Abstract

Abstract We present a detailed study of a scotogenic model accommodating dark matter, neutrino masses and the anomalous magnetic moment of the muon while being consistent with the existing constraints on flavour violating decays of the leptons. Moreover, this model offers the possibility to explain the baryon asymmetry of the Universe via leptogenesis. We determine the viable regions of the model’s parameter space in view of dark matter and flavour constraints using a Markov Chain Monte Carlo setup combined with a particular procedure to accommodate neutrino masses and the anomalous magnetic moment of the muon at the same time. We also discuss briefly the resulting collider phenomenology.

Abstract In the general Two-Higgs Doublet Model it has been shown that the Higgs potential can be expressed in terms of gauge-independent quantities that transform covariantly under unitary mixing of the doublets. In particular, stability, electroweak symmetry breaking, and CP symmetry can be understood in a concise way, avoiding unphysical gauge degrees of freedom. We complete this program and show how all the masses, the trilinear and quartic scalar interactions, the gauge-boson-Higgs interactions, as well as the Yukawa couplings in the general THDM can be expressed in a gauge-invariant way.

Abstract

Heavy metals in model conditions as well as industrial pollution launch disturbances in neural cells of different animals and human beings. The neurotoxicity of mercury, which is one of the most toxic heavy metals, has been studied for several decades. However, its low doses chronic exposure effects for neural tissue cells are still poorly understood. Therefore, the basic molecular mechanisms of mercury should be clarified. The purpose of our research is to clarify the mechanism of mercury genotoxicity, the role of the DNA repair protein apurinic/apyrimidinic endonuclease 1 (APE1) in neural tissue cells, and the response to inorganic mercury-induced neurotoxicity. In our model, we used juvenile rainbow trout exposed to mercury chloride with a range of doses 9‑36 µg/L for 60 days to study the cytotoxicity of chronic exposure. We detected the reactive oxygen species (ROS) production as an index of oxidative stress and APE1 as a marker of cellular DNA damage response in a neural cell. The ROS level was measured by using the fluorometric method based on 2',7'-dichlorofluorescein diacetate reaction. The analyses of markers of the DNA repair (APE1) and apoptosis (B cell lymphoma-2 anti-apoptotic protein – Bcl-2) were carried out with western blotting. The mercury chloride chronic exposure induced statistically significant upregulation of the ROS production in the fish brain. Contrary, the mercury low doses stimulated the downregulation of APE1 expression in the brain tissue. Furthermore, mercury chronic exposure inhibited the expression of Bcl-2 in the animals treated with 18 and 36 µg/L mercury chloride. The harmful effect of mercury could be promoted by oxidative stress generation. The downregulation of APE1 expression could lead to a lack of DNA damage response efficacy and initiate the decline in neural cell functioning. Obtained data on the APE1 expression have shown that the neurotoxic effect of mercury could be mediated, at least partially, by the decline in cellular DNA damage response in the brain. The evaluation of decrease in DNA repair response via detection of the APE1 expression can be a prospective tool to reveal the deleterious effects of toxicants in terms of their neuro- and genotoxicity.

4. A new flavour imprint of SU(5)-like grand unification and its LHC signatures

Abstract

We point out that the hypothesis of an SU(5)-like supersymmetric Grand Unified Theory (GUT) implies a generic relation within the flavour structure of up-type squarks. Contrary to other well-known SU(5) relations between the down-quark and charged lepton sectors, this relation remains exact in the presence of any corrections and extra operators. Moreover it remains valid to a good precision at the electroweak scale, and opens thus new possibilities for testing SU(5)-like GUTs. We derive the low-energy effective theory of observable light up-type squarks, that also constitutes a useful tool for squark phenomenology. We use this effective theory to determine how to test SU(5) relations at the LHC. Focusing on scenarios with light stops, compatible with Natural SUSY, it appears that simple tests involving ratios of event rates are sufficient to test the hypothesis of an SU(5)-like GUT theory. The techniques of charm-tagging and top-polarimetry are a crucial ingredient of these tests.

Abstract

Catabacter hongkongensis was isolated and cultured from human blood for the first time in Scandinavia. The patient, an 83-year-old man from Dalarna, Sweden, recovered without antibiotic treatment, although a high mortality rate associated with C. hongkongensis infection had been reported from China, Canada and France. The genome of the strain ABBA15k was sequenced, assembled and analysed. In contrast to the type strain of the species HKU16T, no antibiotic resistance was observed in Scandinavian strain ABBA15k. The strain was deposited as CCUG 68271, and the draft genome sequence is available from the DNA Data Bank of Japan (DDBJ), the European Molecular Biology Laboratory (EMBL), and GenBank under the accession number LLYX00000000.

Abstract

Improved data on biosphere-atmosphere exchange are fundamental to understanding the production and fate of ammonia (NH3) in the atmosphere. The GRAMINAE Integrated Experiment combined novel measurement and modelling approaches to provide the most comprehensive analysis of the interactions to date. Major inter-comparisons of micrometeorological parameters and NH3 flux measurements using the aerodynamic gradient method and relaxed eddy accumulation (REA) were conducted. These showed close agreement, though the REA systems proved insufficiently precise to investigate vertical flux divergence. Grassland management had a large effect on fluxes: emissions increased after grass cutting (−50 to 700 ng m−2 s−1 NH3) and after N-fertilization (0 to 3800 ng m−2 s−1) compared with before the cut (−60 to 40 ng m−2 s−1). Effects of advection and air chemistry were investigated using horizontal NH3 profiles, acid gas and particle flux measurements. Inverse modelling of NH3 emission from an experimental farm agreed closely with inventory estimates, while advection errors were used to correct measured grassland fluxes. Advection effects were caused both by the farm and by emissions from the field, with an inverse dispersion-deposition model providing a reliable new approach to estimate net NH3 fluxes. Effects of aerosol chemistry on net NH3 fluxes were small, while the measurements allowed NH3-induced particle growth rates to be calculated and aerosol fluxes to be corrected. Bioassays estimated the emission potential Γ = [NH4+]/[H+] for different plant pools, with the apoplast having the smallest values (30–1000). The main within-canopy sources of NH3 emission appeared to be leaf litter and the soil surface, with Γ up to 3 million and 300 000, respectively. Cuvette and within-canopy analyses confirmed the role of leaf litter NH3 emission, which, prior to cutting, was mostly recaptured within the canopy. Measured ammonia fluxes were compared with three models: an ecosystem model (PaSim), a soil vegetation atmosphere transfer model (SURFATM-NH3) and a dynamic leaf chemistry model (DCC model). The different models each reproduced the main temporal dynamics in the flux, highlighting the importance of canopy temperature dynamics (Surfatm-NH3), interactions with ecosystem nitrogen cycling (PaSim) and the role of leaf surface chemistry (DCC model). Overall, net above-canopy fluxes were mostly determined by stomatal and cuticular uptake (before the cut), leaf litter emissions (after the cut) and fertilizer and litter emissions (after fertilization). The dynamics of ammonia emission from leaf litter are identified as a priority for future research.

Abstract

A new biophysical model SURFATM-NH3, simulating the ammonia (NH3) exchange between terrestrial ecosystems and the atmosphere is presented. SURFATM-NH3 consists of two coupled models: (i) an energy budget model and (ii) a pollutant exchange model, which distinguish the soil and plant exchange processes. The model describes the exchanges in terms of adsorption to leaf cuticles and bi-directional transport through leaf stomata and soil. The results of the model are compared with the flux measurements over grassland during the GRAMINAE Integrated Experiment at Braunschweig, Germany. The dataset of GRAMINAE allows the model to be tested in various meteorological and agronomic conditions: prior to cutting, after cutting and then after the application of mineral fertilizer. The whole comparison shows close agreement between model and measurements for energy budget and ammonia fluxes. The major controls on the ground and plant emission potential are the physicochemical parameters for liquid-gas exchanges which are integrated in the compensation points for live leaves, litter and the soil surface. Modelled fluxes are highly sensitive to soil and plant surface temperatures, highlighting the importance of accurate estimates of these terms. The model suggests that the net flux depends not only on the foliar (stomatal) compensation point but also that of leaf litter. SURFATM-NH3 represents a comprehensive approach to studying pollutant exchanges and its link with plant and soil functioning. It also provides a simplified generalised approach (SVAT model) applicable for atmospheric transport models.

The exchange of ammonia between crop canopies and the atmosphere depends on a range of plant parameters and climatic conditions. However, little is known about effects of management factors. We have here investigated the stomatal ammonia compensation point in response to cutting and fertilization of a grass sward dominated by Lolium perenne. Tall grass had a very low NH3 compensation point (around 1 nmol mol−1), reflecting the fact that leaf nitrogen (N) concentration was very low. During re-growth after cutting, leaf tissue concentrations of NO3−, NH4+, soluble N and total N increased along with apoplastic NH4+ concentrations. In contrast, apoplastic pH decreased resulting in largely unaltered NH3 compensation points. Nitrogen fertilization one week after cutting caused the apoplastic NH4+ concentration of the newly emerging leaves to increase dramatically. The NH3 compensation point peaked between 15 and 25 nmol mol−1 the day after the fertiliser was applied and thereafter decreased over the following 10 days until reaching the same level as before fertilisation. Ammonium concentrations in leaf apoplast, bulk tissue and litter were positively correlated (P=0.001) throughout the experimental period. Bulk tissue NH4+ concentrations, total plant N and soil NH4+ concentrations also showed a positive correlation. A very high potential for NH3 emission was shown by the plant litter.

Species diversity in grasslands usually declines with increasing input of nitrogen from fertilizers or atmospheric deposition. Conversely, species diversity may also impact the build-up of soil and plant nitrogen pools. One important pool is NH3/NH4+ which also can be exchanged between plant leaves and the atmosphere. Limited information is available on how plant-atmosphere ammonia exchange is related to species diversity in grasslands. We have here investigated grass species abundance and different foliar nitrogen pools in 4-year-old intensively managed grassland. Apoplastic pH and NH4+ concentrations of the 8 most abundant species (Lolium perenne, Phleum pratense, Festuca pratensis, Lolium multiflorum, Poa pratensis, Dactylis glomerata, Holcus lanatus, Bromus mollis) were used to calculate stomatal NH3 compensation points. Apoplastic NH4+ concentrations differed considerably among the species, ranging from 13 to 117 μM, with highest values in Festuca pratensis. Also apoplastic pH values varied, from pH 6.0 in Phleum pratense to 6.9 in Dactylis glomerata. The observed differences in apoplastic NH4+ and pH resulted in a large span of predicted values for the stomatal NH3 compensation point which ranged between 0.20 and 6.57 nmol mol−1. Three species (Lolium perenne, Festuca pratensis and Dactylis glomerata) had sufficiently high NH3 compensation point and abundance to contribute to the bi-directional NH3 fluxes recorded over the whole field. The other 5 grass species had NH3 compensation points considerably below the atmospheric NH3 concentration and were thus not likely to contribute to NH3 emission but only to NH3 uptake from the atmosphere. Evaluated across species, leaf bulk-tissue NH4+ concentrations correlated well (r2=0.902) with stomatal NH3 compensation points calculated on the basis of the apoplastic bioassay. This suggests that leaf tissue NH4+ concentrations combined with data for the frequency distribution of the corresponding species can be used for predicting the NH3 exchange potential of a mixed grass sward.

10. Vertical structure and diurnal variability of ammonia exchange potential within an intensively managed grass canopy

Stomatal ammonia compensation points (χs) of grass species on a mixed fertilized grassland were determined by measurements of apoplastic [NH4+] and [H>+] in the field. Calculated χs values were compared with in-canopy atmospheric NH3 concentration (χa) measurements. Leaf apoplastic [NH4+] increased by a factor of two from the lowest level in the canopy to the top level. Bulk leaf [NH4+] and especially [NO3−] slightly increased at the bottom of the canopy and these concentrations were very high in senescent plant litter. Calculated χs values were below atmospheric χs at all canopy levels measured, indicating that the grassland was characterized by NH3 deposition before cutting. This was confirmed by the χa profile, showing the lowest χa close to the ground (15 cm above soil surface) and an increase in χa with canopy height. Neither χs nor χa could be measured close to the soil surface, however, the [NH4+] in the litter material indicated a high potential for NH3 emission. A diurnal course in apoplastic [NH4+] was seen in the regrowing grass growing after cutting, with highest concentration around noon. Both apoplastic and tissue [NH4+] increased in young grass compared to tall grass. Following cutting, in-canopy gradients of atmospheric χa showed NH3 emission but since calculated χs values of the cut grass were still lower than atmospheric NH3 concentrations, the emissions could not entirely be explained by stomatal NH3 loss. High tissue [NH4+] in the senescent plant material indicated that this fraction constituted an NH3 source. After fertilization, [NH4+] increased both in apoplast and leaf tissue with the most pronounced increase in the former compared to the latter. The diurnal pattern in apoplastic [NH4+] was even more pronounced after fertilization and calculated χs values were generally higher, but remained below atmospheric [NH3].

Abstract

ObjectivesThe clinical and public health relevance of widespread case finding by testing for asymptomatic chlamydia infections is under debate. We wanted to explore future directions for chlamydia control and generate insights that might guide for evidence-based strategies. In particular, we wanted to know the extent to which we should pursue testing for asymptomatic infections at both genital and extragenital sites.MethodsWe synthesised findings from published literature and from discussions among national and international chlamydia experts during an invitational workshop. We described changing perceptions in chlamydia control to inform the development of recommendations for future avenues for chlamydia control in the Netherlands.ResultsDespite implementing a range of interventions to control chlamydia, there is no practice-based evidence that population prevalence can be reduced by screening programmes or widespread opportunistic testing. There is limited evidence about the beneficial effect of testing on pelvic inflammatory disease prevention. The risk of tubal factor infertility resulting from chlamydia infection is low and evidence on the preventable fraction remains uncertain. Overdiagnosis and overtreatment with antibiotics for self-limiting and non-viable infections have contributed to antimicrobial resistance in other pathogens and may affect oral, anal and genital microbiota. These changing insights could affect the outcome of previous cost–effectiveness analysis.ConclusionThe balance between benefits and harms of widespread testing to detect asymptomatic chlamydia infections is changing. The opinion of our expert group deviates from the existing paradigm of ‘test and treat’ and suggests that future strategies should reduce, rather than expand, the role of widespread testing for asymptomatic chlamydia infections.

41. Evaluating the Safety of Intra-Articular Mitotherapy in the Equine Model: A Potential Novel Treatment for Osteoarthritis

No current treatments available halt osteoarthritis progression in horses or humans. Intra-articular injection of mitochondria is a novel treatment that has the potential to improve cell metabolism and decrease inflammation, but safety of this treatment has yet to be established in the horse. Autologous blood-derived mitochondria isolated using a commercially available kit were injected into the left carpus joint of 3 horses which were monitored for 28 days. Horses received physical examinations, video recorded gait evaluations, joint diameter measurement, synovial fluid collection, and blood collection on day 0 (baseline prior to mitotherapy, day of mitochondria injection), 1, 3, 7, 14, and 28. Systemic inflammation was assessed via complete blood count, fibrinogen, and plasma serum amyloid A (SAA). Local inflammation was assessed via synovial fluid cytology and physical examination parameters. Physical exam parameters remained stable and no joint swelling was observed after mitotherapy. No change was noted in video recorded gait evaluations as determined by a blinded evaluator. Complete blood counts revealed no significant increase in white blood cells. SAA only increased mildly in 1 horse. Fibrinogen became slightly elevated above reference range in 2 horses at day 7, but later normalized. Mild increases in synovial fluid nucleated cell counts and total protein occurred on day 1 and 3, but resolved within 7 days without intervention. Autologous mitochondria injection into the equine intercarpal joint was well tolerated with no signs of inflammation. This safety information allows for future studies evaluating mitotherapy efficacy.

Abstract

The variation of phase and group velocity dispersion of Love and Rayleigh waves was determined for the continental United States and adjacent Canada. By processing ambient noise from the broadband channels of the Transportable Array (TA) of USArray and several Program for the Array Seismic Studies of the Continental Lithosphere experiments and using some earthquake recordings, the effort was focused on determining dispersion down to periods as short as 2 s. The relatively short distances between TA stations permitted the use of a 25 km×25 km grid for the four independent tomographic inversions (Love and Rayleigh and phase and group velocity). One reason for trying to obtain short-period dispersion was to have a data set capable of constraining upper crust velocity models for use in determining regional moment tensors. The benefit of focusing on short-period dispersion is apparent in the tomography maps—shallow geologic structures such as the Mid-Continent Rift, and the Michigan, Illinois, Anadarko, Arkoma, and Appalachian basins are imaged. In our processing, we noted that the phase velocities were more robustly determined than the group velocities. We also noted that the inability to obtain dispersion at short periods shows distinct regional patterns that may be related to the local upper crust structure.

Abstract

Accepted manuscript version, licensed CC BY-NC-ND 4.0. Achieving effective size selectivity for different species within a fishing gear is a major challenge in mixed fisheries. Fish behaviour may be exploited to separate species into separate codends where different selective properties can then be applied. Within the Basque bottom trawl fishery such a set-up has never been tested despite species with different behaviours being present. In this study, we investigate if species separation can be achieved through the use of a horizontal grid, where species typically found close to the seabed are intended to pass through the horizontal grid into the lower codend, while maintaining other species in an upper codend. Furthermore, the effect of artificial light on grid passage probability was estimated. Results were obtained for five fish species of commercial interest in the Basque bottom trawl fishery. Less than 25% of the individuals of all species analysed passed through the grid and were retained in the lower codend, and no significant differences were found when the grid was illuminated. The specific conditions under which the Basque bottom trawl fishery is conducted, i.e., high turbidity levels, high towing speed, may have influenced the performance of the gear in this fishery. Consequently, the design was found to have limited potential to improve species and size selection in the Basque bottom trawl fishery.
