Abstract

Moringa oleifera is a perennial medicinal and edible plant, and polyphenols as one of its main active ingredients. This study reviews the current status of development of the polyphenol types and contents of different parts, polyphenol extraction methods, polyphenol efficacy, and polyphenol applications of Moringa oleifera. Additionally, the current research on the antioxidant, anti-inflammatory, antibacterial, antiviral, anticancer, and hypoglycemic properties of polyphenols from Moringa oleifera is critically examined. Abstract

Background: Unbalanced gut microbiota is considered to cause dysfunction of the gut barrier function. Lactobacillus johnsonii MG, isolated from mouse feces as gut associating lactobacilli, enhanced gut barrier function in Caco‐2 cells via interaction with JAM‐2 in tight junctions. In this study, the anti‐inflammatory effects of L. johnsonii MG were investigated in a colitis mouse model developed by treating mice with 3% dextran sulfate sodium (DSS) for 9 days. Results: MG treatment of colitis mice resulted in fast recovery of the body weight and showed significant improvement in the disease activity index and histopathological scores. The histomorphological score increased by DSS treatment was significantly improved in the MG‐treated group. In the intestine, the expression of Ocln, Zo1, Itga6, Lama3, and Jam2 genes, which are involved in tight junction functions, were significantly upregulated in MG‐treated colitis mice. In the microflora analysis, the abundance of Ruminococcaceae, Bacteroides, and Lachnospiraceae were reduced in DSS‐treated mice and recovered by MG treatment. Conclusion: We reported the potential of L. johnsonii MG in the regeneration of crypts and integrity of matrix proteins in the gut of mice with colitis through its association with tight junctions. 22. Exploration of cyclooxygenase-2 inhibitory peptides from walnut dreg proteins based on in silicoand in vitroanalysis

Abstract

Walnut dreg protein hydrolysates (WDPHs) exhibit a variety of biological activities, however, the cyclooxygenase-2 (COX-2) inhibitory peptide of WDPHs remain unclear. The aim of this study was to rapidly screen for such peptides in WDPHs through a combination of in silicoand in vitroanalysis. In total, 1262 peptide sequences were observed by nano liquid chromatography/tandem mass spectrometry (nano LC-MS/MS) and 4 novel COX-2 inhibitory peptides (AGFP, FPGA, LFPD, and VGFP) were identified. Enzyme kinetic data indicated that AGFP, FPGA, and LFPD displayed mixed-type COX-2 inhibition, whereas VGFP was a non-competitive inhibitor. This is mainly because the peptides form hydrogen bonds and hydrophobic interactions with residues in the COX-2 active site. These results demonstrate that computer analysis combined with in vitroevaluation allows for rapid screening of COX-2 inhibitory peptides in walnut protein dregs.

Pretreatment with MG enhanced barrier function and showed protective effect against Enterococcus faeciumprovided damage. MG treatment increased the gene expressions of transcriptional regulator NFKBand major tight junction protein, ZO-1.

Abstract

Osteoporosis is characterized by decreased bone mass, microarchitectural deterioration, and increased bone fragility. High-fat diet (HFD)-induced obesity also results in bone loss, which is associated with an imbalanced gut microbiome. However, whether HFD-induced obesity or HFD itself promotes osteoclastogenesis and consequent bone loss remains unclear. In this study, we developed HFD-induced obesity (HIO) and non-obesity (NO) mouse models to evaluate the effect of HFD on bone loss. NO mice were defined as body weight within 5% of higher or lower than that of chow diet fed mice after 10 weeks HFD feeding. NO was protected from HIO-induced bone loss by the RANKL /OPG system, with associated increases in the tibia tenacity, cortical bone mean density, bone volume of cancellous bone, and trabecular number. This led to increased bone strength and improved bone microstructure via the microbiome-short-chain fatty acids (SCFAs) regulation. Additionally, endogenous gut-SCFAs produced by the NO mice activated free fatty acid receptor 2 and inhibited histone deacetylases, resulting in the promotion of Treg cell proliferation in the HFD-fed NO mice; thereby, inhibiting osteoclastogenesis, which can be transplanted by fecal microbiome. Furthermore, T cells from NO mice retain differentiation of osteoclast precursors of RAW 264.7 macrophages ex vivo. Our data reveal that HFD is not a deleterious diet; however, the induction of obesity serves as a key trigger of bone loss that can be blocked by a NO mouse-specific gut microbiome. Abstract

In this work, hot gas filters composed of polyimide nanofiber membrane sandwiched between carbon woven fabrics are fabricated to capture fine particles of PM 2.5 level. Polyimide (PI) nanofibers with an average diameter of 190 nm are electrospun on supporting carbon fabrics as filtration layers. The filtration performance tests show that the maximum filtration efficiency reaches 99.99% for PM 2.5, while the maximum pressure drop is only 251.86 Pa after continuously testing for 25 min under a constant flow rate of 20 L/min. As the areal density of the PI nanofiber membrane increases, the filtration efficiency increases first and then levels off when the areal density reaches 11.64 g/m2. In regeneration performance test with back air flush at a pressure of 500 KPa, the composite filter maintains a filtration efficiency of 99.99% and a pressure drop of about 410 Pa. After heat treatment at 260 and 300 °C, the composite filter shows a relatively high filtration efficiency while the tensile strength of the carbon fabric does not change significantly.

40. Walnut Protein Peptides Ameliorate DSS-Induced Ulcerative Colitis Damage in Mice: An in SilicoAnalysis and in VivoInvestigation

Abstract

Walnut (Juglans regiaL.) is a food with food-medicine homology, whose derived protein peptides have been shown to have anti-inflammatory activity in vitro. However, the effects and mechanisms of walnut protein peptides on ulcerative colitis (UC) in vivohave not been systematically and thoroughly investigated. In this study, we applied virtual screening and network pharmacology screening of bioactive peptides to obtain three novel WPPs (SHTLP, HYNLN, and LGTYP) that may alleviate UC through TLR4-MAPK signaling. In vivostudies have shown that WPPs improve intestinal mucosal barrier dysfunction and reduce inflammation by inhibiting activation of the TLR4-MAPK pathway. In addition, WPPs restore intestinal microbial homeostasis by reducing harmful bacteria (Helicobacterand Bacteroides) and increasing the relative abundance of beneficial bacteria (Candidatus_Saccharimonas). Our study showed that the WPPs obtained by virtual screening were effective in ameliorating colitis, which has important implications for future screening of bioactive peptides from medicinal food homologues as drugs or dietary supplements.
