The aims of this study were to develop topical liposomal hydrogels based on thermal waters (TWs) acquired in the region of Biskra (Northeast Algeria) and also to investigate their rheological properties. Liposomes containing two highly mineralized thermal waters, Baraka (BTW) and Salhine (STW), were prepared by probe sonication using phosphatidylcholine (PC) and cholesterol (Chol), plain or mixed with phosphatidylglycerol (PG). Based on their lipid composition, obtained liposomes presented vesicle sizes of 60 nm, a low polydispersity index, and various negative zeta potentials. Impairment of oligodendrocyte progenitor cell (OPC) differentiation into oligodendrocytes and chronic inflammation are key determinants of poor remyelination observed in diseases such as multiple sclerosis. For many pro-myelinating molecules, the therapeutic potential is hindered by poor solubility or limited access to the targeted cells. A promising approach to improve the delivery of those molecules to OPC is to encapsulate them in functionalized Lipid Nanocapsules (LNC). We aimed to develop the first OPC-targeting LNC, by grafting an anti-PDGFRα antibody on the surface of the LNC using several strategies and evaluating the interaction with PDGFRα via ELISA. We found that only site-selective click-chemistry grafting maintained anti-PDGFRα/PDGFRα association, which was confirmed in vitro on primary rat OPC. In conclusion, we demonstrated that it was possible to produce anti-PDGFRα functionalized LNC, we confirmed the antibody's ability to recognize its receptor after grafting and we optimized techniques to characterize antibody functionalized LNC.

Photolytic reactions allow the optical control of the liberation of biological effectors by photolabile protecting groups. The development of versatile technologies enabling the use of deep-red or NIR light excitation still represents a challenging issue, in particular for light-induced drug release (e.g., light-induced prodrug activation). Here, light-sensitive biocompatible lipid nanocapsules able to liberate an antitumoral drug through photolysis are presented. It is demonstrated that original photon upconverting nanoparticles (LNC-UCs) chemically conjugated to a coumarin-based photocleavable linker can quantitatively and efficiently release a drug by upconversion luminescence-assisted photolysis using a deep-red excitation wavelength. In addition, it is also able to demonstrate that such nanoparticles are stable in the dark, without any drug leakage in the absence of light. These findings open new avenues to specifically liberate diverse drugs using deep-red or NIR excitations for future therapeutic applications in nanomedicine.

The main objectives of this work were to formulate liposomes encapsulating highly mineralized thermal waters (TWs) and to study anti-inflammatory effect of free and encapsulated thermal waters on RAW 264.7 macrophage cells stimulated with lipopolysaccharide (LPS). TWs-loaded conventional and deformable liposomes (TWs-Lip and TWs-DLip) were prepared by sonication and extrusion, respectively. They were considered for their vesicle size, zeta potential, entrapment efficiency, physical stability and in vitro anti-inflammatory effect. Formulated liposome suspensions have a low polydispersity and nanometric size range with zeta potential values close to zero. The vesicle size was stable for 30 days. Entrapment efficiency of TWs was above 90% in conventional liposomes and 70% in deformable liposomes. Pretreatment of LPS-stimulated murine macrophages, with free and liposome-encapsulated TWs, resulted in a significant reduction in nitric oxide (NO) production and modulated tumor necrosis factor-α (TNF-α) production suggesting an anti-inflammatory effect which was even more striking with TWs-Lip and TWs-DLip. Liposome formulations may offer a suitable approach for transdermal delivery of TWs, indicated in inflammatory skin diseases.

Liposomes are powerful tools for the optimization of peptides and adjuvant composition in cancer vaccines. Here, we take advantage of a liposomal platform versatility to develop three vaccine candidates associating a peptide from HA influenza virus protein as CD4 epitope, a peptide from HPV16 E7 oncoprotein as CD8 epitope and TLR4, TLR2/6 or NOD1 agonists as adjuvant. Liposomal vaccine containing MPLA (TLR4 liposomes), are the most effective treatment against the HPV-transformed orthotopic lung tumor mouse model, TC-1. This vaccine induces a potent Th1-oriented antitumor immunity, which leads to a significant reduction in tumor growth and a prolonged survival of mice, even when injected after tumor appearance. This efficacy is dependent on CD8+ T cells. Subcutaneous injection of this treatment induces the migration of skin DCs to draining lymph nodes. Interestingly, TLR2/6 liposomes trigger a weaker Th1-immune response which is not sufficient for the induction of a prolonged antitumor activity. Although NOD1 liposome treatment results in the control of early tumor growth, it does not extend mice survival. Surprisingly, the antitumor activity of NOD1 vaccine is not associated with a specific adaptive immune response. This study shows that our modulable platform can be used for the strategical development of vaccines.

Our understanding of the involvement of the immune system in cancer control has increased over recent years. However, the development of cancer vaccines intended to reverse tumor-induced immune tolerance remains slow as most current vaccine candidates exhibit limited clinical efficacy. The skin is particularly rich with multiple subsets of dendritic cells (DCs) that are involved to varying degrees in the induction of robust immune responses. Transcutaneous administration of cancer vaccines may therefore harness the immune potential of these DCs, however, this approach is hampered by the impermeability of the stratum corneum. Innovative vaccine formulations including various nanoparticles, such as liposomes, are therefore needed to properly deliver cancer vaccine components to skin DCs. Areas covered: The recent insights into skin DC subsets and their functional specialization, the potential of nanoparticle-based vaccines in transcutaneous cancer vaccination and, finally, the most relevant clinical trial advances in liposomal and in cutaneous cancer vaccines will be discussed. Expert commentary: To define the optimal conditions for mounting protective skin DC-induced anti-tumor immune responses, investigation of the cellular and molecular interplay that controls tumor progression should be pursued in parallel with clinical development. Therapeutic cancer vaccines need thoughtful design to efficiently deliver appropriate antigens and adjuvants to the immune system. In the current study, we took advantage of the versatility of a liposomal platform to conceive and customize vaccines containing three elements needed for the induction of efficient antitumor immunity: i) a CD4 epitope peptide able to activate CD4+ T helper cells, ii) a CD8 tumor-specific epitope peptide recognized by CD8+ T cytotoxic cells and iii) Pattern Recognition Receptor (PRR) agonists which stand as adjuvants. Each type of component, conjugated to liposomes, was evaluated individually by comparing their vaccine efficacy after immunization of naive mice. These screening steps resulted in the optimization of three liposomal constructs bearing a peptide from HA influenza virus protein as CD4 epitope, a peptide from HPV16 E7 oncoprotein as CD8 epitope and TLR4, TLR2/6 or NOD1 agonists as adjuvant, which displayed antitumor efficiency against a mouse model of disseminated tumors transformed by HPV16. Our results validated the interest of our customizable liposomal platform as delivery system for cancer vaccination. We also demonstrated its interest as tool for vaccine design allowing the strategical selection of components, and the evaluation of epitope-adjuvant association.

International audience; Multilayered coated liposomes were prepared using the layer-by-layer (LbL) technique in an effort to improve their stability in biological media. The formulation strategy was based on the alternate deposition of two biocompatible and biodegradable polyelectrolytes – poly(L-lysine) (PLL) and poly(L-glutamic acid) (PGA) – on negatively charged small unilamellar vesicles (SUVs). Some parameters of the formulation process were optimized such as the polyelectrolyte concentration and the purification procedure. This optimized procedure has allowed the development of very homogeneous formulations of liposomes coated with up to 6 layers of polymers (so-called layersomes). The coating was characterized by dynamic light scattering (DLS), zeta potential measurements and Förster resonance energy transfer (FRET) between two fluorescently labeled polyelectrolytes. Studies on the stability of the formulations at 4 °C in a buffered solution have shown that most structures are stable over 1 month without impacting their encapsulation capacity. In addition, fluorophore release experiments have demonstrated a better resistance of the layersomes in the presence of a non-ionic detergent (Triton™ X-100) as well as in the presence of phospholipase A2 and human plasma. In conclusion, new multilayered liposomes have been developed to increase the stability of conventional liposomes in biological environments.

Ultracentrifugated compact polyelectrolyte complexes (uCoPECs) represent a new class of materials that are obtained by ultracentrifugation of solutions of polyanion/polycation complexes in the presence of salt. In the present study, two polysaccharides, chitosan and alginate, were used to form such complexes, thus providing a solid material uniquely composed of polysaccharides. The conditions for obtaining the uCoPEC material were optimized: the optimal salt concentration and polysaccharide concentrations were assessed, and the ultracentrifugation speed proved to be a key parameter to obtain compact and homogeneous materials. The Young's modulus, E, of the material was of the order of 12 MPa, which is the highest E value measured for a uCoPEC. The material contained nanometer-sized crystals of chitosan as indicated by X-ray diffraction. Most strikingly, this material proves to be totally cell- and bacteria-resistant. International audience; The potent antitumor effect of α-galactosylceramide (α-GalCer) is based on its recognition by invariant Natural Killer T cells (iNKT) after its capture and presentation by antigen presenting cells including dendritic cells (DCs). Synthetic α-GalCer has already been tested in advanced cancer patients but no or only moderate clinical responses were obtained.To optimize α-GalCer efficacy, we have postulated that alternative formulations impacting its molecular organization in aqueous medium could modify DC uptake and iNKT-based immune responses. To this end, we have developed two strategies: (1) the formulation of α-GalCer in non-cationic liposomes and (2) the synthesis of a water-soluble α-GalCer analogue by anchoring a polyethyleneglycol moiety on its sugar head. The biological activities of these new preparations were compared to that induced by the classically used Polysorbate 20 α-GalCer micelles. Both formulations retained their uptake by DCs and activated iNKT cells both in vitro and in vivo. Despite a lower cytokine production, the formulations induced a potent immune response able to control lung murine carcinoma. In conclusion, it is possible to increase α-GalCer solubility in aqueous solution without limiting its antitumor properties.

Mucosal surfaces are promising routes for vaccination. Among mucosa, airway mucosa provides the opportunity to develop non-invasive approaches for vaccine delivery. In the current study, nasal route was used to investigate the potency of highly versatile di-epitopic liposomal constructs of different size, structure and composition to exhibit antitumor efficiency after prophylactic vaccination in mice. Well-characterized small unilamellar (SUV), multilamellar (MLV), reverse-phase evaporation (REV) and ultraflexible small unilamellar vesicles (Uf-SUV), containing the ErbB2 T-cytotoxic epitope, the influenza-derived HA T-helper epitope and the lipopeptide adjuvant Pam2CAG, were formulated. These vaccines were administered into the nasal cavity of BALB/c mice, followed by i.v. or s.c. implantation of ErbB2-surexpressing cancer cells. Nasal vaccination with the SUV vaccine resulted in an efficient antitumor activity against lung tumors and a non-significant protection against s.c. tumors. Size, structure and flexibility of liposomes did not impact vaccine immunity and antitumoral efficiency against lung tumors, in contrast to total dose of vaccine or dose of adjuvant. These results showed an undeniable interest of liposomes as lipid-based carriers for antitumor vaccine delivery by the nasal route, opening new perspectives for cancer treatment.

Influenza A virus infection can predispose to potentially devastating secondary bacterial infections. Invariant natural killer T (iNKT) cells are unconventional, lipid-reactive T lymphocytes that exert potent immunostimulatory functions. Using a mouse model of postinfluenza invasive secondary pneumococcal infection, we sought to establish whether α-galactosylceramide (α-GalCer [a potent iNKT cell agonist that is currently in clinical development]) could limit bacterial superinfection. Our results highlighted the presence of a critical time window during which α-GalCer treatment can trigger iNKT cell activation and influence resistance to postinfluenza secondary pneumococcal infection. Intranasal treatment with α-GalCer during the acute phase (on day 7) of influenza virus H3N2 and H1N1 infection failed to activate (gamma interferon [IFN-γ] and interleukin-17A [IL-17A]) iNKT cells; this effect was associated with a strongly reduced number of conventional CD103+ dendritic cells in the respiratory tract. In contrast, α-GalCer treatment during the early phase (on day 4) or during the resolution phase (day 14) of influenza was associated with lower pneumococcal outgrowth and dissemination. Less intense viral-bacterial pneumonia and a lower morbidity rate were observed in superinfected mice treated with both α-GalCer (day 14) and the corticosteroid dexamethasone. Our results open the way to alternative (nonantiviral/nonantibiotic) iNKT-cell-based approaches for limiting postinfluenza secondary bacterial infections., IMPORTANCE Despite the application of vaccination programs and antiviral drugs, influenza A virus (IAV) infection is responsible for widespread morbidity and mortality (500,000 deaths/year). Influenza infections can also result in sporadic pandemics that can be devastating: the 1918 pandemic led to the death of 50 million people. Severe bacterial infections are commonly associated with influenza and are significant contributors to the excess morbidity and mortality of influenza. Today’s treatments of secondary bacterial (pneumococcal) infections are still not effective enough, and antibiotic resistance is a major issue. Hence, there is an urgent need for novel therapies. In the present study, we set out to evaluate the efficacy of α-galactosylceramide (α-GalCer)—a potent agonist of invariant NKT cells that is currently in clinical development—in a mouse model of postinfluenza, highly invasive pneumococcal pneumonia. Our data indicate that treatment with α-GalCer reduces susceptibility to superinfections and, when combined with the corticosteroid dexamethasone, reduces viral-bacterial pneumonia.

International audience; Objectives: We assessed the analytical performance of the TSH and FT4 assays on ADVIA Centaur in a multicenter national evaluation. Design and methods: A precision study and a method comparison were performed. Reference values stated by the manufacturer were checked from 379 normal subjects. Results: For TSH and FT4, the intra-assay CVs were below 2.3 and 5.2%, respectively, and the inter-assay CVs below 4.4% and 7.2%, respectively. Therefore, the precision and reproducibility were acceptable. Bland-Altman bias plots revealed good correlation and agreement with Cobas assays. TSH and FT4 data yielded reference ranges of 0.64-3.24 mIU/L and 10.5-18.9 pmol/L, respectively. Conclusion: These assays demonstrate reliable characteristics. The reference ranges obtained can be used for interpretation of thyroid function.

With the discovery of tumor-associated antigens such as ErbB2, vaccination is considered as a promising strategy to prevent the development of cancer or treat the existing disease. Among routes of immunization, the respiratory route provides the opportunity to develop non-invasive approach for vaccine delivery. In the current study, this administration route was used in order to investigate the potency of a highly versatile di-epitopic liposomal construct to exhibit local or distant antitumoral efficiency after prophylactic or therapeutic vaccination in mice. Well-characterized liposomes, containing the ErbB2 (p63-71) TCD8(+) and HA (p307-319) TCD4(+) peptide epitopes and the Pam2CAG adjuvant, were formulated and administered into the airway of naïve BALB/c mice. The nanoparticle vaccine candidate induced local and specific systemic immune response, as measured by immune cell infiltration and chemokine and cytokine production in BALF or lung tissue, and by spleen T-cell activation ex vivo, respectively. This potent immune response resulted in an efficient antitumor activity against both lung and solid s.c. tumors. Interestingly, the antitumor efficacy was observed after both prophylactic and therapeutic vaccinations, which are the most judicious ones to fight cancer. Our data showed an undeniable interest of liposomal peptide-based vaccines in antitumor vaccination by the respiratory route, opening new perspectives for cancer treatment.

Item does not contain fulltext Synthetic and molecularly defined constructs containing the minimal components to mimic and amplify the physiological immune response are able to induce an efficient cytotoxic response. In the current study this approach was applied to the development of highly versatile liposomal constructs to co-deliver peptide epitopes in combination with TLR agonists in order to induce a specific anti-tumor cellular immune response against ErbB2 protein-expressing tumor cells. Liposomes containing ErbB2 p63-71 cytotoxic T lymphocyte (CTL) and HA307-319 T- helper (Th) peptide epitopes associated to innovative synthetic TLR2/1 (Pam(3)CAG) or TLR2/6 agonists (Pam(2)CAG and Pam(2)CGD), were injected in mice bearing ErbB2 protein-expressing tumor cells. Mannosylated ligands were also incorporated into the constructs to target antigen-presenting cells. We showed that the TLR2/6 agonists were more efficient than the TLR2/1 agonists for the eradication of tumors expressing ErbB2 protein. Furthermore, mannose-targeted liposomes displayed higher therapeutic efficiency against tumor allowing treatment with decreased quantities of both TLR ligands and peptide epitopes. Our results validated that antigen-associated mannosylated liposomes combined with efficient TLR ligands are effective vectors for vaccination against tumor. In this study we developed useful tools to evaluate the vaccination efficiency of various adjuvants and/or targeting molecules and their potential synergy.

Targetting an encapsulated drug towards a diseased organ, with the possibility to control its release at the right place and at the right time, is one among numerous questions asked to the new nanotechnologies. We present a partial answer with the example of recently elaborated lipidic nanocapsules. Liquid crystals play a role in the preparation techniques, and possibly in the delivery mechanisms of the active principle in situ. We also know that liquid crystalline behaviours are involved at the membrane level and are essential in the cell machinery. Drug nanocarriers could find their way, as do viruses, in the liquid crystalline context of cell organization.

Click-based reactions were conducted at the surface of small unilamellar vesicles (SUVs) to provide onto-vesicle chemistry with efficient innovative ready-for-use tools. For that purpose, four amphiphilic molecules were designed to insert into bilayers while presenting a reactive functional head. In this manner, a dioleylglycero-ethoxy-ethoxy-ethoxy-ethanamine (DOG-PEG4-NH2) was chosen as a common platform while the reactive amine head was converted into several electrophilic functions. Thus, two dioleylglycerol-based cyclooctyne anchors were prepared: cyclooct-1-yn-3-glycolic acid-based anchor (DOG-COA) and 1-fluorocyclooct-2-ynecarboxylic acid-based anchor (DOG-FCOA). The last one differed from the first one in that a fluorine atom reinforces the electrophilic properties of the unsaturated bond. In addition, a third dioleylglycerol-based triphenylphosphine (DOG-PPh3) was synthesized for the first time. These three innovative amphiphilic anchors were designed to react with any azide-based biomolecule following copper-free Huisgen 1,4-cycloaddition and Staudinger ligation, respectively. A fourth anchor bearing a 3,4-dibromomaleimide ring (DOG-DBM) was also unprecedentedly synthesized, to be further substituted by two thiols. Model reactions conducted in solution with either model biotinyl azide or model biotinyl disulfide gave good to total conversions and excellent isolated yields. The four new anchors were inserted into SUVs whose formula is classically used in in vivo biology. Stability and surface overall electrostatic charge were in the expected range and constant over the study. Then, the functionalized liposomes were ligated to biotin-based reagents and the experimental conditions were finely tuned to optimize the conversion. The biotinyl liposomes were demonstrated functional and totally accessible in an affinity test based on biotin scaffold quantification. Finally, DOG-FCOA’s reactivity was confronted to that of DOG-DBM in a ‘one-pot’ orthogonal reaction. (Biotin-S)2 and TAMRA-N3 (tetramethylcarboxyrhodamine azide) were successively conjugated to the liposome suspension in a successful manner. These data implement and reinforce the interest of bioorthogonal click-like reactions onto lipid nanoparticles.

Recent advances in nanoparticle systems for improved drug delivery display a great potential for the administration of active molecules. Generally, the lipid systems presented the advantage of their low toxicity due to their composition of physiological lipids compared to polymeric particles. The physico-chemical stability of the lipid carriers showed variations due to their numerous compositions and structures. This review consequently focuses on the physico-chemical stability of dispersions in the nanometer range where the lipids are the main or the only components. It highlights on the destabilization mechanisms, the techniques used to detect this destabilization and the inductors of the destabilization. Finally, the methods used to optimize the stability of lipid nanoparticle systems are described in the last part.

In order to obtain information on the structure and the stability of lipid nanoparticles, interfacial films, made from their spreading, were studied at the air/water interface. The related results were compared with the spreading of their individual components at the same interface. The interfacial behaviour of the particles indicated a nanocapsule structure with an oily core surrounded by a surfactant layer. Unlike liposomes or high density lipoproteins (HDL), no destruction leading to the release of their core was observed during their spreading and compression. Moreover, the sampling of the nanoparticle films at the air/water interface deposited on mica plate depicted by atomic force microscopy (AFM) showed entire nanocapsules. This stability is attributed to their surface cohesion related to their surfactant shell. Indeed, a partial particle surface “erosion” occurred leading to the spread of a certain amount of less attached hydrophilic surfactant molecules without releasing the nanocapsule lipid content. This study illustrates that the Langmuir balance can be used as a tool for studying the general organisation of lipid particles. Moreover, this method may allow the determination of the release profile of some encapsulated drugs.

This study proposed a method to understand the surfactant role in the first step of the formulation of a novel generation of lipidic nanocapsules. A dynamic rheological protocol was applied using a pendant drop tensiometer in order to determine the interfacial properties of the initial mixture implied in the first formulation step. The response, in terms of interfacial elasticities, described how this mixture led to monodisperse nanometer size range structures after a physico-chemical constraint.

Background: Over the last few decades, new synthetic insulin analogues have been developed. Their measurement is of prime importance in the investigation of hypoglycaemia, but their quantification is hampered by variable cross-reactivity with many insulin assays. For clinical analysis, it has now become essential to know the potential cross-reactivity of analogues of interest. Methods: In this work, we performed an extensive study of insulin analogue cross-reactivity using numerous human insulin immunoassays. We investigated the cross-reactivity of five analogues (lispro, aspart, glulisine, glargine, detemir) and two glargine metabolites (M1 and M2) with 16 commercial human insulin immunoassays as a function of concentration. Results: The cross-reactivity values for insulin analogues or glargine metabolites ranged from 0% to 264%. Four assays were more specific to human insulin, resulting in negligible cross-reactivity with the analogues. However, none of the 16 assays was completely free of cross-reactivity with analogues or metabolites. The results show that analogue cross-reactivity, which varies to a large degree, is far from negligible, and should not be overlooked in clinical investigations. Conclusions: This study has established the cross-reactivity of five insulin analogues and two glargine metabolites using 16 immunoassays to facilitate the choice of the immunoassay(s) and to provide sensitive and specific analyses in clinical routine or investigation.

International audience; Invariant Natural Killer T (iNKT) cells have potent immunostimulatory activities that could be exploited for human therapies. The high-affinity CD1d antigen α-galactosylceramide analogue KRN7000 (KRN) activates a cascade of anti-tumor effector cells and clinical studies have already had some initial success. To improve the efficacy of the treatment, strategies that aim to vectorize KRN would be valuable. In this study, we intended to characterize and compare the effect of KRN encapsulated in poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles (NPs, 90nm) and microparticles instead of macroparticles (MPs, 715nm) on the iNKT cell response. Our data show that whatever the size of the particles, vectorized KRN induced potent primary activation of iNKT cells in vitro and in vivo. We show that endocytosis of PLGA-based particles by dendritic cells is mediated by a clathrin-dependent manner and that this event is important to stimulate iNKT cells. Finally, we report that KRN vectorized in NPs and MPs exhibited different behaviours in vivo in terms of iNKT cell expansion and responsiveness to a recall stimulation. Collectively, our data validate the concept that KRN encapsulated in PLGA-based particles can be used as delivery systems to activate iNKT cells in vitro and in vivo.

Among the particulate systems that have been envisaged in vaccine delivery, liposomes are very attractive. These phospholipid vesicles can indeed deliver a wide range of molecules. They have been shown to enhance considerably the immunogenicity of weak protein antigens or synthetic peptides. Also, they offer a wide range of pharmaceutical options for the design of vaccines. In the past decade, the nasal mucosa has emerged as an effective route for vaccine delivery, together with the opportunity to develop non-invasive approaches in vaccination.This review focuses on the recent strategies and outcomes that have been developed around the use of liposomes in nasal vaccination.The various formulation parameters, including lipid composition, size, charge and mucoadhesiveness, that have been investigated in the design of liposomal vaccine candidates dedicated to nasal vaccination are outlined. Also, an overview of the immunological and protective responses obtained with the developed formulations is presented.This review illustrates the high potential of liposomes as nasal vaccine delivery systems.

They all induce mouse dendritic cell maturation and B cell proliferation. We demonstrate that in spite of the chemical modification, the four compounds remain TLR2 agonists.

To design and evaluate liposomal constructs capable of inducing a potent systemic and airway humoral response to Pseudomonas aeruginosaLiposomes contained a peptide derived from P. aeruginosa pilin protein as B epitope, a peptide derived from Influenza hemagglutinin protein as Th epitope, the TLR agonist Pam3CAG or Pam2CAG as adjuvant, and a mannosylated lipid as dendritic cell targeting agent. These constructions were administered to mice intraperitoneally (i.p.) or intranasally (i.n.). Their immunogenicity was evaluated by measuring B epitope-specific immunoglobulins in the serum and the airways by ELISA.The B epitope, in its native form or after substitution of a cysteine by a serine, induced high systemic IgG titers when formulated in the presence of Pam3CAG or Pam2CAG and administered i.p.. No IgA response was observed in the airways upon injection of candidate vaccines by i.p. route, whatever the B epitope or the adjuvant. However, i.n. vaccination resulted in a significant local production of IgA. Finally, the production of IgG was more rapid when mannose was incorporated.All liposomal candidate vaccines tested induced the production of IgG and/or IgA directed against an immunogenic peptide from P. aeruginosa. Liposomal constructs could be attractive in the vaccination against P. aeruginosa.

An important challenge for the development of new generations of vaccines is the efficient delivery of antigens to antigen presenting cells such as dendritic cells. In the present study we compare the interaction of plain and targeted liposomes, containing mono-, di-, and tetraantennary mannosyl lipid derivatives, with human monocyte-derived immature dendritic cells (iDCs). Whereas efficient mannose receptor-mediated endocytosis by iDCs was observed for the mannosylated liposomes, in contrast, only nonspecific interaction with little uptake was observed with plain liposomes. In accordance with the clustering effect, liposomes prepared with multibranched mannosylated lipids displayed higher binding affinity for the mannose receptor than vesicles containing the monomannosylated analogs. Importantly, we have found that dimannosylated ligands present at the surface of the liposomes were as efficient as tetramannosylated ones to engage in multidentate interactions with the mannose receptor of iDCs, resulting in both cases in an effective uptake/endocytosis. This result will greatly facilitate, from a practical standpoint, the design of mannose-targeted vaccination constructs. Moreover, we showed that mannose-mediated uptake of liposomes did not result in an activation of iDCs. Altogether, our results suggest that antigen-associated targeted liposomes containing diantennary mannosylated lipids could be effective vectors for vaccines when combined with additional DC activation signals.

A new antibacterial coating made of poly(L-lysine)/hyaluronic acid (PLL/HA) multilayer films and liposome aggregates loaded with silver ions was designed. Liposomes filled with an AgNO 3 solution were first aggregated by the addition of PLL in solution. The obtained micrometer-sized aggregates were then deposited on a PLL/HA multilayer film, playing the role of a spacer with the support. Finally, HA/PLL/HA capping layers were deposited on top of the architecture to form a composite AgNO 3 coating. Release of encapsulated AgNO 3 from this composite coating was followed and triggered upon temperature increase over the transition temperature of vesicles, found to be equal to 34 degrees C. After determination of the minimal inhibitory concentration (MIC) of AgNO 3 in solution, the antibacterial activity of the AgNO 3 coating was investigated against Escherichia coli. A 4-log reduction in the number of viable E. coli cells was observed after contact for 120 min with a 120 ng/cm (2) AgNO 3 coating. In comparison, no bactericidal activity was found for PLL/HA films previously dipped in an AgNO 3 solution and for PLL/HA films with liposome aggregates containing no AgNO 3 solution. The strong bactericidal effect could be linked to the diffusion of silver ions out of the AgNO 3 coating, leading to an important bactericidal concentration close to the membrane of the bacteria. A simple method to prepare antibacterial coatings loaded with a high and controlled amount of AgNO 3 is therefore proposed. This procedure is far superior to that soaking AgNO 3 or Ag nanoparticles into a coating. In principle, other small bactericidal chemicals like antibiotics could be encapsulated by this method. This paper describes the development of stable drug delivery systems named layersomes. The layersomes are conventional liposomes coated with one or multiple layers of biocompatible polyelectrolytes in order to stabilise their structure. The formulation strategy is based on an alternative coating procedure of positive poly(lysine) (pLL) and negative poly(glutamic acid) (pGA) polypeptides on initially charged small unilamellar liposomes (SUVs). The size distribution and the zeta potential of the final entity depend on the number of polyelectrolyte layers and the charge of the last coating layer. Morphological studies were achieved by flux cytometry and cryo electron microscopy. Release studies of encapsulated hydrophilic 5(6)-carboxyfluorescein (5,6CF) in the presence of Triton® or ethanol showed an increased membrane resistance of the layersomes compared to classical SUVs. Finally, encapsulation of piroxicam (PX) was performed with success.

We have developed liposome-based synthetic constructs incorporating peptide epitope(s) (ErbB2 p63-67 CTL which is overexpressed in many tumors and/or HA 307-319 T-helper) and lipopeptide adjuvants (Pam3CysSerSer, Pam3CysAlaGly) in order to elicit an anti-tumor immune response. The epitopes, derivatized with a linker containing a cysteine residue, were conjugated on preformed vesicles (dia. approximately 100 nm) containing lipopeptides functionalized with thiol reactive groups (maleimide or bromoacetyl). The therapeutic efficacy of these constructs was evaluated on a Balb/c mice tumor model inoculated with syngenic murine renal carcinoma (Renca) cells expressing human ErbB2 (Her2/neu) receptor. A successful therapeutic vaccination was obtained which was antigen specific. Furthermore, it appeared that the nature of the polar head group of the lipopeptide adjuvant and also its type of functionalization influence the efficacy of the construct. In our study, the best results were obtained with formulations containing a Pam3CSS anchor in association with the CTL and Th epitopes. Considering these promising results studies are in progress with a new generation of liposomes that incorporate a neutral lipid--lacking adjuvant properties--that serves as anchor of the peptide epitopes and new adjuvants synthesized in our laboratory, which are screened for their antitumour activity in a therapeutic setting.

A formulation process, based on the inversion phase of an emulsion, was used to prepare lipid nanocapsules. Triglycerides, lecithin, salted water and hydroxy stearate of poly(ethylene glycol) were used in the preparation. The amounts of each that allowed nanocapsules to be formed described a feasibility domain within a ternary diagram. The size distribution of various nanoparticulate carriers has already been shown to influence their applications. An experimental mixture design inside the feasibility domain has been used in order to approximate, through an empirical model, the influence of the quantitative composition of nanocapsules on their size distribution. Reduced cubic polynomial equations successfully modelled the evolution of responses in terms of particle average diameters and coefficients of variation. The results were presented using an analysis of response surface showing a scale of possible particle sizes between 20 and 95 nm and a coefficient of variation between 11 and 40%. Furthermore, this technique showed that the proportion of hydrophilic surfactant had a major influence on the average diameter and the size distribution of the particles decreasing when its proportion increased. On the contrary, the coefficient of variation and the average diameter slightly increase with the proportion of triglycerides. Such a tool offers major advantages to design the formulation of particles as a function of the required size distribution.

The aim of the present work was to study the biodistribution of a radiolabeled lipidic nanocapsule formulation after intravenous administration in rat by scintigraphy and γ counting. This formulation is expected to be used as anticancer agent delivery devices and as transfection complexes. For this purpose, 99m Tc-oxine was incorporated in the lipidic core, while 125 I labeled tensioactive shell of the nanocapsule. First, in vitro stability of radiolabeled nanocapsules was evaluated by dialysis against distilled water and size measurements. Second, the nanocapsule biodistribution was followed after intravenous administration for 3 h by dynamic scintigraphic acquisition and up to 24 h by determining the gamma activity in blood and tissues. Radiolabeling was efficient and stable in vitro. After intravenous injection blood radioactivity decreased with an early half disappearance time of about 45 min for both radioisotopes. Liver and intestine radioactivities raised up to 24 h. The relatively long remanence in blood of the tracers which is probably due to the presence of PEG at the nanocarrier surface seems promising for the use of these solvent free lipidic nanocapsules as carrier of lipophilic drugs.

To develop and subsequently evaluate a novel phase inversion-based method used to formulate lipidic nanocapsules.Mechanical properties of emulsions prepared by multiinversion phase processes were investigated using a drop tensiometer. Based on the results obtained, a formulation process was developed and a new type of nanocarrier was prepared. These particulates were sized by photon correlation spectroscopy and were visualized by atomic force microscopy and transmission electronic microscopy. Differential scanning calorimetry was also performed.The marginally cohesive but stable interfacial properties of the initial system led to the formulation of lipidic nanocapsules that were composed of a liquid core surrounded by a cohesive interface and were dispersed in an aqueous medium. These related suspensions were stable upon dilution for several months. The control of the formulation parameters allowed an adjustment of the particle mean diameter in the range of 25-100 nm with a monodisperse size distribution.A novel and convenient process for the preparation of lipidic nanocapsules is described. The structure of these particulates resembles a hybrid between polymeric nanocapsules and liposomes. Such nanocapsules display a strong potential for drug delivery.

This paper describes the preparation of colloidal lipid particles containing magnetite from warm emulsions. A two step method was used to obtain the nanoparticles: (i) formulation of a transparent phase by heating a O/W emulsion (aqueous surfactant solution melted with a lipid phase, containing the ethyl oleate and soybean lecithin) in which modified lipophilic magnetite is incorporated, and (ii) preparation of the nanoparticles by dispersing the warm transparent phase in cold water (7 degrees C) under mechanical stirring. The latter method gives spherical nanoparticles of a mean size of 62 nm measured by Photon Correlation Spectroscopy and Transmission Electronic Microscopy. The magnetite entrapment efficiency was determined by use of a magnetophoretic sedimentation method.

The interactions of naked and surface-modified poly(D,L-lactic acid) (PLA) nanocapsules (NC), where polyethyleneglycol (PEG) was adsorbed or covalently attached, have been studied with a macrophage-like cell line. The fluorescent oil marker, DiD, was successfully encapsulated in NCs in order to follow their interactions with cells. The cell-associated fluorescence obtained with PEG-PLA NC was about 3- to 13-fold lower than that obtained with naked-PLA NC. The effects of PEG chain length, its content as a percentage of total polymer and NC concentration in the culture medium were evaluated. PEG-PLA NC showed dramatically reduced fluorescence association with cells during an 18 h incubation compared with naked-PLA NC, showing that covalent attachment of PEG is important for the persistence of low uptake. The best results in reducing cell-associated fluorescence were obtained with a surface-modified PEG-PLA NC bearing a chain with 20000 MW. Increasing the percentage of PEG produced a reduction in marker association for a given PEG chain length. Moreover, when the PEG-containing poloxamer was simply adsorbed, marker association was dependent on the extent of dilution and the type of serum in the culture medium. Serum proteins, especially immunoglobulins, increased cell-associated fluorescence for PEG-adsorbed NC, but had very little effect on PEG-PLA NC. Marker association was only partially inhibited in the presence of cytochalasin B. The mechanisms of cell-NC interaction depended on the characteristics of the NC surface in each formulation. When the NC was physically separated from cells no diffusion of fluorescent marker in aqueous medium occurred. Nevertheless, collision-mediated transfer of DiD from NC to J774 cells was a non-negligible route of marker transfer, mainly for naked NC. However, this collision-mediated transfer was reduced for the PEG-PLA NC probably due to the restricted contact between NC and cells afforded by PEG steric hindrance at the surface.

The Layer-by-Layer (LbL) technique has emerged as a simple and effective method for surface modification and functionalization, especially of nanoparticles. We have explored this coating technique to increase the resistance of conventional liposomes. We have developed a layer-by-layer formulation procedure using two biodegradable and biocompatible polyelectrolytes with opposite charges: poly(L-lysine) (PLL) and poly(L-glutamic) acid (PGA). This procedure has allowed the development of very homogeneous formulations of liposomes coated with up to 6 layers of polymers (layersomes) and liposomes coated with two layers of cross-linked polyelectrolytes. The coating was characterized by dynamic light scattering (DLS), quartz crystal microbalance technique (QCM), and energy transfer between two fluorescent molecules (FRET). Studies on the stability of formulations at 4°C in a buffered solution have shown that some structures are stable over 2 months without impacting their encapsulation capacity. The results suggest a better resistance of the coated liposomes in the presence of a non-ionic detergent (Triton™ X-100) as well as in the presence of plasma. In a second step, this procedure was adapted to coat inactivated H5N1 virus particles in order to develop a "delayed" form of the virus. Viral particles coated with up to 4 polymer layers as well as particles coated with 2 layers of cross-linked polyelectrolytes were obtained and characterized by DLS, laser diffraction (LD) as well as confocal and transmission electron microscopy. Stability studies have shown that the coating of viruses stored at 4°C is stable for at least 2 months. We showed that the LbL assembly had no impact on the immunogenicity of the viral particles and that a delayed release of the virus was likely. This work has demonstrated the adaptability of layer by layer assembly of nanoparticles for various pharmaceutical applications.; La technique du recouvrement couche par couche (« Layer-by-Layer », LbL) s’est imposée en tant que méthode simple et efficace pour la modification et la fonctionnalisation de surfaces, notamment de nanoparticules. Nous avons exploré cette technique de recouvrement afin d’augmenter la résistance de liposomes conventionnels. Nous avons mis au point une procédure de formulation couche par couche en utilisant deux polyélectrolytes biodégradables et biocompatibles de charges opposées : la poly(L-lysine) (PLL) et l’acide poly(L-glutamique) (PGA). Cette procédure a permis de développer des formulations très homogènes de liposomes recouverts jusqu’à 6 couches de polymères (layersomes) et de liposomes recouverts de deux couches de polyélectrolytes réticulés. Le recouvrement a été caractérisé par diffusion dynamique de la lumière (DLS), par la technique de microbalance à cristal de quartz (QCM), ainsi que par transfert d’énergie entre deux molécules fluorescentes (FRET). Des études de stabilité des formulations à 4°C dans une solution tamponnée ont démontré que certaines structures sont stables sur 2 mois sans impacter leur capacité d’encapsulation. Les résultats suggèrent une meilleure résistance des liposomes recouverts en présence d’un détergent non-ionique (Triton™ X-100) ainsi qu’en présence de plasma. Dans un second temps, cette procédure a été adaptée au recouvrement de particules virales inactivées de type H5N1, dans le but de développer une forme « retard » du virus. Des particules virales recouvertes jusqu’à 4 couches de polymères ainsi que des particules recouvertes de 2 couches de polyélectrolytes réticulés ont été obtenues et caractérisées par DLS, par diffraction laser (LD) ainsi que par microscopies confocale et électronique à transmission. Des études de stabilité ont démontré que le recouvrement des virus conservés à 4°C est stable au moins 2 mois. Nous avons montré que le recouvrement n’a pas eu d’impact sur l’immunogénicité des particules virales et qu’un relargage différé du virus était probable. Ces travaux ont ainsi démontré l’adaptabilité de l’assemblage couche par couche de nanoparticules pour des applications pharmaceutiques variées.

Currently, a challenging goal in the area of cancer treatment is the development of innovative targeted antitumoral immunotherapies with a long-term efficiency. In this context, we took advantage of liposomal nanoparticles properties for the conception of a tunable vaccine platform allowing the strategical conception of vaccines containing: i) a CD4 epitope peptide able to stimulate CD4+ T helper cells ii) a tumor CD8 epitope peptide, which induces the differentiation of CD8+ T cells in cytotoxic T cells and iii) Toll or Nod-Like receptor agonist(s), which act as adjuvant for the activation of dendritic cell. After several screening steps, three vaccines, specific tardeting an orthotopic lung tumor model (TC-1) and differing only by their adjuvant composition, were successfully developed. Subsequently, a comparative study of their efficacy time limit and their immunologic mode of action was performed. This study showed the therapeutic supremacy of liposomal vaccines containing a TLR4 agonist (MPLA). In this work, we demonstrated the value of a customizable liposomal platform for the conception of personalized vaccines and we highlighted the necessity of immune monitoring for a better understanding of vaccines impact and the prediction of their efficacy., Un des enjeux majeurs de la thérapie contre le cancer est le développement d’immunothérapies innovantes ciblées et efficaces sur le long terme. Dans ce but, nous avons tiré profit de la versatilité des liposomes pour concevoir une plateforme vaccinale modulable associant i) un épitope peptidique CD4 universel capable de stimuler les lymphocytes T CD4+, ii) un épitope peptidique CD8 dérivé de la tumeur, induisant la différenciation des lymphocytes T CD8+ en lymphocytes T cytotoxiques et iii) un ou des agonistes des récepteurs Toll-like ou NOD-like qui agissent comme des adjuvants en activant les cellules dendritiques. Après plusieurs étapes de criblage, trois vaccins ont été développés, spécifiques d’un modèle orthotopique de cancer du poumon (TC-1) et différant uniquement par leur adjuvant. Une étude comparative dans un modèle murin a été réalisée. Ces travaux ont permis de comparer leur limite temporelle d’efficacité, de mettre en évidence leur mécanismes immunitaires respectifs et de démontrer la supériorité thérapeutique des liposomes contenant un agoniste du TLR4 (MPLA). Ces travaux ont montré l’intérêt d’une plateforme liposomique modulable pour la conception de vaccins personnalisés.

With the identification of tumor antigens and the better understanding of the mucosal immune response, the vaccination by the respiratory route has become a promising field of investigation for cancer treatment. The purpose of this study was to develop nanoparticulate peptide-based liposomal vaccines for antitumor vaccination by respiratory or nasal route. Thus, we have prepared liposomes associating ErbB2 TCD8+ and HA TCD4+ peptide epitopes with an adjuvant molecule. This construct was then optimized by varying its physicochemical characteristics (size, structure, composition) or its formulation (viscosity). The antitumor efficacy of the various vaccines obtained thereby was evaluated in a model of pulmonary or subcutaneous tumors in mice after prophylactic or therapeutic, nasal or respiratory immunization. All our data showed an undeniable interest of peptide vaccines based on liposomes in the antitumor vaccination by the respiratory and nasal routes, opening new perspectives for cancer treatment., Avec l’identification des antigènes tumoraux et la compréhension de la réponse immunitaire mucosale, la vaccination par voie respiratoire est devenue un champ d’investigation prometteur pour le traitement du cancer. L’objectif de cette thèse était de concevoir des vaccins peptidiques nanoparticulaires à base de liposomes, destinés à la vaccination antitumorale par voie respiratoire ou nasale. Ainsi, nous avons formulé des liposomes vectorisant un épitope peptidique TCD8+ dérivé de l’oncogène ErbB2, un épitope TCD4+ et une molécule adjuvante. Cette construction a ensuite été optimisée en faisant varier les caractéristiques physicochimiques du vecteur liposomique (taille, structure, composition) ou de la formulation (viscosité). L’efficacité antitumorale des différents vaccins ainsi obtenus a été évaluée après administration respiratoire ou nasale, prophylactique ou thérapeutique, dans un modèle de tumeurs pulmonaires ou de tumeurs sous-cutanées chez la souris. L’ensemble de ces travaux a montré un intérêt indéniable des vaccins peptidiques à base de liposomes dans la vaccination antitumorale par voie respiratoire et nasale, ouvrant de nouvelles perspectives pour le traitement du cancer.
