The purpose of this study was to evaluate the retentive forces over time of removable partial denture clasps fabricated by the digital method. Occlusal rest seats were fabricated on three premolar teeth fixed in acrylic blocks (9 × 20 × 40 mm). Digitization of the teeth was performed using a laboratory scanner (Zirkonzahn Scanner S600 GmbH, Gais, Italy). After the analysis and determination of the insertion axis, two types of clasps with mesial occlusal rests were designed per tooth: the back-action and the reverse back-action clasps, using the Partial Planner Zirkonzahn program. The file was sent for fabrication of six metal clasps from a cobalt-chromium SP2 alloy in the EOSINT M270 system by a direct laser sintering process. The Instron 5544 universal testing machine was used to perform 20,000 cycles of clasp insertion and removal in the corresponding tooth with a load cell of 100 N and a speed of 2.5 mm/s. The retentive force was recorded for each of the 1000 cycles, and the change in retention over time was calculated. Statistical analysis was performed using the nonparametric Mann–Whitney test and a significance level of 5%. At 16,000 cycles, a maximum change in retention of 3.74 N was recorded for the back-action clasps and a minimum of −24.28 N at 1000 cycles for the reverse back-action clasps. The reverse back-action clasps exhibited statistically significant lower change in retention than the reverse-action clasps at 4000 and 5000 cycles. No differences were observed in the remaining cycles. During the 20,000 cycles, the change in retention was low regardless of the type of clasp. For most cycles, there were no differences in the change in retention between the two types of clasps.

Vascular mineralization is a hallmark of enzootic calcinosis. Histopathological, ultrastructural, and immunohistochemical investigations were performed on the external carotid arteries of seven sheep naturally poisoned by Nierembergia veitchii. Histologically, moderate to marked hyperplasia of the tunica intima was observed without mineralization. The tunica media exhibited mild to severe mineralization and osteochondroid metaplasia. Sheep with enzootic calcinosis showed arterial overexpression of osteopontin and tissue-nonspecific alkaline phosphatase and immunolabeling for osteonectin and osteocalcin in both intima and media layers of the tested arteries. The main ultrastructural finding in the tunica media was a marked phenotypic change of vascular smooth muscle cells from a contractile phenotype (VSMC-C) into a synthetic phenotype (VSMC-S). In the tunica media, VSMC-S produced matrix and extracellular vesicles, forming mineralizable granules associated with arterial mineralization. VSMC-S were also present in the tunica intima, but matrix and extracellular vesicles and mineralization were not observed. The absence of matrix and extracellular vesicles in the intimal hyperplasia, even in the presence of noncollagenous bone proteins, tissue-nonspecific alkaline phosphatase, and vitamin D receptors, reinforces the hypothesis that the presence of matrix and extracellular vesicles are crucial for the development of vascular mineralization in enzootic calcinosis. It is proposed that the two different VSMC-S phenotypes in calcinosis are due to the expression of at least two genetically different types of these cells induced by the action of 1,25(OH)2D3.

Human alphaherpesvirus 1 (HuAHV1) causes fatal neurologic infections in captive New World primates. To determine risks for interspecies transmission, we examined data for 13 free-ranging, black-tufted marmosets (Callithrix penicillata) that died of HuAHV1 infection and had been in close contact with humans in anthropized areas in Brazil during 2012-2019. We evaluated pathologic changes in the marmosets, localized virus and antigen, and assessed epidemiologic features. The main clinical findings were neurologic signs, necrotizing meningoencephalitis, and ulcerative glossitis; 1 animal had necrotizing hepatitis. Transmission electron microscopy revealed intranuclear herpetic inclusions, and immunostaining revealed HuAHV1 and herpesvirus particles in neurons, glial cells, tongue mucosal epithelium, and hepatocytes. PCR confirmed HuAHV1 infection. These findings illustrate how disruption of the One Health equilibrium in anthropized environments poses risks for interspecies virus transmission with potential spillover not only from animals to humans but also from humans to free-ranging nonhuman primates or other animals.

Background Growing evidence suggests a role for gut bacteria and their metabolites in host-signaling responses along the gut-brain axis which may impact mental health. Meditation is increasingly utilized to combat stress, anxiety, and depression symptoms. However, its impact on the microbiome remains unclear. This study observes the effects of preparation and participation in an advanced meditation program (Samyama) implemented with a vegan diet including 50% raw foods, on gut microbiome and metabolites profiles. Methods There were 288 subjects for this study. Stool samples were collected at 3-time points for meditators and household controls. Meditators prepared for 2 months for the Samyama, incorporating daily yoga and meditation practices with a vegan diet including 50% raw foods. Subjects were requested to submit stool samples for 3 time points – 2 months before Samyama (T1), right before Samyama (T2), and 3 months following Samyama (T3). 16 s rRNA sequencing was used to study participants' microbiome. Alpha and beta diversities along with short-chain fatty acid (SCFA) were assessed. Metabolomics were performed on a mass spectrometer coupled to a UHLPC system and analyzed by El-MAVEN software. Results Alpha diversity showed no significant differences between meditators and controls, while beta diversity showed significant changes (padj = 0.001) after Samyama in meditators’ microbiota composition. After the preparation phase, changes in branched short-chain fatty acids, higher levels of iso-valerate (padj = 0.02) and iso-buytrate (padj = 0.019) were observed at T2 in meditators. Other metabolites were also observed to have changed in meditators at timepoint T2. Conclusion This study examined the impact of an advanced meditation program combined with a vegan diet on the gut microbiome. There was an increase in beneficial bacteria even three months after the completion of the Samyama program. Further study is warranted to validate current observations and investigate the significance and mechanisms of action related to diet, meditation, and microbial composition and function, on psychological processes, including mood. Insight into the plasma properties of high-speed ionized flows is key for the validation of predictive aerothermodynamic tools for atmospheric entry applications, namely plasma radiation which will be highly dependent on the flow electron densities, which accordingly need to be accurately estimated. We performed an extensive review on past re-entry microwave diagnostics, including both in-flight and ground-test experiments. This work assesses the peculiar characteristics of entry plasmas, and derives functional requirements for microwave diagnostics tailored for such applications. A preliminary analysis, including a simulation of electromagnetic environment for the RAM-C II flight experiment, obtained through a CFD code coupled to an electromagnetic propagation code, is presented. The obtained results outline the adequacy of such diagnostics for improving verification and validation for high-speed entry plasma flows.

β-galactosidase is an enzyme capable of hydrolysing lactose, used in various branches of industry, mainly the food industry. As the efficient industrial use of enzymes depends on their reuse, it is necessary to find an effective method for immobilisation, maintaining high activity and stability. The present work proposes cross-linked magnetic cross-linked enzyme aggregates (mCLEAs) to prepare heterogeneous biocatalysts of β-galactosidase. Different concentrations of glutaraldehyde (0.6%, 1.0%, 1.5%), used as a cross-linking agent, were studied. The use of dextran-aldehyde as an alternative cross-linking agent was also evaluated. The mCLEAs presented increased recovered activity directly related to the concentration of glutaraldehyde. Modifications to the protocol to prepare mCLEAs with glutaraldehyde, adding a competitive inhibitor or polymer coating, have not been effective in increasing the recovered activity of the heterogeneous biocatalysts or its thermal stability. The biocatalyst prepared using dextran-aldehyde presented 73.6% recovered activity, aside from substrate affinity equivalent to the free enzyme. The thermal stability at 60 °C was higher for the biocatalyst prepared with glutaraldehyde (mCLEA-GLU-1.5) than the one produced with dextran-aldehyde (mCLEA-DEX), and the opposite happened at 50 °C. Results obtained for lactose hydrolysis, the use of its product to produce a rare sugar (D-tagatose) and operational and storage stability indicate that heterogeneous biocatalysts have adequate characteristics for industrial use.

The special issue entitled “Mathematical modelling, nonlinear dynamics, bifurcation, synchronization and control of mechanisms driven by power supply,” published by The European Physical Journal: Special Topics (EPJST), aims to bring into discussion works involving nonlinear and dynamic and chaotic behaviors, which are frequently found in mechanical and electromechanical systems, nonlinear circuits, and biologically inspired systems, stimulating the integration of findings on the nonlinear dynamics that can be observed in various systems in the physical sciences and engineering. This special issue presents 27 papers concerning the dynamics and control of flexible structure systems, nonideal systems, energy harvesting problems, and vibration absorbers. In addition, the papers address problems involving chaos control of nonlinear oscillations, phase-locked loops, synchronization of nonlinear oscillators, dynamics of bio-inspired and biostructural systems, nonlinear methods, and nonlinear circuits.

Bee surveys bring valuable information regarding species distribution, phenology, and their interactions with host plants, and thus are important to bee conservation, especially in the present scenario of drastic declines reported worldwide. The Brazilian bee fauna has been surveyed intensively since the late 1960s, but the state-of-the-art of this research topic has not been explored in detail. Our main goal is to analyze the scientific literature through a systematic review of Brazilian bee surveys, describing (i) spatial sampling gaps; (ii) preferred sampling frequency and methods; (iii) species identification and vouchering practices; (iv) gender participation; and (v) how the studies are distributed among scientific journals. Our review includes 213 published studies, of which half targeted the entire bee fauna (Apoidea: Apidae sensu lato) while the other half targeted the orchid bees (Euglossini). Sampling sites are predominant in eastern Brazil, particularly within the Atlantic Forest. Hand netting bees for 1 year long was the most frequent sampling protocol to capture bees. Taxonomic identification and voucher deposition practices were mentioned in most of the papers. Men and women published almost the same number of papers, but women were less frequent as lead or senior authors and appeared less as prolific researchers. Most papers were published in a few Brazilian journals. We highlight that surveys in understudied areas are needed, especially in Amazon. Monitoring bees in previously surveyed locations is also encouraged to investigate temporal changes in bee assemblages, such as population declines, impacts of land-use changes, and effects of climate change.

Summary Accept-reject-based Markov chain Monte Carlo algorithms have traditionally utilized acceptance probabilities that can be explicitly written as a function of the ratio of the target density at the two contested points. This feature is rendered almost useless in Bayesian posteriors with unknown functional forms. We introduce a new family of Markov chain Monte Carlo acceptance probabilities that has the distinguishing feature of not being a function of the ratio of the target density at the two points. We present two stable Bernoulli factories that generate events within this class of acceptance probabilities. The efficiency of our methods relies on obtaining reasonable local upper or lower bounds on the target density, and we present two classes of problems where such bounds are viable: Bayesian inference for diffusions, and Markov chain Monte Carlo on constrained spaces. The resulting portkey Barker’s algorithms are exact and computationally more efficient that the current state of the art.

No prior studies have evaluated the efficacy and safety of zolpidem and zopiclone to treat insomnia of demented patients. This randomized, triple-blind, placebo-controlled clinical trial used these drugs to treat patients with probable, late onset Alzheimer's dementia (AD) (DSM V and NINCDS-ADRDA criteria) exhibiting insomnia (DSM V criteria and nocturnal NPI scores ≥ 2). Actigraphic records were performed for 7 days at baseline and for 14 days during the treatment period in 62 patients aged 80.5 years in average and randomized at a 1:1:1 ratio for administration of zolpidem 10 mg/day, zopiclone 7.5 mg/day or placebo. Primary endpoint was the main nocturnal sleep duration (MNSD), whereas secondary outcomes were the proportion of the night time slept, awake time after sleep onset (WASO), nocturnal awakenings, total daytime sleep time and daytime naps. Cognitive and functional domains were tested before and after drug/placebo use. Three participants under zopiclone use had intervention interrupted due to intense daytime sedation and worsened agitation with wandering. Zopiclone produced an 81 min increase in MNSD (95% confidence interval (CI): -0.8, 163.2), a 26 min reduction in WASO (95% CI: -56.2, 4.8) and a 2-episode decrease in awakening per night (95% CI: -4.0, 0.4) in average compared to placebo. Zolpidem yielded no significant difference in MNSD despite a significant 22 min reduction in WASO (95% CI: -52.5, 8.3) and a reduction of 1 awakening each night (95% CI: -3.4, 1.2) in relation to placebo. There was a 1-point reduction in mean performance in the symbols search test among zolpidem users (95% CI: -4.1, 1.5) and an almost eight-point reduction in average scores in the digit-symbol coding test among zopiclone users (95% CI: -21.7, 6.2). In summary, short-term use of zolpidem or zopiclone by older insomniacs with AD appears to be clinically helpful, even though safety and tolerance remain issues to be personalized in healthcare settings and further investigated in subsequent trials. Lipase B from Candida antarctica was immobilized on heterofunctional support octyl agarose activated with vinyl sulfone to prevent enzyme release under drastic conditions. Covalent attachment was established, but the blocking step using hexylamine, ethylenediamine or the amino acids glycine (Gly) and aspartic acid (Asp) altered the results. The activities were lower than those observed using the octyl biocatalyst, except when using ethylenediamine as blocking reagent and p-nitrophenol butyrate (pNPB) as substrate. The enzyme stability increased using these new biocatalysts at pH 7 and 9 using all blocking agents (much more significantly at pH 9), while it decreased at pH 5 except when using Gly as blocking agent. The stress inactivation of the biocatalysts decreased the enzyme activity versus three different substrates (pNPB, S-methyl mandelate and triacetin) in a relatively similar fashion. The tryptophane (Trp) fluorescence spectra were different for the biocatalysts, suggesting different enzyme conformations. However, the fluorescence spectra changes during the inactivation were not too different except for the biocatalyst blocked with Asp, suggesting that, except for this biocatalyst, the inactivation pathways may not be so different.

Bacterial biofilm formation in low moisture food processing (LMF) plants is related to matters of food safety, production efficiency, economic loss, and reduced consumer trust. Dry surfaces may appear dry to the naked eye, however, it is common to find a coverage of thin liquid films and microdroplets, known as microscopic surface wetness (MSW). The MSW may favor dry surface biofilm (DSB) formation. DSB formation is similar in other industries, it occurs through the processes of adhesion, production of extracellular polymeric substances, development of microcolonies and maturation, it is mediated by a quorum sensing (QS) system and is followed by dispersal, leading to disaggregation. Species that survive on dry surfaces develop tolerance to different stresses. DSB are recalcitrant and contribute to higher resistance to sanitation, becoming potential sources of contamination, related to the spoilage of processed products and foodborne disease outbreaks. In LMF industries, sanitization is performed using physical methods without the presence of water. Although alternative dry sanitizing methods can be efficiently used, additional studies are still required to develop and assess the effect of emerging technologies, and to propose possible combinations with traditional methods to enhance their effects on the sanitization process. Overall, more information about the different technologies can help to find the most appropriate method/s, contributing to the development of new sanitization protocols. Thus, this review aimed to identify the main characteristics and challenges of biofilm management in low moisture food industries, and summarizes the mechanisms of action of different dry sanitizing methods (alcohol, hot air, UV-C light, pulsed light, gaseous ozone, and cold plasma) and their effects on microbial metabolism.

The ITER Collective Thomson scattering (CTS) diagnostic will measure the dynamics of fusion-born alpha particles in the burning ITER plasma by scattering a 1 MW 60 GHz gyrotron beam off fast-ion induced fluctuations in the plasma. The diagnostic will have seven measurement volumes across the ITER cross section and will resolve the alpha particle energies in the range from 300 keV to 3.5 MeV; importantly, the CTS diagnostic is the only diagnostic capable of measuring confined alpha particles for energies below ∼1.7 MeV and will also be sensitive to the other fast-ion populations. The temporal resolution is 100 ms, allowing the capture of dynamics on that timescale, and the typical spatial resolution is 10–50 cm. The development and design of the in-vessel and primary parts of the CTS diagnostic has been completed. This marks the beginning of a new phase of preparation to maximize the scientific benefit of the diagnostic, e.g., by investigating the capability to contribute to the determination of the fuel-ion ratio and the bulk ion temperature as well as integrating data analysis with other fast-ion and bulk-ion diagnostics.

Leptospirosis is a zoonotic neglected disease of worldwide public health concern. Leptospira species can infect a wide range of wild and domestic mammals and lead to a spectrum of disease, including severe and fatal forms. Herein, we report for the first time a fatal Leptospira interrogans infection in a free-ranging nonhuman primate (NHP), a black-tufted marmoset. Icterus, pulmonary haemorrhage, interstitial nephritis, and hepatocellular dissociation were the main findings raising the suspicion of leptospirosis. Diagnostic confirmation was based on specific immunohistochemical and PCR assays for Leptospira species. Immunolocalization of leptospiral antigens and identification of pathogenic species (L. interrogans species) were important for better understanding the pathogenesis of the disease. One Health-related implications of free-ranging NHPs in anthropized areas and transmission dynamics of human and animal leptospirosis are discussed.

Simulations of human body motion involve different fields of study such as robotics, from nano to macroscale, and biomechanics. The simulation of human motion is a challenging problem from the physical and computational perspectives and several models have been proposed in literature. Using a methodology named predictive dynamics, this work aims to propose a 2D spatial model of human walking during the single stance phase (SSP), using a flat foot adaptation. The model is based on inverse dynamics in which the angular displacements are interpolated by 5th degree B-splines. The equation of motion is developed by the recursive Lagrangian formulation due to its computational efficiency. An optimization problem is set up to obtain the control points of the B-splines using as the objective function the dynamic effort, which is a limited power supply. The constraints imposed on the motion are time-dependent, such as the torque/angle limits, and the dynamic stability criterion defined by the zero moment point (ZMP), or time-independent, such the equation of motion and step length. All the steps are explained in detail to find the dynamics of the body using the smallest number of laboratory results. The solution from the model is favorably compared to Winter’s data, in particular the ground reaction forces, which is important in many engineering applications.

In this study, we investigated which strategy Macrobrachium amazonicum populations use to survive in river and estuary environments. In each environment, abiotic factors were analyzed in situ, and ovigerous females and males of M. amazonicum specimens were captured. To determine ovarian maturity and morphotypes, gonads were removed and processed histologically, and the gonadosomatic index and condition factor (K) were determined. Ovigerous females were analyzed to determine the relationship between ovarian maturation and the embryonic phases of eggs in the brood pouch. Precipitation, turbidity and dissolved oxygen showed differences between the estuary and river. The population of ovigerous females showed that ovarian maturation was simultaneous with embryonic development. However, the freshwater prawn populations in the river were smaller and consisted of a greater number of ovigerous females with maturing and mature ovaries. In the estuary, ovigerous females with spawned and reorganized ovaries were observed more frequently. In males, the GC morphotype was absent in the river, where the TC morphotype predominated and showed similar reproductive conditions as the GC morphotype in the estuary. M. amazonicum has strategies for allocating energy for reproduction or growth in different environments. This description established the trade-off as a strategy used by M. amazonicum to maintain the population in adverse environments.
