Multivariate analysis of thin-layer chromatography (TLC) images was modeled to predict antioxidant activity of Pereskia bleo leaves and to identify the contributing compounds of the activity. TLC was developed in optimized mobile phase using the 'PRISMA' optimization method and the image was then converted to wavelet signals and imported for multivariate analysis. An orthogonal partial least square (OPLS) model was developed consisting of a wavelet-converted TLC image and 2,2-diphynyl-picrylhydrazyl free radical scavenging activity of 24 different preparations of P. bleo as the x- and y-variables, respectively. The quality of the constructed OPLS model (1 + 1 + 0) with one predictive and one orthogonal component was evaluated by internal and external validity tests. The validated model was then used to identify the contributing spot from the TLC plate that was then analyzed by GC-MS after trimethylsilyl derivatization. Glycerol and amine compounds were mainly found to contribute to the antioxidant activity of the sample. Pereskia bleois a species of primitive cactus. In the present study, infrared spectroscopy was used to characterize the antioxidant activity ofP. bleoleaves by multivariate analysis. A total of twenty-four extracts were prepared in different solvents. Antioxidant activities were measured by 1,1-diphenyl-2-picrylhydrazyl assay and fingerprinted by infrared spectroscopy between 4000 and 400 cm−1at a resolution of 2 cm−1. A three component multivariate orthogonal partial least squares model with R2Y of 0.88 and Q2of 0.86 was developed to correlate infrared spectra with antioxidant activity and evaluated by internal cross-validation and a true external test. For external validation, bioactivity of new extracts ofP. bleowas predicted using the model, and -OH, -NH, and -CH were identified as functional groups responsible for the activity. Non-human primates remain the most useful and reliable pre-clinical model for many human diseases. Primate breath profiles have previously distinguished healthy animals from diseased, including non-human primates. Breath collection is relatively non-invasive, so this motivated us to define a healthy baseline breath profile that could be used in studies evaluating disease, therapies, and vaccines in non-human primates. A pilot study, which enrolled 30 healthy macaques, was conducted. Macaque breath molecules were sampled into a Tedlar bag, concentrated onto a thermal desorption tube, then desorbed and analyzed by comprehensive two-dimensional gas chromatography-time of flight mass spectrometry. These breath samples contained 2,017 features, of which 113 molecules were present in all breath samples. The core breathprint was dominated by aliphatic hydrocarbons, aromatic compounds, and carbonyl compounds. The data were internally validated with additional breath samples from a subset of 19 of these non-human primates. A critical core consisting of 23 highly abundant and invariant molecules was identified as a pragmatic breathprint set, useful for future validation studies in healthy primates., (© 2024. The present study was aimed at evaluating mineral contents, B-vitamins (B1, B2, B3, B6), vitamin C, total carotenoids content and carotene composition of unconventional vegetables, comprising 7 leafy and 4 non-leafy, growing in Bangladesh. Investigated unconventional vegetables were found to have varied amount of minerals (0.04 ± 0.01 to 786.41 ± 6.37 mg/100 g), B-vitamins (0.06 ± 0.01 to 1.05 ± 0.13 mg/100 g) and vitamin C (5.23 ± 0.81 to 59.01 ± 2.23 mg/100 g). The study findings also revealed that the selected plant samples exhibited diverse amount of β-carotene ranging from 106.26 ± 5.32 µg/100 g EP in Orhor dal to 994.81 ± 21.01 µg/100 g EP in Orhor Pata whereas lutein content ranged from 7.76 ± 1.02 µg/100 g EP in Pahari Kolar Thor to 566.33 ± 20.63 µg/100 g EP in Kachuripana. It can be suggested that regular intake of these vegetables may contribute to daily minerals and vitamins requirement and can combat micronutrient deficiency.
