Abstract

Endometrial cancer is the most common gynecologic malignancy in developed countries. The antibody–drug conjugate (ADC) sacituzumab govitecan (SG) targets trophoblast cell-surface antigen-2 (Trop-2) – a cell-surface glycoprotein highly expressed in many epithelial tumors – and delivers the active metabolite of irinotecan SN-38 to Trop-2-positive tumor cells. We evaluated Trop-2 expression in endometrial endometrioid carcinoma (EC) tissues and the activity of SG against primary poorly differentiated EC cell lines and xenografts. Trop-2 expression was assessed in 143 formalin-fixed–paraffin-embedded tumors and seven primary tumor cell lines by immunohistochemistry and flow cytometry, respectively. Cell viability of primary tumor cell lines was assessed following exposure to SG, or control antibodies. Antibody-dependent cell cytotoxicity (ADCC) against Trop-2-positive and Trop-2-negative EC cell lines was measured in vitro using 4-h chromium release assays. A Trop-2-positive EC xenograft model was used to determine the in vivo activity of SG. Moderate-to-strong staining was detected in 84% (120/143) of EC samples, whereas 43% (3/7) of the primary EC cell lines tested overexpressed Trop-2. EC cell lines overexpressing Trop-2 were significantly more sensitive to SG compared to control ADC (P = 0.014 and P = 0.005). Both SG and the unconjugated parental antibody hRS7 mediated high ADCC against Trop-2-positive cell lines. Moreover, SG induced significant bystander killing of Trop-2-negative tumors cocultured with Trop-2-positive tumors. In the xenograft model, intravenous administration of SG twice weekly for three weeks was well tolerated and demonstrated impressive tumor growth inhibition against poorly differentiated, chemotherapy-resistant EC xenografts (P = 0.011). Abstract

Background: Epithelial ovarian cancer (EOC) is the most lethal gynecologic malignancy. Sacituzumab govitecan (SG) is a novel antibody-drug-conjugate (ADC) targeting trophoblast-antigen-2 (Trop-2), a cell surface glycoprotein highly expressed in many epithelial tumors, to deliver SN-38, the active metabolite of irinotecan. This study aimed to evaluate Trop-2 expression in EOC tissues and the preclinical activity of SG against primary EOC cell lines and xenografts. Methods: Trop-2 expression was assessed in 90 formalin-fixed-paraffin-embedded tumors and nine primary tumor cell lines by immunohistochemistry (IHC) and flow cytometry, respectively. Trop-2 expression and cell viability after exposure to SG in primary tumor cell lines, non-targeting control ADC, and SG-parental antibody hRS7 were evaluated using flow-cytometry-based-assays. Antibody-dependent-cell-cytotoxicity (ADCC) against Trop-2+ and Trop-2– EOC cell lines was tested in vitro using 4 h Chromium-release-assays. In vivo activity of SG was evaluated against Trop-2+ EOC xenografts. Results: Moderate-to-strong staining was seen in 47% (42/90) of ovarian tumors by IHC while 89% (8/9) of the primary EOC cell lines overexpressed Trop-2 by flow cytometry. EOC Trop-2+ were significantly more sensitive to SG compared to control ADC (p < 0.05). Both SG and hRS7 mediated high ADCC activity against Trop-2+ cell lines. SG also induced significant bystander killing of Trop-2– tumor cells admixed with Trop-2+ EOC cells. In in vivo experiments SG treatment demonstrated impressive anti-tumor activity against chemotherapy-resistant EOC xenografts. Conclusion: SG demonstrates remarkable preclinical activity against biologically aggressive and chemotherapy-resistant EOC cell lines and a significant bystander effect against EOC cell lines with heterogenous Trop-2 expression. Clinical trials are warranted.

Abstract

Background There are several types of cancer, which cause millions of deaths worldwide every year. Many studies have confirmed that plants are adequate natural sources to be examined as anti-cancer drugs with fewer side effects than chemotherapy and radiotherapy. In this study the anti-cancer properties of Lavender aqueous extract on lymphocytes derived from patients with Hodgkin's lymphoma has been studied. Methods In order to determine the cytotoxic effects of the extract on lymphocytes of patients in stages III and IV of Hodgkin's lymphoma and two different cell lines in the presence of different concentrations of aqueous extract of Lavender, MTT colorimetric assay and flow cytometry analysis were used. Results Findings indicated that Lavender inhibited cell proliferation in both lymphocytes and cell lines with different effects. The effective concentration of Lavender that decreased viability of Hodgkin's lymphoma cells below Lethal Concentration 50 (LC50) value was 100 µg/ml and this was half of the therapeutic dose. In addition, apoptosis was the main mechanism the Hodgkin's lymphoma cell encountered when exposed to the aqueous extract of Lavender. Conclusion This experiment proposes that aqueous Lavender extract can be regarded as a potential anti-cancer agent in future studies.

Abstract

This experiment was conducted to investigate the effects of various starvation and refeeding periods on growth, feed utilization and body composition in Asian sea bass (Lates calcarifer) with an average initial weight of 30.26±1.4 g (mean±SE). The fish were exposed to three different regimes: the control group fed twice daily to apparent satiation throughout the experiment (C), the first group starved for 4 days and refed for 16 days, this cycle was repeated two times (T1) and the second group starved for 8 days and refed for 32 days (T2). At the end of experiment, there were not any significant differences in growth and feeding performance among different treatments (p>0.05). Daily feed intake was significantly higher in the deprived fish than in the control fish (p0.05). Abstract

Springs are the important water resources, which thier study is necessary in terms of their management and exploitation. In northern Iran, the karst springs locate in the central Alborz highlands and alluvial springs on the Caspian southern coasts.The Karst and alluvial springs discharge is variable because of different conditions in terms of aquifer, topography and precipitation .This study has been done to simulate the karst and alluvial springs discharge and to compare their hydrologic conditions. So, 80 karst springs and 82 alluvial springs were investigated in central Alborz and on the Caspian southern coasts. Two models were presented to simulate the karst and alluvial springs discharge using Multivariate Regression analysis (SPSS software).Then, their efficiency was considered. Finally, the models and the results of regression analyses were compared. The results showed that the most important factors of the karst springs discharge are aquifer formation (its porosity) and site elevation, but about the alluvial springs, the hydraulic conductivity of aquifer formation and aquifer depth are the most important factors.

Abstract

In the current study, the effects of different levels of probiotics, Lactobacillus plantarum in the diet of Asian sea bass, Lates calcarifer on body composition, serum biochemical parameters (glucose, cholesterol, triglyceride and albumin) and liver enzymes (lactate dehydrogenase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase) were investigated. This experiment was carried out with three treatments and a control group for a period of four weeks. The treatments were included 0, 1×106, 2×106, 3×106 colony forming unit (CFU) gram probiotic in each gram of basic diet. Fish with an average weight of 50.26 ±0.89 grams were randomly distributed in tanks and in each tank stocked 20 pieces of fish. Feeding was done twice a day. The obtained results indicated that the use of different levels of probiotic in diet of Asian sea bass had no significant effects on serum biochemical parameters and liver enzymes. The different levels of probiotic did not affect protein, ash and moisture values between the treatments and the control group (P> 0.05). Moreover, fat value in the fish T2 and T3 was significantly higher (P< 0.05) than the fish control and T1. Also, nitrogen free extract (NFE) value varied significantly (P < 0/05) between the control and the fish T3. Overall, this study indicated that this probiotic had no significant effect on some blood biochemical parameters and liver enzyme, but at the high level can positively influence on body composition of Asian sea bass.
