Abstract The objective of the present experimental study was to gain a better understanding of the foraging activity of Asellus aquaticus during fish egg incubation. A. aquaticus were introduced into experimental setups of dead eggs, viable eggs and hatched larvae of zebrafish (Danio rerio), a commonly used model organism. The amount of A. aquaticus and the duration of their exposure to the eggs significantly affected the proportion of consumed dead eggs in each experimental cycle. A. aquaticus belongs to the group of aquatic detritivores, and no predatory behavior was observed during the experiments. These crustaceans could distinguish between the dead eggs and those containing living embryos. Furthermore, zebrafish larvae remained unharmed by A. aquaticus, even in the absence of alternative food source. These findings underscore the potential sanitary role of these crustaceans in natural waters and offer new perspectives on their possible use as biological control organisms in aquaculture hatcheries. Additionally, our results suggest a potential application of A. aquaticus in combating pathogens by reducing the growth substrates for bacteria and fungi.

Hypophysation is the oldest and most widely used method of induced fish breeding. In practice, common carp (Cyprinus carpio) pituitary is one of the most commonly used agents to induce ovulation. The active ingredients of pituitary (glycoprotein) are very susceptible to fast degradation, therefore the glands must be preserved immediately after collection. Preservation is usually carried out in acetone which dehydrates and defattens the glands.The aim of our experiments was to investigate the efficacy of an extremely long-stored batch of pituitary gland collected in 1967. The biological potential of the glands was assessed through reproductive indices recorded during induced breeding of African catfish (Clarias gariepinus). The experiments were performed in October 2020. One group of females was treated with acetone-dried carp pituitary collected in 1967 and the other group was injected with pituitary collected in the year prior to the experiments. To assess the effectiveness of the pituitary treatment the following reproductive traits were determined: ratio of ovulated females, relative weight of the stripped eggs and the residual ovary, fertilization and hatching rates. The statistical methods used did not indicate a significant difference between the two groups for any of the reproductive indices, which means that the pituitary gland collected in 1967 remained effective after 53 years.

Abstract GBHs are the most widely used herbicides for weed control worldwide that potentially affect microorganisms, but the role of their sublethal exposure in the development of antibiotic resistance of Pseudomonas aeruginosa is still not fully investigated. Here, the effects of glyphosate acid (GLY), five glyphosate-based herbicides (GBHs), and POE(15), a formerly used co-formulant, on susceptibility to imipenem, a potent carbapenem-type antibiotic, in one clinical and four non-clinical environmental P. aeruginosa isolates were studied. Both pre-exposure in broth culture and co-exposure in solid media of the examined P. aeruginosa strains with 0.5% GBHs resulted in a decreased susceptibility to imipenem, while other carbapenems (doripenem and meropenem) retained their effectiveness. Additionally, the microdilution chequerboard method was used to examine additive/antagonistic/synergistic effects between GLY/POE(15)/GBHs and imipenem by determining the fractional inhibitory concentration (FIC) indexes. Based on the FIC index values, glyphosate acid and Total demonstrated a potent antagonistic effect in all P. aeruginosa strains. Dominator Extra 608 SL and Fozat 480 reduced the activity of imipenem in only one strain (ATCC10145), while POE(15) and three other GBHs did not have any effect on susceptibility to imipenem. Considering the simultaneous presence of GBHs and imipenem in various environmental niches, the detected interactions between these chemicals may affect microbial communities. The mechanisms of the glyphosate and GBH-induced imipenem resistance in P. aeruginosa are yet to be investigated.

(1) Background: Microinjection of zebrafish (Danio rerio) embryos offers a promising model for studying the virulence and potential environmental risks associated with Pseudomonas aeruginosa. (2) Methods: This work aimed to develop a P. aeruginosa infection model using two parallel exposition pathways on zebrafish larvae with microinjection into the yolk and the perivitelline space to simultaneously detect the invasive and cytotoxic features of the examined strains. The microinjection infection model was validated with 15 environmental and clinical strains of P. aeruginosa of various origins, antibiotic resistance profiles, genotypes and phenotypes: both exposition pathways were optimized with a series of bacterial dilutions, different drop sizes (injection volumes) and incubation periods. Besides mortality, sublethal symptoms of the treated embryos were detected and analyzed. (3) Results: According to the statistical evaluation of our results, the optimal parameters (dilution, drop size and incubation period) were determined. (4) Conclusions: The tested zebrafish embryo microinjection infection model is now ready for use to determine the in vivo virulence and ecological risk of environmental P. aeruginosa.

In this study, the inseminated sperm method (sperm injection into the ovarian lobe in externally fertilised fish species) combined with hormonal induction was conducted as a novel method to produce hybrid catfish by induced/tank spawning. In the treated group, four Clarias gariepinus ♀ were inseminated equally with Heterobranchus longifilis sperm in the ovaries at a dosage of 1 mL/fish and then introduced into four spawning cages together with four C. gariepinus ♂. Ovulation was induced by extracted carp pituitary (CPE) hormonal administration (♀; 5 mg CPE/BW kg). The hybridization rate was determined on the basis of morphological markers to distinguish between pure African catfish and hybrid in juvenile stage. A control experiment was also designed to determine this and to explore possible interactions between the two genotypes. In the control group, in vitro fertilisation (IVF) was used to produce pure progeny of C. gariepinus and the hybrid progeny between C. gariepinus ♀ × H. longifilis ♂. Exogenously fed larvae from each treatment were raised and compared after 28 days in four replications; the mixed group contained 50 C. gariepinus and 50 hybrids from IVF grown together, while every 100 larvae from each cage spawning pair were grown separately. The hybridisation rate determined based on morphological traits was 98.11 ± 1.59 (mean ± S.D.) for 28-day-old reared offspring. There were no significant differences (P > 0.05) in survival rate (%) between C. gariepinus (45.79 ± 6.82 %) and the hybrid (54.21 ± 6.82 %) in the control treatment; therefore, neither genotype had a negative effect on the other during the rearing period. From this result, we supposed that the hybridisation rate was also over 95 % at the moment of fertilisation. This approach could be an alternative method for producing hybrid catfish or other fish species without in vitro fertilisation.

In the experiments, defatted black soldier fly meal reared on vegetable byproducts was used in the fry rearing of two economically important fish species, African catfish and rainbow trout. Both fish species were reared in a recirculation system and 0–33–66–100% of the complex fry feed was replaced by a defatted prepupae meal of black soldier flies during a 28-day feeding experiment. African catfish was reared at 25 ± 1 °C while rainbow trout was reared at 12 ± 1 °C. The results showed that the growth of African catfish was not significantly reduced when 66% of the feed was replaced by soldier fly meal (mean weight in the control fish group at the end of the experiment was 0.4632 ± 0.2469 g, while the 66% group resulted mean weights of 0.4150 ± 0.1886 g) and the survival did not show any statistically different results (mean survival in control group was 57.48 ± 13.76% while it was 56.6 ± 7.763% in the 66% group). In the case of rainbow trout, replacing the feed entirely with insect meal did not cause a decrease in weight gain (final mean weight in the control group was measured at 1.9640 ± 0.4154 g, while in the group consuming only insect meal, it was 1.9410 ± 0.4248 g) or in survival (in the control group 98.5%, while in the group consuming only insect meal 99.5%). All these preliminary results indicate that black soldier fly meal can be used directly as a nursery feed in fish farming as a partial or total replacement of complete feeds. The results showed that black soldier fly meal could replace 66% of the complex brood feed of African catfish and up to 100% of rainbow trout feed without deterioration of production results. Our experiments have therefore opened the way for further experiments on insect meal in larval rearing.

Successful propagation of fish depends on many factors including the optimal administration routes and delivery vehicles of spawning agents. In our experiments, the effects of different routes of administration and delivery vehicles of Ovopel (mammalian GnRH-analouge and metoclopramide) on ovulation were compared in African catfish (Clarias gariepinus). Seven females per treatment were used (BW = 606–1088 g). In experiment 1, fish in treatment 1 and 2 were given intramuscular (IM) and intraperitoneal injections (IP) of Ovopel suspended in fish physiological saline, respectively. For treatment 3 and 4, Ovopel was suspended in fish physiological saline (NaCl) and fish sperm (SP), respectively. The suspensions were introduced into the ovary of the females by a catheter. In experiment 2 (verification or clarification experiment), two treatments were administrated identically as experiment 1 for IM and SP. As a new treatment (SI) sperm was introduced into the ovary by a catheter, the Ovopel were administrated intramusculary. In both experiments, all females ovulated in all treatment groups. The relative number of stripped eggs was also similar among treatments within experimental cycles. Hatching rate was significantly lower for treatments where fish sperm was introduced to the ovary by a catheter whether it contained Ovopel (39.1 ± 18.3 %; SP, experiment 1 and 35.1 ± 21.2 %; SP, experiment 2.) or not (SI: 46.0 ± 9.2 %). While in other treatments, hatching rates were significantly higher and varied between 55.1 ± 16.1 % (IM), 66.7 ± 9.3 % (IP), and 63.4 ± 9.0 % (NaCl) in experiment 1 and 61.9 ± 12.0 % (IM) in experiment 2. The result of ovarian lavage with hormone preparations in saline have shown that the effects this novel method do not differ from the effects of reproduction with the use of traditionally injection by needle.

Absorbance and computer-assisted sperm analysis (CASA) was used to assess the sperm concentration of common carp (Cyprinus carpio) as an alternative to hemocytometer counting. Also the effect of different sperm concentrations on post-thaw sperm parameters and fertilizing capacity were tested. The main goal of the research was to find out whether the tested methods are suitable for fast measurement of sperm concentration and if the pre-setting of sperm concentration before cryopreservation offers any advantages over dilution of sperm to a given ratio. A linear relationship of sperm concentration measured with CASA (N = 12) as well as sperm absorbance assessed with a microplate reader with spectrophotometry module (N = 18) with sperm concentration counted in a Bürker-Türk type hemocytometer was established. For cryopreservation, 4 sperm concentrations were used (0.5; 1; 2; 4 × 109 spermatozoa per ml) and compared to dilution to a ratio of 1:9. Surrogate production through germline stem cell (GSC) transplantation is becoming a very promising technology in species management and ex situ species conservation. In the present study, we have evaluated if the triploid rainbow trout is a suitable surrogate parent for the production of other salmonid gametes. In three independent trials conducted in two countries, we have transplanted brown trout and grayling GSCs into triploid rainbow trout larvae as recipients. Two months after transplantation, dissection of recipients displayed that both spermatogonial (SSCs) and oogonial (OSCs) stem cells of both species were able to colonize recipient gonads. After three years of rearing, neither male nor female recipients displayed signs of gametogenesis progression. Species-specific (for both donor and recipient species) amplification of mtDNA control region on the other hand displayed a presence of donor-derived germ cells within recipient gonads. This indicated that even though donor cells were able to colonize recipient gonads, they remained in a dormant or quasi-dormant state, and did not progress with gametogenesis. This study displays that the rainbow trout is not a suitable recipient for all salmonid species, and that careful selection of recipients is a crucial step in developing the surrogate production technology.

Fumonisins are frequent food contaminants. The high exposure to fumonisins can cause harmful effects in humans and animals. Fumonisin B1 (FB1) is the most typical member of this group; however, the occurrence of several other derivatives has been reported. Acylated metabolites of FB1 have also been described as possible food contaminants, and the very limited data available suggest their significantly higher toxicity compared to FB1. Furthermore, the physicochemical and toxicokinetic properties (e.g., albumin binding) of acyl-FB1 derivatives may show large differences compared to the parent mycotoxin. Therefore, we tested the interactions of FB1, N-palmitoyl-FB1 (N-pal-FB1), 5-O-palmitoyl-FB1 (5-O-pal-FB1), and fumonisin B4 (FB4) with human serum albumin as well as the toxic effects of these mycotoxins on zebrafish embryos were examined. Based on our results, the most important observations and conclusions are the following: (1) FB1 and FB4 bind to albumin with low affinity, while palmitoyl-FB1 derivatives form highly stable complexes with the protein. (2) N-pal-FB1 and 5-O-pal-FB1 likely occupy more high-affinity binding sites on albumin. (3) Among the mycotoxins tested, N-pal-FB1 showed the most toxic effects on zebrafish, followed by 5-O-pal-FB1, FB4, and FB1. (4) Our study provides the first in vivo toxicity data regarding N-pal-FB1, 5-O-pal-FB1, and FB4.

Abstract Zebrafish is one of the most commonly used model organisms in biomedical, developmental and genetic research. The production of several thousands of transgenic lines is leading to difficulties in maintaining valuable genetic resources as cryopreservation protocols for eggs and embryos are not yet developed. In this study, we utilized testis cryopreservation (through both slow-rate freezing and vitrification) and spermatogonia transplantation as effective methods for long-term storage and line reconstitution in zebrafish. During freezing, utilization of 1.3 M of dimethyl sulfoxide (Me2SO) displayed the highest spermatogonia viability (~60%), while sugar and protein supplementation had no effects. Needle-immersed vitrification also yielded high spermatogonia viability rates (~50%). Both optimal slow-rate freezing and vitrification protocols proved to be reproducible in six tested zebrafish lines after displaying viability rates of >50% in all lines. Both fresh and cryopreserved spermatogonia retained their ability to colonize the recipient gonads after intraperitoneal transplantation of vasa::egfp and actb:yfp spermatogonia into wild-type AB recipient larvae. Colonization rate was significantly higher in dnd-morpholino sterilized recipients than in non-sterilized recipients. Lastly, wild-type recipients produced donor-derived sperm and donor-derived offspring through natural spawning. The method demonstrated in this study can be used for long-term storage of valuable zebrafish genetic resources and for reconstitution of whole zebrafish lines which will greatly improve the current preservation practices.

In the Central European region, there is a long tradition of breeding fish in artificially constructed ponds. As the area belongs to the temperate zone, farmed fish need to survive cold winter periods. Common carp (Cyprinus carpio L.), which is an omnivorous, bioturbating species, is well adapted to warm and cold periods and the alluvial water environment. Since the Middle Ages, a large scale, efficient carp farming methodology has been developed in the region, where production is based on natural resources (protein and fatty acid sources) of renewable water ecosystems. This summary aims to present this well-developed breedi:ng method through discussing aspects of hydrobiology and energy transfer through the food chain as well. Capabilities and effects of agro-technical treatments such as liming and organic manuring, zooplankton management and possible supplementary feedings are also reviewed. Analysing chemical processes of waters uncovers that biological production has no carbon footprint; no carbon dioxide is released into the atmosphere. In contrast, gaseous carbon dioxide diffuses into pond water containing calcium and/or magnesium, then it accumulates in algae production and, through energy migration to upper trophic levels, increases carp production. Thus, it can be declared that pond-farmed carp provides an environmentally friendly, delicious meat among products of animal origin.

The impact of pharmaceuticals on non-target organisms in the environment is of increasing concern and study. Pharmaceuticals and other pollutants are often present as mixtures in an environmental compartment. Studies on the toxicological implications of these drugs on fish, particularly as mixtures at environmentally relevant concentrations, are very limited. Thus, this study aimed to evaluate the chronic effects of the anticonvulsant drug carbamazepine (CBZ) and progesterone (P4) at environmentally relevant concentrations, individually and in binary mixtures, applying a suite of biomarkers at the molecular level in zebrafish (Danio rerio). The effects on biotransformation enzymes 7-ethoxyresorufin O-deethylase (EROD) and glutathione-S-transferase (GST), antioxidant enzymes catalase (CAT), superoxide dismutase (SOD), glutathione peroxidases (GPxSe and GPxTOT), and glutathione reductase (GR), and markers of damage, such as DNA strand breaks (DNAsb), lactate dehydrogenase (LDH), lipid peroxidation (LPO), and vitellogenin-like proteins (VTG), were evaluated. Analyses of the biochemical markers indicated that a synergistic dose-ratio-dependent effect of CBZ and P4 in zebrafish occurs after chronic exposure regarding VTG, biotransformation enzymes (EROD, GST), and oxidative stress marker (DNAsb). The results suggest a synergistic effect regarding VTG, thus indicating a high risk to the reproductive success of fish if these pharmaceuticals co-occur.

Worldwide, the anticonvulsant drug carbamazepine (CBZ) is the most frequently identified pharmaceutical residue detected in rivers. Reported chronic effects of CBZ in non-target freshwater organisms, particularly fish, include oxidative stress and damage to liver tissues. Studies on CBZ effects in fish are mostly limited to zebrafish and rainbow trout studies. Furthermore, there are only a few chronic CBZ studies using near environmental concentrations. In this study, we provide data on subacute effects of CBZ exposure (28 days) to common carp (Cyprinus carpio), employing a set of biochemical markers of damage and exposure. CBZ was found to induce a significant change in the hepatic antioxidant status of fish subjected to 5 µg/L. Moreover, with increasing concentrations, enzymatic and non-enzymatic biomarkers of oxidative defence (catalase (CAT), superoxide dismutase (SOD), glutathione reductase (GR), DNA strand breaks)), toxicant biotransformation (ethoxyresorufin-o-demethylase (EROD), glutathione-S-transferase (GST)), and organ and tissue damage (lactate dehydrogenase (LDH), cetylcholinesterase (AChE)) were altered. The AChE, LDH, and lipid peroxidation (LPO) results indicate the occurrence of apoptotic process activation and tissue damage after 28 days of exposure to CBZ. These findings suggest significant adverse effects of CBZ exposure to common carp at concentrations often found in surface waters.

The objective of our study was to investigate the effects of heavy metals on the fertilizing capacity of exposed zebrafish sperm, on embryonic survival, and on occurrence of embryonic deformities following fertilization with exposed sperm. It is important to test heavy metals because they are well-known pollutants. Sperm of externally fertilizing species can get in contact with pollutants found in aquatic environment. Zebrafish sperm, despite its advantages, has seldom been used in in vitro toxicological studies and no reports are available regarding the fertilizing capacity of exposed sperm. Zebrafish sperm was stripped and exposed to concentrations of the tested heavy metals (Zn 2+ , Cd 2+ , Cr 3+ , Cu 2+ , Ni 2+ , Hg 2+ , As 3+ ) for 30 or 120 minutes. Calculated half-maximal effective concentration (EC 50 ) values do not differ significantly from those calculated for motility for any of the tested heavy metals, which means fertilization rate can indicate the toxicity of the given substance following exposure of sperm. Thus, its application as in vitro toxicological end point is reasonable. The survival of embryos and embryonic development have not been affected by the exposure of spermatozoa, which means all alterations in spermatozoa caused by heavy metals have been expressed before 24 hours post fertilization.

We compared the effectiveness of the ovarian sperm injection method with that of the traditional one over the artificial propagation of jundia. Three experimental groups were set up containing seven females each. Females in Group 1 received common carp pituitary extract (CPE) homogenized in saline to induce ovulation and the stripped eggs were fertilized with the traditional dry method. In Group 2 ovulation was induced as in Group 1, however, sperm for fertilization was injected to the ovarian cavity of the females. In Group 3, CPE was first homogenized in sperm which was then injected to the ovarian cavity of females. All females received the same dose of 5.0 mg kg−1 bodyweight. Females in Group 2 and 3 were inseminated by a catheter. To fertilize the eggs, sperm samples were pooled from six males and they were used for all groups. There were no significant differences among the three Groups (P < 0.05) in the investigated parameters, such as ovulation ratio, pseudo-gonadosomatic index and fertilization rate. Tench is a cyprinid fish that has undergone human-induced translocations. The natural populations of the species are on the decline due to habitat loss and spawning grounds degradation. The genetic diversity of seven natural populations was investigated to establish the genetic knowledge base for successful conservation efforts and for selective breeding. Twelve microsatellite markers, the sequencing of a 615 bp section of mtDNA (Cytb) and PCR-RFLP analysis of two nuclear markers (Act) and (RpS7) were used to analyze the genetic variation and structure among 175 individuals. All microsatellite loci were found to have moderate levels of polymorphism. The pairwise Fst values between population pairings were moderate; the populations were aligned to four clusters. The Cytb gene showed 20 haplotypes; 67.1% of individuals were categorized as Eastern, while 32.9% to the Western haplogroup. Analysis of the Act and RpS7 genes showed that the level of hybridization among the two haplogroups is high within the sampled populations. Hungarian Tench populations are genetically less diverse compared to natural populations in Western-Europe, but they still represent valuable genetic resources and Lake Fertő, Lake Kolon and Csörnöc-Herpenyő populations can be optimal candidates for future selective breeding programs.

Aflatoxin B1 (AFB1) is a potent mycotoxin and natural carcinogen. The primary producers of AFB1 are Aspergillus flavus and A. parasiticus. Sterigmatocystin (STC), another mycotoxin, shares its biosynthetic pathway with aflatoxins. While there are abundant data on the biological effects of AFB1, STC is not well characterised. According to published data, AFB1 is more harmful to biological systems than STC. It has been suggested that STC is about one-tenth as potent a mutagen as AFB1 as measured by the Ames test. In this research, the biological effects of S9 rat liver homogenate-activated and non-activated STC and AFB1 were compared using two different biomonitoring systems, SOS-Chromotest and a recently developed microinjection zebrafish embryo method. When comparing the treatments, activated STC caused the highest mortality and number of DNA strand breaks across all injected volumes. Based on the E. coli SOS-Chromotest, the two toxins exerted the same genotoxicities. Moreover, according to the newly developed zebrafish microinjection method, STC appeared more toxic than AFB1. The scarce information correlating AFB1 and STC toxicity suggests that AFB1 is a more potent genotoxin than STC. Our findings contradict this assumption and illustrate the need for more complex biomonitoring systems for mycotoxin risk assessment.

The effects of a simple saline solution prepared using two different pH (4.4 and 8.5) on sperm motility in burbot were investigated. Results were recorded during a 96-hour chilled storage (4°C) in 24-hour intervals. Measurements were focused on the detailed characteristics of motility using 12 parameters obtained from the Computer-assisted Sperm Analysis (CASA). Significantly higher progressive motility (pMOT), distance average path (DAP), distance curved line, distance straight line (DSL), average path velocity (VAP), curvilinear velocity, straight line velocity, and beat cross frequency (BCF) were observed with the activating solution buffered at pH 8.5 in comparison with pH 4.4. Already after 24 h a significant reduction was measured in pMOT (0 h: 49 ± 24%, 24 h: 12 ± 7%). Similar decreasing tendency was recorded only after 72 h in DAP (0 h: 26 ± 4 µm/s, 72 h: 19 ± 9 µm/s), DSL (0 h: 21 ± 5 µm/s, 72 h: 17 ± 8 µm/s), VAP (0 h: 59 ± 9 µm/s, 72 h: 43 ± 21 µm/s), and BCF (0 h: 28 ± 2 Hz, 72 h: 18 ± 10 Hz). The response of different investigated CASA parameters to different treatments varied in our experiments. According to our studies, numerous burbot sperm motility parameters are sensitive to chilled storage and to low pH of the activating solution. Our results could support the effective sperm quality assessment and successful artificial propagation process in burbot.

Scardinius racovitzai appeared to be locally adapted to an extreme high temperature environment (26–35 °C). As a species of conservation concern, due to declining ecological condition of the solitary thermal lake it could be found in (Lake Petea, Oradea, Romania) and eventual near complete abstraction of this lake, 60 adults were saved in 2014. In this study we investigated methods for induced spawning and sperm cryopreservation to aid recovery and reintroduction to restored habitats. Induced spawning was introduced successfully by intraperitoneal induction of dried carp pituitary extract and human chorion gonadotropin. Larvae and juveniles were successfully reared to age 3 months using standard methods for cyprinids. Our experiments on the cryopreservation of S. racovitzai sperm show that the extender consisting 40 mM KCl 200 mM glucose, 30 mM Tris buffered with HCL pH 8.0 is suitable for freezing warm water rudd sperm together with methanol as a cryoprotectant. By rescuing the remaining wild stock into captivity and undertaking propagation research we have prevented extinction of this rare species – more so S. racovitzai will be important in the study of freshwater fishes and how they can adapt to significant warming. Histopathological alterations in various fish organs have a pronounced value in aquatic toxicology and are widely used in environmental monitoring. The aim of this study was to evaluate whether histopathological alterations in fish gills can discriminate seasonal variations in environmental conditions within the same aquatic ecosystem, and if so, which alterations contributed the most to seasonal differentiation. Microscopic examination of common bream Abramis brama gills displayed various alterations in gill structure, including epithelial hypertrophy, hyperplasia, mucous and chloride cell alterations, epithelial lifting, necrosis, hyperemia and aneurism. These alterations were subsequently quantified by a semi-quantitative analysis in order to detect differences in the intensity of the mentioned alterations. Epithelial hypertrophy, hyperplasia, epithelial lifting and necrosis varied significantly between seasons with only necrosis being significantly higher in the first season. Discriminant canonical analysis displayed that epithelial hyperplasia, mucous cell alterations, epithelial lifting and necrosis contributed the most to discrimination between seasons. Overall, this study demonstrates that histopathological biomarkers in fish gills can be used in discriminating seasonal variations in water quality within the same aquatic ecosystem.

Chinese carps, namely silver carp (Hypophthalmichthys molitrix), bighead carp (Aristichthys nobilis) and grass carp (Ctenopharyngodon idella) have the largest annual production in freshwater aquaculture globally. Induced breeding of these species enlarged the geographical range for their culture as well as permitted more reliable seed supply. For the assessment of efficiency, long-term data from 18 spawning seasons on induced breeding of Chinese carps at a large-scale hatchery were evaluated. Assessment of ovulation was carried out by determining the ovulation ratio (number of ovulated females / number of injected females) × 100. The relative amount of stripped eggs (pseudo-gonadosomatic index, PGSI) was calculated as follows: (weight of stripped egg mass / body weight before stripping) × 100. We also analyzed the ovulation ratio and the PGSI relative to three different weight classes of females (small, medium and large).Over the 18 spawning seasons, 555 silver carp, 300 bighead carp and 1175 grass carp females were selected for propagation. Almost 80% of silver carp females selected belonged to categories between 4 and 7 kg. For grass carp and especially for bighead carp, breeders preferred higher weight categories.Ovulation ratios were similar between silver carp (80.9%), bighead carp (77.3%) and grass carp (79.1%). Mean PGSI for grass carp (9.3 ± 4.26%) was significantly lower than that for silver carp (10.5 ± 4.85%) and bighead carp (10.1 ± 3.96). In silver carp and bighead carp, there were no significant differences in ovulation ratios and mean PGSI values among different weight classes. Mean PGSI values were also similar among different weight classes of grass carp females. Ovulation ratios, however, were different among the three different weight classes in grass carp being the lowest in the “small” weight class and the highest in the “large” weight class. These results will provide guidelines for Chinese carp breeders to select optimal brood fish for ovulation induction and calculate their hatchery's larval production in advance. Bigheaded carps (bighead carp, Hypophthalmichthys molitrix, and silver carp, Hypophthalmichthys nobilis) and their hybrids play an important ecological and economic role in their original habitat, while their introduced stocks may pose serious ecological risks. To address questions about the persistence and invasiveness of these fish, we need to better understand their population structures. The genetic structures of bigheaded carp populations inhabiting Lake Balaton and the Tisza River were examined with ten microsatellite markers and a mitochondrial DNA marker (COI). The Lake Balaton stock showed higher genetic diversity compared with the Tisza River stock. Based on hierarchical clustering, the Tisza population was characterized only by only silver carps, while the Balaton stock included hybrid and silver carp individuals. All COI haplotypes originated from the Yangtze River. Based on the high genomic and mitochondrial diversity, along with the significant deviation from H–W equilibrium and the lack of evidence of bottleneck effect, it can be assumed that bigheaded carps do not reproduce in Lake Balaton. The present stock in Balaton may have originated from repeated introductions and escapes from the surrounding fishponds. The Tisza stock consists solely of silver carp individuals. This stock appears to have significant reproductive potential and may become invasive if environmental factors change due to climate change.

Lake Balaton is the largest shallow lake in Central Europe. Its water quality is affected by its biggest inflow, the Zala River. During late 20th century, a wetland area named the Kis-Balaton Water Protection System (KBWPS) was constructed in the hopes that it would act as a filter zone and thus ameliorate the water quality of Lake Balaton. The aim of the present study was to test whether the KBWPS effectively safeguards Lake Balaton against toxic cyanobacterial blooms. During April, May, July and September 2018, severe cyanobacterial blooming was observed in the KBWPS with numbers reaching up to 13 million cells/mL at the peak of the bloom (July 2018). MC- and STX-coding genes were detected in the cyanobacterial biomass. Five out of nine tested microcystin congeners were detected at the peak of the bloom with the concentrations of MC-LR reaching 1.29 µg/L; however, accumulation of MCs was not detected in fish tissues. Histopathological analyses displayed severe hepatopancreas, kidney and gill alterations in fish obtained throughout the investigated period. In Lake Balaton, on the other hand, cyanobacterial numbers were much lower; more than 400-fold fewer cells/mL were detected during June 2018 and cyanotoxins were not detected in the water. Hepatic, kidney and gill tissue displayed few alterations and resembled the structure of control fish. We can conclude that the KBWPS acts as a significant buffering zone, thus protecting the water quality of Lake Balaton. However, as MC- and STX-coding genes in the cyanobacterial biomass were detected at both sites, regular monitoring of this valuable ecosystem for the presence of cyanobacteria and cyanotoxins is of paramount importance.

Pesticides may accumulate in freshwater fish due to contamination from the environment. This paper reports on a risk assessment of DDT and DDT metabolites in carp. A survey was conducted about dietary habits among fish consumers. Cluster analysis was accomplished based on the frequency and amount of carp consumption. Classical and carcinogenic risk assessments were performed for the clusters. While DDT contamination was present, it was not found to be risky concerning the complete diet of the clusters (carcinogenic risk was also negligible), moreover, carp consumption did not contribute significantly to the risk level even in the case of the extreme consumers.

T-2 mycotoxin degradation and detoxification efficiency of seven bacterial strains were investigated with zebrafish microinjection method in three steps ((1) determination of mycotoxin toxicity baseline, (2) examination of bacterial metabolites toxicity, (3) identification of degradation products toxicity). Toxicity of T-2 was used as a baseline of toxic effects, bacterial metabolites of strains as control of bacterial toxicity and degradation products of toxin as control of biodegradation were injected into one-cell stage embryos in the same experiment. The results of in vivo tests were checked and supplemented with UHPLC-MS/MS measurement of T-2 concentration of samples. Results showed that the Rhodococcus erythropolis NI1 strain was the only one of the seven tested (R. gordoniae AK38, R. ruber N361, R. coprophilus N774, R. rhodochrous NI2, R. globerulus N58, Gordonia paraffinivorans NZS14), which was appropriated to criteria all aspects (bacterial and degradation metabolites of strains caused lower toxicity effects than T-2, and strains were able to degrade T-2 mycotoxin). Bacterial and degradation metabolites of the NI1 strain caused slight lethal and sublethal effects on zebrafish embryos at 72- and 120-h postinjection. Results demonstrated that the three-step zebrafish microinjection method is well-suited to the determination and classification of different bacterial strains by their mycotoxin degradation and detoxification efficiency.

Common carp (Cyprinus carpio) is one of the most cultured fish species over the world with many different breeds and plenty of published protocols for sperm cryopreservation. However, data regarding preservation of gonadal tissue and surrogate production is still missing. A protocol for freezing common carp spermatogonia was developed through varying different factors along a set of serial subsequent experiments. Among the six cryoprotectants tested, the best survival was achieved with dimethyl sulfoxide (Me2SO). In the next experiment, a wide range of cooling rates (0.5-10°C/min) and different concentrations of Me2SO were tested resulting in the highest survival achieved using 2 M Me2SO and cooling rate of -1°C/min. When testing different tissue sizes and incubation times in the cryomedia, the highest viability was observed when incubating 100 mg tissue fragments for 30 min. Finally, sugar supplementation did not yield significant differences. When testing different equilibration (ES) and vitrification solutions (VS) used for needle-immersed vitrification, no significant differences were observed between the tested groups. Additionally, varied exposure time to VS did not improve the vitrification outcome where the viability was 4-fold lower than that of freezing. The functionality of cryopreserved cells was tested by interspecific transplantation into sterilized goldfish recipients. The exogenous origin of the germ cells in gonads of goldfish recipient was confirmed by molecular markers and incorporation rate was over 40% at 3 months post-transplantation. Results of this study can serve for long-term preservation of germplasm in carp which can be recovered in a surrogate recipient.

Background Analyses of the control region sequences of European brown trout populations’ mitrochondrial DNA have revealed five main evolutionary lineages (Atlantic, Danubian, Mediterranean, Adriatic, Marble) mostly relating to the main water basins; however, the hybridization between lineages were increasingly reported. Due to the hydrogeography of Hungary, wild populations should theoretically belong to the Danubian lineage, however, this has not been verified by genetic studies. Methods In our study multiple molecular marker sets (mitochondrial sequence, microsatellites, PCR-RFLP of nuclear markers and sex marker) were used to investigate the genetic composition and population genetics of the brown trout populations in two broodstocks, six wild streams in Hungary and one Serbian population. Results The admixture of Atlantic and Danubian lineages in these populations, except the Serbian population with pure Danubian origin, was observed by control region sequences of mitochondrial DNA and PCR-RFLP markers in the nuclear genome, and one unpublished Danubian haplotype was found in Hungarian populations. A sex-specific marker revealed equal gender ratio in broodstocks and Kemence stream, whereas in other wild streams the proportion of female individuals were less than 50%. Structure and principal component analyses based on the alleles of microsatellite loci also revealed overlapping populations, however the populations were still significantly different from each other and were mostly in Hardy-Weinberg equilibrium. Discussion Stocking and migration can have a significant genetic impact on trout populations of wild streams, however there are no guidelines or common practices for stocking of small streams in Hungary, thus the genetic background of these populations should be considered when developing conservation actions.

Since three bullhead catfish species were introduced to Europe in the late 19th century, they have spread to most European countries. In Hungary, the brown bullhead (Ameiurus nebulosus) was more widespread in the 1970s–1980s, but the black bullhead (Ameiurus melas) has gradually supplanted since their second introduction in 1980. The introgressive hybridization of the two species has been presumed based on morphological examinations, but it has not previously been supported by genetic evidence. In this study, 11 different Hungarian habitats were screened with a new species-specific nuclear genetic, duplex PCR based, marker system to distinguish the introduced catfish species, Ameiurus nebulosus, Ameiurus melas, and Ameiurus natalis, as well as the hybrids of the first two. More than 460 specimens were analyzed using the above markers and additional mitochondrial sequence analyses were also conducted on >25% of the individuals from each habitat sampled. The results showed that only 7.9% of the specimens from two habitats belonged to Ameiurus nebulosus, and 92.1% were classified as Ameiurus melas of all habitats, whereas the presence of Ameiurus natalis was not detected. Two specimens (>0.4%) showed the presence of both nuclear genomes and they were identified as hybrids of Ameiurus melas and Ameiurus nebulosus. An additional two individuals showed contradicting results from the nuclear and mitochondrial assays as a sign of a possible footprint of introgressive hybridization that might have happened two or more generations before. Surprisingly, the level of hybridization was much smaller than expected based on the analyses of the North American continent’s indigenous stock from the hybrid zones. This phenomenon has been observed in several invasive fish species and it is regarded as an added level of complexity in the management of their rapid adaptation.

BACKGROUND:Cryopreservation of zebrafish embryos is still an unsolved problem despite market demand and massive efforts to preserve genetic variation among numerous existing lines. Chilled storage of embryos might be a step towards developing successful cryopreservation, but no methods to date have worked. METHODS:In the present study, we applied a novel strategy to improve the chilling tolerance of zebrafish embryos by introducing a preconditioning hydrostatic pressure treatment to the embryos. In our experiments, 26-somites and Prim-5 stage zebrafish embryos were chilled at 0°C for 24 hours after preconditioning. Embryo survival rate, ability to reach maturation and fertilizing capacity were tested. RESULTS:Our results indicate that applied preconditioning technology made it possible for the chilled embryos to develop normally until maturity, and to produce healthy offspring as normal, thus passing on their genetic material successfully. Treated embryos had a significantly higher survival and better developmental rate, moreover the treated group had a higher ratio of normal morphology during continued development. While all controls from chilled embryos died by 30 day-post-fertilization, the treated group reached maturity (~90-120 days) and were able to reproduce, resulting in offspring in expected quantity and quality. CONCLUSIONS:Based on our results, we conclude that the preconditioning technology represents a significant improvement in zebrafish embryo chilling tolerance, thus enabling a long-time survival. Furthermore, as embryonic development is arrested during chilled storage this technology also provides a solution to synchronize or delay the development.
