In this work, we examine the use of environment-sensitive fluorescent dyes in fluorescence lifetime imaging microscopy (FLIM) biosensors. We screened merocyanine dyes to find an optimal combination of environment-induced lifetime changes, photostability, and brightness at wavelengths suitable for live-cell imaging. FLIM was used to monitor a biosensor reporting conformational changes of endogenous Cdc42 in living cells. The ability to quantify activity using phasor analysis of a single fluorophore (e.g., rather than ratio imaging) eliminated potential artifacts. We leveraged these properties to determine specific concentrations of activated Cdc42 across the cell.

The neurotransmitter serotonin plays a pivotal role in mood and depression. It also acts as a vasoconstrictor within blood vessels and is the main neurotransmitter in the gastrointestinal system. In neurotransmission, released serotonin is taken up by serotonin transporters, which are principal targets of antidepressants and the psychostimulant, ecstasy. The investigation of serotonin transporters have relied almost exclusively on the use of radiolabeled serotonin in heterogenous end-point assays. Here we adapt the genetically encoded fluorescent biosensor, iSeroSnFR, to establish and validate the Serotonin (5-HT) Fluorescence Assay for Transport and Release (5-HT_FAsTR) for functional and pharmacological studies of serotonin transport and release. We demonstrate the applicability of the method for the study of a neuronal, high-affinity, low-capacity serotonin transporter (SERT) as well as an extraneuronal low-affinity, high-capacity organic cation transporter and mutants thereof. 5HT_FAsTR offers an accessible, versatile and reliable semi-homogenous assay format that only relies on a fluorescence plate reader for repeated, real-time measurements of serotonin influx and efflux. 5HT_FAsTR accelerates and democratizes functional characterization and pharmacological studies of serotonin transporters and genetic variants thereof in disease states such as depression, anxiety and ADHD.

Exosomes are critical tumor biomarkers, which can be detected by label‐free sensing of plasmonic metasurfaces with expensive antibody functionalization. Due to their low cost and small size, aptamers should be promising to replace antibodies in plasmonic metasensing of exosomes. However, research on aptamer‐based plasmonic metasensing of exosomes is still unexplored. Here, the optical near‐field sensing mechanism of aptamer‐based plasmonic metasurface (APM) is investigated. Based on the study of the evanescent field of APM, using thiol‐modified aptamer boosts the utilization rate of resonance wavelength shift from 27.5% to 83.3% compared to biotinylated aptamers, attributed to a significant reduction in the near‐field occupation of the biofunction layer. Moreover, APM biosensors effectively detect clinical serum exosomes for breast cancer (BC) diagnosis and classification. The receiver operating characteristic analysis shows an area under the curve of 99.3% for APM diagnosis, surpassing the 79.1% for hematological tests of the conventional BC biomarker CA153. The results also indicate that APM biosensors can be a noninvasive tool for molecular classification of BC with great distinguishment of P < 0.0001. The research demonstrates that APM biosensors are enabling a reliable and convenient platform to facilitate clinical hematological tests of exosomes for auxiliary diagnosis of cancer. In food safety analysis, the detection and control of foodborne pathogens and their toxins are of great importance. Monitoring of virus transmission is equally important, especially in light of recent findings that coronaviruses have been detected in frozen foods and packages during the current global epidemic of coronavirus disease 2019. In recent years, field-effect transistor (FET) biosensors have attracted considerable scholarly attention for pathogenic microorganisms and toxins detection and sensing due to their rapid response time, high sensitivity, wide dynamic range, high specificity, label-free detection, portability, and cost-effectiveness. FET-based biosensors can be modified with specific recognition elements, thus providing real-time qualitative and semiquantitative analysis. Furthermore, with advances in nanotechnology and device design, various high-performance nanomaterials are gradually applied in the detection of FET-based biosensors. In this article, we review specific detection in different biological recognition elements are immobilized on FET biosensors for the detection of pathogenic microorganisms and toxins, and we also discuss nonspecific detection by FET biosensors. In addition, there are still unresolved challenges in the development and application of FET biosensors for achieving efficient, multiplexed, in situ detection of pathogenic microorganisms and toxins. Micro-opto-electro-mechanical systems (MOEMS) biosensors are employed in various applications such as disease monitoring, drug investigation, detection of pollutants, and biological fluid studies. In this paper, a novel MOEMS biosensor based on a differential folded-flexure structure is introduced. The designed device is employed to detect prostate-specific antigen (PSA) protein and Hepatitis DNA. The target molecules cause a mechanical deflection in the folded-flexure; subsequently, the transmitted optical power across the finger, attached to the flexure, is modulated in proportion to the input concentration. Then, a photodiode power sensor measures the modulated optical power, where the output of the sensor is simply a current related to the target molecules' concentrations. The employed readout circuit operates at a wavelength of λ = 1550 nm with a laser power of 1 µW. The dimensions of the proposed biosensor are considered to be 365 × 340 × 2 μm³, making this sensor small enough and suitable for integration. The designed biosensor provides notable features of mechanical deflection sensitivities of 0.2053 nm/(ng/ml) and 7.2486 nm/nM, optical transmittance sensitivities of 0.535504 × 10−3 1/(ng/ml) and 18.91 × 10−3 1/nM, total output sensitivities of 0.5398 (mA/W)/(ng/ml) and 19.059 (mA/W)/nM, and measurement ranges of 0-1000 ng/ml and 0-28.33 nM for PSA and Hepatitis DNA, respectively. Summary: Gelatine is obtained from the hydrolysis of collagen in bones, skin, fins, fish scales and insect bodies of cattle, pigs, camels, poultry, fish and insects. Consequently, the diversity and availability of gelatine sources, along with gelatine authentication, are concerns for those consuming gelatine products from religious, health and economic perspectives. Therefore, rapid and reliable protein‐ or DNA‐based detection methods have been developed for gelatine authentication. This review discusses and evaluates biosensor methods, comparing them with various analytical methods in terms of their popularity and precision. Additionally, the development of biosensor methods has been explored to identify a reference for a rapid, accurate, portable and simple method for gelatine detection with a low detection limit and high specificity. Actinobacteria cells Rhodococcus erythropolis 4-1 and Rhodococcus erythropolis 11-2 and Proteobacteria Alcaligenes faecalis 2, which have amidase activity, were immobilized by entrapping barium alginate and agarose into the gel structure, as well as by obtaining biofilms on thermally expanded graphite (TEG). The operational stability of such immobilized biocatalysts after storage in frozen and dehydrated form was determined, and a prototype of a conductometric acrylamide biosensor based on such a bioselective agent was developed. The most preferred method for storing immobilized cells was freezing at temperatures from –20 to –80°C; long-term storage is also possible wet at 4–25°C. It was shown that these cells were most preferable for the biodetection of acrylamide A. faecalis 2, immobilized in an agarose gel structure. An agarose gel with bacterial cells immobilized in its structure had greater mechanical strength and stability during successive cycles of conversion of acrylamide into acrylic acid compared to barium alginate gel. The mechanical strength of barium alginate gel can be enhanced by the addition of carbon nanomaterials during cell immobilization. Growing biofilms on carbon materials used for manufacturing electrodes is also promising. Biofilms of R. erythropolis 11-2 on TEG are capable of converting acrylamide into acrylic acid in more than 20 reaction cycles while maintaining at least 50% amidase activity. [ABSTRACT FROM AUTHOR]

Nanozymes are a new kind of material which has been applied since the beginning of this century, and its birth has promoted the development of chemistry, materials science, and biology. Nanozymes can be used as a substitute for natural enzyme and has a wide range of applications; therefore, it has attracted extensive attention from all sectors of the community, and the number of studies has constantly increasing. In this paper, we introduced the outstanding achievements in the field of nanozymes in recent years from the main function, the construction of nanozyme-based biosensors, and the treatment of ischemic stroke, and we also illustrated the internal mechanism and the catalytic principle. This study developed an innovative biosensor strategy for the sensitive and selective detection of canine mammary tumor biomarkers, cancer antigen 15–3 (CA 15–3) and mucin 1 (MUC-1), integrating green silver nanoparticles (GAgNPs) with machine learning (ML) algorithms to achieve high diagnostic accuracy and potential for noninvasive early detection. The GAgNPs-enhanced electrochemical biosensor demonstrated selective detection of CA 15–3 in serum and MUC-1 in tissue homogenates, with limits of detection (LODs) of 0.07 and 0.11 U mL−1, respectively. The nanoscale dimensions of the GAgNPs endowed them with electrochemically active surface areas, facilitating sensitive biomarker detection. Experimental studies targeted CA 15–3 and MUC-1 biomarkers in clinical samples, and the biosensor exhibited ease of use and good selectivity. Furthermore, ML algorithms were employed to analyze the electrochemical data and predict biomarker concentrations, enhancing the diagnostic accuracy. The Random Forest algorithm achieved 98% accuracy in tumor presence prediction, while an Artificial Neural Network attained 76% accuracy in CA 15–3-based tumor grade classification. The integration of ML techniques with the GAgNPs-based biosensor offers a promising approach for noninvasive, accurate, and early detection of canine mammary tumors, potentially revolutionizing veterinary diagnostics. As widely used commercial sensors, amperometric biosensors based on electrochemical (EC) reactions exhibit good linearity and reproducibility but suffer from a low dynamic range and poor limit of detection (LoD). In contrast, biosensors utilizing field‐effect transistors (FETs) demonstrate a wide detection range and ultralow LoDs but are susceptible to interferences or ionic strength influences and have poor quantitative accuracy due to their nonlinear response mechanism. Here, EC and carbon nanotube (CNT) FET‐based biosensors are integrated into one chip via a compatible fabrication process. The dual‐mode biosensing chip (DMSC) provides the advantages from both the EC biosensors and FET biosensors; these advantages include high reliability and accuracy for quantitatively detecting glucose, a dynamic detection range spanning 6 orders of magnitude, and a detection limit of 1 fM. The introduction of a time‐division multiplexing detection method enables the DMSC to carry out complementary detection of the same target, providing early warning of specific abnormal biomarkers and accurate determination of their concentration in real time. The DMSC approach is generally applicable to the detection of other biomarkers and provides exciting opportunities for more accurate self‐monitoring diagnostics in real situations. Reactive oxygen species (ROS) including the superoxide anion (O2•−) are typically studied in cell cultures using fluorescent dyes, which provide only discrete single-point measurements. These methods lack the capabilities for assessing O2•− kinetics and release in a quantitative manner over long monitoring times. Herein, we present the fabrication and application of an electrochemical biosensor that enables real-time continuous monitoring of O2•− release in cell cultures for extended periods (> 8 h) using an O2•− specific microelectrode. To achieve the sensitivity and selectivity requirements for cellular sensing, we developed a biohybrid system consisting of superoxide dismutase (SOD) and Ti3C2Tx MXenes, deposited on a gold microwire electrode (AuME) as O2•− specific materials with catalytic amplification through the synergistic action of the enzyme and the biomimetic MXenes-based structure. The biosensor demonstrated a sensitivity of 18.35 nA/μM with a linear range from 147 to 930 nM in a cell culture medium. To demonstrate its robustness and practicality, we applied the biosensor to monitor O2•− levels in human leukemia monocytic THP-1 cells upon stimulation with lipopolysaccharide (LPS). Using this strategy, we successfully monitored LPS-induced O2•− in THP-1 cells, as well as the quenching effect induced by the ROS scavenger N-acetyl-l-cysteine (NAC). The biosensor is generally useful for exploring the role of oxidative stress and longitudinally monitoring O2•− release in cell cultures, enabling studies of biochemical processes and associated oxidative stress mechanisms in cellular and other biological environments. l‐2‐Hydroxyglutarate (l‐2‐HG) is a functionally compartmentalized metabolite involved in various physiological processes. However, its subcellular distribution and mitochondrial transport remain unclear owing to technical limitations. In the present study, an ultrasensitive l‐2‐HG biosensor, sfLHGFRH, composed of circularly permuted yellow fluorescent protein and l‐2‐HG‐specific transcriptional regulator, is developed. The ability of sfLHGFRH to be used for analyzing l‐2‐HG metabolism is first determined in human embryonic kidney cells (HEK293FT) and macrophages. Then, the subcellular distribution of l‐2‐HG in HEK293FT cells and the lower abundance of mitochondrial l‐2‐HG are identified by the sfLHGFRH‐supported spatiotemporal l‐2‐HG monitoring. Finally, the role of the l‐glutamate transporter SLC1A1 in mitochondrial l‐2‐HG uptake is elucidated using sfLHGFRH. Based on the design of sfLHGFRH, another highly sensitive biosensor with a low limit of detection, sfLHGFRL, is developed for the point‐of‐care diagnosis of l‐2‐HG‐related diseases. Food contamination has emerged as a significant global health concern, posing substantial challenges to the food industry. Bacteria are the primary cause of foodborne diseases. Consequently, it is crucial to develop accurate and efficient sensing platforms to detect foodborne bacteria in food products. Among various detection methods, biosensors have emerged as a promising solution due to their portability, affordability, simplicity, selectivity, sensitivity, and rapidity. Electrospun nanofibers have gained increasing popularity in enhancing biosensor performance. These nanofibers possess a distinctive three-dimensional structure, providing a large surface area and ease of preparation. This review provides an overview of the electrospinning technique, nanofibers and nanofiber-based biosensors. It also explores their mechanisms and applications in the detection of foodborne bacteria such as Salmonella, Listeria monocytogenes (L. monocytogenes), Escherichia coli (E. coli), Staphylococcus aureus (S. aureus) and Pseudomonas putida (P. putida). Breast cancer stands as the most prevalent form of cancer amongst females, constituting more than one-third of all cancer cases affecting women. It causes aberrant cell development, which can assault or spread to other sections of the body, perhaps leading to the patient's death. Based on research findings, timely detection can diminish the likelihood of mortality and enhance the quality of healthcare provided for the illness. However, current technologies can only identify cancer at an advanced stage. Consequently, there is a substantial demand for rapid and productive approaches to detecting breast cancer. Researchers are actively pursuing precise and timely methods for the diagnosis of breast cancer, aiming to achieve enhanced accuracy and early detection. Biosensor technology can allow for the speedy and accurate diagnosis of cancer-related cells, as well as a more sensitive and specialized technique for generating them. Additionally, numerous treatments for breast cancer are depicted such as herbal therapy, nanomaterial-based drug delivery, miRNA targeting, CRISPR technology, immunotherapy, and precision medicine. Catalytic hairpin assembly (CHA) is an efficient nucleic acid amplification method that does not depend on enzyme biocatalysis and temperature cycling, and is characterized by good biocompatibility, simple operation, and mild reaction conditions. By combining CHA with various signal output modes, highly sensitive detection of various biological targets (nucleic acids, small molecules, proteins, cells) can be realized, which shows great potential for application in disease diagnosis. Therefore, this paper focuses on classical CHA, modified CHA, and several cascade reactions based on the CHA strategy with other isothermal enzyme-free amplification methods, and briefly reviews their applications in disease diagnosis. AbstractPathogen detection is increasingly applied in medical diagnosis, food processing and safety, and environmental monitoring. Rapid, sensitive, and accurate pathogen quantification is the most critical prerequisite for assessing protocols and preventing risks. Among various methods evolved, those based on clustered regularly interspaced short palindromic repeats (CRISPR)-associated proteins (Cas) have been developed as important pathogen detection strategies due to their distinct advantages of rapid target recognition, programmability, ultra-specificity, and potential for scalability of point-of-care testing (POCT). However, arguments and concerns on the quantitative capability of CRISPR-based strategies are ongoing. Herein, we systematically overview CRISPR-based pathogen quantification strategies according to the principles, properties, and application scenarios. Notably, we review future challenges and perspectives to address the of precise pathogen quantification by CRISPR-Cas. A Ca2+ sensor based on DNA nanospheres was developed. The DNA nanospheres were obtained by complementary pairing of Y-shaped DNA and DNAzyme, in which Y-shaped DNA is a triple-branched DNA with three sticky ends formed by three partially complementary single-stranded DNAs. DNAzyme is a recently reported RNA-cleaving DNAzyme, EtNa, that has high selectivity for Ca2+ and was used as linker in DNA nanosphere preparation. When EtNa DNAzyme was formed by enzyme chain (E) and substrate chain (S), the fluorescence signal was quenched significantly due to the proximity of fluorophore and quencher. In the presence of Ca2+, it bound to the E strand of EtNa DNAzyme, catalyzing the cleavage of S strand, which led to the separation of S from E and then the DNA nanosphere was disintegrated. At the same time, the fluorescence signal was restored and Ca2+ assay was realized. This detection system based on DNA nanospheres determined Ca2+ across a linear range from 20 to 400 μM with a detection limit of 1.6 μM and excellent selectivity. In addition, this proposed Ca2+ assay has been successfully applied to the assay of Ca2+ in human serum, thus demonstrating clinical application. [ABSTRACT FROM AUTHOR]

Surface plasmon resonance (SPR) is a key technique in developing sensor platforms for clinical diagnostics, drug discovery, food quality, and environmental monitoring applications. While prism‐coupled (Kretschmann) SPR remains a “gold‐standard” for laboratory work‐flows due to easier fabrication, handling and high through put, other configurations such as grating‐coupled SPR (GC‐SPR) and wave‐guide mode SPR are yet to fulfil their technology potential. This work evaluates the technical aspects influencing the performance of GC‐SPR and reviews recent progress in the fabrication of such platforms. In principle, the GC‐SPR involves the illumination of the plasmonic metal film with periodic gratings to excite the surface plasmons (SP) via diffraction‐based phase matching. The real performance of the GC‐SPR is, however, heavily influenced by the topography of the grating structures produced via top‐down lithography techniques. This review discusses latest in approaches to achieve consistent plasmonic gratings with uniform features and periodicity over a large scale and explores the choice of plasmon‐active and substrate material for enhanced performance. The review also provides insights into the different GC‐SPR measurement configurations and highlights on opportunities with their potential applications as biosensors with translational capabilities. Stimuli‐responsive polymers have received increasing attention for various applications due to their ability to adapt physical and chemical properties in response to external environmental stimuli. In this regard, poly(N‐isopropylacrylamide) (PNIPAM) is the most extensively studied stimuli‐responsive polymer and, consequently has been prominently featured in (bio)‐sensor development, adaptive coating technology, drug delivery, wound healing, tissue regeneration, artificial actuator design, sensor technology, responsive coatings, and soft robotics. This success can be mainly attributed to the accessible and versatile nature of the PNIPAM platform, thus allowing the synthesis of a wide variety of copolymer architectures, topologies and compositions. Within this review, the structural and compositional features of PNIPAM‐based materials in sensor and biosensor applications are discussed with a focus on the literature from 2016 until now. The reader is provided with the current state of the art regarding PNIPAM‐based sensor development and their molecular design. Finally, the challenges ahead in the successful implementation of PNIPAM‐based sensors are highlighted, as well as the opportunities in the rational design of improved PNIPAM‐based sensors. Altogether, this review provides comprehensive insights into the exciting and rapidly expanding field of PNIPAM‐based sensing systems, which will benefit the chemical, pharmaceutical, textile, and biotech industries is believed. Soil microbiome characterization is typically achieved with next-generation sequencing (NGS) techniques. Metabarcoding is very common, and meta-omics is growing in popularity. These techniques have been instrumental in microbiology, but they have limitations. They require extensive time, funding, expertise, and computing power to be effective. Moreover, these techniques are restricted to controlled laboratory conditions; they are not applicable in field settings, nor can they rapidly generate data. This hinders using NGS as an environmental monitoring tool or an in-situ checking device. Biosensing technology can be applied to soil microbiome characterization to overcome these limitations and to complement NGS techniques. Biosensing has been used in biomedical applications for decades, and many successful commercial products are on the market. Given its previous success, biosensing has much to offer soil microbiome characterization. There is a great variety of biosensors and biosensing techniques, and a few in particular are better suited for soil field studies. Aptamers are more stable than enzymes or antibodies and are more ready for field-use biosensors. Given that any microbiome is complex, a multiplex sensor will be needed, and with large, complicated datasets, machine learning might benefit these analyses. If the signals from the biosensors are optical, a smartphone can be used as a portable optical reader and potential data-analyzing device. Biosensing is a rich field that couples engineering and biology, and applying its toolset to help advance soil microbiome characterization would be a boon to microbiology more broadly. Nature inspires technological innovation through unique micronanostructured surfaces, such as fish fins, lotus leaves, butterfly wings, rose petals, a bird's spongy bone, etc. These structures exhibit capabilities beyond conventional engineering, making biomimetics a key focus of scientific research. Integrating nanotechnology, biology, and bioengineering has significantly propelled the development of nanomaterials with distinct functions and properties. This interdisciplinary synergy has extensively advanced biomimetic nanomaterials. This review highlights biomimetic nanomaterials, with an emphasis on state-of-the-art sensing devices. In addition to this, it furnishes an extensive compendium of investigations into synthesis and fabrication of biomimetic nanomaterials, such as metal–organic frameworks (MOFs), molecularly imprinted polymers (MIPs), carbon-based biomimetic nanomaterials (graphene, carbon nanotubes), and gold nanoparticles (AuNPs), for biosensor development. We have focused on their applications in disease diagnostics within healthcare while also addressing their role in environmental monitoring and agriculture. This comprehensive review aims to impart an insightful understanding of the scientific complexities associated with these technologies and concludes with an appraisal of current challenges and future developments. Optical sensors face challenges when detecting ultralow amounts of analytes in whole blood, including signal quenching due to optical absorption and false positives due to nonspecific binding. This study introduces gold nanoscale array features termed nanoledges (NLs), which interact with incident white light to produce a transmitted surface plasmon resonance (tSPR) signal. This extraordinary optical transmission (EOT) spectrum occurs in the near-infrared (NIR) region, thereby minimizing signal quenching caused by visible-light absorption from blood proteins and pigments. To develop a sensitive, selective, and label-free optical biosensor for detecting various levels of cardiac troponin I (cTnI) in very small volumes of whole blood samples, DNA aptamers are tethered to the NL surface, specifically binding to the cTnI biomarker. This biological binding activity alters the refractive index at the NL surface, causing a peak shift in the EOT spectrum and enabling quantification of cTnI levels. The NL array chip demonstrated high sensitivity for cTnI detection in buffer, human serum (HS), and human whole blood (HB), with detection limits of 0.079, 0.084, and 0.097 ng/mL, respectively. Control measurements using blank target mediums and those containing up to 125 ng/mL of other proteins, such as myoglobin, creatine kinase, and heparin, showed minimal interference and high specificity. Recently, the concept of introducing disorder into ordered metasurfaces or periodic metastructures has shown great potential in improving their performance for light extraction, scattering, reflection, and radiation. However, its use in optical biosensing enhancement is still barely reported. Here, a kind of plasmonic biosensors based on disorder‐enhanced nanograin metasurfaces (DENMs) are proposed, and utilized for high‐sensitivity detection of SARS‐CoV‐2 Omicron variant and vaccine‐induced total antibodies. With the aim to elucidate the physics of short‐range‐disordered meta‐elements in long‐range‐ordered metastructures, the meta‐atom evolution is deduced from periodic nanohole metasurfaces to DENMs and totally disordered nanograin metasurfaces. It is found that the disorder of nanograin plays a critical role in elevating the DENM surface sensitivity of biomolecules. The DENM‐based biosensing demonstrates an extremely high diagnostic specificity with the probability P < 0.0001 on distinguishing the Omicron variant from other respiratory viruses. Moreover, these biosensors are used as a convenient tool to monitor vaccine efficacy for inoculators with the third booster injections. This study implies the promise of disorder‐enhanced metasurfaces on biomedical detection and will guide their applications on virus early discovery and prevention for future mobile healthcare. This study presents a terahertz (THz) resonance gas sensor design incorporating graphene, black phosphorus, and MXene materials in a metasurface structure. The sensor leverages the unique properties of these advanced two-dimensional materials to achieve enhanced sensitivity and versatility in gas detection applications. The proposed design consists of elliptical and square-shaped resonators arranged on a SiO2 substrate with a ground plane back reflector. Comprehensive simulations using COMSOL Multiphysics were conducted to analyze the sensor's performance across various structural parameters and operating conditions. The sensor demonstrates a maximum sensitivity of 400 GHzRIU−1 and a figure of merit up to 0.816 RIU−1 within a refractive index range of 1–1.07 RIU. Electric field distribution analysis validates the sensor's transmittance response at different frequencies. Notably, the design shows potential for 2-bit encoding applications based on transmittance characteristics under varying graphene chemical potential values. Compared to existing studies, the senso's performance is particularly better in terms of sensitivity, offering advantages such as room temperature operation and fast response times. In this paper, an optical refractive index sensor based on a hybrid plasmonic-photonic crystal (P-PhC) topology for detection of various blood components is proposed. The sensor's structure is composed of a gold spiral resonator inside a rod-type hexagonal Si PhC cavity. The finite-difference time-domain method is used for the numerical investigation of the structures. The proposed hybrid sensor provides an astonishing figure of merit (FoM) of 2991 RIU-1 and fairly high sensitivity of 1675 nm/RIU. The maximum FoM achieved in this case is found to be 5 times higher than the conventional plasmonic sensors reported in the literature. For the proposed sensor, combining the stronger plasmonic light-matter interaction and the extraordinary confining properties of the PhC cavity contributes to the FoM enhancement. The development of biosensors for determining the most diverse biomolecules is a constant focus of many research groups. There is a latent need to propose sensors that combine portability, simple measurements, and good analytical performance. Here, we propose an electrochemical immunosensor that is fully portable and energy-independent for diagnosing antibodies against SARS-CoV-2 (the virus that causes COVID-19). Initially, disposable screen-printed carbon electrodes (SPEs) were covered by gold microblobs (AuMBs), which were synthesized amperometrically from Au3+ ions. Then, the SPE-AuMBs were coated with cysteamine, which allowed the N-hydroxysuccinimide-activated SARS-CoV-2 antigen (spike protein) to be immobilized. The antigen-activated electrode was used to detect COVID-19 antibodies from current measurements obtained by differential pulse voltammetry. The AuMBs synthesis time was optimized, and the presence of gold structures improved the electrochemical responses of the SPE. It was possible to quantitatively determine antibodies in the concentration range of 0.25 to 10 µg mL−1. This range includes concentrations found in biological fluids from patients at any stage of the disease. An analysis took approximately the same time as traditional rapid nasal tests (20 min) and costed less, considering all the steps necessary to prepare a disposable antigen-functionalized SPE. Optical biosensors have emerged as a powerful tool in analytical biochemistry, offering high sensitivity and specificity in the detection of various biomolecules. This article explores the advancements in the integration of optical biosensors with microfluidic technologies, creating lab-on-a-chip (LOC) platforms that enable rapid, efficient, and miniaturized analysis at the point of need. These LOC platforms leverage optical phenomena such as chemiluminescence and electrochemiluminescence to achieve real-time detection and quantification of analytes, making them ideal for applications in medical diagnostics, environmental monitoring, and food safety. Various optical detectors used for detecting chemiluminescence are reviewed, including single-point detectors such as photomultiplier tubes (PMT) and avalanche photodiodes (APD), and pixelated detectors such as charge-coupled devices (CCD) and complementary metal–oxide–semiconductor (CMOS) sensors. A significant advancement discussed in this review is the integration of optical biosensors with pixelated image sensors, particularly CMOS image sensors. These sensors provide numerous advantages over traditional single-point detectors, including high-resolution imaging, spatially resolved measurements, and the ability to simultaneously detect multiple analytes. Their compact size, low power consumption, and cost-effectiveness further enhance their suitability for portable and point-of-care diagnostic devices. In the future, the integration of machine learning algorithms with these technologies promises to enhance data analysis and interpretation, driving the development of more sophisticated, efficient, and accessible diagnostic tools for diverse applications. A highly selective biosensor for glycine betaine (GB) based on the curcumin derivative (1BHD) is described, which displays a significant visual change and a lower detection limit (76 nM). The GB sensor displays a 1:1 binding ratio and a binding constant of 9.042 × 107 M−1. The binding mechanism of 1BHD for glycine betaine was confirmed by 1H NMR titration, mass spectrometry, and DFT calculation. The fluorescence intensity of 1BHD was selectively quenched by GB, and an obvious color change from yellow to orange was observed by the naked eye. Using a chemiluminescence reaction between luminol and H2O2 in basic solution, an ultrasensitive electrochemiluminescence (ECL) aptasensor was developed for the determination of tobramycin (TOB), as an aminoglycoside antibiotic. Ti3C2/Ni/Sm-LDH-based nanocomposite effectively catalyzes the oxidation of luminol and decomposition of H2O2, leading to the formation of different reactive oxygen species (ROSs), thus amplifying the ECL signal intensity of luminol, which can be used for the determination of TOB concentration. To evaluate the performance of the electrochemiluminescence aptasensor and synthesized nanocomposite, different methods such as cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) analyses were performed. The considerable specific area, large number of active sites, and enhanced electron transfer reaction on this nanocomposite led to the development of an ECL aptasensor with high sensitivity and electrocatalytic activity. After optimizing the preparation method and analysis conditions, the aptasensor revealed a wide linear response ranging from 1.0 pM to 1.0 μM with a detection limit of 18 pM, displaying outstanding accuracy, specificity, and response stability. The developed ECL sensor was found to be applicable to the determination of TOB in human serum samples and is anticipated to possess excellent clinical potentials for detecting other antibiotics, as well. A new analytical method, based on SPRi biosensors, has been developed for the simultaneous determination of the pro-angiogenic factors HIF-1α, angiopoietin-2 (ANG-2), and interleukin-1β (IL-1β) in biological fluids. These proteins take part in the process of angiogenesis, i.e., the creation of new blood vessels, which is a key stage of cancer development and metastasis. A separate validation process was carried out for each individual compound, indicating that the method can also be used to study one selected protein. Low values of the limit of detection (LOD) and quantification (LOQ) indicate that the developed method enables the determination of very low concentrations, in the order of pg/mL. The LOD values obtained for HIF-1α, ANG-2, and IL-1β were 0.09, 0.01, and 0.01 pg/mL, respectively. The LOQ values were 0.27, 0.039, and 0.02 pg/mL, and the response ranges of the biosensor were 5.00–100.00, 1.00–20.00, and 1.00–15.00 pg/mL. Moreover, determining the appropriate validation parameters confirmed that the design offers high precision, accuracy, and sensitivity. To prove the usefulness of the biosensor in practice, determinations were made in plasma samples from a control group and from a study group consisting of patients with diagnosed bladder cancer. The preliminary results obtained indicate that this biosensor can be used for broader analyses of bladder cancer. Each of the potential biomarkers, HIF-1α, ANG-2, and IL-1β, produced higher concentrations in the study group than in the control group. These are preliminary studies that serve to develop hypotheses, and their confirmation requires the analysis of a larger number of samples. However, the constructed biosensor is characterized by its ease and speed of measurement, and the method does not require special preparation of samples. SPRi biosensors can be used as a sensitive and highly selective method for determining potential blood biomarkers, which in the future may become part of the routine diagnosis of cancers. Nile tilapia Oreochromis niloticus is an important aquaculture species worldwide, and understanding its behavior and growth performance under variable culture conditions is essential for optimizing culture. Light as an environmental factor profoundly affects fish development, growth, food consumption, hormone secretion, and other life processes. This study investigated the effects of different light colors (blue, green, and red) on the stress response of individually reared Nile tilapia using a wireless biosensor system for real-time glucose monitoring. The biosensor system effectively measured glucose concentration (stress indicator) by enzymatic oxidation, allowing continuous monitoring of fish eyeball interstitial sclera fluid (EISF), providing longer and more accurate data. Air exposure and ammonia exposure were used as stressors, and stress responses were monitored under different light colors. Nile tilapia exhibited lower quiescent blood glucose levels, lower blood glucose elevations, and faster stress recovery under blue light. Green light induced more significant glucose elevations during acute physical stress, but showed similar fluctuations to blue light during chronic chemical stress, indicating potential benefits for fish welfare. In contrast, whether exposed to air or ammonia, red light induced large fluctuations in blood glucose levels, which may be detrimental to the fish. The study highlights the importance of light color in influencing stress responses and recovery in Nile tilapia, providing valuable insights for improving aquaculture practices and promoting fish welfare. MXene is a rapidly emerging group of two-dimensional (2D) multifunctional nanomaterials, drawing huge attention from researchers of a broad scientific field. Reporting the synthesis of MXene was the following breakthrough in 2D materials following the discovery of graphene. MXene is considered the most recent developments of materials, including transition metal carbonitrides, nitrides, and carbides synthesized by etching or mechanical-based exfoliation of selective MAX phases. MXene has a plethora of prodigious properties such as unique interlayer spacing, high ion and electron transport, large surface area, excellent thermal and electrical conductivity, exceptional volumetric capacitance, thermal shock, and oxidation resistance, easily machinable and inherently hydrophilic, and biocompatibility. Owing to the abundance of tailorable surface function groups, these properties can be further enhanced by surface functionalization with covalent and non-covalent modifications via numerous surface functionalization methods. Therefore, MXene finds their way to a plethora of applications in numerous fields including catalysis, membrane separation, energy storage, sensing, and biomedicine. Here, the focus is on reviewing the structure, synthesis techniques, and functionalization methods of MXene. Furthermore, MXene-based detection platforms in different sensing applications are survived. Great attention is given to reviewing the applications of MXene in the detection of biomolecules, pathogenic bacteria and viruses, cancer biomarkers food contaminants and mycotoxins, and hazardous pollutants. Lastly, the future perspective of MXene-based biosensors as a next-generation diagnostics tool is discussed. This study's main focus was on the process by which Ciclopirox adhered to AlN nanostructures, Calculations using the density functional theory were done to reach this goal. The calculations encompassed the assessment of Ciclopirox's adsorption energy, the evaluation of the energy band gap, the analysis of variations in the energy band gap, the examination of charge transfers, and the characterisation of the types of interactions that arise from Ciclopirox's adsorption onto AlN nanostructures. We carried out an investigation using the AIM method to explore deeper into the binding properties between the studied AlN nanostructures and Ciclopirox. Our discoveries provide light on the Ciclopirox/AlN nanosheet interaction's electrostatic characteristics. Moreover, the presence of Ciclopirox led to an augmentation in the electrical conductivity of the AlN nanostructures. This intriguing outcome suggests the potential application of these AlN nanomaterials as chemical sensors, capable of generating an electronic signal upon detection of this chemically modified amino acid. The observed sensitivity sequence indicated that the AlN nanosheet exhibited the highest sensitivity, followed by the AlN nanotube and the AlN nanocluster. Modifying electrical and optical properties in two-carbon materials is essential for electronic devices, energy storage and biosensor applications. We report a comparative study of the effect of systematically intercalated strontium atoms in AA- and AB-stacked bilayer graphene (BG) in the framework of density functional theory (DFT). The electronic band structures, the total and partial electronic density of states and the dielectric function are studied. We show that the intercalation of atoms can influence both electronic properties. Our results show that the Dirac cones, a signature of graphene, are translated into the conduction band. This property may be useful in band-gap tuning applications of nanodevices. The optical properties were calculated parallel and perpendicular to the graphene sheet under polarised electric fields. The calculated optical properties show that the intercalation of Sr in the AA- and AB-stacked BGs promotes IR absorption. The findings provide a basis for developing graphene intercalary compounds, which can be used in optoelectronics to control the absorption of light wavelengths and specifically in biosensor applications. The advantages of rapid amplification of nucleic acid without a template based on terminal deoxyribonucleotidyl transferase (TdT) have been widely used in the field of biosensors. However, the catalytic efficiency of TdT is affected by extension conditions. The sensitivity of TdT- mediated biosensors can be improved only under appropriate conditions. Therefore, in this review, we provide a comprehensive overview of TdT extension characteristics and its applications in biosensors. We focus on the relationship between TdT extension conditions and extension efficiency. Furthermore, the construction strategy of TdT-mediated biosensors according to five different recognition types and their applications in targets are discussed and, finally, several current challenges and prospects in the field are taken into consideration. The development of rapid, accurate, and efficient detection methods for protozoan parasites can substantially control the outbreak of protozoan parasites infection, which poses a threat to global public health. Idealistically, electrochemical biosensors would be able to overcome the limitations of current detection methods due to their simplified detection procedure, on-site quantitative analysis, rapid detection time, high sensitivity, and portability. The objective of this scoping review is to evaluate the current state of electrochemical biosensors for detecting protozoan parasites. This review followed the most recent Preferred Reporting Items for Systematic Review and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) recommendations. Using electrochemical biosensor and protozoan parasite keywords, a literature search was conducted in PubMed, Scopus, Web of Science, and ScienceDirect on journals published between January 2014 and January 2022. Of the 52 studies, 19 were evaluated for eligibility, and 11 met the review's inclusion criteria to evaluate the effectiveness and limitations of the developed electrochemical biosensor platforms for detecting protozoan parasite including information about the samples, biomarkers, bioreceptors, detection system platform, nanomaterials used in fabrication, and limit of detection (LoD). Most electrochemical biosensors were fabricated using conventional electrodes rather than screen-printed electrodes (SPE). The range of the linear calibration curves for the developed electrochemical biosensors was between 200 ng/ml and 0.77 pM. The encouraging detection performance of the electrochemical biosensors demonstrate their potential as a superior alternative to existing detection techniques. Hybridization coupling among plasmon modes is an effective approach to manipulate near-field properties thus optical spectral shapes of plasmonic nanostructures. Generally, mode hybridization coupling is achieved by modifying the topography and dimensions of nanostructures themselves, with few concerns about substrate-induced manipulation. Herein, we propose a composite nanostructure consisting of a gold (Au) nanodisk array and a thin Au film supported by a dielectric substrate. In this configuration, both the refractive index of the dielectric substrate and thin gold film's thickness mediate the interaction of plasmon modes supported by upper and lower interfaces of the composite nanostructure, resulting in two hybridized plasmon modes. We systematically investigate the relationship between optical fields at the top surface of plasmon modes before and after the hybridization coupling. Specifically, the near-field amplitude at the top surface of the unhybridized modes is stronger than that of individual hybridized mode, and lower than the near-field summation of these two hybridized modes. This work not only provides a straightforward strategy for generating two plasmon modes in a nanostructure but also elucidates the variation of the optical field during the hybridization process, which is of crucial significance for applications, such as upconversion enhancement and multi-resonance sensing. Biosensing for diagnostics has risen rapidly in popularity over the past decades. With the discovery of new nanomaterials and morphologies, sensitivity is being constantly improved enough for reliable detection of trace biomarkers in human samples, like serum or sweat. This precision has enabled detailed research on the efficacy of biosensors. However, current biosensors suffer from reduced speed of operation. To make better use of this sensitivity, the development of a conductometric biosensor with in-situ use of an Laser Emitting Device (LED) display can provide rapid determination of sample results, steadily pushing biosensors toward more clinical, point-of-care (POC) applications. In this research, a simple LED was used for facile optical determination and visual output of an ultrasensitive bio-signal amplification circuit was made to interface with a B-type Natriuretic Peptide (BNP) biosensor. Tuning circuit gain enables an elegant method for adjustable separation of concentrations into 3 discrete categories: sub-threshold, analog, and saturation regions. These regions corresponded to 0 < [C] < 500 pg/mL (25, 100, 250 pg/mL, LED off), 500 < [C] < 1000 pg/mL (LED varying intensity), and 1000 pg/mL < [C] (LED full intensity). System efficacy was tested using human blood serum samples from University of Pittsburgh Medical Center patients, which were able to be accurately detected and sorted for rapid low cost and power. determination without need for complex digital elements. The rapid and sensitive detection of bacterial contaminants using low-cost and portable point-of-need (PoN) biosensors has gained significant interest in water quality monitoring. Cell-imprinted polymers (CIPs) are emerging as effective and inexpensive materials for bacterial detection as they provide specific binding sites designed to capture whole bacterial cells, especially when integrated into PoN microfluidic devices. However, improving the sensitivity and detection limits of these sensors remains challenging. In this study, we integrated CIP-functionalized stainless steel microwires (CIP-MWs) into a microfluidic device for the impedimetric detection of E. coli bacteria. The sensor featured two parallel microchannels with three-electrode configurations that allowed simultaneous control and electrochemical impedance spectroscopy (EIS) measurements. A CIP-MW and a non-imprinted polymer (NIP)-MW suspended perpendicular to the microchannels served as the working electrodes in the test and control channels, respectively. Electrochemical spectra were fitted with equivalent electrical circuits, and the charge transfer resistances of both cells were measured before and after incubation with target bacteria. The charge transfer resistance of the CIP-MWs after 30 min of incubation with bacteria was increased. By normalizing the change in charge transfer resistance and analyzing the dose–response curve for bacterial concentrations ranging from 0 to 107 CFU/mL, we determined the limits of detection and quantification as 2 × 102 CFU/mL and 1.4 × 104 CFU/mL, respectively. The sensor demonstrated a dynamic range of 102 to 107 CFU/mL, where bacterial counts were statistically distinguishable. The ability of aquatic mesofauna representatives involved in trophic chains to sorb and accumulate toxicants is important for understanding the functioning of aquatic ecosystems and for fishing industry. This study investigated the capacity of marine amphipod Gammarus oceanicus and freshwater amphipods Eulimnogammarus vittatus and Gammarus lacustris to absorb the DNA-alkylating agent methyl methanesulfonate (MMS). The presence of alkylating agents in the environment and in the tissues of the amphipods was determined using whole-cell lux-biosensor Escherichia coli MG1655 pAlkA-lux, in which the luxCDABE genes from Photorhabdus luminescens, enabling the luminescence of the cell culture, are controlled by the PalkA promoter of DNA glycosylase. It was shown that within one day of incubation in water containing MMS at a concentration above 10 μM, the amphipods absorbed the toxicant and their tissues produce more alkylation damage to biosensor cells than the surrounding water. Concentrations of MMS above 1 mM in the environment caused the death of the amphipods before the toxicant could be significantly concentrated in their tissues. The sensitivity and the capacity to absorb MMS were found to be approximately the same for the marine amphipod G. oceanicus and the freshwater amphipods E. vittatus and G. lacustris. This study observes the MOSFET's performance concerning several biomolecules for use as a biosensor device. The double gate MOSFET with gate stack configuration has been chosen as the suggested device to surpass the limitations of short-channel effects (SCEs). The cavity was created to restrict the passage of charged and uncharged biological molecules so that they could be detected. These molecules fill the cavity, changing the device's electrical properties. The Double Gate MOSFET (DG-MOSFET) biosensor is subject to limitations such as Short Channel Effects (SCEs) and issues with the power supply. The suggested device decreased SCEs and it also demonstrates the potential benefits of having DG-MOSFETs with gate stacking for biosensor applications. Comparison of transconductance, the generating factor for transconductance, and sensitivity parameters such as Id and Vth sensitivity and Ion/Ioff sensitivity has been carried out in this study.Article Highlights: The variation of temperature can influence the mobility of charge carriers within the semiconductor, affecting the conductance of the sensor. Alpha-fetoprotein (AFP) is a key marker for early cancer detection and assessment. However, the current detection methods struggle to balance accuracy with the need for decentralized medical treatment. To address this issue, a new AFP analysis platform utilizing digital image colorimetry has been developed. Functionalized gold nanoparticles act as colorimetric agents, changing from purple-red to light gray-blue when exposed to different AFP concentrations. A smartphone app captures these color changes and calculates the AFP concentration in the sample. To improve detection accuracy, a hardware device ensures uniform illumination. Testing has confirmed that this system can quantitatively analyze AFP using colorimetry. The limit of detection reached 0.083 ng/mL, and the average accuracy reached 90.81%. The extensive use of chemical pesticides has significantly boosted agricultural food crop yields. Nevertheless, their excessive and unregulated application has resulted in food contamination and pollution in environmental, aquatic, and agricultural ecosystems. Consequently, the on‐site monitoring of pesticide residues in agricultural practices is paramount to safeguard global food and conservational safety. Traditional pesticide detection methods are cumbersome and ill‐suited for on‐site pesticide finding. The systematic review provides an in‐depth analysis of the current status and perspectives of nanobiosensors (NBS) for pesticide detection in the agricultural arena. Furthermore, the study encompasses the fundamental principles of NBS, the various transduction mechanisms employed, and their incorporation into on‐site detection platforms. Conversely, the assortment of transduction mechanisms, including optical, electrochemical, and piezoelectric tactics, is deliberated in detail, emphasizing its advantages and limitations in pesticide perception. Incorporating NBS into on‐site detection platforms confirms a vital feature of their pertinence. The evaluation reflects the integration of NBS into lab‐on‐a‐chip systems, handheld devices, and wireless sensor networks, permitting real‐time monitoring and data‐driven decision‐making in agronomic settings. The potential for robotics and automation in pesticide detection is also scrutinized, highlighting their role in improving competence and accuracy. Finally, this systematic review provides a complete understanding of the current landscape of NBS for on‐site pesticide sensing. Consequently, we anticipate that this review offers valuable insights that could form the foundation for creating innovative NBS applicable in various fields such as materials science, nanoscience, food technology and environmental science. In this manuscript, a Trench Gate Engineered Junctionless Accumulation Mode Gate-All-Around (TGE-JAM-GAA) BioFET structure has been proposed and critically analyzed for the label-free identification of biomolecules using extensive numerical device simulations. This device is superior in terms of a variety of performance metrics, including higher sensitivity, a larger shift in threshold voltage, drain current, transconductance, and the ION/IOFF current ratio. These improvements are due to both the triple metal gate engineering and the trench gate. A comparative analysis of TGE-JAM-GAA BioFET has been performed with a triple metal normal gate JAM Gate-All-Around FET (TMNG-JAM-GAAFET) and a conventional single metal normal gate JAM Gate-All-Around FET (SMNG-JAM-GAAFET) biosensor. For instance, in the case of APTES biomolecules, the drift in threshold voltage obtained is 236.24 mV, which is 58.75% and 159.18% higher than that of the TMNG-JAM-GAAFET and SMNG-JAM-GAAFET, respectively, demonstrating that the TGE-JAM-GAA BioFET is more suitable for applications in biosensing. Accurate and timely diagnosis of plant viral infections plays a key role in effective disease control and maintaining agricultural productivity. Recent advances in the diagnosis of plant viruses have significantly expanded our ability to detect and monitor viral pathogens in agricultural crops. This review discusses the latest advances in diagnostic technologies, including both traditional methods and the latest innovations. Conventional methods such as enzyme-linked immunosorbent assay and DNA amplification-based assays remain widely used due to their reliability and accuracy. However, diagnostics such as nextgeneration sequencing and CRISPR-based detection offer faster, more sensitive and specific virus detection. The review highlights the main advantages and limitations of detection systems used in plant viral diagnostics including conventional methods, biosensor technologies and advanced sequence-based techniques. In addition, it also discusses the effectiveness of commercially available diagnostic tools and challenges facing modern diagnostic techniques as well as future directions for improving informed disease management strategies. Understanding the main features of available diagnostic methodologies would enable stakeholders to choose optimal management strategies against viral threats and ensure global food security. The sensitive nonenzymatic sensing of glucose has been made feasible for the first time using a nonthermal plasma (NTP)-synthesized ZnO nanostructure. This work presents an interesting and novel method for surface modification. The effects of flow gas of Argon/Oxygen (20, 30 and 40 l/min) on the Zn foil surface and various properties were investigated. Optical emission spectroscopy OES has been used to characterize transmissions for positive and negative systems of Ar/O plasma. X-ray diffraction showed close adjacent tops and the strongest peak at Zn (101). EDX displays the constants (Ok_α), (Znk_α) and (Znk_β) and change in (ZnL_α). Photoluminescence (PL) Analysis shows a shift at the vertex toward the left and then toward the right confirming the reaction of all PL spectra within a strong UV emission peak. Raman spectroscopy analysis demonstrates two clear peaks gradually shifting to the right with an increase in the percentage of gas flowing compared to the blank metal, and scanning electron microscopy (FE-SEM) images show changes in the shape of nanoparticles due to increasing gas flow, the surface of zinc metal is affected by cold plasma and forms particles with very small diameters ranging from 10.8nm to 123nm. Also altered the surface morphology where the nano shape changed from flower-like particles to sheets with diameters ranging 282.7–643.5nm and the sheets grew with the increase of gas flow to diameters ranging 132–710nm. ZnO nanostructures are employed as biosensor electrodes (nonenzymatic) glucose as the increase in current is proportional to the flowing gas (2.06E-03mA, 2.09E-03mA and 4.34E-03mA). Recent advancements in medical science have ushered in the era of "Personalized Medicine," which tailors treatments to individual genetics, lifestyle, and environmental factors. A cornerstone of this approach is continuous monitoring and adjustment of treatment regimens to optimize effectiveness while minimizing side effects. In therapeutic drug monitoring (TDM), crucial for drugs like doxorubicin (DOX) with severe side effects, traditional methods such as spectrofluorimetry and chromatography are utilized, but they suffer from cost and complexity issues. Biosensors, especially those leveraging Localized Surface Plasmon Resonance (LSPR) and surface-enhanced Raman spectroscopy (SERS), offer a promising alternative. This study focuses on developing a cost-effective plasmonic biosensor using gold nanoislands semiembedded in a glass substrate. The LSPR platform, based on gold nanoislands (Au NIs) with an average diameter of 23 nm, offers a rapid method for ultrasensitive, label-free detection of DOX in biological fluids up to the nanomolar range, even in diluted (1:10) and undiluted blood serum samples from cancer patients receiving the drug. The method capitalizes on the extraordinary adhesion between the nanoislands and the glass matrix, preserving plasmon integrity while enhancing the electric field surrounding the nanoislands for improved sensitivity. Moreover, by exploiting nanoislands with Au/Al2O3 core/shell structures with an average diameter and gap of about 70 and 7 nm, respectively, highly efficient SERS-active platforms are created, enabling the detection of ultralow DOX concentrations down to picomolar range, where the LSPR system is insensitive. The combined use of both systems enables the detection of DOX across a broad range, from low to high nanomolar concentrations, in biological fluids. This integration of semiembedded nanoislands presents a scalable and multifunctional approach suitable for point-of-care diagnostics, offering reproducible and stable characteristics over time.
