Inherited channelopathies are a clinically and heritably heterogeneous group of disorders that result from ion channel dysfunction. The aim of this study was to characterize the clinicopathologic features of a Belgian Blue x Holstein crossbred calf with paradoxical myotonia congenita, craniofacial dysmorphism, and myelodysplasia, and to identify the most likely genetic etiology. The calf displayed episodes of exercise-induced generalized myotonic muscle stiffness accompanied by increase in serum potassium. It also showed slight flattening of the splanchnocranium with deviation to the right side. On gross pathology, myelodysplasia (hydrosyringomielia and segmental hypoplasia) in the lumbosacral intumescence region was noticed. Histopathology of the muscle profile revealed loss of the main shape in 5.3% of muscle fibers. Whole-genome sequencing revealed a heterozygous missense variant in KCNG1 affecting an evolutionary conserved residue (p.Trp416Cys). The mutation was predicted to be deleterious and to alter the pore helix of the ion transport domain of the transmembrane protein. The identified variant was present only in the affected calf and not seen in more than 5200 other sequenced bovine genomes. We speculate that the mutation occurred either as a parental germline mutation or post-zygotically in the developing embryo. This study implicates an important role for KCNG1 as a member of the potassium voltage-gated channel group in neurodegeneration. Providing the first possible KCNG1-related disease model, we have, therefore, identified a new potential candidate for related conditions both in animals and in humans. This study illustrates the enormous potential of phenotypically well-studied spontaneous mutants in domestic animals to provide new insights into the function of individual genes.

Hernias of intervertebral discs are a common canine disease that is usually treated surgically. Recently, a histological scoring system for surgically removed canine intervertebral herniated discs has been developed by scoring the lesions of both the anulus fibrosus (AF) and the nucleus pulposus (NP). Since the proportion of AF and NP in the surgical samples may vary, depending on the surgical approach, the aim of this study is to grade separately the lesions in AF and NP and to modify the previously described scoring system adding three parameters: inflammation, mineralization and neovascularization. Possible association of the modified grading system with clinical parameters were statistically assessed. Herniated disc material was collected from 27 dogs. AF was present in 10/27 cases and was classified as grade 2 in 4 cases, grade 3 in 5 cases and grade 4 in 1 case. The NP was present in all 27 cases and was classified as grade 2 in 1 case, grade 3 in 5 cases, grade 4 in 9 cases and grade 5 in 12 cases. Blood supply is essential for development and growth of tumors and angiogenesis is the fundamental process of new blood vessel formation from preexisting ones. Angiogenesis is a prognostic indicator for a variety of tumors, and it coincides with increased shedding of neoplastic cells into the circulation and metastasis. Several molecules such as cell surface receptors, growth factors, and enzymes are involved in this process. While antiangiogenic therapy for cancer has been proposed over 20 years ago, it has garnered much controversy in recent years within the scientific community. The complex relationships between the angiogenic signaling cascade and antiangiogenic substances have indicated the angiogenic pathway as a valid target for anticancer drug development and VEGF has become the primary antiangiogenic drug target. This review discusses the basic and clinical perspectives of angiogenesis highlighting the importance of comparative biology in understanding tumor angiogenesis and the integration of these model systems for future drug development.

This study compared heat shock proteins Hsp60, Hsp72 and Hsp73, along with p63 and androgen receptor (AR) immunoexpression between 16 cases of benign prostatic hyperplasia (BPH) and 11 prostatic carcinomas (PCa) in dogs. The proportion of Hsp60-positive cells was higher in PCa compared with BPH (P =0.033), whereas the frequency and intensity of Hsp73 immunostaining did not differ significantly between the two groups. Hsp72-immunostained nuclei formed a discontinuous layer along the basement membrane in BPH, whereas cells in this layer in PCa were negative or weakly positive. Hsp72 nuclear score showed significant positive associations with both p63 (P =0.016) and AR (P =0.009) scores. Double immunofluorescence revealed Hsp72-p63 and Hsp72-AR co-expressions in basal cell nuclei. Aberrant cytoplasmic p63 immunolabelling was observed in 3 of 11 PCa cases. These results suggest a role of the combined expression of Hsp72, p63 and AR in basal epithelial cells in canine BPH and PCa.

A 23-year-old female rhesus macaque presented with a 2.5-cm diameter, fi rm, moveable, lobulated subcutaneous mass associated with a supranummary teat on the right side of the chest. Th is animal was a retired breeder, currently in an aging study. No exogenous hormone treatments were noted in the animal’s history. Chest radiographs were within normal limits. Blood screens showed no noteworthy variations from normal. Needle aspirate cytology showed clusters of neoplastic cells. Grossly the mass was well circumscribed, fi rm, and homogeneously tan, with a glandular appearance. Diff erential diagnoses included sebaceous or mammary adenoma, carcinoma in situ, and lobular or ductular carcinoma. Histopathology was consistent with a mammary ductal carcinoma with comedo pattern. Subsequent needle aspirate cytology from an adjacent right axillary lymph node showed tumor cells with a few lymphoid cells, interpreted as lymphatic spread. Chest radiographs 2 and 6 weeks postbiopsy showed no evidence of pulmonary metastasis. After 1 year, there was no marked change on chest radiographs, but a small cluster of new nodules was palpable in the right axillary region. Histopathology of an excisional biopsy of the new nodules indicated tumor growth subjacent to regional lymph nodes. Further treatment was not performed and the animal remained clinically normal fi ve years after the initial diagnosis. Spontaneous mammary neoplasia is a major concern in human medicine, yet it rarely has been reported to occur in nonhuman primates. Th is case is important in documenting an additional case of spontaneous mammary tumor development.

I meccanismi di fisiopatologia respiratoria sono prevalentemente conseguenti a componenti broncospastiche e bronchiolitiche che portano ad alterazione di diversi parametri polmonari (resistenza delle vie aeree, compliance, compliance dinamica, rapporto ventilazione/perfusione, volume residuo) condizionanti, nella manifestazione progredita della malattia, un enfisema alveolare. This paper is a review of the most recent findings concerning the etiopathogenesis and respiratory pathophyslology mechanisms in Recurrent Airway Obstruction (RAO) of the horse. From the etiopathogenetical point of view RAO is characterized by an excessive recruitment and a delay in the elimination of neutrophils that enhances the pulmonary damage and a unbalance in the composition of lymphocyte Th subpopulations deputed to prime the immunopathologic reaction. The pathophysiological mechanisms are due prevalently by to bronchospasm and bronchiolitis, which lead to the alteration of different respiratory parameters (airway resistance, compliance, dynamic compliance, ventilation/perfusion ratio, residual volume) and influence the production of alveolar emphysema in the late stages of the disease.
