Aedes albopictus (Skuse, 1894) has a widespread distribution that includes both temperate and tropical climates. It is a potential vector of several important worldwide arboviruses, including chikungunya, zika, and dengue, and its geographical distribution expands rapidly. Despite its vectorial importance, the bioecological features of this species in newly established environments are still not very well known. Thus, this study investigated phenotypic variations among Ae. albopictus populations from different regions in Turkey. This is the first comparative morphological study of Ae. albopictus populations in Turkey. The procrustes distances phenogram derived from wing data indicates that the shape differences among some populations and the population from the Aegean coast differ from the Black Sea populations. Size differences were also detected between some populations.

Vaccaria hispanica (Mill.) Rauschert (Caryophyllaceae) doğada yaygın şekilde yetişen, özellikle triterpenoid saponin ve kaliteli nişasta içeriği çok zengin bir doğal bitkidir. Bu tez çalışmasında literatürde henüz ele alınmamış bir konu olan V. hispanica'da somatik embriyogenesis ile ilgili olarak somatik embriyogenesisin ilk basamağı olan embriyojenik kallusların elde edilmesi amaçlanmıştır. Bu amaçla biri nişasta (Genotip A), diğeri saponin (Genotip B) içeriği yüksek iki V. hispanica genotipinin hipokotil, kotiledon, boğum arası ve yaprak eksplantları, iki grup denemede çok sayıda besin ortamında kültüre alınmıştır. Bu ortamların bazal besin ortamları I. grup kallus denemelerinde MS ve NN ortamları, II. grup kallus denemelerinde ise sadece MS ortamı olup, bazal besin ortamlarına oksin olarak 2,4-D veya NAA nın 0, 1 ve 2 mg/L ile sitokinin olarak da BA veya TDZ nin 0, 0.5, 1 ve 2 mg/L lik konsantrasyonlarından oluşan bitki gelişim düzenleyicileri (BGD) çeşitli kombinasyonlarda ilave edilmiştir. Çalışmada bu ortamların iki V. hispanica genotipine ait farklı eksplant tiplerinde; kallus (%) ve embriyojenik kallus oluşturma (%) ile çeşitli kallus özellikleri ve ayrıca kalluslardan meydana gelebilecek farklı morfolojik yapıları oluşturma üzerine etkileri araştırılmıştır.Çalışmanın sonuçlarına göre V. hispanica'nın kallus, embriyojenik kallus ve çeşitli morfolojik yapıları oluşturma yönünden çok yüksek rejenerasyon kapasitesine sahip olduğu belirlenmiştir. Bu kapasite; hiçbir hormonun kullanılmadığı kontrol ortamlarında bile çok yüksek oranda kallus, yoğun kök yapıları ve sürgün oluşması nedeniyle, büyük ölçüde V. hispanica'nın yüksek olduğu tahmin edilen içsel hormon seviyelerinden kaynaklanmaktadır. Çeşitli BGD lerin kullanıldığı tüm ortamlarda ise V. hispanica'nın hem A hem de B genotiplerinde içsel oksin düzeyinin de yüksek olması nedeniyle her iki genotipin bütün eksplantlarında oldukça yüksek oranlarda kallus meydana gelmiştir. Bu nedenle V. hispanica'da sadece kallus oluşturma yönünden belirli bir ortam önerisinde bulunmak mümkün değildir. Ancak bu kalluslara embriyojenik özellik kazandırılabilmesi için, hazırlanacak ortamlara oksin olarak 2,4-D ve sitokinin olarak TDZ nin belirli kombinasyonlarda ilave edilmesinin gerekli olduğu belirlenmiştir. Genotip A ve Genotip B'nin tüm eksplantlarında aynı anda en yüksek oranda embriyojenik kallus oluşturabilmek için önerilebilecek en etkili ortamın, MS + 2 mg/L 2.4-D + 0.5 mg/L TDZ içeriğine sahip ortam olduğu tespit edilmiştir. Çalışmada elde edilen morfolojik yapılara gelince; çoğunluğu globular aşamada en yüksek embriyo oluşumu her iki genotipte MS + 2 mg/L 2,4-D içeren ortamda meydana gelmiştir. En fazla embriyo oluşumu miktar olarak Genotip A da yaprak, Genotip B de ise kotiledon ve onu takiben yaprak eksplantlarından elde edilmiştir. En fazla sürgün oluşumu Genotip A da hem I., hem de II. grup denemelerde BGD içermeyen kontrol ortamında kültüre alınan boğum arası eksplantlarında, Genotip B de ise yaprak eksplantlarında gerçekleşmiştir. Farklı yapıdaki köklenmeler yönünden Genotip A ve Genotip B'de; en yüksek orandaki kısa kök oluşumlarının genel olarak sitokininsiz 2,4-D içeren ortamlarda, uzun ve gerçek kök oluşumlarının ise oksinsiz NAA içeren ortamlarda oluştuğu dikkat çekmiştir. Bu çalışmada elde edilen özellikle köklenmelere ait detaylı bulguların, içerik yönünden zengin V. hispanica'da ileride yapılabilek in vitro sekonder metabolit çalışmaları için de faydalı olması umulmaktadır. Vaccaria hispanica (Mill.) Rauschert (Caryophyllaceae) is a natural plant that grows widely in nature, especially rich in triterpenoid saponin and quality starch. In this thesis, it is aimed to obtain embryogenic calluses which is the first step of somatic embryogenesis related to somatic embryogenesis in V. hispanica which has not been yet studied in the literature. For this purpose hypocotyl, cotyledon, internodal and leaf explants of two V. hispanica genotypes with high content of starch (Genotype A) and saponin (Genotype B) were cultured in several culture media in two groups of trials. The basal nutrient media of these media were MS and NN media in I. group of callus experiments and only MS medium in II. group callus experiments. Plant growth regulators (BGDs) consisting of 2,4-D or NAA as auxin at 0, 1 and 2 mg/L and BA or TDZ as cytokinin at 0, 0.5, 1 and 2 mg/L of were added to the basal nutrient media in various combinations. The effects of these media on the formation of callus (%), embryogenic callus (%), different callus characteristics and morphological structures that may occur from callus were investigated in different explant types of two V. hispanica genotypes.According to the results of the study, V. hispanica has a very high regeneration capacity in terms of forming callus, embryogenic callus and various morphological structures. This capacity is largely due to internal hormone levels, which are estimated to be high in V. hispanica, due to the high rate of callus, dense root structures and shoot formation formed even in control cultures in which no hormone was used. In all cultures containing various BGDs, very high rates of callus formed in all explants of both genotypes due to the high internal auxin level of both A and B genotypes of V. hispanica. Therefore, it is not possible to propose a specific culture content in V. hispanica only in terms of callus formation. However, in order to give embryogenic properties to these calluses, it has been determined that 2,4-D as auxin and TDZ as cytokinin should be added in certain combinations. It was specified that the most effective medium was MS + 2 mg / L 2.4-D + 0.5 mg / L TDZ to produce high level of embryogenic callus in all explants of Genotype A and Genotype B.As for the morphological structures obtained in the study, the highest embryo formation mostly in the globular stage occurred in medium containing MS + 2 mg / L 2,4-D in both genotypes. The maximum embryo formation was obtained from the leaf explant in Genotype A and from the cotyledons followed by leaf explants in Genotype B. The highest shoot formation took place in the internodal explants cultured in the control cultures of both I. and II. group of experiments in Genotype A while in the leaf explants in Genotype B. In regard of different types of rooting in Genotype A and Genotype B, it was noted that the highest rate of short root formation occured mostly in the media containing 2,4-D without cytokinin while the highest long and true root formation in the media containing NAA without auxin. It is hoped that the detailed findings especially on rooting obtained in this study will be useful for in vitro secondary metabolite studies in V. hispanica, which has rich content. 104

Doğal sistemlerde sivrisineklerin konak tür tercihlerinin belirlenmesi patojen-vektör-konak üçlüsü arasındaki karmaşık ekolojik ve evrimsel ilişkilerin aydınlatılması ve vektör kaynaklı hastalıkların kontrolünde önemlidir. Sivrisineklerde konak tercihinin genetik temelli olmasına rağmen, konak türlerinin bulunabilirliğine ve ulaşılabilirliğine göre bu konak tercih durumu esneklik gösterebilir.Bu tez çalışması 2012-2013 yılları arasında sivrisinek üreme dönemlerinde, Aras Havzası'ndan örneklenmiş sivrisinek türlerinin konak tür tercihlerinin Polimeraz Zincir Reaksiyonu (PZR) temelli, Reverse Line Blotting (RLB) yöntemi kullanılarak tespit edilmesi amaçlanmıştır. Konak tercihinin belirlenmesine yönelik bu çalışma, Aras Havzası'ndan toplanan sivrisineklerle ilk kez yapılmıştır. Toplanan kan emmiş dişi sivrisineklerin abdomeninden elde edilen DNA'lara, omurgalıların 344 baz çiftlik sitokrom b bölgelerini amplifiye eden biotin ile işaretlenmiş genel primerler ile PZR yapılmıştır. PZR sonucu pozitiflik veren 7 farklı sivrisinek türünden toplam 916 bireyin konak tercihleri Reverse Line Blotting (RLB) yöntemi ile tespit edilmiştir.PZR-RLB hibridizasyon analizlerine göre, pozitif sonuç veren 654 bireyin 323 (%49,4)'ünün tek konak üzerinden beslendiği bulunmuştur. Tek konak üzerinden beslenen sivrisineklerin 219 (%68) tanesininin insan, 97 (%30,1) tanesinin sığır, 5 (%1,5) tanesinin koyun, 1 (%0,3) tanesinin at ve 1 (%0,3) tanesinin güvercin konakları üzerinden beslendiğini tespit edilmiştir. Diğer 331 (%50,6) bireyin karışık beslendiği, 290 (%44,3) tanesinin iki, 30 (%4,6) tanesinin üç, 10 (%1,6) tanesinin dört ve 1 (%0,15) tanesinin de beş konak türü üzerinden beslendikleri belirlenmiştir.Bu çalışma ile alanda bulunan sivrisineklerin insan üzerinden beslenme oranlarının oldukça fazla olduğu, bu durumun sivrisinek kaynaklı patojenlerin bulaşım ve yayılımında ciddi problemlere neden olabileceği öngörülmektedir. Alanda sivrisinek kaynaklı hastalık bulaşımından kaçınmak ya da azaltmak için herhangi bir koruyucu önlem alınmaması bu sonucu doğurmaktadır. Çalışma sonucu olarak alanda sivrisinek kaynaklı hastalıkların araştırılmasının aciliyeti gerekmektedir. Despite the fact that the host preference in mosquitoes is genetical it also shows flexibility due to availability and accessibility of host species.In this project we aimed determination of host preference of the mosquito species of Aras Valley via blood meal analysis using the Polimerase Chain Reaction (PCR) based Reverse Line Blotting method. The proposed study aimed the determination of host preference of mosquitoe species in this location is done for the first time. DNA, extracted from blood fed mosquiotes collected from the field, were subjected to PCR with biotine labelled universal primers that amplify the 344 base pair cytochrome b gene region. Bloodmeal sources identified from 916 mosquitoes representing 7 different species by Reverse Line Blotting (RLB).According to the Reverse Line Blotting (RLB) hybridization results of 654 polymerase chain reaction positive individuals, it was observed that 323 (%49,4) of the mosquitoes fed on single hosts. From these single hosts, it was determined that 219 (%68) fed on human, 97 (%30,1) fed on cow, 5 (%1,5) fed on sheep, 1 (%0,3) fed on equus and 1 (0,3) fed on pigeon. It was determined that other 331 (%50,6) individiuals prefered fed on multiple hosts and 290 (%44,3) of them took double, 30 (%4,6) of them took triple, 10 (%1,6) of them took quadruple and 1 (%0,15) them took quintette blood meal.In this study it was shown that blood feeding habits of mosquitoes on human is quite high in this area and therefore, this may cause a high risk for transmission and spread of the mosquito-borne pathogens to humans. This may be the consequence of humans not taking any precaution to avoid or to reduce the contact with mosquitoes. As a result, this study shows the urgent requirement of investigating mosquito-borne diseases in this area. Türkiye-Ermenistan sınırında, Temmuz 2007'de, birbirini izleyen dokuz gece boyunca (akşam 18:15'den sabah 05:45'e kadar), açık ve kapalı alanda insana saldıran sivrisinekler toplandı. Dokuz sivrisinek türünden oluşan toplam 1005 dişi örnek yakalandı. Yakalanan türler arasında en dominant tür Ochlerotatus dorsalis Meigen, 1830 (yakalanan toplamın %47,46) olup, bunu sırasıyla Anopheles hyrcanus Palas,1771 (%22,89), Culex theileri Theobald, 1903 (%9,25) Aedes vexans Meigen, 1830 (%6,57), Ochlerotatus caspius Palas, 1771 (%4,88), Anopheles maculipennis Meigen, 1818 (%3,08), Culex territans Walker, 1856 (%2,79), Coqullettidia richiardii Ficalbi, 1889 (%1,59) ve Culex pipiens L., 1758 (%1,49) takip etmektedir. Toplam yakalanan örneklerin açık alan/kapalı alan oranı 4,4'tür. Bu oranlar sırasıyla, Cq. richardii'de 15, Ae. vexans'ta 10 ve Oc. dorsalis'te 7,2 iken, An.hyrcanus'ta 1,94, An. maculipennis'te 1,58 olarak bulunmuştur. Oc .caspius, Oc .dorsalis ve Cq .richiardii'nin populasyon yoğunluğu akşam (19:15-19:45) ve sabah alaca kanlığı (04:15-04:45) periyotlarında en yüksek noktaya ulaştı. Ae. vexans'ın populasyon yoğunluğu, karanlık çöktükten sonraki periyotta (20:15-20:45) arttı ve en yüksek pikine sabah alacakaranlığında (04:15-04:45) ulaştı. An. hyrcanus ve Cx. theileri'nin ısırma aktivitesi, akşam karanlık çöker çökmez (20:15-20:45) en yüksek noktaya ulaşmaktadır. An. maculipennis'in büyük çoğunluğu gecenin ikinci yarısında toplandı.Anahtar Kelimeler: Anopheles hyrcanus, ısırma aktivitesi, sıtma, Aras Vadisi During nine consecutive nights (from 18:15 to 05:45 h), landing-biting on man mosquitoes were collected outdoor and indoor in July 2007 in Turkey-Armenia border. A total of 1,005 female specimens were collected consisting of nine species. The most dominant species was Ochlerotatus dorsalis Meigen, 1830 (47.46% of total catch) followed by Anopheles hyrcanus Pallas, 1771 (22.89%), Culex theileri Theobald, 1903 (9.25%), Aedes vexans Meigen, 1830 (6.57%), Ochlerotatus caspius Pallas, 1771 (4.88%), Anopheles maculipennis Meigen, 1818 (3.08%), Culex territans Walker, 1856 (2.79%), Coquillettidia richiardii Ficalbi, 1889 (1.59%) and Culex pipiens L., 1758 (1.49%). Outdoor/indoor ration of total caught specimens was 4.40. While these ratios were 15 for Cq. richiardii, 10 for Ae. vexans and 7.2 for Oc. dorsalis, these ratios were found as 1.94 for An. hyrcanus and 1.58 for An. maculipennis. The population density of Oc. caspius, Oc. dorsalis and Cq. richiardii reached their peaks at dusk (19:15-19:45 h) and dawn (04:15-04:45 h) twilight periods. The population density of Ae. vexans increased soon after getting dark (20:15-20:45 h), and it reached its highest peak at dawn (04:15-04:45 h). The peak biting time for An. hyrcanus and Cx. theileri was the first period immediately after getting dark (20:15-20:45 h). A great many of An. maculipennis was collected at the second half of the night.Key words: Anopheles hyrcanus, biting activity, malaria, Ararat Valley 64
