3. Immunohistochemical Localization of Leptin and Ghrelin in Kidney Tissue of Capsaicin Administered Diabetic and Non-diabetic Rats.

In this study, we investigated the effect of Capsaicin (CAP) on leptin and ghrelin expression in diabetic kidneys by immunohistochemical method. The subjects were separated into 5 groups: control, sham, diabetes, CAP, and D+CAP. Streptozotocin (45 mg/kg, single dose) was administered to each rat in the diabetes and D+CAP groups intraperitoneally. Rats with blood glucose levels of200 mg/dL or higher were considered diabetic. Capsaicin (1 mg/kg) was administered subcutaneously to the rats in the CAP and D+CAP groups every day for 2 weeks. H&E, PAS, and Crossman's Triple staining were performed to examine the histology of the kidney. Leptin and ghrelin in the kidney were examined by immunohistochemical method. Histological examination revealed normal kidney tissue in the control, sham, and CAP groups. The diabetes group showed the glomerulus with scattered and narrowing of the Bowman's capsule. While leptin immunoreactivity was most intense in the diabetes group, it was observed the weakest in the CAP and D+CAP groups. While strong ghrelin immunoreactivity was seen in the control and sham groups, mild immunoreactivity was seen in the CAP group, and weak immunoreactivity was seen in the diabetes and D+CAP groups. In this study, we concluded that the application of CAP could prevent loss of appetite by reducing the leptin expression during diabetes, but could not protect ghrelin from adverse effects of diabetes. 5. Determination of the Protective Impact of Hypericum perforatum Extract Against Ovarian Ischemia and Reperfusion in Rats.

Objectives: Ovarian torsion is an important gynaecological emergency because it causes ischemia in ovary. Our aim is to research the effects of Hypericum perforatum on p53, TNF-α, CAT, SOD, GSH, PTEN and is haPI3K/Akt/mTOR pathway in ovarian of rats with ischemia reperfusion (I/R) injury. Materials and Methods: The study included 56 adult female rats which were allocated to 7 groups; control, ischemia, ischemia 300HP, ischemia 600HP, I/R, I/R 300HP and I/R 600HP. H. perforatum was applied to ischemia 300HP, ischemia 600HP, I/R 300HP and I/R 600HP groups at the dose of 300 mg/kg or 600 mg/kg by oral gavage. Collected ovarian tissues were examined by light microscopy, biochemical and real-time PCR techniques. Results: In histological examination, the least degeneration was seen in 600HP group among other groups, apart from control group. TUNEL results showed that apoptosis level of other groups was lower than that of ischemia and I/R groups. CAT and GSH levels of I/R 600HP group increased. The highest level of PTEN and mTOR was found in the ischemia group. The Akt level was lower in I/R and I/R 300HP groups but its level was close to that of control and I/R 600HP groups. TNF-α and p53 mRNA expressions in I/R group were increased. Conclusions: The result of the study showed that H. perforatum (600 mg/kg) had an effect on tissue degeneration, CAT and GSH levels, and also TNF-α and p53 mRNA expressions levels. We suggest that H. perforatum can be considered as an agent to protect against tissues damage during ischemia reperfusion. [ABSTRACT FROM AUTHOR]

Bu araştırma, bıldırcın içme suyuna oregofarm (nane-kekik-anason yağı) ilavesinin büyüme ve besi performansı, karkas özellikleri ve ileumun histolojik yapısı üzerine etkisini belirlemek amacıyla yapılmıştır. Araştırmada her iki cinsiyette toplam 348 adet Japon bıldırcın (Coturnix coturnix japonica) civcivi kullanılmıştır. Her grupta 116 civciv bulunan 1 kontrol 2 deneme grubu oluşturulmuştur. Gruplar kendi aralarında 4'erli alt gruba ayrılmıştır ve her alt grup 29 adet civcivden oluşturulmuştur. Deneme altı hafta sürdürülmüştür. Bütün deneme grupları temel rasyonla beslenmiştir ve deneme boyunca temiz içme suyu sağlanmıştır. Kontrol grubunun içme suyuna oregofarm ilavesi yapılmamıştır. 1. deneme grubu ve 2. deneme grubu içme suyuna sırasıyla 1.0 mg/5 L ve 1.5 mg/5 L. oregofarm ilavesi yapılmıştır. Tüm deneme gruplarına yem ve su ad-libitum olarak verilmiştir. Araştırma sonunda, yem tüketimi ve yemden yararlanma oranı bakımından 3. haftada istatistiksel açıdan önemli farklılıklar (P0.05). Denemenin sonunda gruplar arasında sıcak ve soğuk karkas parametreleri bakımından istatistiksel olarak farklılıklar bulunmuştur (P0.05). Sonuç olarak, bıldırcın içme suyuna oregofarm ilavesinin performansı artırıcı ilave bir etkisinin olmadığı tespit edilmiştir., This study was conducted in order to define the effects of oregofarm (peppermint, thyme and anise oil) supplementation in the drinking water on the growth performance, carcass quality and histologic structure of terminal ileum in quails. A total of 348 Japanese quail chicks (Coturnix coturnix japonica) of both sexes were included in this study. They were divided into one control group and two experimental groups and each of them contained 116 Japanese quail chicks. Each group was further divided into four subgroups with 29 Japanese quail chicks. This study was finalized in six weeks. All groups were fed with basal diets and received fresh water during the experiment. The control group received non-supplemented water. The group 1 and 2 received 1.0 ml/5 L and 1.5 ml/5 L oil mixture, respectively. All experimental groups were fed with water and ad-libitum. As a result of the study, there were statistically significant differences between the feed consumption and efficiency (P0.05). At the end of the study, there were statistically differences in the warm and cold carcass parameters (P0.05). Conclusively, the supplementation of oregofarm (peppermint + thyme and anise oil) has no additional effect on quail performance.

The aim of the present study was to determine the histological structure and histochemical properties of the pancreatic ducts in the goose Anser anser. Pancreatic ducts of 6 male and 6 female geese were examined. Crossman’s triple stain and Masson’s trichrome stain were used for histological examination, and periodic acid–Schiff, alcian blue, periodic acid–Schiff/alcian blue, and aldehyde fuchsin/alcian blue stainings were performed for histochemical examination. Dorsal and ventral pancreatic ducts were found that transfer pancreatic secretion into the duodenum of the goose, and these channels were found to have a histologically similar structure. No differences were detected between male and female geese. Duct wall was observed to consist of mucosa, muscle, and adventitial layers. In addition to dark principal, light and basal cells were observed in a simple columnar epithelium layer lining the duct. Glands, including centroacinar cells, were also detected in all layers of duct. Furthermore, lymphoid follicles were observed in the connective tissue of the mucosa. Sulfated and carboxylated mucosubstance-secreting cells were found in the duct as well as neutral, acidic, and mixed mucosubstance-secreting cells. We conclude that the results of our study may contribute to future studies.

The aim of this study is to examine the goose (Anser anser) cerebellum histometric and histologically. Six female geese (aged between 10-12 months) were used in this study. After fixation in 10% formalin and following routine histologic processes, the cerebellar tissues of geese were embedded in paraffin blocks and the sections were taken from these blocks. The Kluver-Barrera nerve tissue stain technique was applied to the sections. The ocular micrometer was used for histometric measurements of folia length, diameter of purkinje cells and thicknesses of stratum granulosum and stratum moleculare. SPSS 16.0 was used for calculating mean values. It was observed histological that folia and sulci cerebelli were in the outermost layer of the cerebellum. As a result of Kluver-Barrera staining, the nerve cells in all layers appeared purple and myelin fibers appeared blue. It was found that the average length of the folia was 3397.88 ± 931.32 µm, the average thickness of molecular layer was 348.53 ± 72.92 µm, the average thickness of granular layer was 184.83 ± 48.77 µm and the average diameter of Purkinje cells was 24.23 ± 3.96 µm. 18. Examination of the immunohistochemical localization and gene expression by RT-PCR of the oxytocin receptor in diabetic and non-diabetic mouse testis.

Objective(s): The aim of this study was to determine Oxytocin receptor (OTR) gene expression and localization in diabetic and non-diabetic mouse testes by RT-PCR and immunohistochemistry, respectively. Materials and Methods: In this study, 18 male BALB/c mice (8-12 weeks old) were used and divided into three groups: diabetic, sham, and control. Streptozotocin (STZ) was applied to the diabetic group and sodium citrate was administered to the sham group in the same way, however, the control group was left untouched. The testicular tissues were removed on the thirtieth day of testing; the right testis tissues were passed through a routine histologic process and sections were stained with H&E and PAS staining techniques. The avidin-biotin-peroxidase method was applied to determine OTR immunoreactivity, while the left testis tissues were used for RT-PCR. Results: It was found that the body weight had decreased in the diabetic group and the diameter of the seminiferous tubules in the said group was shorter than those of the other groups. There were no obvious differences with regard to the histologic appearance between the groups. The immunohistochemical examination showed that the OTR immunoreactivity was strong in the control and sham groups but weak in the diabetic group, and the immunoreactivity was only seen in the Leydig cells. In addition, the OTR gene expression was lower in the diabetic group than in the other groups. Conclusion: We concluded that diabetes reduces the OTR expression in the testis. It is suggested that OTR protection should be researched in diabetes for healthy reproduction and sexuality. [ABSTRACT FROM AUTHOR]

Diabetes mellitus hastalığı, dünya üzerinde yaygın olarak seyreden hastalıklar arasında yer almaktadır. Bilim dünyası, DM ve komplikasyonları ile antioksidanlar arasındaki ilişkiyi araştırmaya devam etmektedir. Bu çalışmada; deneysel diabet oluşturulan farelerin böbreklerinde, katalaz gen ekspresyonu düzeyini RT-PCR yöntemiyle, katalaz enziminin lokalizasyonunu immunohistokimyasal ve dokuda meydana gelen yapısal değişiklikleri histolojik olarak incelemek amaçlandı.Çalışmada kullanılan 36 adet swiss albino fare deneme (n=15), sham (n=15) ve kontrol (n=6) olmak üzere üç gruba ayrıldı. Deney hayvanları 30 gün boyunca standart fare yemi ile ad libitum olarak beslendi. Deneme grubuna, Na sitrat tamponu içinde çözdürülen 100 mg/kg doz i.p STZ, sham grubuna Na sitrat tamponu i.p. olarak uygulandı. Deneysel diabet oluşturulan grupta kan glikoz düzeyi 200 mg/dl üzerinde olan fareler diabetik kabul edilerek çalışmaya alındı. Katalaz gen ekspresyonu düzeyini tespit etmek için RT-PCR yöntemi kullanıldı. Katalaz enziminin lokalizasyonu immunohistokimyasal olarak incelendi. Histolojik incelemeler için H.E. ve PAS boyama yöntemleri kullanıldı. Katalaz enzimi protein aktivitesinin tayini için Aebi'nin yöntemi kullanıldı. Ayrıca, grupların canlı ağırlık ve böbrek ağırlıkları da karşılaştırıldı.Diabetik grupta ortalama kan glikoz düzeyi, 3. günden itibaren ölçüm yapılan tüm günlerde 300 mg/dl'nin üzerinde seyretti. Katalaz gen ekpresyonunun ve enzim aktivitesinin deneme grubunda sham ve kontrol gruplarına göre düşük olduğu tespit edildi (P < 0,05). İmmunohistokimyasal olarak tüm gruplarda kortekste belirgin, fakat deneme grubunda diğer gruplara nazaran daha hafif bir katalaz boyamasının olduğu gözlendi. Deneme grubunda, diğer gruplara göre canlı ağırlıkta azalma böbrek ağırlığında ise artış olduğu bulundu (P < 0,05).Diabetik böbreklerin tubulus proksimalislerinde glikojen birikimi gözlendi. Hücre çekirdeklerinin daha bazofilik ve dış hatlarının daha düzensiz olduğu görüldü. Diabetes mellitus (DM) is one of the disease that has wide-spread effect on the world. Scientists have been carrying out the studies elucidating the relationship between antioxidants and complications of DM for decades. The aim of this study was to determine the gene expression pattern of catalase by RT-PCR technique, its tissue localization by immuno-histochemistry method and the structural aberration of DM kidney by routine histological techniques.İn this study, 36 swiss albino mice were used . Animals were divided into three groups as follows; treatment (n = 15), sham (n = 15) and control (n = 6). Animals were feeded with standard mouse feed as ad libitum for 30 days. Streptozotocin (STZ, 100 mg / kg ) prepared within Na citrate buffer solution was apllied to treatment group and Na citrate buffer solution was applied to sham group as intra peritoneal injection. Mice in which blood glucose level were above 200 mg/dl were taken as DM group and considered as treatment or experimental group for the present study. To determine the level of catalase gene expression level, RT-PCR technique was used. Tissue localization of catalase enzyme was analysed by immunohistochemical methods. Hematoxylin Eosin (HE) and Periodic Acid Schiff (PAS) staining methods were used for histological examination. Aebi's method was used for determination of enzyme activity of catalase. In addition, groups were compared with body weight and kidney weight.Blood glucose level of mice from diabetic group was above 300 mg/dl starting on day 3 of the current study and stayed at that level for the rest of the experimental period. Gene expression and enzyme activity of catalase from treatment group were found to be lower than that of sham and control groups (P < 0,05). Catalase staining was mainly observed in the cortex region of kidney from all groups, whereas its staining was lesser in treatment group than that of other groups. The treatment group had lowered body weights along with the increased kidney weights compared that of control and sham groups (P < 0,05).Glycogen accumulation was observed in the proximal tubule of diabetic kidney. Nucleus of the cell was observed to be more basophilic, and the outer frame of nuclei was seen as more irregular. Light hydropic degeneration were found especially in distal tubule of diabetic kidney.This study will make a nice contribution to beter understanding of the relationship between antioxidants and DM and lead the new researchs in the field of DM.Key words: Diabetes Mellitus, Catalase, RT-PCR, İmmunohistochemistry, Kidney 113

The purpose of this study was to examine the comparative differences of eggshell structure between geese and chickens during embryonic development by SEM. Goose eggs and chicken eggs were used for the study. Eggs of each species were divided into three groups: The eggs within first group were used subsequent to collection, those of second group were collected during the mid-point of incubation and those of third group were collected at the end of the incubation. The eggshells samples with dimensions of 0.5 cm2 were taken from the equatorial region of eggshells. After these samples were passed through routine process, they were examined via SEM for structural changes of eggshells and rate of elements. In the examination, there were not changes of eggshell structure between two species for days of the incubation. Eggshells thickness of each species and the rate of calcium, oxygen, nitrogen and carbon of eggshells were not statistically different among groups. It was concluded that structural changes of eggshell did not have significant effect on low yield for hatching of goose. [ABSTRACT FROM AUTHOR]

The aim of the present study was to determine the histological structure and histochemical properties of the pancreatic ducts in the goose Anser anser. Pancreatic ducts of 6 male and 6 female geese were examined. Crossman's triple stain and Masson's trichrome stain were used for histological examination, and periodic acid-Schiff, alcian blue, periodic acid-Schiff/alcian blue, and aldehyde fuchsin/alcian blue stainings were performed for histochemical examination. Dorsal and ventral pancreatic ducts were found that transfer pancreatic secretion into the duodenum of the goose, and these channels were found to have a histologically similar structure. No differences were detected between male and female geese. Duct wall was observed to consist of mucosa, muscle, and adventitial layers. In addition to dark principal, light and basal cells were observed in a simple columnar epithelium layer lining the duct. Glands, including centroacinar cells, were also detected in all layers of duct. Furthermore, lymphoid follicles were observed in the connective tissue of the mucosa. Sulfated and carboxylated mucosubstance-secreting cells were found in the duct as well as neutral, acidic, and mixed mucosubstance-secreting cells. Bu çalışmanın amacı sezeryan ve normal doğum yapmış kadınlardan elde edilen plasentalarda oksitosin reseptörü (OXTR) immünoreaktivitesini karşılaştırmaktır. Çalışma örneklerini Kars Harakani Devlet Hastanesi ve Kafkas Üniversitesi Sağlık Araştırma ve Uygulama Hastanesi'de 16 adet normal doğum yapmış (kontrol) ve 16 adet sezaryen (olgu) doğum yapmış sağlıklı iki gruptan oluşan 32 annenin plasentaları oluşturmaktadır. Toplamda elde edilen 40 plasentadan 5'i kan uyuşmazlığı nedeni ile, 2'si sistemden daha sonra elde edilen kronik rahatsızlığın (troid, Behçet hastalığı) tespiti ile, 1 tanesi ise aksesuar plasenta bulundurması nedeni ile çalışmaya dahil edilmemiş olup, çalışma 32 plasenta üzerinde sürdürülmüştür. İkili değişkenlere ait ortalamalar Bağımsız İki Örneklem T-Testi uygulanarak yapılmıştır. Değişkenler arasındaki ilişkiyi incelemek için ise Pearson Korelasyon analizi kullanılmıştır. Plasentalardan elde edilen doku örneklerine histolojik incelemler için Periyodik asit-Schiff Boyama, H&E ve Triple boyama teknikleri uygulandı. İmmunohistokimyasal inceleme için OXTR primer antikoru kullanıldı. Histolojik açıdan gruplar arasında belirgin bir fark gözlenmedi. Çalışmada uterusa yakın olan maternal kısmında bulunan desidua hücrelerinde ve villus stromasında OXTR'ü immünoreaksiyonunun daha fazla olduğu tespit edildi. Villus stromasında normal doğum grubunda sezaryen doğum grubuna göre daha güçlü bir OXTR immunoreaktivitesi tespit edilidi. Sinsityotrofoblast hücrelerinde ve sinsityal düğümlerde reaksiyona her iki grupta da rastlanmazken fetal endotel hücreleri ve fetal kapilleri etrafında güçlü immünoreaksiyon gözlemlendi. Sonuç olarak, yavru ile ilgili bilgi vermesi nedeniyle plasentanın her yönüyle araştırmalara konu edilmesi ayrıca plasenta patolojisinin incelenmesinin her doğum için rutin hale getirilmesi gerektiği düşünülmektedir. The aim of this study was to compare oxytocin receptor (OXTR) immunoreactivity in placentas obtained from women having cesarean and natural birth. The study samples consisted of placentas of 32 mothers who had 16 natural birth (control) and 16 cesarean (case) birth in Kars Harakani State Hospital and Kafkas University Health Research and Application Hospital. At the beginning of the study, 40 placentas obtained in total, 5 were not included in the study due to mismatched blood, 2 were not included in the study after the chronic disease (thyroid, Behçet's disease) detected, and 1 was not included in the study due to the presence of accessory placenta. The means of binary variables were determined by applying the Independent Two-Sample T-Test. Pearson Correlation analysis was used to examine the relationship between the variables. Periodic acid-Schiff staining, H&E and Triple staining techniques were applied to tissue samples obtained from placentas for histological examination. OXTR primary antibody was used for immunohistochemical examination. There was no significant difference between the groups in terms of histological examination. In the study, it was found that OXTR immunoreactivity was strong in decidual cells and stromal cells in villi. A stronger OXTR immunoreactivity was detected in villous stroma of placenta of the natural birth group compared to the cesarean birth group. Reaction was not observed in syncytotrophoblast cells and syncytial nodes in both groups, but strong immunoreaction was observed fetal endothelial cells and fetal capillaries in both groups.As a result, it is thought that the placenta pathology should be made routine for every birth because it gives information about the offspring. 99

Bu çalışma, sezaryen ve normal doğum yapmış kadınlardan elde edilen plasentalarda katalaz enzimi immunoreaktivitesinin karşılaştırılması amacıyla yapılmıştır. Çalışma için 18-49 yaş aralığında sağlıklı miadında normal (16 adet) ve sezaryen (16 adet) doğum yapan ve dışlama kriterlerinden herhangi bir özelliği taşımayan kadınların plasentaları kullanıldı. Çalışma grupları; Kontrol (normal doğum) ve Olgu (sezaryen doğum) grubu olarak belirlendi. Alınan plasentaların merkez (kord çıkış yeri), perifer ve orta kısmından kesitler alındı. Alınan plasenta dokuları rutin histolojik işlemlerden geçirilerek parafinde bloklanıp örnekler alındı. Alınan kesitlere histolojik olarak incenlenmek için H&E, PAS ve Crossman'ın üçlü boyaması yapıldı. Katalaz'ın immunoreaktivitesini belirlemek için Avidin-Biotin-Peroksidaz Kompleks (ABC) metodu uygulandı. Grupların Anne, Bebek ve Plasenta Bilgi Formu'ndaki verileri, plasenta doku örneklerinde Katalaz'ın immünohistokimyasal lokalizasyonu ve plasentanın histolojik yapısı karşılaştırıldı. Anne yaşı arttıkça gravida sayısının da arttığı, anne yaşı arttıkça anne beden kitle indeksinin arttığı, bebek kilosu arttıkça plasenta ağrılığının da arttığı ve normal doğumlara oranla, sezaryen doğumlarda kan kaybının daha fazla olduğu görüldü. Histolojik incelemeler sonucunda gruplar arasında histolojik açıdan belirgin farklar görülmedi. İmmunohistokimyasal incelemeler sonucunda gruplar arasında Katalaz'ın aynı yapılarda immunoreaktivite gösterdiği, immunoraktivitenin fetal ve maternal damarlarda (intima, media, adventisya), fetal ve maternal kanda, villöz stromada, desidualize hücrelerde, desidua stromasında, sitotrofoblastlarda ve hafbauer hücrelerinde olduğu görüldü. Katalaz immunoreaktivitesinin normal doğum grubunda sezaryen doğum grubuna göre daha güçlü olduğu tespit edildi. Sinsisyotrofoblastlar ve sinsisyal düğümlerde ise her iki grupta da katalaz immunoraktivitesinin olmadığı görüldü. The aim of this study was to compare catalase enzyme immunoreactivity in placentas obtained from women who had cesarean and natural birth. Placentas of women who had natural (16) and cesarean (16) birth between 18-49 years of age and did not have any exclusion criteria were used for the study. Working groups were divided into control (natural birth) and case (cesarean birth) groups. Slices were taken from the central (cord outlet), periphery and middle parts of the placentas. The placenta tissues were subjected to routine histological procedures and they were blocked in the paraffin. To obtain histological examination of the sections, H&E, PAS and Crossman's triple techniques were performed. Avidin-Biotin-Peroxidase Complex (ABC) method was used to determine the immunoreactivity of catalase. Data of the groups of the Mother, Baby and Placenta Information Forms, immunohistochemical localization of Catalase in placenta and histological structure of placenta were compared. It was seen that the number of gravida increased with increasing maternal age, maternal body mass index increased with increasing maternal age, placental weight increased with increasing baby weight and blood loss was higher in cesarean birth compared to natural birth group. There were no significant histological differences between the groups. As a result of immunohistochemical studies, catalase showed immunoreactivity in the same structures among the groups, and immunoractivity was observed in fetal and maternal vessels, fetal and maternal blood, villous stroma, decidualized cells, decidual stroma, cytotrophoblasts and hafbauer cells. Catalase immunoreactivity was found to be stronger in natural birth group than cesarean birth group. Amaç:Bu çalışmada, trombositten zengin fibrin (TZF) membran implantasyonu uygulamasının, deneysel alkali yanıklarda ortaya çıkacak inflamasyon, yeni damar oluşumu, korneal ülser ve opasite oluşumu üzerine etkisinin, morfolojik, histolojik ve immunohistokimyasal olarak incelenmesi, ayrıca medikal tedavi ve amniyotik membran transplantasyonu (AMT) tedavilerinin etkinliği ile karşılaştırılması amaçlanmıştır. Gereç ve Yöntem: Çalışmaya dahil edilen 24 adet Yeni Zelanda ırkı tavşanın 24 sağ gözü, biri kontrol grubu olmak üzere (1. Grup, n=6) 4 eşit gruba ayrıldı. İlk gün deneklerin kornealarında topikal anestezi (Alcaine % 0,5 damla; Alcon) altında 1 normal (N) NaOH ile alkali yanık oluşturuldu. Tedavi olarak 2. Gruba (n=6) topikal antibiyotik ve steroid, 3. Gruba (n=6) topikal tedaviyle birlikte AMT, 4. Gruba (n=6) topikal tedaviyle birlikte TZF membran implantasyonu yapılması planlandı. Birinci, 7, 14 ve 21. günlerde kli-nik muayeneleri yapıldı. Yirmibirinci günde denekler sakrifiye edilerek korneaları limbus sınırından çıkarılarak, dokular histopatolojik incelemeye gönderildi. Bulgular: Alkali yanık sonrası ilk vizitte gruplar arasında epitel defekti, kornea kalınlığı ve opasite açısından fark yoktu (p>0,05). Santral kornea kalınlığında vizitler ve gruplar arasında herhangi bir faklılık izlenmedi (p>0,05). Epitel defektin büyüklüğünde, TZF membran ve AMT grubunda, tedavi sonrası ilk vizite göre anlamlı iyileşme görüldü (p 0.05). Bu çalışmanın amacı; anneler, bebekler ve plasentalardan toplanan veriler ile sezaryen ve normal doğumla elde edilen plasentalarda histolojik ve melatonin reseptör 1B immunoreaktivetesi karşılaştırmaktır.Temel bir endokrin organ görevini gören plasenta gebelik sırasında anne ile fetüs arasındaki besin transferini sağlar. Çalışmamızda Kars Harakani Devlet Hastanesi'nde ve Kars Kafkas Üniversitesi Eğitim Araştırma ve Uygulama Hastanesi'nde 01/10/18-18/12/2018 tarihleri arasında miadında sağlıklı normal vajinal (16 kadın) ve sezaryen (16 kadın) doğum yapan, 18-49 yaş aralığında olan ve çalışmaya katılmayı kabul eden toplam 32 sağlıklı kadının uterin kaviteden normal ve sezaryen doğum sonrası attıkları plasentaları topladık. Kadınlardan ve bebeklerden alınan bilgilerle istatistiksel analiz yapıldı. İkili değişkenlere ait ortalamaların karşılaştırması bağımsız iki örneklem için Student t-testi uygulanarak yapıldı. Değişkenler arasındaki ilişkiyi incelemek için ise pearson korelasyon analizi kullanıldı. Toplanan plasentaların perifer, orta ve merkezinden kesitler alınarak Triple, Periodic Asit Schiff ve Hematoksilen-Eozin boyama ve melatonin reseptörüyle immün boyama yapıldı. Plasentanın perifer, orta ve merkezinden alınan kesitlerde plasentanın histolojik yapısındaki koryon villus ve desidua hücrelerinin dağılımı ve görünümü açısından herhangi bir değişiklik gözlenmedi. İstatistiksel verilerde doğum şekline göre gravida, parite, abortus, küretaj, yaşayan çocuk sayıları arasında ve cinsiyet dağılımında anlamlı bir fark saptanmadı. Literatürde maternal kanda artan melatoninin plasentadan kaynaklandığını öne süren çalışmalar olduğunu gördük fakat normal vajinal ve sezaryen ile doğum yapan kadınların plasentalarında melatonin reseptör 1B immünoreaktivitesi ile ilgili çalışmaya rastlamadık. Melatonin reseptör 1B immünoreaktivitesinin normal doğum plaseantalarının desidua hücrecelerinde güçlü, sezaryen doğum plasentalarının desidua hücrelerinde hafif düzeyde olduğu gözlendi. Histolojik yapı bakımından her iki grup arasında belirgin bir fark olmadığı görüldü. Sonuç olarak, sezaryen doğum ve normal doğum plasentaları arasında desidua hücrelerindeki melatonin reseptör 1B immuonreaktivitesi bakımından bir farklılık dışında belirgin bir farklılık gözlenmemesine rağmen plasentanın gebeliğin her döneminde değerlendirilmesi ve incelenmesinin rutin hale gelmesinin önemli olduğu düşünülmektedir. In this study, it was compared some data obtained from mothers, infants and placenta, and melatonin receptor 1B and histological structure of placenta between caesarean section and normal birth. Placenta has an important role as a basic endocrine organ and provides nutrient transfer between mother and her fetus during the prenatal period. In our study, we collected placenta excreted from uterine cavity of healthy and ful term pregnant women who are between 18-49 years old, accept to participate the study, and have birth by normal vaginal (16 female) and cesarean section (16 female) in Kars Harakani State Hospital and Kars Kafkas University Educational Research and Application Hospital between 01.10.2018-18.12.2018. Statistical analysis was performed with data obtained from women and infants. The comparison between the mean of two variables were performed using the Student t-test. Pearson correlation analysis was used to evaluate the relationship between the variables. Peripheral, middle and central sections of the placentas were taken and Triple, Periodic Acid Schiff and Hematoxylin-Eosin staining and immunostaining with melatonin receptor 1B were performed. It was not observed any difference in the distribution and appearance of the chorionic villus and decidua cells in terms of the histological structure of the placenta in the sections taken from the peripheral, middle and central sections of the placenta. There was no significant difference between the numbers of gravida, parity, abortus, and curettage, number of living children and gender distribution of infants according to the type of birth. There are studies suggesting that melatonin increased in maternal blood originates from the placenta, but we did not encountered any study on melatonin receptor 1B immunoreactivity in placentas of women who had normal vaginal and cesarean birth. Melatonin receptor 1B immunoreactivity was found to be strong in decidua cells of normal vaginal birth and mild in decidua cells of cesarean birth. There was no significant difference in histological structure between the two groups. In conclusion, although there is no significant difference between cesarean birth and normal vaginal birth placentas except of melatonin receptor 1B immuonreactivity in decidua cells, it is considered that placenta evaluation and examination becomes routin in every period of pregnancy. 85

ÖZETBu çalışmada, diyabetik ve non-diyabetik fare kalp dokusunda oksitosin reseptörünün immunohistokimyasal lokalizasyonunun incelenmesi amaçlanmıştır. Çalışmada 18 adet Balb-c cinsi erkek fare kullanıldı. Deney grupları; kontrol, sham ve diyabet grubu olarak belirlendi. Diyabet grubuna i.p. enjeksiyonla Streptozotosin (STZ) verildi ve diyabet oluşturuldu. Sham grubuna yine i.p. yolla sodyum sitrat çözeltisi uygulandı. Kontrol grubundaki hayvanlara ise herhangi bir uygulama yapılmadı. STZ uygulandıktan 30 gün sonra farelere eter anestezi altında servikal dislokasyon yapıldıktan sonra kalp dokuları alındı. Alınan her bir kalp dokusu düşey olarak ikiye bölünerek rutin histolojik işlemlerden geçirilerek parafinde bloklanıp kesitler alındı. Alınan kesitlere, histolojik olarak incenlenmek üzere Crossman'ın üçlü boyaması (triple), H&E ve PAS yapıldı. Oksitosin reseptörü'nün immunoreaktivitesi Avidin-Biotin-Peroksidaz Kompleks (ABC) metodu uygulanarak belirlendi. Deney süresi bitiminde; grupların canlı ağırlıkları, kan glikoz seviyesi, doku ağırlıkları ve kalp doku örneklerinde oksitosin reseptörünün immunohistokimyasal lokalizasyonu ve dokunun histolojik yapısı karşılaştırıldı. Diyabet grubunda ilk ve son gün canlı ağırlığı karşılaştırıldığında son gün canlı ağırlığında azalma belirlendi. Kalp dokusunun ağırlıkları gruplar arasında karşılaştırıldığında gruplar arasında istatistiksel düzeyde bir fark olmadığı. Histolojik incelemeler sonucunda diyabet grubunda diğer gruplardan histolojik açıdan belirgin bir farklar olduğu tespit edildi. İmmunohistokimyasal incelemeler sonucunda, sham ve kontrol gruplarında oksitosin reseptörünün benzer özellikte immunoreaktivite gösterdiği görüldü. Diyabet grubunda ise diğer gruplara göre oksitosin reseptörünün immunoreaktivitesinin endotel ve kapiller alanlarda benzer, hücre membranı, sitoplazma ve purkinje hücrelerinde daha az olduğu görüldü. Anahtar Sözcükler: Diyabet, immünohistokimya, Kalp, Oksitosin, Oksitosin Reseptörü SUMMARYThe aim of this study was to investigate the immunohistochemical localization of oxytocin receptor in diabetic and non-diabetic mouse heart tissue. Eighteen male Balb-c mice were used in the study. Animals were divided into three groups; control, sham and diabetes. The diabetes group was given STZ by i.p injections and diabetes was induced. Sham group was again treated with sodium citrate solution by ip. The animals in the control group did not receive any treatment. After 30 days of STZ application, mice were cervical dislocated under ether anesthesia and their heart tissues were removed. Each heart tissue was vertically divided into two parts and routine histological procedures were applied and then tissues were blocked in paraffin and sections were taken. For histological examination, Crossman triple staining, H&E and PAS were applied to the sections. Immunoreactivity of OTR was determined by Avidin-Biotin-Peroxidase Complex (ABC) method. At the end of the study period; live weight of the groups, blood glucose level, tissue weights and immunohistochemical localization of OTR in heart tissue samples and histological structure of tissue were compared. When the first and last day body weight were compared in the DM group, it was determined a decrease in the last day and in control group, it was determined an increase in the last day. When weights of heart tissue were compared between the groups, there was no statistically significant difference between the groups. As a result of histological examinations, it was found that there was a significant difference histologically in the diabetes group from the other groups. Immunohistochemical examinations showed that oxytocin receptor showed similar immunoreactivity in sham and control groups. In the diabetic group, the immunoreactivity of oxytocin receptor was similar in endothelial and capillary areas, but less in cell membrane, cytoplasm and purkinje cells.Keywords: Diabetes, immunohistochemistry, Heart, Oxytocin and Oxytocin Receptor 63
