirendum , is confirmed as valid and is rediagnosed. Bayesian inference analysis of 28S rDNA sequences representing 28 species from nine genera and four subfamilies of bucephalid, indicates that i) subfamily classifications previously based on morphological characters are broadly robust, ii) the sequence representing H. perardua n. sp. is resolved as distinct, and basal, to sequences representing the Bucephalinae, the Prosorhynchinae, the Paurorhynchinae, and the Dolichoenterinae, iii) the Dolichoenterinae and the Prosorhynchinae are monophyletic sister clades, basal to the Bucephalinae and the Paurorhynchinae, iv) sequences representing Grammatorcynicola , Prosorhynchus , and Dollfustrema are also monophyletic, v) the Bucephalinae is paraphyletic relative to the Paurorhynchinae, and vi) the bucephaline genera Prosorhynchoides , Rhipidocotyle , and Bucephalus are each polyphyletic. The morphological and molecular differences observed among the four previously recognised subfamilies in this study lead us to propose Heterobucephalopsinae n. subfam. to accommodate the genus Heterobucephalopsis . We describe the life cycle of the bucephalid Prosorhynchoides carvajali from the intertidal rocky zone of central Chile. To elucidate the life cycle of this digenean, two mytilid bivalves, Semimytilus algosus and Perumytilus purpuratus, and ten intertidal fish species belonging to the families Blenniidae, Tripterygiidae, Labrisomidae, Kyphosidae and Gobiesocidae were analysed for natural infections. In addition, experimental infections of fish were undertaken and molecular analyses were performed of several developmental stages of the digeneans in various host species. Experimental infections of fish were made from infected mytilids to determine which fish species were suitable for the metacercarial stage of Prosorhynchoides. We also determined the abundance and prevalence of metacercariae in natural infections in fish and found that they were lower than in the experimental infections. A molecular analysis showed that sporocysts from S. algosus were identical to metacercariae from five fish species and P. carvajali adults. Sporocysts isolated from P. purpuratus were similar to metacercaria found in one fish species only (G. laevifrons) but were different from P. carvajali, with 1.9–2.0% genetic divergence. Therefore, the complete life cycle of P. carvajali consists of the mytilid species S. algosus as the first intermediate host, at least five intertidal fish species as second intermediate hosts (Scartichthys viridis, Auchenionchus microcirrhis, Hypsoblennius sordidus, Helcogrammoides chilensis and Gobiesox marmoratus), two carnivorous fish as definitive hosts (Auchenionchus microcirrhis and A. variolosus) and one occasional definitive host (Syciases sanguineus). The bivalve Perumytilus purpuratus is a common species that is widely distributed throughout rocky intertidal zones in Chile. This bivalve is the first intermediate host for three trematode species: one bucephalid (an undetermined species) and two fellodistomids (Proctoeces lintoni and one undetermined species). A few studies based on morphological comparisons, experimental infection and molecular analyses have been performed to ascertain the taxon (at least at the family level) to which these trematodes belong; yet, there remains no clarification about the specific identity of these trematodes. Therefore, in this study, we compared the V4 region nucleotide sequences of the 18S rRNA of these three sporocyst species, classified as morphotypes, found in P. purpuratus and nine adult trematode species from intertidal fishes that are likely definitive hosts for these parasites. The sequences from two of the sporocyst morphotypes matched with adult trematodes from the intertidal fish: type 1 sporocyst was similar to Prosorhynchoides carvajali (Bucephalidae), with a mean genetic divergence of 0.78%, and type 2 sporocyst was similar to Proctoeces sp. (but not P. lintoni), with 0% genetic divergence. The third species (type 3 sporocyst) was classified to the family Fellodistomidae; however, the sequence from this species differed greatly from the three other fellodistomid species documented in the marine fish of Chile and from other fellodistomids in public databases. Moreover, this morphotype has a particular cercarial morphology that greatly differs from other fellodistomid species described thus far. Therefore, this intriguing trematode remains a mystery. We examined four species of Plectropomus Oken, 1817 (Serranidae: Epinephelinae), Plectropomus areolatus (Rüppell), Plectropomus laevis (Lacepède), Plectropomus leopardus (Lacepède) and Plectropomus maculatus (Bloch) from sites off Heron Island and Lizard Island on the Great Barrier Reef, Australia (GBR), and the Gambier Islands, French Polynesia. Three new species of Neidhartia Nagaty, 1937, five new species of Prosorhynchus Odhner, 1905, and one previously described species, Prosorhynchus freitasi Nagaty, 1937, are characterised. The three species of Neidhartia, Neidhartia haywardi n. sp., Neidhartia plectropomi n. sp. and Neidhartia tyleri n. sp. are readily distinguishable morphologically. Two of the six species of Prosorhynchus ( Prosorhynchus lesteri n. sp. and Prosorhynchus wrightae n. sp.) are easily distinguished from their other congeners by morphology but the other four species ( P. freitasi, Prosorhynchus heronensis n. sp., Prosorhynchus munozae n. sp. and Prosorhynchus plectropomi n. sp.) are generally similar in morphology and were only distinguished initially by comparing their ITS2 rRNA sequences. Three additional taxa, one from the GBR and two from French Polynesia, were recognised as distinct on the basis that their ITS2 rRNA sequences differed from those of the new taxa described here; these species remain unnamed for the present. Inter-specific divergence observed within these genera in the ITS2 rRNA ranged from 10 to 42 base pairs (4-16 %) for species of Neidhartia and 2-57 base pairs (3-21 %) for species of Prosorhynchus. Inter-generic divergences were 42-55 base pairs (17-21 %). No intraspecific variation in the ITS2 rRNA region was observed for any of the six species for which multiple sequence replicates were obtained. Phylogenetic analysis of 12 operational taxa from Plectropomus together with sequences of three other species from epinepheline serranids demonstrated that Neidhartia and Prosorhynchus were reciprocally monophyletic with the exception that P. wrightae n. sp. fell either within or basal to the Neidhartia species. The richness of bucephalids in species of Plectropomus appears to be exceptional within the Serranidae relative to that observed in other serranid genera in the tropical Indo-West Pacific. Perumytilus purpuratus is an abundant bivalve located in the intertidal rocky zone of South America that has been considered as a key species of the ecosystem. There are few studies of the host-parasite relationship of this bivalve; thus, this research aims to analyse the spatial and temporal variation in the prevalence of trematodes in P. purpuratus. Bivalves were collected from three localities (El Tabo, Las Cruces and Montemar) of central Chile (33°S, 71°W) during different seasons of 2010. The bivalves were also collected every metre, from the lowest to the highest level of the intertidal rocky zone, to determine the parasite distribution within the localities. Three species of trematodes as sporocyst stages were found: Prosorhynchoides carvajali, Proctoeces sp. and an undetermined fellodistomid species. Of the 37,692 bivalve specimens collected, 2.68% were parasitised. The undetermined fellodistomid species was the most prevalent parasite observed (1.69%). There were little detected differences in the prevalence of some trematode species between seasons. The prevalence of P. carvajali varied between localities, being most prevalent at Montemar. The distribution of trematodes along the rocky zone within the localities was variable, with P. carvajali being more prevalent in the mid-lowest level of the intertidal zone and the undetermined fellodistomid species being more prevalent in the mid-highest level. Both the abundance of definitive hosts and the environmental conditions likely result in different levels of infection by trematodes in P. purpuratus between and within the localities. This paper reports the adult stage of Prosorhynchoides borealis (Digenea) from Lophius piscatorius in Icelandic waters and infections with the larval stages (sporocysts and cercariae) found for the first time in the bivalve Abra prismatica (Semelidae). The previously known first intermediate host was Abraalba (Semelidae). Ribosomal DNA sequencing studies on all three life stages of the parasite (cercariae, metacercariae, adults) were performed to confirm their identites. Morphometric measurements confirmed that the adult worms belong to the newly described species P. borealis. Prosorhynchoides borealis sporocysts filled with cercariae were found in 16% of A. prismatica bivalves sampled at depths between 34 and 93 m off South Iceland. Prevalence ranged from 0 to 44% between different localities. The parasite was found only in the larger bivalves. Extensive sporocyst infection in the haemocoel of the foot caused mechanical muscle damage with subsequent degeneration and necrosis. Other tissues, including the digestive gland, nephridia, gills and intestine, were less heavily infected. Only focal necrosis was observed in the digestive gland, nephridia and gills, and local atrophy in the intestine. Cercariae were also observed in the lumen of both the stomach and intestine. This is the first report of A. prismatica as an alternative first intermediate host for P. borealis. Ribosomal DNA sequence data reveals 100% homology in the data between cercariae, metacercariae and adult digeneans, supporting the morphological data suggesting that all stages belong to the same species. The article presents the study which described the eight prosorhynchine parasite species found in epinepheline fishes from New Caledonia. Among the species are the Neidhartia lochepintade n. species (sp.) in Epinephelus chlorostigma, Neidhartia haywardi Bott, Miller & Cribb, 2013 in Plectropomus leopardus, and Prosorhynchus robertsthomsoni Bott & Cribb, 2009 in Cephalopholis argus.

Sporocysts of Prosorhynchoides borealis were obtained from the marine bivalves Abra prismatica and studied using transmission electron microscopy. The sporocyst body consists of a mass of branching and intertwining hollow tubules that ramify through the host's digestive gland and gonads. This study investigated the ultrastructure of the sporocyst branches which comprise alternate distended areas (brood chambers) with a relatively thin body wall, narrower portions with a thicker body wall (constricted areas) and terminal regions. Pronounced differences between these areas were revealed in the structure of their tegument and body cavity lining, as well as in the cellular composition of the subtegumental layers. Body wall composition in distended areas was consistent with the specialization for cercarial nurture in the brood chambers. The structure of the constrictions suggested a dual role of nutrient absorption and physical separation of adjacent brood chambers. Two types of terminal region were identified, one specialized for the investigation and penetration of host tissues and the other, in which the germinal cells are formed, for cercarial production. The overall structure of the sporocyst branches helps explain why this linear modular system, i.e. brood chambers and constrictions continuously growing into the host tissue, enables the sporocyst's long-term existence and can continuously produce cercariae in numbers comparable with those produced by rediae and/or daughter sporocyst infrapopulations in other digeneans. A polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique was developed for the molecular identification of 2 introduced bucephalid trematodes, Prosorhynchoides ozakii and Parabucephalopsis parasiluri. The method was applied for sporocysts and cercariae obtained from the golden mussel Limnoperna fortunei collected in the Uji River, Japan. The PCR-RFLP method showed that L. fortunei is the intermediate host of both trematode species. The present study thus recognizes the risk of L. fortunei, an invasive molluscan species, as a potential host for pathogenic trematodes. [ABSTRACT FROM AUTHOR]

new bucephalid species, Prosorhynchoides carvajali sp. nov. is described. This parasite was found in three marine fish, Auchenionchus microcirrhis (type-host), A. variolosus and Sicyases sanguineus (other-hosts), collected from the intertidal rocky zones of central Chile. P. carvajali sp. nov. is characterized by a pharynx in a post-equatorial position, a large cirrus sac length (half of the total worm length) and rounded caecum extending dorsally and anteriorly from pharynx. Although Prosorhynchoides carvajali sp. nov. closely resembles P. labiata; the latter has an elongated, narrow and inverted-U-shape caecum, contrasting to P. carvajali sp. nov. which has a larger rounded caecum, directed anteriorly. Podvyaznaya I. 2011. An ultrastructural study of alimentary tract development in the cercariae of Prosorhynchoides borealis (Digenea, Bucephalidae). - Acta Zoologica (Stockholm) : 170-178. The development of digestive system in Prosorhynchoides borealis cercariae was studied using transmission electron microscopy. The foregut and caecum primordia arise in early cercarial embryos as two adjoining cellular cords. The primordial pharynx appears as a cluster of myoblasts in the mid-part of the foregut primordium whose proximal end abuts onto the ventral embryonic tegument. Later, a lumen develops within the gut primordia and their component cells form the embryonic cellular epithelium with an essentially similar structure in the foregut and caecal regions. Subsequently, the foregut epithelial cells merge to form a syncytium. This process proceeds asynchronously and the most proximal foregut area remains cellular for the longest time. The syncytial lining of the foregut establishes syncytial connections with secretory cytons differentiating in the surrounding parenchyma. These cytons produce secretory granules, which are transported through cytoplasmic connections to the foregut syncytium. Before cercariae reach maturity, their foregut epithelium becomes anucleate and continuous with the external tegument. By the end of cercarial development, numerous short lamellae appear on the luminal surface of the caecal epithelium. Abstract: This paper reports the parasites found in three commercially exploited bivalve molluscs (Mytella guyanensis, Anomalocardia brasiliana and Iphigenia brasiliana) of an estuarine region of Ilhéus, south of Bahia, Brazil (14°48′23′′S; 39°02′47′′W). Samples of 20 individuals of each species were collected fortnightly from August 2005 to August 2006. A total of 1480 individuals was collected and processed by standard histologic techniques; the histologic sections were stained with Harris haematoxylin and eosin and examined with light microscope. The water temperature in the study area varied from 24 to 30.5°C and the salinity from 0 to 23ppt. Remarkable differences were found in the parasitic community between the three mollusc species involved in the study, which occupied different habitats in the estuarine region of the Cachoeira river. The following parasites were found: intracellular rickettsia-like colonies in digestive epithelia; intracellular gregarine Nematopsis sp. in gills, mantle, gonad, digestive gland and foot muscle; sporocysts of a Bucephalidae trematode in gonads, mantle, gills, digestive gland and foot; unidentified digenetic metacercariae in digestive gland and gonad; metacestodes of Tylocephalum sp. in connective tissue in the digestive gland and in gonad; and an unidentified metazoan in mantle and intestinal lumen. No significant temporal variation in the prevalence of any parasite was detected, which could be due to the narrow temperature range of the region and the absence of patterns of salinity and rainfall variation through the year. The infestation by sporocyst was the only pathological threat detected for the studied populations because of its potential for host castration. The low infection intensity and/or prevalence of the other parasites and the lack of obvious lesions suggest that there is no other serious pathological risk for the studied mollusc populations. The cercaria of Bucephalus minimus infects the digestive gland and gonads of its first intermediate host, the edible cockle, Cerastoderma edule. Light microscopy (LM) and scanning electron microscopy (SEM) of the cercaria showed a tail formed by a central stem, with 2 long contractile arms presenting distinct morphological surfaces. The encysted metacercaria naturally infected the flathead grey mullet, Mugil cephalus. The cysts found in the heart, liver, and spleen were shown to be identical by the internal transcribed spacer (ITS1) sequence and morphological features and were associated with encapsulation, recruitment of cell infiltrates, and presence of melanomacrophages and adipose tissue. To establish the life cycle, we compared the ITS1 sequence in an adult from the known definitive host, Dicentrarchus labrax; encysted metacercariae from the liver, heart, and spleen of M. cephalus; and a cercaria from C. edule. With this comparison, we determined that they had a 100% similarity. Therefore, the ITS1 sequence data clearly indicate that these 3 parasitic stages belong to the same species, i.e., B. minimus. An introduced pathogenic fish trematode, Parabucephalopsis parasiluri Wang, 1985 (Digenea: Bucephalidae), was studied to determine the seasonal fluctuation in its metacercariae abundance. The timing of the recruitment period of the metacercariae and the relationship between yearly fluctuation in metacercarial abundance and some environmental factors were also examined to develop an effective plan for disease control. Monthly changes in metacercarial abundance in the caudal fin of pale chub ( Zacco platypus) and of the prevalence of the first intermediate host (golden mussel Limnoperna fortunei) suggest that P. parasiluri generally has an annual life cycle. The metacercarial abundance in chub increased when the water temperature fell below 7°C in winter, but the rapidity of the increase varied between 2007 and 2008. The fluctuations in the abundance of metacercariae in winter from 2001 to 2008 showed that mean flow rate and mean water level in January were negatively related to the infection level each year, and positively related to the mean water temperature. Based on these results, we propose a control plan for parabucephalopsiosis involving discharge control; that is, more than 50 m3/s (corresponding to 150 m3/s in the Yodo River) of water should be flushed from upstream dams for a period of at least one week starting on the day when the water temperature falls below 7°C. Three arbitrary primers produced 114 RAPD markers for 37 cercariae from nine Bucephalus polymorphus sporocysts obtained from three Dreissena polymorpha mollusks, which were collected in two water reservoirs of the Volga basin. Analysis of the RAPD patterns established a unique genotype for each cercaria. The topology of an UPGMA dendrogram did not reliably differentiate the cercaria according to the corresponding sporocysts. However, three groups of genotypes were isolated and corresponded to the host mollusks, indicating that each cercaria clone had a different genotype set. A within-sporocyst variation made the greatest contribution (53.0%) to the total RAPD diversity, while the contributions of within-host and between-host variations to the total diversity were equal (23.5%). Cercariae isolated from two mollusks of the Rybinskoe Water Reservoir were more similar to each other than to cercariae from the geographically distant Gor’kovskoe Water Reservoir. The article focuses on the Rhipidocotyle tridecapapillata which is drawn from the intestine of the white bass Morone chrysops from the Luxapalila River in Mississippi. It has 13 papillae that protude from the anterior rim of the dorsal lobe of the rynchus. Meanwhile, the Prosorhynchoides potamoenis is drawn from the pyloric ceca and intestine of the white crappie which has the anterior half of the body.

Two new species of bucephalids (Digenea: Bucephalidae) were found parasitizing freshwater fishes of Argentina. Prosorhynchoides cambapuntaensis sp. n. from Salminus maxillosus (Characidae) is characterised by the tegument covered with two kinds of spines, scale-like spines anteriorly and fine spines posteriorly; uterine loops reaching the vitelline arch in the pre-oral region; and the Laurer''s canal short, opening dorsally immediately posterior to the ovary. Rhipidocotyle santanaensis sp. n. from Acestrorhynchus pantaneiro (Acestrorhynchidae) is characterised by the rhynchus with two lateral projections; the testes arranged in tandem or slightly diagonal; and the vitelline follicles forming an arch in the pre-oral region. Nucleotide variation in a portion of the mitochondrial cytochrome c oxidase subunit1 (cox1) gene from asexual stages of bucephalids of southern Australian scallops (Chlamys asperrima, Chlamys bifrons and Pecten fumatus) was investigated using a mutation scanning–sequencing approach. Single-strand conformation polymorphism (SSCP) analysis revealed three main profile types (A, B and C) for parasites isolated from scallops. Sequence analysis revealed that samples represented by profiles B and C had a high degree (97.3%) of sequence similarity, whereas they were ∼21% different in sequence from those represented by profile A. These findings suggested that at least two types or species (represented by profile A, or profile B or C) of bucephalid infect scallops, of which both were detected in South Australia, while only one was found in Victoria. The prevalence of bucephalids (and their SSCP haplotypes) appeared to differ among the three species of scallop in South Australia as well as between the two scallop species in Victoria, indicating a degree of host specificity. Adult bucephalids were collected from Eastern Australian Salmon (Arripis trutta), in an attempt to match them with the asexual stages from the scallop hosts. Neither of the two taxa of adult bucephalid (Telorhynchus arripidis and an un-named Telorhynchus species) shared SSCP profiles with the bucephalids from scallops, but were genetically similar, suggesting that the asexual stages from scallops may represent the genus Telorhynchus. This study, which assessed nucleotide sequence variation in a portion of the mitochondrial cox1 gene for bucephalids found in scallops and arripid fish, illustrates the usefulness of the mutation scanning approach to elucidate complex life-cycles of marine parasites. This study reports the prevalence of Bucephalus sp. in Perna perna populations from a culture station of southern Brazil and its effect on the mussel reproductive tissue and immune system. The prevalence of Bucephalus sp. in P. perna (n=1871) was considered low (3.1%) and did not seasonally vary. Histological sections of the mantle of infected mussels revealed a marked (80%) reduction of the reproductive tissue that was severe even in mussels exhibiting a moderate infection degree. The total (THC) and differential (DHC) hemocyte counts were lower in infected mussels (3.9×106 hem/ml; granular hemocytes=33%) as compared with non-infected animals (5.5×106 hem/ml; granular hemocytes=40%). The plasma protein concentration did not vary upon infection. Hemocyte infiltration was significantly higher only in mussels with a very heavy infection degree. The parasite sporocysts were never seen encapsulated by the host hemocytes. Our results indicate that Bucephalus sp. promotes a severe castration in its host and apparently evades the mussel immune system. [Copyright &y& Elsevier]
