Práce je zaměřena na odstraňování dusičnanů z modelových odpadních vod metodou autotrofní denitrifikace bakteriemi Thiobacillus denitrificans. Jako elektronový donor je zde využita elementární síra. V teoretické části práce je popsán vliv zvýšených koncentrací dusičnanů ve vodách na různé složky ekosystému. Dále jsou shrnuty a porovnány metody odstraňování dusičnanů z vod, včetně metod biologických, kam patří heterotrofní a autotrofní denitrifikace. Stěžejní část je zaměřena na bakteriální kmen Thiobacillus denitrificans a mechanismy jeho působení při autotrofním čištění vod obsahujících dusičnany. Experimenty byly zaměřeny na systematické studium odstraňování dusičnanů autotrofní denitrifikací z uměle připravovaných odpadních vod, které jsou chudé na obsah organických látek a zároveň v nich koncentrace dusičnanových iontů nepřesahuje hodnoty 100 mg.l-1. První část experimentů se zabývala vsádkovým uspořádáním, kde byl k regulaci pH denitrifikačních procesů přidáván do reakčních směsí vápenec. Pozornost byla věnována studiu vlivu řady faktorů, jako je teplota, velikost použité frakce náplně, míchání, přídavek fosforu a trojmocného železa do reakční směsi. Druhá část experimentů byla zaměřena na dlouhodobé studium autotrofní denitrifikace v průtočné koloně, kde byla jako nehybná náplň použita pouze elementární síra. Kolona byla nepřetržitě provozována celkem 428 dní a byl do ní čerpán v různých fázích optimalizace procesu modelový pracovní roztok obsahující od 15 do 50 mg.l-1 dusičnanů. The second part of the experiments was focused on a long-term study of autotrophic denitrification in a continuous arrangement, i.e. flow column, where only elemental sulfur was used as a fixed bed. The column was operated continuously for a total of 428 days and a model working solution containing from 15 to 50 mg.l-1 of nitrates was pumped into it in various phases of process optimization. Degradation of the active pharmaceutical ingredient diclofenac (DCL) was studied using a laboratory-scale photoreactor equipped with highly efficient UV-A LEDs. Three photocatalysts (Aeroxide P25, Hombikat UV 100 and Precheza AV01) were tested and compared. In addition to deionized water, the other types of water (water matrices), were then tap water, unfiltered surface water, and the primary effluent of a wastewater from the municipal treatment plant. The experimental results indicated that the UV-A LED source operating at a wavelength of 368 nm had been much more efficient when combined with Precheza AV-01 photocatalyst of the microstructured anatase type. A negligible contribution of photolysis was noticed, while UV-A/TiO2 photocatalysis was more efficient in DCL removal in both the tap and the surface water matrices. The removal rate of DCL from the wastewater matrix was more dependent on the matrix itself than on the chemical structure of the micropollutant. The addition of hydrogen peroxide to the photocatalytic system enhanced the removal rates. The results of the algal growth inhibition test revealed a significant reduction of toxicity to Parachlorella kessleri after UV-A LED irradiation of the DCL solution. The comparative experimental study has been performed of removal of pharmaceuticals such as paracetamol and ibuprofen from aqueous solution by ozonation and photochemical degradation in several types of reactors. Ozone, oxygen, and hydrogen peroxide were used as oxidizing agents. Heterogeneous photocatalysis was carried out in the presence of anatase type TiO2 catalyst. To determine the amount of radiation emitted by the UV lamps and transferred into the reactor, chemical feroxioxalate actinometric experiments were performed. The degree of the pollutant removal was monitored by HPLC, COD, BOD5, and TOC. An ecotoxic biological test of process products with Parachlorella kessleri was also evaluated. The results indicate that the removal of both pharmaceuticals is most effective by heterogeneous photocatalysis. In the UV processes, the degradation rate increased considerably when H2O2 was added. However, the addition of peroxide into the ozonation system improved the ozonation only slightly. Při použití UV záření se míra degradace výrazně zvýšila přídavkem H2O2. Oproti tomu přidání H2O2 k ozonizaci vedlo jen k nepatrnému zvýšení účinnosti degradace. Všechny rozkladné produkty z testovaných procesů prokázaly lepší biologickou rozložitelnost na poměr BOD5 / COD než výchozí léčiva a nízkou toxicitu vůči Parachlorella kessleri.

These substances are very often found in outlets of WWTP or in surface waters, because among all classes of pharmaceuticals, they are frequently prescribed and purchased. These substances have by their nature a strong biological effect, so non-target organisms are affected. For example diclofenac causes cytological malformations of liver tissue within a concentration 28 μg/L (O. mykkis). Sensitive organisms to naproxen are crustaceans, where the EC50 for (C. dubia) is 330 μg/L, but degradation products of this drug are significantly more toxic. EC50 for degradation products of naproxen to C. dubia are less than 100 μg/L. Diclofenac and naproxen (500 μg/L) were from model water samples removed by heterogeneous photocatalysis in 4L batch reactor. After irradiation of the catalyst, the system produces •OH radicals, which causes destruction of the organic substance. As the catalyst used was 0.5 g/L titanium dioxide (TiO2, Precheza a.s.). As a source of UV radiation was used UV LED diode Luminus, with peak wavelength at 365 nm. At the same time the effect of hydrogen peroxide (H2O2) or effect of pH was determined. Samples were preconcentrated by solid phase extraction (SPE) and analyzed by high performance liquid chromatography (HPLC). Po ozáření katalyzátoru tento systém produkuje •OH radikály, které vedou k rozkladu organické hmoty. Jako katalyzátoru bylo použito 0,5 g/l oxidu titaničitého (TiO2, Precheza a.s.). Jako zdroj UV záření byla použita UV LED dioda Luminus, s emisním maximem při 365 nm. V souběžném experimentu čase byl testován účinek přídavku peroxidu vodíku (H2O2) nebo vliv pH. Vzorky reakční směsi před analýzou byly překoncentrovány extrakcí na pevné fázi (SPE) a analyzovány pomocí vysokoúčinnou kapalinovou chromatografií (HPLC). Také byly diskutovány charakteristiky a možnosti odstranění léčiv ze vstupu odpadní vody do ČOV.

Thiobacillus denitrificans patří mezi chemolitoautotrofní, fakultativně anaerobní bakterie, které získávají energii oxidací elementární síry a některých jejích dalších anorganických sloučenin za současného uvolňování elektronů, jejichž akceptorem se za anoxických podmínek stávají dusičnanové či dusitanové ionty. Dochází tak k redukci na plynný dusík a probíhá tzv. denitrifikace. Díky denitrifikačním schopnostem mohou být tyto organismy využity v procesech sloužících jako alternativní metody odstraňování dusičnanů z kontaminovaných vod. Pro správný chod denitrifikačních procesů je potřeba zajistit optimální nastavení nutričních podmínek pro bakteriální buňky. V diplomové práci je sledován vliv několika vybraných faktorů na rychlost a průběh autotrofní denitrifikace činností bakterií Thiobacillus denitrificans. Je prokázáno, že na denitrifikaci nemá žádný vliv sluneční světlo. Tato bakalářská práce se zabývá glycerolem, jeho vlastnostmi, metodami výroby, biochemickým významem a analytickými metodami jeho stanovení. Glycerol vzniká jako vedlejší produkt při výrobě mýdel, bionafty a vyšších mastných kyselin, také je možné ho vyrobit syntetickou a mikrobiologickou cestou. V práci je také popsáno čištění vyrobeného glycerolu, např. pomocí destilace nebo iontovou výměnou. Diplomová práce se zabývá problematikou interní komunikace na Městském úřadě v Bystřici pod Hostýnem. Teoretická část obsahuje charakteristiku vnitroorganizační komunikace a jednotlivých komunikačních nástrojů se zaměřením na instituce veřejné správy. Praktická část této práce obsahuje analýzy současného stavu interní komunikace na městském úřadě. Součástí analýz je i dotazníkové šetření mezi zaměstnanci.
