1. Intrinsic and Instrumental Care in Pen Pal Letters: Recognizing Care in STEM Classrooms

Abstract

Studies of both professional science practice and children's science learning show that care is not merely ancillary to disciplinary work but a core and generative constituent of science practice. In science education research, however, students' care is often overlooked. In this paper, we describe the expression of care across two STEM classrooms (6th and 9th grade) studying biology and ecology and participating in a pen pal exchange. We analyze artifacts from the pen pal exchange as well as students' retrospective interviews and written reflections. Two ways of expressing care surfaced in students' letters: caring for guppies and caring for pen pals. We describe each form of care using examples from our data. We find that students' care for guppies and pen pals was both instrumental (in service of their investigations) and intrinsic (positioning guppies and pen pals as inherently valuable). We then connect these findings to studies of care in children's science learning and in professional science. We discuss methodological and practical implications for recognizing and analyzing how students' care manifests in classrooms and for designing learning activities that cultivate care.

2. Widening University Participation in Learning Using Students' Contextualised Storytelling in General Chemistry

Abstract

Many students find introductory general chemistry courses difficult because they feel alienated by traditional approaches to teaching and learning. This can become particularly problematic in laboratory sessions where students simply follow processes and procedures that students can view as being mundane and lacking creativity. Contextualised storytelling offers a novel pedagogical approach to help students connect and make sense of chemistry ideas in the context of their own life experiences. The current study implemented the CLEAR (chemistry learning "via" experiential academic reflection) approach to contextualised storytelling as a sequence of four assignments across a laboratory course for first-year students. The research explored students' experiences writing contextualised stories to make sense of and learn chemistry. Using hermeneutics as a methodology, the data collected included participants' written contextualised stories, semi-structured interview recordings, and field notes. While the CLEAR approach differs from other approaches to storytelling in chemistry education, the current study suggests that CLEAR can make positive contributions to student learning. The findings showed that, although many students initially resisted or felt confused by the new approach, CLEAR helped students see the connection and relevance of chemistry concepts to their lives. Students also recognized the importance of self-directed learning while writing their CLEAR stories, which suggests that CLEAR engaged students in learning that was active and organic. Furthermore, writing CLEAR stories supported students in talking to people about scientific concepts they learned in class, which suggests that writing the CLEAR stories: (a) helped students find the relevancy of the ideas to the degree that they felt they could share the ideas in their own words outside of class and (b) increased students' interest in the course and what they were learning to the degree that they wanted to share it. Implementing CLEAR as multiple assignments across the course appears important and valuable because students refined their thinking and writing skills through iteration within and across CLEAR stories.

3. Different Strategies to Facilitate Meaningful Reflections among Post-Secondary Students in a Community Service Learning Water Project

Abstract

This study explored how continuous diverse reflective exercises embedded in a Community Service Learning chemistry lab support science students' meaningful learning. The findings of this study are intended for those involved in teaching natural science in higher education, as well as those interested in Community Service Learning, self-directed learning, and reflective strategies. Fourteen students in a second-year Analytical Chemistry II lab participated in this study. Reflective exercises representing multiple modes of reflection were purposefully designed and embedded across the lab curriculum. Qualitative content analysis of data from reflective writings, scrapbook reflections, and reflective discussions demonstrates that students were able to articulate their self-directed learning from the perspective of academic enhancement, personal growth, and civic engagement in the different reflective exercises. Students indicated a high level of satisfaction, agreed that the integration of diverse continuous reflective strategies can enhance their transformative learning practice in an engaging way, and would like to continue this practice for other science laboratory courses.

4. Food fraud and the perceived integrity of European food imports into China.

Abstract

Persistent incidents of food fraud in China have resulted in low levels of consumer trust in the authenticity and safety of food that is domestically produced. We examined the relationship between the concerns of Chinese consumers regarding food fraud, and the role that demonstrating authenticity may play in relieving those concerns.A two-stage mixed method design research design was adopted. First, qualitative research (focus groups n = 7) was conducted in three Chinese cities, Beijing, Guangzhou and Chengdu to explore concerns held by Chinese consumers in relation to food fraud. A subsequent quantitative survey (n = 850) tested hypotheses derived from the qualitative research and theoretical literature regarding the relationship between attitudinal measures (including risk perceptions, social trust, and perceptions of benefit associated with demonstrating authenticity), and behavioral intention to purchase "authentic" European products using structural equation modelling.Chinese consumers perceive food fraud to be a hazard that represents a food safety risk. Food hazard concern was identified to be geographically influenced. Consumers in Chengdu (tier 2 city) possessed higher levels of hazard concern compared to consumers in Beijing and Guangzhou (tier 1). Structural trust (i.e. trust in actors and the governance of the food supply chain) was not a significant predictor of attitude and intention to purchase authenticated food products. Consumers were shown to have developed 'risk-relieving' strategies to compensate for the lack of trust in Chinese food and the dissonance experienced as a consequence of food fraud. Indexical and iconic authenticity cues provided by food manufacturers and regulators were important elements of product evaluations, although geographical differences in their perceived importance were observed.Targeted communication of authenticity assurance measures, including; regulations; enforcement; product testing; and actions taken by industry may improve Chinese consumer trust in the domestic food supply chain and reduce consumer concerns regarding the food safety risks associated with food fraud. To support product differentiation and retain prestige, European food manufactures operating within the Chinese market should recognise regional disparities in consumer risk perceptions regarding food fraud and the importance of personal risk mitigation strategies adopted by Chinese consumers to support the identification of authentic products.

8. Prevalence of Mobile Phone vs. Child-Related Driver Distraction in a Sample of Families With Young Children

Abstract

Motor vehicle crashes are the leading cause of accidental death in Australia, with substantial societal costs. Unlike crash test dummies, child vehicle passengers rarely sit still and their behaviour can often be unpredictable. Analysis of naturalistic driving video data from journeys undertaken by 12 families with young children revealed that children accounted for 12% of all potentially distracting activities, with drivers in this study interacting with rear seat child occupants 12 times as often as they did with mobile phones. Educational interventions to reduce driver distraction are discussed and the use of the naturalistic driving methodology is proposed to investigate the potential benefits of a novel, best practices-based road safety education program targeting child-related driver distraction. Outcomes of such an evaluation could be used to inform and refine future education strategies designed to minimise child-related driver distraction and crash risk, and to improve overall road safety in Australasia.

Abstract

In the ~1000 sols since the Mars 2020 Perseverance rover landed on the floor of Jezero crater, it has traversed >23 km, carrying out analyses of the crater floor and western fan. The fan is comprised of sediments transported and deposited by streams that once flowed into Jezero crater in the late Noachian to early Hesperian[1]. Detailed investigation of the sediments and rocks of the western fan can thus provide insights into ancient fluvial to lacustrine environments on Mars, whether they were habitable, and/or if biosignatures maybe preserved.

Abstract

Social media is commonly used by the public during election campaigns to express their opinions regarding different issues. Among various social media channels, Twitter provides an efficient platform for researchers and politicians to explore public opinion regarding a wide range of topics such as the economy and foreign policy. Current literature mainly focuses on analyzing the content of tweets without considering the gender of users. This research collects and analyzes a large number of tweets and uses computational, human coding, and statistical analyses to identify topics in more than 300,000 tweets posted during the 2020 U.S. presidential election and to compare female and male users regarding the average weight of the discussed topics. Our findings are based upon a wide range of topics, such as tax, climate change, and the COVID-19 pandemic. Out of the topics, there exists a significant difference between female and male users for more than 70% of topics.

Abstract

Oral and gut Bacteroidetes produce unique classes of serine-glycine lipodipeptides and glycine aminolipids that signal through host Toll-like receptor 2. These glycine lipids have also been detected in human arteries, but their effects on atherosclerosis are unknown. Here, we sought to investigate the bioactivity of bacterial glycine lipids in mouse models of atherosclerosis. Lipid 654 (L654), a serine-glycine lipodipeptide species, was first tested in a high-fat diet (HFD)-fed Ldlr−/− model of atherosclerosis. Intraperitoneal administration of L654 over 7 weeks to HFD-fed Ldlr−/− mice resulted in hypocholesterolemic effects and significantly attenuated the progression of atherosclerosis. We found that L654 also reduced liver inflammatory and extracellular matrix gene expression, which may be related to inhibition of macrophage activation as demonstrated in vivo by lower major histocompatibility complex class II gene expression and confirmed in cell experiments. In addition, L654 and other bacterial glycine lipids in feces, liver, and serum were markedly reduced alongside changes in Bacteroidetes relative abundance in HFD-fed mice. Finally, we tested the bioactivities of L654 and related lipid 567 in chow-fed Apoe−/− mice, which displayed much higher fecal glycine lipids relative to HFD-fed Ldlr−/− mice. Administration of L654 or lipid 567 for 7 weeks to these mice reduced the liver injury marker alanine aminotransferase, but other effects seen in Ldlr−/− were not observed. Therefore, we conclude that conditions in which gut microbiome-derived glycine lipids are lost, such as HFD, may exacerbate the development of atherosclerosis and liver injury, whereas correction of such depletion may protect from these disorders.

Abstract

Better methods to interrogate host-pathogen interactions during Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infections are imperative to help understand and prevent this disease. Here we implemented RNA-sequencing (RNA-seq) using Oxford Nanopore Technologies (ONT) long-reads to measure differential host gene expression, transcript polyadenylation and isoform usage within various epithelial cell lines permissive and non-permissive for SARS-CoV-2 infection. SARS-CoV-2-infected and mock-infected Vero (African green monkey kidney epithelial cells), Calu-3 (human lung adenocarcinoma epithelial cells), Caco-2 (human colorectal adenocarcinoma epithelial cells) and A549 (human lung carcinoma epithelial cells) were analyzed over time (0, 2, 24, 48 hours). Differential polyadenylation was found to occur in both infected Calu-3 and Vero cells during a late time point (48 hpi), with Gene Ontology (GO) terms such as viral transcription and translation shown to be significantly enriched in Calu-3 data. Poly(A) tails showed increased lengths in the majority of the differentially polyadenylated transcripts in Calu-3 and Vero cell lines (up to ~101 nt in mean poly(A) length, padj = 0.029). Of these genes, ribosomal protein genes such as RPS4X and RPS6 also showed downregulation in expression levels, suggesting the importance of ribosomal protein genes during infection. Furthermore, differential transcript usage was identified in Caco-2, Calu-3 and Vero cells, including transcripts of genes such as GSDMB and KPNA2, which have previously been implicated in SARS-CoV-2 infections. Overall, these results highlight the potential role of differential polyadenylation and transcript usage in host immune response or viral manipulation of host mechanisms during infection, and therefore, showcase the value of long-read sequencing in identifying less-explored host responses to disease.

31. Using Gravity Recovery and Climate Experiment data to derive corrections to precipitation data sets and improve modelled snow mass at high latitudes

Abstract

The amount of lying snow calculated by a land surface model depends in part on the amount of snowfall in the meteorological data that are used to drive the model. We show that commonly used data sets differ in the amount of snowfall, and more generally precipitation, over four large Arctic basins. An independent estimate of the cold-season precipitation is obtained by combining water balance information from the Gravity Recovery and Climate Experiment (GRACE) with estimates of evaporation and river discharge and is generally higher than that estimated by four commonly used meteorological data sets. We use the Joint UK Land Environment Simulator (JULES) land surface model to calculate the snow water equivalent (SWE) over the four basins. The modelled seasonal maximum SWE is 38 % less than observation-based estimates on average, and the modelled basin discharge is significantly underestimated, consistent with the lack of snowfall. We use the GRACE-derived estimate of precipitation to define per-basin scale factors that are applied to the driving data and increase the amount of cold-season precipitation by 28 % on average. In turn this increases the modelled seasonal maximum SWE by 30 %, although this is still underestimated compared to observations by 19 % on average. A correction for the undercatch of precipitation by gauges is compared with the the GRACE-derived correction. Undercatch correction increases the amount of cold-season precipitation by 23 % on average, which indicates that some, but not all, of the underestimation can be removed by implementing existing undercatch correction algorithms. However, even undercatch-corrected data sets contain less precipitation than the GRACE-derived estimate in some regions, and it is likely that there are other biases that are not currently accounted for in gridded meteorological data sets. This study shows that revised estimates of precipitation can lead to improved modelling of SWE, but much more modest improvements are found in modelled river discharge. By providing methods to better define the precipitation inputs to the system, the current study paves the way for subsequent work on key hydrological processes in high-latitude basins.

Abstract

In this whitepaper, we call for the concept of biomanufacturing to be expanded and widely adopted as a support function of human Space-travel. To demonstrate the impact and value of this strategy, we identify the specific offworld scenarios where the concept is most applicable, as well as the vital inventories that can be made available thereby. This will serve to increase capabilities of human operations beyond Earth-orbit and allow for extended mission design through greater autonomy while minimizing risks through redundancy. To this end, we sketch the potential routes and systems to arrive at these goals, in the form of specialized microbial cell factories that can most meaningfully leverage the resources available along the journey. The strategic vision presented here relies heavily on Synthetic Biology as it integrates with major plans for In Situ Resource Utilization and highlights applications that engineered biology is uniquely suited to address. We finish by advocating for the research and development investments that need to be made in order to significantly increase readiness of these technologies over the coming decade. This dovetails with current efforts to return humans to the Moon with Mars on the horizon. Besides ensuring the feasibility and sustainability of crewed Space exploration and habitation, the advancement of these technologies may spawn a new scalable microgravity-based biotechnology industry that contributes to the creation of a circular economy on Earth.

Abstract

Abstract Background Multiple sclerosis (MS) is a central nervous system (CNS) autoimmune disease characterized by both inflammatory demyelination and impaired remyelination. Studies indicate that Toll-like receptor 2 (TLR2) signaling contributes to both the inflammatory component and the defective remyelination in MS. While most MS therapeutics target adaptive immunity, we recently reported that reducing TLR2 signaling in innate immune cells by inducing TLR2 tolerance attenuates adoptively transferred experimental autoimmune encephalomyelitis. Given that previous reports suggest TLR2 signaling also inhibits myelin repair, the objective of this study was to assess how reducing TLR2 signaling through TLR2 tolerance induction affects CNS myelin repair. Methods Chow containing 0.2% cuprizone was fed to male and female wild-type (WT) C57BL/6 mice or TLR2-deficient (TLR2−/−) mice for 5 weeks to induce demyelination. During a 2-week remyelination period following discontinuation of cuprizone, WT mice received either low dose TLR2 ligands to induce systemic TLR2 tolerance or vehicle control (VC). Remyelination was evaluated via electron microscopy and immunohistochemical analysis of microglia and oligodendrocytes in the corpus callosum. Statistical tests included 2-way ANOVA and Mann-Whitney U analyses. Results Inducing TLR2 tolerance in WT mice during remyelination significantly enhanced myelin recovery, restoring unmyelinated axon frequency and myelin thickness to baseline levels compared to VC-treated mice. Mechanistically, enhanced remyelination in TLR2 tolerized mice was associated with a shift in corpus callosum microglia from a pro-inflammatory iNOS+ phenotype to a non-inflammatory/pro-repair Arg1+ phenotype. This result was confirmed in vitro by inducing TLR2 tolerance in WT microglia cultures. TLR2−/− mice, without TLR2 tolerance induction, also significantly enhanced myelin recovery compared to WT mice, adding confirmation that reduced TLR2 signaling is associated with enhanced remyelination. Discussion Our results suggest that reducing TLR2 signaling in vivo by inducing TLR2 tolerance significantly enhances myelin repair. Furthermore, the enhanced remyelination resulting from TLR2 tolerance induction is associated with a shift in corpus callosum microglia from a pro-inflammatory iNOS+ phenotype to a non-inflammatory/pro-repair Arg1+ phenotype. While deletion of TLR2 would be an impractical approach in vivo, reducing innate immune signaling through TLR2 tolerance induction may represent a novel, two-pronged approach for treating both inflammatory and myelin repair components of MS.

Abstract

Objective: We aimed to identify factors independently associated with the need for inotropic support for low cardiac output or haemodynamic instability after pulmonary artery banding surgery for CHD. Methods: We performed a retrospective chart review of all neonates and infants who underwent pulmonary banding between January 2016 and June 2019 at our institution. Bivariate and multivariable analyses were performed to identify factors independently associated with the use of post-operative inotropic support, defined as the initiation of inotropic infusion(s) for depressed myocardial function, hypotension, or compromised perfusion within 24 hours of pulmonary artery banding. Results: We reviewed 61 patients. Median age at surgery was 10 days (25%,75%:7,30). Cardiac anatomy was biventricular in 38 patients (62%), hypoplastic right ventricle in 14 patients (23%), and hypoplastic left ventricle in 9 patients (15%). Inotropic support was implemented in 30 patients (49%). Baseline characteristics of patients who received inotropic support, including ventricular anatomy and pre-operative ventricular function, were not statistically different from the rest of the cohort. Patients who received inotropic support, however, were exposed to larger cumulative doses of ketamine intraoperatively – median 4.0 mg/kg (25%,75%:2.8,5.9) versus 1.8 mg/kg (25%,75%:0.9,4.5), p < 0.001. In a multivariable model, cumulative ketamine dose greater than 2.5mg/kg was associated with post-operative inotropic support (odds ratio 5.5; 95% confidence interval: 1.7,17.8), independent of total surgery time. Conclusions: Inotropic support was administered in approximately half of patients who underwent pulmonary artery banding and more commonly occurred in patients who received higher cumulative doses of ketamine intraoperatively, independent of the duration of surgery.

Abstract

Management of complex renal cysts is guided by the Bosniak classification system, which may be inadequate for risk stratification of patients for intervention. Fractional tumor vascularity (FV) calculated from volumetric contrast-enhanced ultrasound (CEUS) images may provide additional useful information.To evaluate CEUS and FV calculation for risk stratification of patients with complex renal cysts.This was a pilot prospective study with institutional review board approval involving patients undergoing surgery for Bosniak IIF-IV complex renal cysts. CEUS was performed preoperatively on the day of surgery with two-dimensional (2D) and three-dimensional (3D) imaging and sulfur hexafluoride lipid-type A microspheres as the ultrasound contrast agent.A custom MATLAB program was used to select regions of interest on CEUS scans. FV was calculated according to FV = 1 - (total nonenhancing area/total lesion area). We assessed the ability of 2D- and 3D-derived percentage FV (2DFV%, and 3DFV%) and Bosniak classification schemes (pre-2019 [P2019B] and post-2019 [B2019]) to predict malignancy, aggressive histology, and upstaging on surgical pathology. Performance was assessed as area under the receiver operating characteristic curve (AUC).Twenty eligible patients were included in final analysis, of whom 85% (n = 17) had Bosniak IV cysts and 85% (n = 17) had malignant disease on final pathology. Four (24%) of the malignant lesions were International Society of Urological Pathology grade 3-4. The AUC for predicting malignancy was 0.980, 0.824, 0.863, and 0.824 with P2019B, B2019, 2DFV%, and 3DFV%, respectively. When the Bosniak classification was combined with FV%, three models had an AUC of 1, while the combined 2DFV% + B2019 model had AUC of 0.980.FV is a novel metric for evaluating complex cystic renal masses and enhances the ability of the Bosniak classification system to predict malignancy. This metric may serve as an adjunct in risk stratification for surgical intervention. Further prospective evaluation is warranted.Cysts in the kidney are currently classified using a scheme called the Bosniak system. We assessed measurement of the percentage of vascular tissue (called fractional vascularity) in cysts on a special type of ultrasound scan. This promising test adds information when combined with the Bosniak system and can help in guiding appropriate treatment.

Dr. Jonathan B. Clark is an adjunct associate professor of Neurology and Space Medicine at Baylor College of Medicine. From 1997 to 2005, he worked at NASA as a six-time Space Shuttle crew surgeon and was chief of the Medical Operations Branch at the Johnson Space Center. During this event, Dr. Clark delivered a historic review of Extravehicular Activity (EVA) events with an emphasis on EVA system hazards. This is one event of a medical series delivered to the SKC Program by Dr. Clark. This series focuses on spacesuit medical lessons learned and medical safety aspects of the suit.
