Purpose: To evaluate four decontamination strategies utilizing enzymatic agents available in most clinical settings to determine (1) the amount of biomaterial that can be removed in a group of previously used healing abutments (uHAs) and (2) the degree to which the decontaminated HAs are capable of inducing an inflammatory response in vitro compared to new HAs. Materials and Methods: In total, 50 HAs were collected following 2 to 4 weeks of intraoral use and distributed randomly into five test groups (groups A-E; n = 10 per group). Group A used enzymatic cleaner foam and an autoclave. Group B used an ultrasonic bath with enzymatic cleaner and an autoclave. Group C used a prophy jet, enzymatic cleaner foam, and an autoclave. Group D used a prophy jet, an ultrasonic bath with enzymatic cleaner, and an autoclave. Lastly, group E used a prophy jet and an autoclave. The control group consisted of 10 new and sterile HAs. Residual protein concentration was determined by a Micro BCA protein assay (Thermo Fisher Scientific), while HAs from each group were stained with Phloxine B and macroscopically examined for the presence of debris. To examine the inflammatory potential, human primary macrophages were exposed to HAs, and supernatant levels of nine cytokine and chemokine profiles were analyzed using a multiplex bead assay. Results: All test groups showed differences in the degree of visual decontamination compared to controls. Groups D and E displayed the most effective surface debris removal and reduced protein concentration, while group A was the least effective. However, compared to controls, all test groups showed high levels of inflammatory cytokine secretion via multiplex assay for up to 5 days. Conclusions: Our study found that decontamination of uHAs utilizing enzymatic cleaners failed to reestablish inert HA surfaces and prevent an inflammatory immune response in vitro. Background: Autologous tissues such as buccal mucosa (BM) are widely used for reconstruction of urethral strictures; however, limitations such as donor site morbidity and scarce tissue supply require the development of alternative biomaterials for urethral repair. The goals of this study were to determine the safety and efficacy of bi-layer silk fibroin (BLSF) matrices for urethral stricture repair and compare histological and functional outcomes to the standard approach, BM urethroplasty under good laboratory practices.Material and methods: A total of 13 rabbits exhibiting urethral stricture formation following electrocoagulation injury were treated with onlay urethroplasty with either acellular BLSF (N = 7) or autologous BM (N = 6) grafts for 3 months. Uninjured control rabbits were maintained in parallel (N = 4).Results and conclusion: Animals receiving BLSF implants were demonstrated to be functionally equivalent to BM grafts in their ability to restored strictured calibers, support micturition and promote tissue regeneration with minimal inflammation.

The objective of this preclinical study was to evaluate the feasibility and safety of transcatheter endocardial alginate hydrogel injection (TEAi) in a large animal model, utilizing the high-stiffness XDROP® alginate hydrogel in combination with the dedicated EndoWings® catheter-based system. All swine (n = 9) successfully underwent TEAi without complications. Acute results from a subset of animals (n = 5) demonstrated the ability of the catheter to access a wide range of endomyocardial areas and achieve consecutive circumferential hydrogel distribution patterns within the mid-left ventricular wall. Histological examinations at 6 months (n = 4) demonstrated that the XDROP® remained localized within the cardiac tissue. In addition, serial echocardiographic imaging showed that XDROP® had no adverse impacts on LV systolic and diastolic functions. In conclusion, this innovative combination technology has the potential to overcome the translational barriers related to alginate hydrogel delivery to the myocardium.

While oropharyngeal cancer treatment regimens, including surgical resection, irradiation, and chemotherapy, are effective at removing tumors, they lead to muscle atrophy, denervation, and fibrosis, contributing to the pathogenesis of oropharyngeal dysphagia - difficulty swallowing. Current standard of care of rehabilitative tongue strengthening and swallowing exercises is ineffective. Here, we evaluate an alternative approach utilizing an acellular and injectable biomaterial to preserve muscle content and reduce fibrosis of the tongue after injury. Skeletal muscle extracellular matrix (SKM) hydrogel is fabricated from decellularized porcine skeletal muscle tissue. A partial glossectomy injury in the rat is used to induce tongue fibrosis, and SKM hydrogels along with saline controls are injected into the site of scarring two weeks after injury. Tissues are harvested at 3 and 7 days post-injection for gene expression and immunohistochemical analyses, and at 4 weeks post-injection to evaluate histomorphological properties. SKM hydrogel reduces scar formation and improves muscle regeneration at the site of injury compared to saline. SKM additionally modulates the immune response towards an anti-inflammatory phenotype. This study demonstrates the immunomodulatory and tissue-regenerative capacity of an acellular and minimally invasive ECM hydrogel in a rodent model of tongue injury.

BackgroundAutologous tissues derived from bowel, buccal mucosa and skin are primarily used to repair or replace diseased vaginal segments as well as create neovaginas for male-to-female transgenders. These grafts are often limited by scarce tissue supply, donor site morbidity and post-operative complications. Bi-layer silk fibroin (BLSF) biomaterials represent potential alternatives for vaginoplasty given their structural strength and elasticity, low immunogenicity, and processing flexibility. The goals of the current study were to assess the potential of acellular BLSF scaffolds for vaginal tissue regeneration in respect to conventional small intestinal submucosal (SIS) matrices in a rat model of vaginoplasty.MethodsInlay vaginoplasty was performed with BLSF and SIS scaffolds (N = 21 per graft) in adult female rats for up to 2 months of implantation. Nonsurgical controls (N = 4) were investigated in parallel. Outcome analyses included histologic, immunohistochemical and histomorphometric evaluations of wound healing patterns; µ-computed tomography (CT) of vaginal continuity; and breeding assessments.ResultsAnimals in both scaffold cohorts exhibited 100% survival rates with no severe post-operative complications. At 2 months post-op, µ-CT analysis revealed normal vaginal anatomy and continuity in both graft groups similar to controls. In parallel, BLSF and SIS grafts also induced comparable constructive remodeling patterns and were histologically equivalent in their ability to support formation of vascularized vaginal neotissues with native tissue architecture, however with significantly less smooth muscle content. Vaginal tissues reconstructed with both implants were capable of supporting copulation, pregnancy and similar amounts of live births.ConclusionsBLSF biomaterials represent potential "off-the-shelf" candidates for vaginoplasty.

Purpose: To evaluate the influence of initial implant protrusion within the subantral space on hard tissue gain for implants placed simultaneously with transcrestal sinus floor elevation (TSFE) with a biomaterial. Materials and Methods: A total of 50 implants were placed after TSFE in 44 patients using either a human demineralized bone matrix or a deproteinized bone mineral matrix. Intraoral radiographs were obtained before and immediately after surgery. CBCT scans were obtained at the last follow-up (mean: 6.6 years). Results: The initial bone crest height was 4.6 ± 1.4 mm, and the initial protrusion of the implants above the sinus floor was 3.5 ± 1.4 mm. At the follow-up assessments, the hard tissue mean gain was 2.5 ± 1.5 mm, resulting in a mean residual protrusion of 1.1 ± 1.3 mm. Only 10 implants did not protrude above the apical level of hard tissue. Positive correlations were found between hard tissue gain and initial protrusion (r = 0.55; 95% CI: 0.32 to 0.72; P = .0001), between the initial and final protrusions (r = 0.38; 95% CI: 0.10 to 0.60; P = .007), and between the follow-up period and final protrusion (r = 0.35; 95% CI: 0.07 to 0.58; P = .012). Conclusions: The higher the initial protrusion was, the higher were the hard tissue gain and the final protrusion of the implant above the apical level of the hard tissue. [ABSTRACT FROM AUTHOR]

BACKGROUND: At present, the treatment of infected bone defects has the problems of long course of disease, poor treatment effect and high cost. The osteogenic effect of personalized bone replacement materials in clinical treatment is limited. Therefore, a 3D-printed bone graft material with both good osteogenic effect and antibacterial effect is urgently needed for clinical treatment. OBJECTIVE: To summarize the research status of 3D-printed scaffolds loaded with antimicrobial agents for the treatment of infected bone defects. METHODS: PubMed, Web of Science, Elsevier, and CNKI databases from January 2010 to June 2022 were searched for related articles. The Chinese search terms were “bone defect, 3D printing, scaffold material, antibacterial, animal experiments, in vitro experiments”. English search terms were “bone defect, 3D printing, scaffold, antibiosis, animal experiment, in vitro”. Finally, 60 articles were included for review and analysis. RESULTS AND CONCLUSION: The 3D scaffolds made of titanium, magnesium, tantalum and other metals and their alloys have certain osteogenic properties, but do not have antibacterial function. Hydroxyapatite and other bioceramic materials have good biocompatibility and are prone to be degraded, whereas due to the lack of strength, they are usually combined with artificial polymer materials to form composite materials, which respectively mimic the inorganic and organic components in natural bone, and play their respective excellent functions. Antibiotics, silver/copper nanoparticles, antimicrobial peptides, gallium and other antibacterial agents play an antibacterial role by destroying bacterial cell membrane, producing reactive oxygen species to interfere with bacterial DNA replication, inhibiting iron absorption and other mechanisms. As a result, the 3D-printed scaffold has both antibacterial and osteogenic effects. However, there are still some problems such as drug resistance and difficult to control effective concentrations. 3D-printed scaffolds are often loaded with antibacterial agents by loading drug-loaded microspheres on scaffolds, preparing antibacterial coating on the scaffold surface, and participating in joint 3D printing with drugs. The loading mode of antibacterial coating prepared on the scaffold surface is the most widely used, and its antibacterial effect is more stable. Nonetheless, the selection of the most suitable loading mode for antibacterial agents needs to be further discussed and summarized. It is a future research prospect to optimize the mechanical properties of composite scaffolds and prepare biomimetic bone scaffolds so that the degradation rate is consistent with the bone reconstruction rate in infected bone defects. The ideal antibacterial agents may play a role through a variety of antibacterial mechanisms, thus being expected to play a good antibacterial effect through low antibacterial concentration, which should be a hot spot of anti-bone infection research. After loading antibacterial agents on the surface of the scaffold, antibacterial agents can “intelligently” react to the local microenvironment, achieving controlled release, and regulating the osteogenesis, vascularization and immune response of the microenvironment, which is the focus of current research. Background: Treatment of articular cartilage injury remains a challenging clinical problem in orthopedics. Chitosan-derived biomaterial could be a potential adjuvant treatment to improve cartilage repair. In the current study, we examined the effects of two potential chitosan-derived materials on cartilage regeneration of osteochondral defects in rabbits. Methods: An osteochondral defect was created over the rabbit knee and treated using three approaches: group A received no material (n = 24), group B received chitosan membranes with glucose absorption (CGA; n = 25), and group C received chitosan-glucose derivative membranes obtained via the Maillard reaction (CGMR; n = 25). Cartilage repair over the osteochondral defect was analyzed 12 weeks post-surgery via histological analysis, immunostaining, and reverse transcription-qualitative polymerase chain reaction (RT-qPCR) for type-I and type-II collagen mRNA. Results: According to histological analysis, CGMR-treated defects showed significantly improved modified O'Driscoll scoring when compared with no material- and CGA-treated defects (20.9 ± 4.3 vs. 13.00 ± 2.5 and 17.7 ± 4.6, p < 0.001). Moreover, group C exhibited higher intensity of type-II collagen immunohistochemical staining over the regenerated cartilage than groups A and B, along with increased expression of type-II collagen mRNA by RT-qPCR. This review comprehensively explores the multifunctional applications of pectins derived from food waste and by-products, emphasizing their role as versatile biomaterials in the medical-related sectors. Pectins, known for their polyelectrolytic nature and ability to form hydrogels, influence the chemical composition, sensory properties, and overall acceptability of food and pharmaceutical products. The study presents an in-depth analysis of molecular parameters and structural features of pectins, such as the degree of esterification (DE), monosaccharide composition, galacturonic acid (GalA) content, and relative amounts of homogalacturonan (HG) and rhamnogalacturonan I (RG-I), which are critical for their technofunctional properties and biological activity. Emphasis is placed on pectins obtained from various waste sources, including fruits, vegetables, herbs, and nuts. The review also highlights the importance of structure–function relationships, especially with respect to the interfacial properties and rheological behavior of pectin solutions and gels. Biological applications, including antioxidant, immunomodulatory, anticancer, and antimicrobial activities, are also discussed, positioning pectins as promising biomaterials for various functional and therapeutic applications. Recalled pectins can also support the growth of probiotic bacteria, thus increasing the health benefits of the final product. Biomaterials have been the subject of extensive research, and their applications in medicine and pharmacy are expanding rapidly. Collagen and its derivatives stand out as valuable biomaterials due to their high biocompatibility, biodegradability, and lack of toxicity and immunogenicity. This review comprehensively examines collagen from various sources, its extraction and processing methods, and its structural and functional properties. Preserving the native state of collagen is crucial for maintaining its beneficial characteristics. The challenges associated with chemically modifying collagen to tailor its properties for specific clinical needs are also addressed. The review discusses various collagen-based biomaterials, including solutions, hydrogels, powders, sponges, scaffolds, and thin films. These materials have broad applications in regenerative medicine, tissue engineering, drug delivery, and wound healing. Additionally, the review highlights current research trends related to collagen and its derivatives. These trends may significantly influence future developments, such as using collagen-based bioinks for 3D bioprinting or exploring new collagen nanoparticle preparation methods and drug delivery systems. Spinal cord injury (SCI) results from various causes, including sports‐related incidents, degenerative cervical myelopathy, traffic accidents, and falls. SCI typically leads to sensory and motor dysfunction and even paralysis. Current treatments for SCI include systemic administration of high‐dose steroids and surgical decompression and stabilization. However, excessive use of glucocorticoids may increase susceptibility to infections and systemic bleeding. The long‐term effect of surgery intervention remains unclear, with ongoing debates regarding its timing, efficacy, and safety. Therefore, innovative approaches are urgently needed to alleviate secondary injuries and promote spinal recovery. One emerging therapeutic approach for SCI is modulating the microenvironments to achieve neuroprotection and neurogenesis during recovery. Several biomaterials with favorable physicochemical properties have been developed to enhance therapeutic effects by regulating microenvironments. This Review first discusses the pathology of SCI microenvironments and then introduces biomaterials‐based regulatory strategies targeting various microenvironmental components, including anti‐inflammation, anti‐oxidation, reduction of excitotoxicity, revascularization, neurogenesis, and scar density reduction. The high-throughput method based on the micron-level structure that 3D bioprinting technology offers for various environmental microbiological engineering applications is made possible by its several printing paths and precision programming control. This versatility makes it an on-demand manufacturing technology. A novel 3D manufacturing technique called 3D bioprinting may be used to precisely uptake and disperse bacteria to create microbial active substances with a variety of intricate functionalities for environmental applications. The technological challenges that the current 3D bioprinting technology must face include the mechanical properties of materials, the creation of specific bioinks to adapt to different strains, and the exploration of 4D bioprinting for intelligent applications. Therefore, this analysis delves deeply into the core technological ideas of 3D bioprinting, bioink materials, and their environmental applications. It also offers recommendations about the challenges and opportunities associated with 3D bioprinting. Combined with the present advancements in microbe enhancement technology, 3D bioprinting will provide an enabling platform for multifunctional microorganisms and facilitate the management of in situ directional responses in the environmental domain. Triboelectric nanogenerators, known as TENGs, offer great potential as versatile energy harvesting devices. In recent years, there has been a rise in TENG designs that prioritize compatibility with sustainable biomaterials, leading to new possibilities in green technology. The novelty of this work lies in its pioneering exploration of utilizing sustainable biomaterials, particularly coconut husk, within the field of Tribo-electric nanogenerators (TENGs). This study focuses on evaluating and characterizing coconut husk as TENG material considering factors such as rotational speed, vane count, and coarseness, all of which influence the output potential of the B-TENG. The BTENG model employed in this research operates on a rotational sliding mode, featuring a biobased material layer of coconut husk, a layer of PTFE, and copper as electrodes. The B-TENG has a diameter of 100 mm with varying vane configurations (3-vane, 4-vane, and 5-vane). The sliding mode demonstrated impressive versatility, yielding output voltages spanning from 0.73 V to 4.0 V across rotational speeds of 200 RPM to 1400 RPM. Remarkably, the 5-vane fine-grained coconut husks achieved a maximum power of 121.10 mW at 10 Ohm and a power density of 3.84 mW/cm². This research carries global significance, contributing to the advancement of energy harvesting technology. The insect cuticle, which serves as both a protective barrier and an efficient lever system for locomotion, is an extracellular matrix primarily composed of chitin and protein. The cuticle protein CPCFC characterized by a “CFC” motif containing 2 Cys split by the insertion of 5 residues is distributed across most insect species and specifically localized in the hard part of the cuticle. However, their physiological function is not fully understood. Here, we report 2 CPCFC proteins, TcCPCFC1 and TcCPCFC2, derived from the Coleopteran insect Tribolium castaneum. We revealed that TcCPCFC1 and TcCPCFC2 were predominantly expressed during the larval and adult stages of T. castaneum, respectively. The transcription downregulation of TcCPCFC1 significantly decreased the modulus and toughness of the elytral cuticle. We found that TcCPCFC proteins have high binding affinity to chitin. We cloned and produced recombinant TcCPCFC proteins and demonstrated that the addition of TcCPCFC proteins to chitin hydrogel greatly enhanced the hydrogel's modulus and toughness by forming denser chitin fibrous networks. Our findings reveal the functional role of CPCFC proteins in enhancing mechanical properties of insect cuticle, and we validate this process in vitro, and offer a protein candidate for fabrication of advanced chitin‐based materials. Dentin tissue can act as a reservoir for bioactive molecules that create signals for cellular proliferation and differentiation to initiate tissue regeneration. Therefore, the aim of this study is to compare the cell viability, inflammatory response, and antimicrobial activity of bovine dentin grain-added calcium-hydroxide (CH-BDG) with different pulp-capping materials.ProRoot MTA, Biodentine, Dycal, TheraCal-LC, and an experimental material, CH-BDG, were examined. Cell viability was determined via the WST-1 assay. The inflammatory response was analysed by the monocyte chemoattractant protein-1 (MCP-1/CCL2) and macrophage inflammatory protein-1α (MIP-1α/CCL3) levels. The antimicrobial activity was evaluated by agar-diffusion.The cell viability of CH-BDG was analogous with Biodentine and control at 24 h. The cell viability of CH-BDG decreased at 48 h, but the rate was higher than ProRoot MTA and Dycal (p<0.05). For MCP-1 and MIP-1α values, there was no significant difference between the control and CH-BDG. The MCP-1 level of CH-BDG was lower compared to other pulp-capping materials (p<0.05). The MIP-1α level of CH-BDG was lower compared to ProRoot MTA, Biodentine, and TheraCal-LC (p<0.05). No inhibition zone was detected against oral microorganisms for CH-BDG.The experimentally developed CH-BDG showed competing properties and additional advantages compared to the existing pulp-capping materials. Repairing or reconstructing significant bone defects is typically challenging. In the present study, two composite cements were used as scaffolds in a sub-critical femoral defect in rats. A control group and two experimental batches were used to compare the outcomes. This research aimed to investigate the osteogenic potential and toxicological tolerance of the bioproducts through histopathology and computed tomography imaging analysis at 14, 28, 56, and 90 days post-implantation. The biomaterials used in the investigation consisted of a 65% bioactive salinized inorganic filler and a 25% weight organic matrix. The organic part of the biomaterial was composed of Bis-GMA (bisphenol A-glycidyl methacrylate), UDMA (urethane dimethacrylate), HEMA (2-Hydroxyethyl methacrylate), and TEGDMA (triethylene glycol dimethacrylate), while the inorganic filler was composed of silica, barium glass, hydroxyapatite, and fluor aluminosilicate glass. The first findings of this research are encouraging, revealing that there is a slight difference between the groups treated with biomaterials, but it might be an effective approach for managing bone abnormalities. Material C1 exhibited a faster bone defect healing time compared to material C2, where bone fractures occurred in some individuals. It is unclear if the fractures were caused by the presence of the biomaterial C2 or whether additional variables were to blame. Studies aiming to understand the effects of storage on whole blood (WB) clotting often rely on characterizing coagulation under static conditions. Minimal work has explored the effects of physiologic shear on clot formation and thrombolysis utilizing fractionated and reconstituted whole blood (rWB) products. WB was fractionated into platelet‐free plasma, packed red blood cells, and platelets storing each component under its ideal conditions—including platelet cryopreservation. Recombination at their native ratios was accomplished over 91 days of storage and clotting/thrombolysis was analyzed utilizing thromboelastography and Chandler loop. rWB preserved clot strength through 91 days with minimal deviation from baseline, in contrast to WB stored at 4°C which experienced a significant decline by storage Day‐42. Clot formation under shear for both rWB and WB led to increased clot mass through storage. No significant deviation from baseline was appreciated until Day 70 of storage in rWB. Increasing degrees of thrombolysis were seen in both groups, with rWB significantly deviating from baseline at Day 70. No significant changes in overall clot architecture occurred throughout storage and recombination. This fractionation and recombination protocol serves as a method to further develop reproducible in vitro clot analogs for preclinical thrombolytic therapy screening. [ABSTRACT FROM AUTHOR]

The demand for novel tissue grafting and regenerative wound care biomaterials is growing as traditional options often fall short in biocompatibility, functional integration with human tissue, associated cost(s), and sustainability. Salmon aquaculture generates significant volumes of waste, offering a sustainable opportunity for biomaterial production, particularly in osteo-conduction/-induction, and de novo clinical/surgical bone regeneration. Henceforth, this study explores re-purposing salmon waste through a standardized pre-treatment process that minimizes the biological waste content, followed by a treatment stage to remove proteins, lipids, and other compounds, resulting in a mineral-rich substrate. Herein, we examined various methods—alkaline hydrolysis, calcination, and NaOH hydrolysis—to better identify and determine the most efficient and effective process for producing bio-functional nano-sized hydroxyapatite. Through comprehensive chemical, physical, and biological assessments, including Raman spectroscopy and X-ray diffraction, we also optimized the extraction process. Our modified and innovative alkaline hydrolysis–calcination method yielded salmon-derived hydroxyapatite with a highly crystalline structure, an optimal Ca/P ratio, and excellent biocompatibility. The attractive nano-scale cellular/tissular properties and favorable molecular characteristics, particularly well-suited for bone repair, are comparable to or even surpass those of synthetic, human, bovine, and porcine hydroxyapatite, positioning it as a promising candidate for use in tissue engineering, wound healing, and regenerative medicine indications. Notably, Sericin, a silk-derived protein, and Chelidonium majus L. (C. majus), a botanical agent, have emerged as compelling candidates, providing a unique combination of natural elements that may revolutionize conventional wound care approaches. Sericin, renowned for its diverse properties, displays unique properties that accelerate the wound healing process. Simultaneously, C. majus, with its diverse pharmacological compounds, shows promise in reducing inflammation and promoting tissue regeneration. As the demand for innovative wound care solutions increases, understanding the therapeutic potential of natural products becomes imperative. This review synthesizes current knowledge on Sericin and C. majus, envisioning their future roles in advancing wound management strategies. The exploration of these natural substances as constituents of wound dressings provides a promising avenue for developing sustainable, effective, and biocompatible materials that could significantly impact the field of wound healing. With the increasing shortage of water resources and the aggravation of water pollution, solar-driven interfacial steam generation (SISG) technology has garnered considerable attention because of its low energy consumption, simple operation, and environmental friendliness. The popular multi-layer SISG evaporator is composed of two basic structures: a photothermal layer and a support layer. Herein, the support layer underlies the photothermal layer and carries out thermal management, supports the photothermal layer, and transports water to the evaporation interface to improve the stability of the evaporator. While most research focuses on the photothermal layer, the support layer is typically viewed as a supporting object for the photothermal layer. This review focuses on the support layer, which is relatively neglected in evaporator development. It summarizes existing progress in the field of multi-layer interface evaporators, based on various polymers and biomaterials, along with their advantages and disadvantages. Specifically, mainly polymer-based support layers are reviewed, including polymer foams, gels, and their corresponding functional materials, while biomaterial support layers, including natural plants, carbonized biomaterials, and other innovation biomaterials are not. Additionally, the corresponding structure design strategies for the support layer were also involved. It was found that the selection and optimal design of the substrate also played an important role in the efficient operation of the whole steam generation system. Their evolution and refinement are vital for advancing the sustainability and effectiveness of interfacial evaporation technology. The corresponding potential future research direction and application prospects of support layer materials are carefully presented to enable effective responses to global water challenges. In this review, we systematically elaborated the synopsis of the cGAS-STING signaling pathway as well as involved immunological effects in cancer immune cycle, and thoroughly overviewed the summary of chemical or engineering strategies to heighten the potential application of the cGAS-STING signaling pathway-related agonists. Oral fungal infections pose a threat to human health and increase the economic burden of oral diseases by prolonging and complicating treatment. A cost-effective strategy is to try to prevent these infections from happening in the first place. With this purpose, biomaterials with antifungal properties are a crucial element to overcome fungal infections in the oral cavity. In this review, we go through different kinds of biomaterials and coatings that can be used to functionalize them. We also review their potential as a therapeutic approach in addition to prophylaxis, by going through traditional and alternative antifungal compounds, e.g., essential oils, that could be incorporated in them, to enhance their efficacy against fungal pathogens. We aim to highlight the potential of these technologies and propose questions that need to be addressed in prospective research. Finally, we intend to concatenate the key aspects and technologies on the use of biomaterials in oral health, to create an easy to find summary of the current state-of-the-art for researchers in the field. This study reported the biological changes occurring after γ-irradiation of in vivo rat model and the osteochondral protective effect of Gelatine-Chitosan-Ginger (GEL-CH-GING). The results indicated that Electron Paramagnetic Resonance (EPR) Spectroscopy of GEL-CH-GING identified two paramagnetic centers with g-values of 2.19 and 2.002. Fourier Transform Infrared Spectroscopy (FTIR) analysis demonstrated an increased intensity in the peaks corresponding to C-H chains and C=O carbonyl groups. Additionally, X-ray Diffraction (XRD) analysis revealed no alteration in crystallinity. After gamma ray exposure, the rat groups have received an osteochondral defect and then were treated with GEL-CH-GING composite. Sixty days post-surgery, a significant reduction in thiobarbituric acid-reactive compounds (TBARs) was seen when compared to non-implanted rat group. Concerning oxidative stress status, GEL-CH-GIN significantly improved Superoxide Dismutase (SOD) 76 nmol/l, Catalase (CAT) 0.79 nmol/l, and Glutathione Peroxidase (GPx) 1.77 nmol/l activities in osteochondral tissue. Regarding the histomorphometric parameters of cartilaginous tissue (nCg.Th, µm), (cCg.Th, µm), (Cg. Th, µm), irradiated-GEL-CH-GIN group showed a significant increase as compared to irradiated group with 116, 74 and 188 µm, respectively (p< 0.01). The microanalysis showed a high percentage of O and C in the regenertaed osteochondral tissue and indicated the deposition of novel collagen matrix. The biomechanical behaviour showed a significantly enhanced hardness measurement (1.73±0.029 VH, p< 0.05) when compared with that of irradiated group Biochemical markers suggested an osteocartilage repair capacity. In fact, the levels of IL-1ß, IL-6, TNF-a and VEGF in the implanted rat with GEL-CH-GING composite exhibited 51±3.48, 30.05±5.18, 65.12±4.33 and 40.42±3.32 ng/l, respectively. A hallmark of angiogenesis is the sprouting of endothelial cells. To replicate this event in vitro, biomaterial approaches can play an essential role in promoting cell migration. To study the capacity of a scaffold of fibrin (fibrinogen:thrombin mix) to support the movement of the endothelial cells, the migration area of spheroids formed with the HULEC cell line was measured. The cells were first allowed to form a spheroid using the hanging drop technique before being encapsulated in the fibrin gel. The cells' migration area was then measured after two days of embedding in the fibrin gel. Various conditions affecting fibrin gel polymerization, such as different concentrations of fibrinogen and thrombin, were evaluated alongside rheology, porosity, and fiber thickness analysis to understand how these factors influenced cell behavior within the composite biomaterial. Data point toward thrombin's role in governing fibrin gel polymerization; higher concentrations result in less rigid gels (loss tangent between 0.07 and 0.034) and increased cell migration (maximum concentration tested: 5 U/mL). Chitosan is a very promising material for tissue model printing. It is also known that the introduction of chemical modifications to the structure of the material in the form of methacrylate groups makes it very attractive for application in the bioprinting of tissue models. The aim of this work is to study the characteristics of biomaterials containing chitosan (BCH) and its methacrylated equivalent (BCM) in order to identify differences in their usefulness in 3D bioprinting technology. It has been shown that the BCM material containing methacrylic chitosan is three times more viscous than its non-methacrylated BCH counterpart. Additionally, the BCM material is characterized by stability in a larger range of stresses, as well as better printability, resolution, and fiber stability. The BCM material has higher mechanical parameters, both mechanical strength and Young's modulus, than the BCH material. Both materials are ideal for bioprinting, but BCM has unique rheological properties and significant mechanical resistance. In addition, biological tests have shown that the addition of chitosan to biomaterials increases cell proliferation, particularly in 3D-printed models. Moreover, modification in the form of methacrylation encourages reduced toxicity of the biomaterial in 3D constructs. Our investigation demonstrates the suitability of a chitosan-enhanced biomaterial, specifically methacrylate-treated, for application in tissue engineering, and particularly for tissues requiring resistance to high stress, i.e., vascular or cartilage models. Distal ulna locking bone plates (DLBPs) are commonly employed in the treatment of distal ulna fractures. However, commercially available metallic bone plates experience stress shielding and lack corrosion resistance. Poly lactic acid (PLA) is highly favored biopolymer due to its biocompatible and bioabsorbable nature with human tissues. The use of additive layer manufacturing (ALM) is gaining attention for creating customized implants with intricate structures tailored to patient autonomy. ALM‐based PLA bone plates must provide high resistance against impact and torsional forces, necessitating the adjustment of printing process parameters. This study focuses on examining the influence of key printing parameters, on the impact strength and torque‐withstanding capability of DLBPs. Experimental results, along with microscopic images, reveal that an increase in infill density (IF) and wall thickness imparts strong resistance to layers against crack propagation under impact and torsional loads. On the contrary, an increase in layer height and printing speed leads to delamination and early fracture of layers during impact and torsional testing. IF significantly contributes to improving the impact strength and torque‐withstanding capability of DLBPs by 70.53% and 80.65%, respectively. Hazardous pollution of water sources is a major impediment to the supply of usable and potable water around the world. Economical, innovative, renewable and environmentally friendly technologies are needed for the treatment of wastewater. Untreated waste materials, like date palm flower stalks (PFS) collected directly from trees, can serve as adsorbents for removing textile dyes. Batch kinetic and equilibrium experiments performed to investigate the effects of various factors, including contact time (0 – 180 min), adsorbent dose (10–50 mg / 25 mL), initial pH (2 – 12), initial concentration of dye (0 – 200 mg/L) and temperature (15 – 60 °C). The equilibrium capability achieved after 90 min of biosorption was 38.522 mg/g. The Freundlich and Langmuir isotherm models were fitted to the experimental data and their goodness of fit for adsorption was compared. Langmuir isotherm (R2 ~ 0.998) best described the equilibrium of SRL sorption on PSF. Adsorption was found to be greater under highly basic conditions (pHpzc = 6.3). The thermodynamic parameters such as Gibbs free energy ΔG°, enthalpy change ΔH°, and entropy ΔS° were calculated. Adsorption Gibbs free energy (ΔG°) was calculated to be in the range -8.49 to -8.62 kJ / mol. The values of activation parameters such as enthalpy (ΔH°) and entropy (ΔS°) were also determined as -7.76 kJ / mol and 2.49 J / mol.K, respectively. Alginates are salts of alginic acid derived mainly from sea algae of the genus brown algae. They are also synthesized by some bacteria. They belong to negatively charged polysaccharides exhibiting some rheological properties. High plasticity and the ability to modify the structure are the reasons for their application in numerous industries. This work involves additively manufactured Ti-6Al-4V alloys, which are widely used in automobile, biomedical, and aircraft components for a comparison of the microstructure–properties relationship between electron beam melted (EBM) and selective laser melted (SLM) alloys after hot isostatic pressing treatment. We carried out microstructural, mechanical, and electrochemical measurements on both alloys. They showed comparable α and β phase contents with slightly higher lattice parameters in the EBM sample compared to the SLM. The EBM sample showed higher yield strength and uniform elongation due to the activation of multistage defects-driven strengthening and strain hardening mechanisms. Cracking during the tensile test nucleated mainly at the α phase near high-mechanical mismatch α/β interfaces. This mechanism was consistent with the reported generation of hetero-deformation-induced strengthening and strain hardening. Both alloys showed similar electrochemical behavior, but the SLM sample was more susceptible to corrosion than the EBM alloy. [ABSTRACT FROM AUTHOR]

Teeth have excellent mechanical properties, with high wear resistance and excellent fracture resistance. This is due to their well-organized multilevel hierarchical structure. While a number of studies in the last decades have revealed the relationship between tooth structure and mechanical properties, there is still no general agreement on how different orientations of hydroxyapatite (HAp) fibers affect the mechanical properties of enamel. With a scanning electron microscope and nanoindenter, the orientations of HAp fibers and their properties were investigated. HAp fibers have two different orientations: parallel and perpendicular to the surface. Fibers oriented parallel to the surface exhibited higher hardness, elastic modulus and wear resistance. Under applied force, fibers oriented perpendicular to the surface suffered deeper shearing in the protein along the long axis, resulting in lower mechanical properties. Teeth resist damaging fractures by combining hard and soft structures. Background: The mandible of the rabbit is considered a reliable model to be used to study bone regeneration in defects. The aim of the present study was to evaluate the formation of new bone around implants installed in defects of either 5 or 10 mm in the mandible of rabbits. Materials and methods: In 12 rabbits, 3 mm deep circumferential defect, either 5 or 10 mm in diameter, were prepared bilaterally and an implant was placed in the center. A collagen membrane was placed to close the entrance. After 10 weeks, biopsies were taken, histological slides were prepared, and different regions of the defects were analyzed. Results: Similar amounts of new bone were found in both defects. However, most of the 5 mm defects were filled with new bone. New bone was observed closing the entrance of the defect and laid onto the implant surface. Only in a few cases the healing was incomplete. Despite a similar percentage of new bone found within the 10 mm defects, the healing was incomplete in most of the cases, presenting a low rate of bone formation onto the implant surface within the defect. Only one case presented the closure of the entrance. Conclusions: The dimensions of the defect strongly influenced the healing so that a circumferential marginal defect of 10 mm around an implant in the mandible body should be considered a critical-sized defect. In recent years, many different biomedical implants have been created for prolonged usage within the human body. The number of these implants has been steadily expanding. It is a common practice to use metallic bioimplants as substitutes when attempting to replace human body components in an anatomically acceptable manner. But the metallic bioimplant was mostly made biocompatible by putting a biocompatible coating on its surface and then using the right surface modification techniques. This was the primary method by which the biocompatibility of the bioimplant was achieved. Thermal spraying is a method that is frequently employed in surface modification treatments to modify metallic bioimplants. These methods can be found in the medical field. This is due to the adaptability of the thermal spraying method. Because of their capacity to respond and adapt to physiochemical circumstances, coatings that are based on hydroxyapatite (HAp) are the most popular type to be used in thermal spraying. This is due to the fact that HAp is a naturally occurring mineral. On the other hand, the majority of the HAp coatings are unsuccessful as a result of the coating’s weak mechanical qualities and insufficient adhesive strength. Supplying the coating in question with reinforcement in the form of hard particles and doing so in the appropriate proportions makes it feasible to alter the qualities of HAp-based coatings to fulfill a variety of requirements. This can be accomplished by providing the coating with reinforcement. The purpose of this inquiry was to deconstruct and analyze a variety of thermally sprayed (TS) coatings. The musculoskeletal system, which is responsible for weight‐bearing, movement, and organ protection, faces many disorders arising from injuries, diseases, and trauma that affect millions of people worldwide, resulting in a decreased quality of life and socioeconomic burden. Tissue engineering is at the forefront of current research on tissue regeneration and demonstrates great potential for musculoskeletal tissue repair. Among the numerous grafts available, adhesive hydrogels have demonstrated potential for tissue applications. Despite the surge in the development of bioadhesive hydrogel formulations in recent years, the absence of an evaluation protocol for their formulation has led to the emergence of numerous similar products that do not fully meet the clinical requirements for applicability in musculoskeletal tissue regeneration. This review aims to address this gap by first discussing the design considerations for an ideal bioadhesive hydrogel relevant to successful musculoskeletal tissue repair. By thoroughly reviewing recent research advances in bioadhesive hydrogels, with a particular focus on their applications in facilitating musculoskeletal tissue repair, improvements are proposed in the current evaluation criteria for the development of novel bioadhesive hydrogels for musculoskeletal tissue applications, and several key challenges and research directions for their implementation are summarized. Granular hydrogels have emerged as a promising class of biomaterials in medical research, enabling independent control of matrix stiffness within a porous biomaterial. Such microgel packings comprise interconnected pores and high surface‐to‐volume ratios. These features facilitate cell viability and nutrient diffusion, which are critical in enabling tissue regeneration. Despite the current interest in granular hydrogels for tissue engineering applications, only a few in vitro platforms are used to investigate cell interactions, limiting their design, and translation. In this study, microfluidic platforms able to reproducibly confine and immobilize microgels without the need for secondary cross‐linking are developed. Protocols are established for the generation of human bone marrow‐derived mesenchymal stem/stromal cells (hBM‐MSC)‐infiltrated microporous substrates and early‐time responses of cells to their environment are studied. Further, a tissue invasion assay is established, where cells infiltrate granular materials at different rates depending on growth factor presence or material properties. This platform is compatible with a range of different granular materials, and it is envisioned to have significant utility as a pre‐clinical tool for the rational design of materials for tissue healing applications. This research delves into the development of a hydrogel based on sodium alginate (Na‐Alg), CMTKG and PSA for the controlled release of Metformin Hydrochloride (Metformin HCl), a commonly used drug for the treatment of type 2 diabetes. The hydrogel was synthesized via a free radical mechanism using N, N′‐methylene‐bis(acrylamide) (MBA) as a crosslinker and potassium persulphate (KPS) as an initiator. The hydrogels were analyzed using techniques such as ATR‐FTIR, and SEM. The research also investigated swelling behaviour, drug loading, drug encapsulation efficiency, and release kinetics of Metformin HCl. The finding revealed that the hydrogel exhibited pH‐responsive metformin HCl release, with higher drug release perceived at pH 7.4 compared to pH 1.2. Mathematical models were employed to study the drug release mechanism, indicating diffusion as a mechanism liable for the drug release from the hydrogel. Moreover, the synthesized metformin HCl‐loaded Na‐Alg/PSA/CMTKG hydrogel shows excellent anti‐diabetic activity on α‐amylase and α‐glucosidase enzyme. Henceforth, the formulated hydrogel reveals potential for facilitating the targeted release of Metformin HCl, offering opportunities for improving patient amenability and lessening side effects related to frequent dosing. Human hair, composed primarily of keratin, represents a sustainable waste material suitable for various applications. Synthesizing keratin nanoparticles (KNPs) from human hair for biomedical uses is particularly attractive due to their biocompatibility. In this study, keratin was extracted from human hair using concentrated sulfuric acid as the hydrolysis agent for the first time. This process yielded KNPs in both the supernatant (KNPs-S) and precipitate (KNPs-P) phases. Characterization involved scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), Zeta potential analysis, X-ray diffraction (XRD), and thermogravimetric analysis (TG). KNPs-S and KNPs-P exhibited average diameters of 72 ± 5 nm and 27 ± 5 nm, respectively. The hydrolysis process induced a structural rearrangement favoring β-sheet structures over α-helices in the KNPs. These nanoparticles demonstrated negative Zeta potentials across the pH spectrum. KNPs-S showed higher cytotoxicity (CC50 = 176.67 µg/mL) and hemolytic activity, likely due to their smaller size compared to KNPs-P (CC50 = 246.21 µg/mL), particularly at concentrations of 500 and 1000 µg/mL. In contrast, KNPs-P did not exhibit hemolytic activity within the tested concentration range of 32.5 to 1000 µg/mL. Both KNPs demonstrated cytocompatibility with fibroblast cells in a dose-dependent manner. Compared to other methods reported in the literature and despite requiring careful washing and neutralization steps, sulfuric acid hydrolysis proved effective, rapid, and feasible for producing cytocompatible KNPs (biomaterials) in single-step synthesis. The natural physiological response to skin injury is wound healing. However, to restore skin continuity, wound healing is a complicated process that involves the collaboration of a variety of cell types and other mediators. This process ultimately results in tissue regeneration and the restoration of skin barrier function. Hydrogels are appealing dosage forms for biomedical regenerative medicine since they are composed of 3D networks with high water content and flexible rheological features. Hydrogels that can self-heal are particularly interesting for wound treatment because they can autonomously restore their original functionalities and repair structural damage. Recently, the use of self-healing hydrogels as biomedical materials has attracted increased interest. In this review, the self-healing systems used in tissue regeneration, especially wound healing, will be explored. A focus on the fabrication methods, characterization tests, and mechanism of self-healing will be introduced, along with the biomedical applications of self-healing hydrogels loaded with conventional and therapeutic biomaterials. Amphiphilic peptides hold great potential as drug delivery systems. A popular peptide design approach has been to place amino acids in the peptide sequence based on their known properties. On the other hand, the directed discovery approach aims to screen a sequence space for a desired property. However, screening amphiphilic peptides for desirable drug delivery properties is not possible without a quantity that is predictive of these properties. We studied the predictive power of critical aggregation concentration (CAC) values on the drug delivery performance of a series of amphiphilic peptides with different hydrophobic tails and close CAC values. The CAC values were predicted by our previously developed model and doxorubicin was used as a model hydrophobic drug. All peptides showed close drug loading, entrapment efficiency, and release profile. They also formed similar spherical particles by assembling in reverse β -sheet arrangements regardless of drug presence. Moreover, the assembled particles were able to accumulate doxorubicin inside ordinary as well as drug-resistant breast cancer cells and enhance its toxicity up to 39 and 17 folds, respectively. It can be concluded that similar drug delivery properties displayed by the peptides can be attributed to their similar hydrophilic-lipophilic balance as reflected in their close CAC values. Preclinical and clinical studies have demonstrated that precision therapy has a broad variety of treatment applications, making it an interesting research topic with exciting potential in numerous sectors. However, major obstacles, such as inefficient and unsafe delivery systems and severe side effects, have impeded the widespread use of precision medicine. The purpose of drug delivery systems (DDSs) is to regulate the time and place of drug release and action. They aid in enhancing the equilibrium between medicinal efficacy on target and hazardous side effects off target. One promising approach is biomaterial-assisted biotherapy, which takes advantage of biomaterials' special capabilities, such as high biocompatibility and bioactive characteristics. When administered via different routes, drug molecules deal with biological barriers; DDSs help them overcome these hurdles. With their adaptable features and ample packing capacity, biomaterial-based delivery systems allow for the targeted, localised, and prolonged release of medications. Additionally, they are being investigated more and more for the purpose of controlling the interface between the host tissue and implanted biomedical materials. This review discusses innovative nanoparticle designs for precision and non-personalised applications to improve precision therapies. We prioritised nanoparticle design trends that address heterogeneous delivery barriers, because we believe intelligent nanoparticle design can improve patient outcomes by enabling precision designs and improving general delivery efficacy. We additionally reviewed the most recent literature on biomaterials used in biotherapy and vaccine development, covering drug delivery, stem cell therapy, gene therapy, and other similar fields; we have also addressed the difficulties and future potential of biomaterial-assisted biotherapies. Recent developments in biomaterials have resulted in the creation of cement composites with potential wound treatment properties, even though they are currently mainly employed for bone regeneration. Their ability to improve skin restoration after surgery is worth noting. The main purpose of this research is to evaluate the ability of composite cement to promote wound healing in a rat experimental model. Full-thickness 5 mm skin defects were created, and the biomaterials were applied as wound dressings. The hybrid light-cured cement composites possess an organic matrix (Bis-GMA, TEGDMA, UDMA, and HEMA) and an inorganic phase (bioglasses, silica, and hydroxyapatite). The organic phase also contains γ-methacryloxypropyl-trimethoxysilane, which is produced by distributing bioactive silanized inorganic filler particles. The repair of the defect is assessed using a selection of macroscopic and microscopic protocols, including wound closure rate, histopathological analysis, cytotoxicity, and biocompatibility. Both composites exerted a favorable influence on cells, although the C1 product demonstrated a more extensive healing mechanism. Histological examination of the kidney and liver tissues revealed no evidence of toxicity. There were no notable negative outcomes in the treated groups, demonstrating the biocompatibility and efficacy of these bioproducts. By day 15, the skin of both groups had healed completely. The progress in developing biomaterials for cartilage replacement is still not sufficient, and researchers face the challenge of developing materials that can imitate the physical, chemical, and mechanobiological characteristics of natural cartilage. In this research, the graphene oxide was blended with polyvinyl alcohol/chitosan composite to fabricate composite hydrogels (PVA/Chitosan/GO) using different concentrations of graphene oxide (0, 0.75, 1.5, 2.25, and 3 wt%). The images taken with a scanning electron microscope (SEM) displayed that adding graphene oxide affected the hydrogel structure by reducing the porosity. The composite hydrogel with 3 wt% graphene oxide exhibited the highest compressive strength of 2.15 MPa, and the storage modulus was significantly greater than that of polyvinyl alcohol/chitosan, which satisfied the application requirements. Furthermore, the composite hydrogels displayed excellent antimicrobial activity against Escherichia coli and Staphylococcus aureus bacterial species. Composites are fabricating by reinforcing waste bambara nut shell powder (BNSP) as a bio-filler into an epoxy resin matrix. The composite is fabricated by varying the mass fractions of filler in the range of 5–35 wt%. The effects of bio-filler content on physical, mechanical, and thermal properties are evaluated. The studies reveal that the tensile strength, tensile modulus, flexural strength, and impact strength increased with increasing bio-filler content. The highest mechanical properties of BNSP-loaded epoxy composites are achieved at bio-filler mass content of 15 wt%, whereas hardness increased for 5–35 wt% with peak value at 35 wt%. The water absorption and thickness swelling of the BNSF/epoxy composites increased with increasing bio-filler contents, while the density decreased with an increase in bio-filler content. The TGA results revealed that an increasing bio-filler content decreased thermal stability, whereas the Tg and Tm of the composites increased with increasing bio-filler content. In this research, hydrogels based on chitosan, pectin, and salt (NaCl) were synthesized through the formation of polyelectrolyte complexes (PECs). The synthesis parameters, including pH, salinity, and polymer concentration, were varied to explore their influence. Weight and texture analysis revealed differences in hydrogel morphology. Swelling behavior studies showed hydrogels synthesized at pH 4 exhibiting higher swelling capacities. Additionally, the presence of salt affected the formation process. Thermal characterization showed a first decomposition step occurring around 180–224 °C. Morphological testing using SEM highlighted differences in pore size and distribution, notably when salt was included in the formulation (pore wall diameter without NaCl, 2.2 ± 1.1 um, with NaCl, 4.7 ± 1.2 um). Physico-chemical tests, including Zeta potential, FTIR, and XRD, provided insights into interactions within the hydrogels: hydrogen bonds and electrostatic interactions. Moreover, antibacterial tests demonstrated efficacy against Escherichia coli and Staphylococcus aureus, with varying inhibition degrees correlated with NaCl content (halo for E. coli without NaCl, 8 and 10 mm; with NaCl, 10 and 15 mm). Further assessments, including water vapor transmission rate (WVTR) and lidocaine release assays, highlighted hydrogel potential for wound dressing applications, with suitable moisture retention properties and controlled drug release capabilities. The release percentage achieved by the hydrogel with 0.15 M NaCl was higher than without salt (111.1% ± 9.5% and 31.16% ± 15.13%, respectively). Porous materials are pivotal in emerging fields like tissue engineering, scaffold, and drug delivery due to their distinctive porosity-driven functional properties. This paper describes how to achieve multiscale porosity in food-based composites through a thermally activated gelatinization process of amylopectin molecules coupled with 3D printing. By controlling printing paths, macropores are engineered, while degree of gelatinization governs micro- and nanopores formation. Process-microstructure relationship reveals that longer preheating treatments at higher gelatinization temperatures significantly reduce micro-pore area by over twofold and nanopore surface area by over threefold. This study unveils a sustainable, easily recyclable biocomposite, leveraging the dynamic nature of covalently adaptive bonds in a vitrimer matrix. The fabrication involved a fatty acid-derived vitrimer as the polymer matrix and multi-layered, nonwoven flax mat as reinforcing scaffold. The incorporation of these fibers significantly improved the mechanical performance of the vitrimer matrix uniformly. The ester-based covalently adaptive network plays a crucial role in enabling exceptional fiber-matrix bonding, as well as recyclability. The vitrimer matrix dissolves in ethylene glycol through transesterification, facilitating complete material recovery and biocomposite recycling without compromising the original properties of the matrix and reinforcing fibers. Extrusion is extensively used due to its ability to produce intricate products that maintain a uniform cross-sectional shape throughout their complete length. Beyond manufacturing sectors, the extrusion process extends its application into the biomedical domain. Collagen, a biocompatible substance, is prominently used in biomedical applications due to its inherent properties. The biocompatible nature of magnesium facilitates its application in production of bone fixtures, screws, bone plates and stents. Material extrusion presents a unique approach towards advancement in additive manufacturing by utilization of metals, polymers, ceramics, composites and smart materials for implant fabrication. The extrusion technique's ability to fabricate patient specific and low-cost implants makes it an important element in enhancing patient outcomes. In orthopedics, this process is utilized to fabricate various types of bone plates, screws, intramedullary nails and other implants used in fracture fixation and joint replacement surgeries. This study provides an insight knowledge about the materials used in extrusion process of biomedical implants. It also discusses about various orthopedic and orthodontic implants produced using conventional and extrusion based additive manufacturing processes. The extrusion based additive manufacturing technique poses a variety of challenges such as proper configuration of printing parameters, methodology for effective utilization of diverse material availability, long printing time for layer-by-layer fabrication, post processing requirements, cross contamination between materials on application of multi extruder system and continuous alignment of tools during the fabrication process. New vaccine platforms that activate humoral immunity and generate neutralizing antibodies are required to combat emerging pathogens, including influenza virus. A slurry of antigen-loaded hydrogel microparticles that anneal to form a porous scaffold with high surface area for antigen uptake by infiltrating immune cells as the biomaterial degrades is demonstrated to enhance humoral immunity. Antigen-loaded-microgels elicited a robust cellular humoral immune response, with increased CD4+ T follicular helper (Tfh) cells and prolonged germinal center (GC) B cells comparable to the commonly used adjuvant, aluminum hydroxide (Alum). Increasing the weight fraction of polymer material led to increased material stiffness and antigen-specific antibody titers superior to Alum. Vaccinating mice with inactivated influenza virus loaded into this more highly cross-linked formulation elicited a strong antibody response and provided protection against a high dose viral challenge. By tuning physical and chemical properties, adjuvanticity can be enhanced leading to humoral immunity and protection against a pathogen, leveraging two different types of antigenic material: individual protein antigen and inactivated virus. The flexibility of the platform may enable design of new vaccines to enhance innate and adaptive immune cell programming to generate and tune high affinity antibodies, a promising approach to generate long-lasting immunity.

Abstract Background Treatment of articular cartilage injury remains a challenging clinical problem in orthopedics. Chitosan-derived biomaterial could be a potential adjuvant treatment to improve cartilage repair. In the current study, we examined the effects of two potential chitosan-derived materials on cartilage regeneration of osteochondral defects in rabbits. Methods An osteochondral defect was created over the rabbit knee and treated using three approaches: group A received no material (n = 24), group B received chitosan membranes with glucose absorption (CGA; n = 25), and group C received chitosan-glucose derivative membranes obtained via the Maillard reaction (CGMR; n = 25). Cartilage repair over the osteochondral defect was analyzed 12 weeks post-surgery via histological analysis, immunostaining, and reverse transcription-qualitative polymerase chain reaction (RT-qPCR) for type-I and type-II collagen mRNA. Surface changes of biomaterials are essential for aligning with the biological system’s dynamics and enhancing the effectiveness of bioimplants. Customized surface alterations based on the material’s bonding ability, biocompatibility, and interactions with host cells may have a substantial impact. This investigation uses hydroxyapatite (HAp) with 0, 10, 20, 30, and 40 wt% silver as a thin coating layer on stainless steel (SST) 316 L by electrophoretic deposition preparation at 30 V and 30 min coating duration. The coating’s crystallinity, morphology, and microstructure have been investigated using structural characterization methods such as X-ray diffraction, Scanning electron microscopy, and energy dispersive spectroscopy. The resistance to corrosion of uncoated and coated SST substrates has been assessed using potentiodynamic polarization experiments. The results show that the HAp-nanosilver coating layer increases the SST’s resistance to corrosion in Ringer solution. The HAp 10% silver-coated SST displays a reduction in corrosion current density. These further demonstrate the potential for using HAp and silver-coated stainless steel as a surgical instrument to increase biocompatibility and resistance to corrosion.

This study reported the biological changes occurring after γ-irradiation of in vivo rat model and the osteochondral protective effect of Gelatine-Chitosan-Ginger (GEL-CH-GING). The results showed that Electron Paramagnetic Resonance (EPR) Spectroscopy of GEL-CH-GING showed two paramagnetic centres which correspond to g=2.19 and g= 2.002. The Fourier transform infrared spectroscopy (FTIR) analyses revealed an increase in peak intensity at C–H chains, as well as, C=O carbonyl groups. The X-ray diffraction (XRD) analysis showed no change of crystallinity. After gamma ray exposure, the rat groups have received an osteochondral defect and then were treated with GEL-CH-GING composite. Sixty days post-surgery, a significant reduction in thiobarbituric acid-reactive compounds (TBARs) was seen when compared to non-implanted rat group. Concerning oxidative stress status, GEL-CH-GIN significantly improved Superoxide Dismutase (SOD) 76 nmol/l, Catalase (CAT) 0.79 nmol/l, and Glutathione Peroxidase (GPx) 1.77 nmol/l activities in osteochondral tissue.
