Abstract Background Effluents from Food Services Establishments (FSEs) contain primarily Fats, Oil and Grease (FOG) which severely impact on sewers and the environment when released in high concentrations. In Trinidad & Tobago, it is estimated that approximately 231,304 kg/day of unaccounted for FOG bearing wastewaters from FSEs, are released into the environment with no viable treatment in the country. This research explored the optimization of physico-chemical processes for the treatment of FOGs for subsequent release into sewers. Results Bench-scale studies analysed the characteristics of FSE’s effluents from three popular sources, conducted the treatment of these effluents using Jar Tests, and subsequently confirm results via a pilot plant study. Characterization showed the mean concentration of the parameters examined to be; FOG (511 mg/l ± 116 mg/l), Suspended Solids (446 mg/l ± 146 mg/l), Chemical Oxygen Demand (2229 mg/l ± 963 mg/l) and pH (6 ± 0.3). Jar Tests were conducted using Poly-aluminium Chloride (PACl) as coagulant, anionic and cationic polyelectrolytes as flocculant aids with suitable pH adjustments of samples to determine the isoelectric point for the coagulant. Effluent results showed FOG removal levels of 99.9% and final effluent concentration of 0.17 mg/l. This was attained using PACl concentration of 250 mg/l, a 0.1% low cationic polyelectrolyte (CP 1154) at 4 mg/l with the pH of sample adjusted to 8. The pilot plant achieved a 97.4% removal of FOG (residual of 16.8 mg/l) using the same coagulant dosing, and pH value, but increasing the strength of the flocculant aid to 0.1% medium cationic (CP1156) at 5 mg/l. Conclusion Experimentation showed high concentrations of emulsified FOG can be efficiently removed to levels below the permissible requirements (20 mg/l) for entry into sewer systems in Trinidad and Tobago using coagulation, flocculation and sedimentation techniques. Pilot scale study also revealed that a higher strength and/or dose of the cationic polyelectrolyte and increased times in primary and final tanks were required to attain the desired results as in the bench level study, where equipment limitations in the flocculation tank were faced. This is in alignment with theory where factors critical for agglomeration is equipment type and density charge. It is, concluded that the optimum combination of chemicals and the respective dosages attained at the bench level study should prove effective should the right equipment be made available.

Background: Aeromonas hydrophila is a widely recognized broad-spectrum pathogen that primarily targets the gastrointestinal tract. Type IV pili (T4P) are proteinaceous nano-machines located on the bacterial cell surface, playing a crucial role in host colonization and infection. Regrettably, the T4P systems of A. hydrophila remain largely underexplored. Methods: A. hydrophila genomes with complete genome assembly and annotation reports up to 31 March 2023, were obtained from the NCBI Genome database or KEGG genome database, followed by a global search for T4P secretion system genes. Protein sequences of these manually curetted genes were used as secondary quarry for Synteny analysis. Protein–protein interaction analysis was performed by string analysis and in silico study of genomic islands. Results: We identified 27 orthologs of type IV pili (T4P) nano-machine components in A. hydrophila. These orthologs are primarily distributed across three operons: pilABCD, pilMNOPQ, and pilVWXY. While the first two operons are commonly found in all experimental genomes, the presence of the pilVWXY operon, coding for 11 orthologs, is reported here for the first time in A. hydrophila. Notably, the complete pilVWXY operon is absent in nonvirulent strains. A genomic islands study between a nonvirulent and hypervirulent strain also confirms absence of most of the genes coded by pilVWXY in nonvirulent strain. Interestingly, among the 51 experimental genomes analyzed, the pilVWXY operon was completely absent in 10 strains, most of which are categorized as nonvirulent; Conclusions: The distribution of two major type IV pili (T4P) nano-machines, PilABCDMNOPQ and PilVWXY, is reported here for the first time in A. hydrophila. The digestion process in fish is a complex phenomenon and is accomplished with the help of both endogenous and exogenous enzymes. A wide range of enzymes have been reported in fish and their secretion and concentrations highly depend on feeding habits, gut structure and environmental conditions. In this present investigation, we have demonstrated the activity of two endogenous enzymes (protease and amylase) in four economically important air-breathing fish species (Clarias batrachus, Channa punctatus, Anabas testudineus and Heteropneustes fossilis) at different seasonal temperatures. Our results demonstrated that protease activity was higher in all fish during the month of May, followed by September and December. It was also detected that enzyme activity was higher in the hindgut region (HG) compared to the foregut (FG), which suggests that HG is more active in digestion compared to FG. The optimization of feeding efficiency in aquaculture industries is very important, as the cost of feed is high and its availability is also limited. The information presented may be of importance when developing optimized diets and feeding regimes for the species considered.

The composition of resident microbes in the human body is linked to various diseases and their treatment outcomes. Although studies have identified pancreatic ductal adenocarcinoma (PDAC)-associated bacterial communities in the oral and gut samples, herein, we hypothesize that the prevalence of microbiota in pancreatic tumor tissues is different as compared with their matched adjacent, histologically normal appearing tissues, and these microbial molecular signatures can be highly useful for PDAC diagnosis/prognosis. In this study, we performed comparative profiling of bacterial populations in pancreatic tumors and their respective adjacent normal tissues using 16S rRNA-based metagenomics analysis. This study revealed a higher abundance of Proteobacteria and Actinomycetota in tumor tissues compared with adjacent normal tissues. Interestingly, the linear discriminant analysis (LDA) scores unambiguously revealed an enrichment of Delftia in tumor tissues, whereas Sphingomonas, Streptococcus, and Citrobacter exhibited a depletion in tumor tissues. Furthermore, we analyzed the microbial composition between different groups of patients with different tumor differentiation stages. The bacterial genera, Delftia and Staphylococcus, were very high at the G1 stages (well differentiated) compared with G2 (well to moderate/moderately differentiated) and G3/G4 (poorly differentiated) stages. However, the abundance of Actinobacter and Cloacibacterium was found to be very high in G2 and G3, respectively. Additionally, we evaluated the correlation of programmed death-ligand (PDL1) expression with the abundance of bacterial genera in tumor lesions. Our results indicated that three genera such as Streptomyces, Cutibacterium, and Delftia have a positive correlation with PD-L1 expression. Collectively, these findings demonstrate that PDAC lesions harbor relatively different microbiota compared with their normal tumor adjacent tissues, and this information may be helpful for the diagnosis and prognosis of PADC patients. Introduction: The worldwide higher prevalence of obesity is currently one of the most important reasons for the occurrence of several health disorders, along with an increased risk of mortality and morbidity. However, the magnitude of the effect of sub-maximal aerobic exercise on cardiac autonomic and electrical function among Indian obese individuals has yet to be established. Aim: To evaluate the effect of exercise on ECG components and their correlation with Body Mass Index (BMI) among young asymptomatic obese and non-obese individuals, in order to assess the risk for developing Cardiovascular Diseases (CVD) in the future. Materials and Methods: An observational cross-sectional study was conducted at the Autonomic Research Laboratory of the Physiology Department at RG Kar Medical College and Hospital, Kolkata, between January 2017 to December 2017. Two hundred healthy medicos were divided into two groups: Obese (BMI =25 kg/m²; n=88) and non-obese (BMI =18.5 to =23 kg/m²; n=112). Resting Blood Pressure (BP) was manually recorded, followed by the recording of long lead-II ECG in the supine posture both at rest and ten minutes after submaximal (50% VO2max) aerobic exercise on a treadmill using the modified Bruce protocol and following the Astrand-Rhyming nomogram. The HR, QT interval, and RR interval were recorded from the lead-II ECG. The measured QT interval was corrected using Bazett's formula (QTC=QT/vRR). The results were analysed using both unpaired and paired t-tests and the Pearson correlation coefficient in Graphpad-Quickcalc software. Results: Significant prolongation of the QTc interval both at rest (p-value: 0.019) and after exercise (p-value: 0.044), along with delayed Heart Rate Recovery (HRR) (p-value: 0.001), were observed among the obese compared to the non-obese population. Additionally, a significantly positive linear correlation was observed between BMI and ECG components at rest (BMIRHR: r:+0.293; p: 0.005 and BMI-QTc Interval: r:+0.226; p-value: 0.034) and during the recovery state (BMI-HRR: r:+0.42; p-value: 0.00004 and BMI-QTc Interval: r:+0.365; p-value: 0.0004), only among the obese. Conclusion: At rest, obese individuals had an elevated QTc, which could be due to alterations in cardiac autonomic function, along with a significant positive correlation between BMI and RHR, indicating lower aerobic fitness in this population. Prolongation of the QTc interval, delayed decrease in HR, and a significant positive correlation with BMI after exercise in obese individuals indicated myocardial repolarisation instability, autonomic dysfunction, and an increased risk of out-of-hospital sudden cardiac events in these individuals. A laboratory-scale investigation at ambient temperature was performed to examine the applicability of a four-stage across-the-flow rotating biological contactor (RBC) reactor for the treatment of synthetic wastewater containing phenol and thiocyanate. The composition was so maintained that the phenol to thiocyanate ratio approximated that usually found in coal carbonization effluent. In the combined treatment of the toxicants, it was noted that the toxicants were removed in sequence. Phenol was mostly removed in the earlier (first and second) stages, while thiocyanate-fed microbes acted more efficiently in the latter stages (third and fourth). The overall reaction rate coefficient for phenol biodegradation ([K.sub.PH]) varied in the range of 10.20-3.50 g/[m.sup.2] [multiplied by] d in presence of 0-240 g/[m.sup.3] of influent thiocyanate concentration. The same coefficient for thiocyanate biodegradation ([K.sub.TH]) was found to be in the range of 8.20-0.30 g/[m.sup.2] [multiplied by] d in presence of 0-240 g/[m.sup.3] of influent phenol concentration. The effects of various operating and process parameters such as disk rotational speed, temperature of the reactor liquid, and substrate load on the removal of phenol and thiocyanate were also examined. The microbes, predominantly found in the earlier stages, were identified as Pseudomonas putida, Pseudomonas stutzeri and Candida tropicalis, whereas in the latter stages, Thiobacillus thioparus and Nocardia sp. populations predominated.
