Abstract: This paper presents the project Comprehensive spAce wEather Studies for the ASPIS prototype Realization (CAESAR), which aims to tackle the relevant aspects of Space Weather (SWE) science and develop a prototype of the scientific data centre for Space Weather of the Italian Space Agency (ASI) called ASPIS (ASI SPace Weather InfraStructure). To this end, CAESAR involves the majority of the SWE Italian community, bringing together 10 Italian institutions as partners, and a total of 92 researchers. The CAESAR approach encompasses the whole chain of phenomena from the Sun to Earth up to planetary environments in a multidisciplinary, comprehensive, and unprecedented way. Detailed and integrated studies are being performed on a number of well-observed “target SWE events”, which exhibit noticeable SWE characteristics from several SWE perspectives. CAESAR investigations synergistically exploit a great variety of different products (datasets, codes, models), both long-standing and novel, that will be made available in the ASPIS prototype: this will consist of a relational database (DB), an interface, and a wiki-like documentation structure. The DB will be accessed through both a Web graphical interface and the ASPIS.py module, i.e., a library of functions in Python, which will be available for download and installation. The ASPIS prototype will unify multiple SWE resources through a flexible and adaptable architecture, and will integrate currently available international SWE assets to foster scientific studies and advance forecasting capabilities.

11. Transpiration Responses of Herbicide-Resistant and -Susceptible Palmer Amaranth (Amaranthus palmeri (S.) Wats.) to Progressively Drying Soil

Abstract: Drought events are predicted to increase in the future. Evaluating the response of herbicide-resistant and -susceptible weed ecotypes to progressive drought can provide insights into whether resistance traits affect the fitness of resistant weed populations. Two experiments were conducted in the greenhouse between January and May 2021 to evaluate drought tolerance differences between Palmer amaranth accessions resistant to S-metolachlor or glyphosate and their susceptible counterparts. The accessions used were S-metolachlor-resistant (17TUN-A), a susceptible standard (09CRW-A), and glyphosate-resistant (22–165 EPSPS copies) and glyphosate-susceptible (3–10 EPSPS copies) plants from accession 16CRW-D. Daily transpiration of each plant was measured. The daily transpiration rate was converted to normalized transpiration ratio (NTR) using a double-normalization procedure. The daily soil water content was expressed as a fraction of transpirable soil water (FTSW). The threshold FTSW (FTSWcr), after which NTR decreases linearly, was estimated using a two-segment linear regression analysis. The data showed differences between S-metolachlor-resistant and -susceptible accessions (p ≤ 0.05). The FTSW remaining in the soil at the breakpoint for the S-metolachlor-susceptible accession (09CRW-A) was 0.17 ± 0.007. The FTSW remaining in the soil at the breakpoint for the S-metolachlor-resistant accession (17TUN-A) was 0.23 ± 0.004. The FTSW remaining in the soil at the breakpoint for the glyphosate-resistant and glyphosate-susceptible plants (16CRW-D) was 0.25 ± 0.007 and 0.25 ± 0.008, respectively. Although the mechanism endowing resistance to S-metolachlor might have contributed to increased drought tolerance, follow-up experiments are needed in order to verify this finding. Increased EPSPS copy numbers did not improve the drought tolerance of Palmer amaranth. As droughts are predicted to increase in frequency and severity, these results suggest that S-metolachlor-resistant and glyphosate-resistant Palmer amaranth populations will not be at a competitive disadvantage compared to susceptible genotypes. Alternative and diverse management strategies will be required for effective Palmer amaranth control, regardless of herbicide resistance status.

12. Quantification of palmer amaranth seed number using a computerized particle analyzer

Abstract: Abstract We evaluated the accuracy of a computerized particle analyzer (CPA) for high‐throughput counting of Palmer amaranth (Amaranthus palmeri S. Watson) seeds and subsequently used the CPA to verify the accuracy of two subsampling methods for estimation of Palmer amaranth seed production. To determine accuracy of the CPA, 55 hand‐counted samples, ranging from 500 to 5000 Palmer amaranth seeds, were drawn from field samples and counted with the CPA. The relationship between hand and CPA seed counts was described by a linear model (R2 = 0.99) with a slope of 0.987 and a y‐intercept of 3.49. Thus, very little discrepancy exists between seed counts conducted by hand or the CPA. Additionally, two published methods for estimation of Palmer amaranth seed production were compared to the CPA and proven to be highly accurate. We conclude from these findings that the CPA offers a high‐throughput alternative for weed scientists who frequently count large quantities of seed.

15. Resistance of Palmer amaranth (Amaranthus palmeri) to S-metolachlor in the midsouthern United States

Abstract: Palmer amaranth (Amaranthus palmeri S. Watson) is one of the most problematic weeds in many cropping systems in the midsouthern United States because of its multiple weedy traits and its propensity to evolve resistance to many herbicides with different mechanisms of action. In Arkansas, A. palmeri has evolved metabolic resistance to S-metolachlor, compromising the effectiveness of an important weed management tool. Greenhouse studies were conducted to evaluate the differential response of A. palmeri accessions from three states (Arkansas, Mississippi, and Tennessee) to (1) assess the occurrence of resistance to S-metolachlor among A. palmeri populations, (2) evaluate the resistance level in selected accessions and their resistant progeny, (3) and determine the susceptibility of most resistant accessions to other soil-applied herbicides. Seeds were collected from 168 crop fields between 2017 and 2019. One hundred seeds per accession were planted in silt loam soil without herbicide for >20 yr and sprayed with the labeled rate of S-metolachlor (1,120 g ai ha−1). Six accessions (four from Arkansas and two from Mississippi) were classified resistant to S-metolachlor. The effective doses (LD50) to control the parent accessions ranged between 73 and 443 g ha−1, and those of F1 progeny of survivors were 73 to 577 g ha−1. The resistance level was generally greater among progeny of surviving plants than among resistant field populations. The resistant field populations required 2.2 to 7.0 times more S-metolachlor to reduce seedling emergence 50%, while the F1 of survivors needed up to 9.2 times more herbicide to reduce emergence 50% compared with the susceptible standard.

Abstract: The objectives were to evaluate the effects of increasing metal micronutrient concentrations and silicon (Si) concentrations on plant growth and susceptibility to Pythium root rot with hydroponically grown lettuce (Lactuca sativa). In the first experiment, lettuce was grown in hydroponic solutions with metal micronutrients iron (Fe), manganese (Mn), copper (Cu), and zinc (Zn) supplied at either 0, 2.5, 5, or 10 mg∙L−1. A standard commercial hydroponic solution was also included as a control, with metal micronutrients supplied at 2 Fe, 1 Mn, 0.5 Cu, and 0.5 Zn mg∙L−1. In the second experiment, hydroponic lettuce was grown with Si at 0, 7, 14, 28, and 56 mg∙L−1. Hydroponic treatment solutions for both experiments were either dosed with Pythium myriotylum (Pythium treatment) at 1.80 × 104 oospores per L or deionized water as a non-Pythium control. Data were collected on leaf SPAD chlorophyll content, shoot height and width, total plant fresh mass, and root disease severity. Increasing the Cu concentration in solution decreased Pythium disease severity but reduced lettuce growth and yield. Increasing the concentration of the other metal micronutrients also tended to reduce lettuce growth but had no significant influence on root disease. Supplementing the hydroponic solution with Si had no effect on Pythium root disease severity and slightly decreased lettuce growth at 56 mg∙L−1 Si. Results of this study suggest that the management of micronutrients and Si nutrition is not an effective strategy and, at best, a risky strategy for controlling Pythium in hydroponic lettuce. Growers would likely benefit from maintaining metal micronutrient and Si concentrations within the ranges of (in mg∙L−1) 0.5 to 5.5 for Fe, 0.1 to 2.0 for Mn, 0.1 to 0.6 for Cu, 0.1 to 0.6 for Zn, and 0 to 28 for Si for many hydroponic crops. Supplementing Si has the potential to negatively influence plant growth and quality for certain plant species, and testing is necessary to evaluate phytotoxicity risks prior to implementing in commercial practice. Overall, successful mitigation of root rot pathogens in commercial hydroponic production requires the combination of proper sanitation, best management and cultural practices, appropriate hydroponic system design, and the implementation of a water treatment system with proper design and a multi-barrier approach.

Abstract: A deep survey of the Large Magellanic Cloud at ~0.1-100TeV photon energies with the Cherenkov Telescope Array is planned. We assess the detection prospects based on a model for the emission of the galaxy, comprising the four known TeV emitters, mock populations of sources, and interstellar emission on galactic scales. We also assess the detectability of 30 Doradus and SN 1987A, and the constraints that can be derived on the nature of dark matter. The survey will allow for fine spectral studies of N157B, N132D, LMC P3, and 30 Doradus C, and half a dozen other sources should be revealed, mainly pulsar-powered objects. The remnant from SN 1987A could be detected if it produces cosmic-ray nuclei with a flat power-law spectrum at high energies, or with a steeper index 2.3-2.4 pending a flux increase by a factor >3-4 over ~2015-2035. Large-scale interstellar emission remains mostly out of reach of the survey if its >10GeV spectrum has a soft photon index ~2.7, but degree-scale 0.1-10TeV pion-decay emission could be detected if the cosmic-ray spectrum hardens above >100GeV. The 30 Doradus star-forming region is detectable if acceleration efficiency is on the order of 1-10% of the mechanical luminosity and diffusion is suppressed by two orders of magnitude within, Accepted for publication in MNRAS. 24. Biomaterial Engineering for Controlling Pluripotent Stem Cell Fate

Abstract: Pluripotent stem cells (PSCs) represent an exciting cell source for tissue engineering and regenerative medicine due to their self-renewal and differentiation capacities. The majority of current PSC protocols rely on 2D cultures and soluble factors to guide differentiation; however, many other environmental signals are beginning to be explored using biomaterial platforms. Biomaterials offer new opportunities to engineer the stem cell niches and 3D environments for exploring biophysical and immobilized signaling cues to further our control over stem cell fate. Here, we review the biomaterial platforms that have been engineered to control PSC fate. We explore how altering immobilized biochemical cues and biophysical cues such as dimensionality, stiffness, and topography can enhance our control over stem cell fates. Finally, we highlight biomaterial culture systems that assist in the translation of PSC technologies for clinical applications.

25. Evaluating Species-Specific Replenishment Solution Effects on Plant Growth and Root Zone Nutrients with Hydroponic Arugula (Eruca sativa L.) and Basil (Ocimum basilicum L.)

Abstract: Managing nutrients in recirculating solutions can be challenging in hydroponic production, and poor management practices can rapidly result in root zone nutrient imbalances in reduced yield. Using mass balance principles to formulate hydroponic replenishment solutions is a proposed strategy to reduce nutrient imbalances and the need to periodically replace the hydroponic solution. Objectives were to (1) formulate species-specific nutrient replenishment solutions for arugula (Eruca sativa L.) and basil (Ocimum basilicum L.) using mass balance principles and (2) evaluate the effects of using these replenishment solutions on plant growth and root zone nutrients over time. In the first experiment, arugula and basil tissues were analyzed over 42 d for macronutrient concentrations which were used to custom formulate species-specific replenishment solutions for both species. In the second experiment, nutrients were resupplied for hydroponic arugula and basil over time using either the species-specific or a standard commercial hydroponic replenishment solution. Species-specific replenishment solutions resulted in decreased solution EC and concentrations of most macronutrients over time for both species. In contrast, replenishment with the standard hydroponic solution resulted in increased solution EC and concentrations of calcium, magnesium, and sulfate and decreased nitrogen, phosphorus, and potassium. Replenishment treatment had no effect on yield for arugula and basil; however, results suggested root zone imbalances may have still occurred eventually for both treatments and species. Species-specific replenishment solutions would be most effective in scenarios where solutions could be formulated for a specific crop, mixed using high-quality irrigation water, and where environmental conditions are controlled and stable, such as indoor farming systems. This study highlighted several practical challenges and considerations regarding the formulation of hydroponic solutions using mass balance.

Abstract: Infiltrative growth is a major cause of high lethality of malignant brain tumors such as glioblastoma (GBM). We show here that GBM cells upregulate guidance receptor Plexin-B2 to gain invasiveness. Deletion of Plexin-B2 in GBM stem cells limited tumor spread and shifted invasion paths from axon fiber tracts to perivascular routes. On a cellular level, Plexin-B2 adjusts cell adhesiveness, migratory responses to different matrix stiffness, and actomyosin dynamics, thus empowering GBM cells to leave stiff tumor bulk and infiltrate softer brain parenchyma. Correspondingly, gene signatures affected by Plexin-B2 were associated with locomotor regulation, matrix interactions, and cellular biomechanics. On a molecular level, the intracellular Ras-GAP domain contributed to Plexin-B2 function, while the signaling relationship with downstream effectors Rap1/2 appeared variable between GBM stem cell lines, reflecting intertumoral heterogeneity. Our studies establish Plexin-B2 as a modulator of cell biomechanics that is usurped by GBM cells to gain invasiveness., Huang et al demonstrate that glioblastoma cells upregulate axon guidance molecule Plexin-B2 to gain invasiveness and that Plexin-B2 promotes glioblastoma cell infiltration along axon fiber tracts in intracranial transplant models by modulating cellular biomechanics.

Abstract: Very-high Energy (VHE) gamma-ray astroparticle physics is a relatively young field, and observations over the past decade have surprisingly revealed almost two hundred VHE emitters which appear to act as cosmic particle accelerators. These sources are an important component of the Universe, influencing the evolution of stars and galaxies. At the same time, they also act as a probe of physics in the most extreme environments known - such as in supernova explosions, and around or after the merging of black holes and neutron stars. However, the existing experiments have provided exciting glimpses, but often falling short of supplying the full answer. A deeper understanding of the TeV sky requires a significant improvement in sensitivity at TeV energies, a wider energy coverage from tens of GeV to hundreds of TeV and a much better angular and energy resolution with respect to the currently running facilities. The next generation gamma-ray observatory, the Cherenkov Telescope Array Observatory (CTAO), is the answer to this need. In this talk I will present this upcoming observatory from its design to the construction, and its potential science exploitation. CTAO will allow the entire astronomical community to explore a new discovery space that will likely lead to paradigm-changing breakthroughs. In particular, CTA has an unprecedented sensitivity to short (sub-minute) timescale phenomena, placing it as a key instrument in the future of multi-messenger and multi-wavelength time domain astronomy. Abstract: Vasculature plays a vital role in human biology as blood vessels transport nutrients and oxygen throughout the body. Endothelial cells (ECs), specifically, are key as they maintain barrier functions between the circulating blood and the surrounding tissues. ECs derived from human pluripotent stem cells (hPSCs) are utilized to study vascular development and disease mechanisms within in vitro models. Additionally, ECs derived from induced pluripotent stem cells (iPSCs) hold great promise for advancing personalized medicine, cell therapies, and tissue-engineered constructs by creating patient-specific cell populations. Here, we describe a xeno-free, serum-free differentiation protocol for deriving ECs from hPSCs. In brief, mesoderm progenitor cells are derived via WNT pathway activation. Following this, EC maturation is achieved with exogenous vascular endothelial growth factor A (VEGFA) and basic fibroblast growth factor 2 (bFGF2). We have characterized these cells as expressing mature EC markers and have illustrated their functionality in vitro.

Abstract: A large-area, solid-state detector with single-hit precision timing measurement will enable several breakthrough experimental advances for the direct measurement of particles in space. Silicon microstrip detectors are the most promising candidate technology to instrument the large areas of the next-generation astroparticle space borne detectors that could meet the limitations on power consumption required by operations in space. We overview the novel experimental opportunities that could be enabled by the introduction of the timing measurement, concurrent with the accurate spatial and charge measurement, in Silicon microstrip tracking detectors, and we discuss the technological solutions and their readiness to enable the operations of large-area Silicon microstrip timing detectors in space.

41. Large crabgrass (Digitaria sanguinalis) and Palmer amaranth (Amaranthus palmeri) intraspecific and interspecific interference in soybean

Abstract: Field studies were conducted in 2016 and 2017 at Clinton, NC, to quantify the effects of season-long interference of large crabgrass [Digitaria sanguinalis (L.) Scop.] and Palmer amaranth (Amaranthus palmeri S. Watson) on ‘AG6536’ soybean [Glycine max (L.) Merr.]. Weed density treatments consisted of 0, 1, 2, 4, and 8 plants m−2 for A. palmeri and 0, 1, 2, 4, and 16 plants m−2 for D. sanguinalis with (interspecific interference) and without (intraspecific interference) soybean to determine the impacts on weed biomass, soybean biomass, and seed yield. Biomass per square meter increased with increasing weed density for both weed species with and without soybean present. Biomass per square meter of D. sanguinalis was 617% and 37% greater when grown without soybean than with soybean, for 1 and 16 plants m−2 respectively. Biomass per square meter of A. palmeri was 272% and 115% greater when grown without soybean than with soybean for 1 and 8 plants m−2, respectively. Biomass per plant for D. sanguinalis and A. palmeri grown without soybean was greatest at the 1 plant m−2 density. Biomass per plant of D. sanguinalis plants across measured densities was 33% to 83% greater when grown without soybean compared with biomass per plant when soybean was present for 1 and 16 plants m−2, respectively. Similarly, biomass per plant for A. palmeri was 56% to 74% greater when grown without soybean for 1 and 8 plants m−2, respectively. Biomass per plant of either weed species was not affected by weed density when grown with soybean due to interspecific competition with soybean. Yield loss for soybean grown with A. palmeri ranged from 14% to 37% for densities of 1 to 8 plants m−2, respectively, with a maximum yield loss estimate of 49%. Similarly, predicted loss for soybean grown with D. sanguinalis was 0 % to 37% for densities of 1 to 16 m−2 with a maximum yield loss estimate of 50%. Soybean biomass was not affected by weed species or density. Results from these studies indicate that A. palmeri is more competitive than D. sanguinalis at lower densities, but that similar yield loss can occur when densities greater than 4 plants m−2 of either weed are present.

42. Efficacy of fall-applied residual herbicides on weedy rice control in rice (Oryza sativa L.)

Abstract: Field experiments were initiated near Colt, AR, in the fall of 2016 and continued through the summer of 2018 to evaluate rice tolerance and weedy (or red) rice control after fall-applied very-long-chain fatty acid (VLCFA)-inhibiting herbicides. A split-plot design was used for the experiment, with the whole-plot factor being winter condition (flooded or non-flooded) and the split-plot factors being herbicide and rate. Herbicide treatments included acetochlor, dimethenamid-P, pethoxamid, pyroxasulfone, and S-metolachlor applied at 1,050, 525, 420, 205, and 1,070 g ai ha-1 and at 2,100, 1,050, 840, 410, and 2,140 g ha-1 for low rates and high rates, respectively. Herbicides were applied in the fall, then ‘CL172’ rice was drill seeded in the spring of the following calendar year. Weedy rice control differed between years, but acetochlor and pyroxasulfone consistently provided the greatest levels of control across rates and flood conditions. Consequently, herbicides that best controlled weedy rice also caused the greatest injury to cultivated rice. Rice injury did not exceed 13% regardless of herbicide treatment at 3 wk after planting (WAP). However, the high rate of pyroxasulfone caused 20% rice injury at 5 WAP in 2018. Although it was expected that winter condition may affect residual activity of the VLCFA-inhibiting herbicides, herbicide selection and application rate both had much greater effects on rice injury and on weedy rice control. Based on these results, rice injury would be minimal or nonexistent after fall applications of the tested VLCFA inhibitors, and intermediate levels of weedy rice control may be achieved. The implementation of VLCFA-inhibiting herbicides in rice production systems would offer a novel herbicide site of action and offer a degree of selective control of weedy rice.Nomenclature: Acetochlor; dimethenamid-P; pethoxamid; pyroxasulfone, S-metolachlor; red rice, Oryza sativa L. Abstract: Watermelon [Citrullus lanatus(Thunb.) Matsum & Nakai] grafting is commonly used for management of diseases caused by soilborne pathogens; however, little research exists describing the effect of grafting on the weed-competitive ability of watermelon. Field experiments determined the response in yield, fruit number, and fruit quality of grafted and nongrafted watermelon exposed to increasing densities of Palmer amaranth (Amaranthus palmeriS. Watson). Grafting treatments included ‘Exclamation’ triploid (seedless) watermelon grafted on two interspecific hybrid squash rootstocks ‘Carnivor’ and ‘Kazako’, with nongrafted Exclamation as the control. Weed treatments includedA. palmeriat densities of 1, 2, 3, and 4A. palmeriplants per watermelon planting hole (0.76-m row) and a weed-free control. IncreasingA. palmeridensities caused significant reductions (P A. palmeriplants planting hole−1reduced marketable yield 41%, 38%, and 65% for Exclamation, Carnivor, and Kazako, respectively. Neither grafting treatment norA. palmeridensity had a biologically meaningful effect on soluble solids content or on the incidence of hollow heart in watermelon fruit.Amaranthus palmeriseed and biomass production was similar across weed population densities, but seed number per femaleA. palmeridecreased according to a two-parameter exponential decay equation. Thus, increasing weed population densities resulted in increased intraspecific competition amongA. palmeriplants. While grafting may offer benefits for disease resistance, no benefits regarding weed-competitive ability were observed, and a consistent yield penalty was associated with grafting, even in weed-free treatments.

Abstract: Field experiments determined the critical period for weed control (CPWC) in grafted and nongrafted watermelon [Citrullus lanatus(Thumb.) Matsum. & Nakai] grown in plasticulture. Transplant types included ‘Exclamation’ seedless watermelon as the nongrafted control as well as Exclamation grafted onto two interspecific hybrid squash (ISH) rootstocks, ‘Carnivor’ and ‘Kazako’. To simulate weed emergence throughout the season, establishment treatments (EST) consisted of two seedlings each of common purslane (Portulaca oleraceaL.), large crabgrass [Digitaria sanguinalis(L.) Scop.], and yellow nutsedge (Cyperus esculentusL.) transplanted in a 15 by 15 cm square centered on watermelon plants at 0, 2, 3, 4, and 6 wk after watermelon transplanting (WATr) and remained until the final watermelon harvest at 11 WATr. To simulate weed control at different times in the season, removal treatments (REM) consisted of two seedlings of the same weed species transplanted in a 15 by 15 cm square centered on watermelon plants on the same day of watermelon transplanting and allowed to remain until 2, 3, 4, 6, and 11 WATr, at which time they were removed. Season-long weedy and weed-free controls were included for both EST and REM studies in both years. For all transplant types, aboveground biomass of weeds decreased as weed establishment was delayed and increased as weed removal was delayed. The predicted CPWC for nongrafted Exclamation and Carnivor required only a single weed removal between 2.3 and 2.5 WATr and 1.9 and 2.6 WATr, respectively, while predicted CPWC for Kazako rootstock occurred from 0.3 to 2.6 WATr. Our study results suggest that weed control for this mixed population of weeds would be similar between nongrafted Exclamation and Exclamation grafted onto Carnivor. But the observed CPWC of Exclamation grafted onto Kazako suggests that CPWC may vary with specific rootstock–scion combinations.

45. Comparison of Root System Morphology of Cucurbit Rootstocks for Use in Watermelon Grafting

Abstract: Grafting of watermelon (Citrullus lanatus) is an established production practice that provides resistance to soilborne diseases or tolerance to abiotic stresses. Watermelon may be grafted on several cucurbit species (interspecific grafting); however, little research exists to describe root systems of these diverse rootstocks. A greenhouse study was conducted to compare root system morphology of nine commercially available cucurbit rootstocks, representing four species: pumpkin (Cucurbita maxima), squash (Cucurbita pepo), bottle gourd (Lagenaria siceraria), and an interspecific hybrid squash (C. maxima × C. moschata). Rootstocks were grafted with a triploid watermelon scion (‘Exclamation’), and root systems were compared with nongrafted (NG) and self-grafted (SG) ‘Exclamation’. Plants were harvested destructively at 1, 2, and 3 weeks after transplant (WAT), and data were collected on scion dry weight, total root length (TRL), average root diameter, root surface area, root:shoot dry-weight ratio, root diameter class proportions, and specific root length. For all response variables, the main effect of rootstock and rootstock species was significant (P < 0.05). The main effect of harvest was significant (P < 0.05) for all response variables, with the exception of TRL proportion in diameter class 2. ‘Ferro’ rootstock produced the largest TRL and root surface area, with observed values 122% and 120% greater than the smallest root system (‘Exclamation’ SG), respectively. Among rootstock species, pumpkin produced the largest TRL and root surface area, with observed values 100% and 82% greater than those of watermelon, respectively. These results demonstrate that substantial differences exist during the initial 3 WAT in root system morphology of rootstocks and rootstock species available for watermelon grafting and that morphologic differences of root systems can be characterized using image analysis.

We used human induced pluripotent stem cell (iPSC)-derived cerebral organoids expressing tau-V337M and isogenic corrected controls to discover early alterations due to the mutation that precede neurodegeneration. At 2 months, mutant organoids show upregulated expression of MAPT, and glutamatergic signaling pathways and regulators including the RNA-binding protein ELAVL4. Over the following 4 months, mutant organoids accumulate splicing changes, disruption of autophagy function and build-up of tau and P-tau S396. By 6 months, tau-V337M organoids show specific loss of glutamatergic neurons of layers affected in patients. Mutant neurons are susceptible to glutamate toxicity which was rescued pharmacologically by treatment with the PIKFYVE kinase inhibitor apilimod. Our results demonstrate a sequence of events that precede cell death, revealing molecular pathways associated with glutamate signaling as potential targets for therapeutic intervention in FTD.

Abstract: Multimessenger astrophysics is based on the detection, with the highest possible accuracy, of the cosmic radiation. During the last 20 years, the advent space-borne magnetic spectrometers in space (AMS-01, Pamela, AMS-02), able to measure the charged cosmic radiation separating matter from antimatter, and to provide accurate measurement of the rarest components of Cosmic Rays (CRs) to the highest possible energies, have become possible, together with the ultra-precise measurement of ordinary CRs. These developments started the era of precision Cosmic Ray physics providing access to a rich program of high-energy astrophysics addressing fundamental questions like matter-antimatter asymmetry, indirect detection for Dark Matter and the detailed study of origin, acceleration and propagation of CRs and their interactions with the interstellar medium.In this paper we address the above-mentioned scientific questions, in the context of a second generation, large acceptance, superconducting magnetic spectrometer proposed as mission in the context of the European Space Agency’s Voyage2050 long-term plan: the Antimatter Large Acceptance Detector In Orbit (ALADInO) would extend by about two orders of magnitude in energy and flux sensitivity the separation between charged particles/anti-particles, making it uniquely suited for addressing and potentially solving some of the most puzzling issues of modern cosmology.

Abstract: The Schwarzschild-Couder Telescope (SCT) is a telescope concept proposed for the Cherenkov Telescope Array. It employs a dual-mirror optical design to remove comatic aberrations over an $8^{\circ}$ field of view, and a high-density silicon photomultiplier camera (with a pixel resolution of 4 arcmin) to record Cherenkov emission from cosmic ray and gamma-ray initiated particle cascades in the atmosphere. The prototype SCT (pSCT), comprising a 9.7 m diameter primary mirror and a partially instrumented camera with 1536 pixels, has been constructed at the Fred Lawrence Whipple Observatory. The telescope was inaugurated in January 2019, with commissioning continuing throughout 2019. We describe the first campaign of observations with the pSCT, conducted in January and February of 2020, and demonstrate the detection of gamma-ray emission from the Crab Nebula with a statistical significance of $8.6\sigma$., Comment: 13 pages, 12 figures, 3 tables, submitted to Astroparticle Physics
