The present research aims to enhance the antimicrobial activity of kaolinite surfaces by a one-step cost-effective and energy-efficient dry thermal reaction, producing an antibacterial and antifungal silver-kaolinite (Ag-Kao) nanocomposite agent. Pharmaceutical grade kaolin powder samples, with variable kaolinite structural order–disorder degree, were homogeneously mixed with silver nitrate in a proportion 1:4 AgNO3:kaolin (w/w) and sintered at 400 °C for 30 min. The composition, microstructure, microtexture and surface characteristics of the pyro-fabricated nanocomposites were characterized by XRD/XRF diffractometry, differential scanning calorimetry DSC, FT-IR spectroscopy, TEM/EDX, zeta potential (mV) measured within the 2–12 pH range, and BET method. Physicochemical stability was evaluated by silver dissociation testing under close-neutral and acidic conditions with Ag content assay using ICP-OES. The resulting Ag-Kao nanocomposites exhibited bulk silver contents ranging from 9.29% to 13.32% with high physicochemical stability in both neutral and acidic mediums (Ag dissociation rate <0.5% in 5 days). Ag nanocrystals exhibited particle sizes ranging from 5 to 30 nm, which were embedded and reinforced within the kaolinite matrix. The sizes of the Ag nanocrystals and their distribution patterns on the edges and faces of kaolinite platelets were controlled by the structural order–disorder degree. Highly ordered kaolinites (Hinckley Index, HI > 1) produced platelet edge-clustered silver nanocrystals due to the abundance of the dangling hydroxyls on platelet edges, while the highly disordered kaolinite (HI < 1) provided homogeneous platelet basal-doped silver nanocrystals due to the presence of some residual charges by exposed basal hydroxyl groups with interplatelet silver diffusivity. The present work aimed at assessing the pharmaceutical tableting properties of some Egyptian kaolin samples belong to the Abu Zenima kaolin deposits (estimated at 120 million tons). Four representative samples were selected based on kaolinite richness and their structural order-disorder degree, and after purification, they were dried at 70 C and heated from room temperature up to 400 C (10 C/min). Mineralogy, micromorphology, microtexture, granulometry, porosimetry, moisture content, bulk and tapped density, direct and indirect flowability, and tableting characteristics are studied. Results indicated that purified kaolin samples were made up of 95–99% kaolinite, <3% illite, 1% quartz and 1% anatase. The powder showed mesoporous character (pore diameters from 2 to 38 nm and total pore volume from 0.064 to 0.136 cm3/g) with dominance of fine nanosized particles (<1 m–10 nm). The powder flow characteristics of both the ordered (Hinckley Index HI > 0.7, crystallite size D001 > 30 nm) and disordered (HI < 0.7, D001 < 30 nm) kaolinite-rich samples have been improved (Hausner ratio between 1.24 and 1.09) as their densities were influenced by thermal treatment (with some observed changes in the kaolinite XRD reflection profiles) and by moisture content (variable between 2.98% and 5.82%). The obtained tablets exhibited hardness between 33 and 44 N only from the dehydrated powders at 400 C, with elastic recovery (ER) between 21.74% and 25.61%, ejection stress (ES) between 7.85 and 11.45 MPa and tensile fracture stress (TFS) between 1.85 and 2.32 MPa, which are strongly correlated with crystallinity (HI) and flowability (HR) parameters. These findings on quality indicators showed the promising pharmaceutical tabletability of the studied Egyptian kaolin powders and the optimization factors for their manufacturability and compactability.

The present work aims to increase the alumina percentage recovery (APR) extracted from kaolin via the addition of 0.5-4.0 wt.% charcoal as a thermochemical fluxing agent in the lime-sintering process at 1260-1360°C. The transformation, microstructural and microtextural changes and self-disintegration performance were characterized using thermogravimetric analysis and differential scanning calorimetry, X-ray diffraction/X-ray fluorescence, scanning electron microscopy coupled with energy-dispersive spectroscopy and laser diffraction particle-size distribution analysis. The optimum enhancement of APR, from 77.7% to 87.40%, was obtained by sintering at 1360°C with the addition of 1.5% charcoal. With further increase of the charcoal content to 4%, the APR reduced to 75.6%. Combustion of ≤1.5% charcoal provided additional heat that amorphized the crystalline calcium aluminate into highly leachable amorphous phases with improved self-disintegration efficiency. Sintering at temperatures of >1360°C or with charcoal contents >4% led to mullite crystallization and decreased alumina leachability, thereby reducing the APR. Charcoal is a cost-effective and energy-efficient activator to increase the APR extracted from kaolin.

This work aimed at studying the potentiality of interactions between kaolinite surfaces and a protein-fragment (350–370 amino acid units) extracted from the glycoprotein E1 in the transmembrane domain (TMD) of hepatitis C virus capsid. A computational work was performed for locating the potential electrostatic interaction sites between kaolinite aluminol and siloxane surfaces and the residues of this protein-fragment ligand, monitoring the possible conformational changes. This hydrated neutralized kaolinite/protein-fragment system was simulated by means of molecular modeling based on atomistic force fields based on empirical interatomic potentials and molecular dynamic (MD) simulations. The MD calculations indicated that the studied protein-fragment interacted with the kaolinite surfaces with an exothermic process and structural distortions were observed, particularly with the hydrophilic aluminol surface by favorable adsorption energy. The viral units isolation or trapping by the adsorption on the kaolinite nanoparticles producing structural distortion of the peptide ligands could lead to the blockage of the entry on the receptor and hence a lack of viral activity would be produced. Therefore, these findings with the proposed insights could be an useful information for the next experimental and development studies in the area of discovering inhibitors of the global challenged hepatitis and other pathogenic viruses based on the phyllosilicate surface activity. These MD studies can be extended to other viruses like the COVID-19 interacting with silicate minerals surfaces.

This work contributes to the knowledge of colloidal and thermal properties of some important primary-originated kaolin deposits from Bolivar State, Venezuela, and their possible use as semisolid formulations in medicinal muds for topical applications. Eight selected high purity kaolin samples were characterized. Rheological and thermal properties were correlated to physicochemical characteristics of the clay suspensions (pH, Ca2+, and Mg2+ cation desorption and surface charge). Most of the studied kaolin pastes showed adequate viscosities, acceptable skin safe pH, and good thermal properties for pelotherapeutic uses. Three of the studied samples, in particular, showed very high kaolinite purities (>92% kaolinite), elevated viscosities (>1 Pa¿s), and good thermal and pH performances for topical application. © 2019 by the authors. The application of clay minerals in therapeutics is becoming important due to their structural and surface physicochemical properties. 5-aminosalicylic acid (5-ASA) is a very common pharmaceutical drug and is used worldwide. The interactions between the 5-ASA molecule and both the aluminol and siloxane surfaces of kaolinite are studied by means of atomistic calculations using force fields based on empirical interatomic potentials and quantum mechanics calculations based on density functional theory. A conformational analysis of 5-ASA has been performed and the anion of 5-ASA was also studied. The calculated adsorption energy values indicate that 5-ASA is likely to be adsorbed on the kaolinite surfaces with greater affinity to the aluminol surface. Hence, kaolinite may be considered as a promising pharmaceutical carrier of 5-ASA.

El presente estudio tiene como objetivo evaluar, por primera vez, la potencialidad de los depósitos de caolín egipcio de Abu Zenimacon una reserva estimada de 120 millones de toneladas, para su empleo en ámbito farmacéutico. El distrito del caolín estudiado cubre aproximadamente 333 Km2 de tierras cultivadas y accesibles que se encuentra en la parte oeste de la península central del Sinaí, entre la longitud 33˚ 14 00˝ y 33˚ 24 00˝ E y la latitud 28˚ 52 00˝ y 29˚ 10 00˝ N. Los objetivos del estudio se centran en conocer las características de calidad farmacéutica, que incluyen parámetros mineralógicos y tecnológicos destinados a establecer la identidad, pureza y riqueza de las muestras de caolines y sus posibilidades de empleo farmacéutico y aplicaciones biomédicas. Paralelamente al trabajo de recogida de muestras se llevó a cabo una profunda revisión bibliográfica del empleo farmacéutico de la caolinita, para comprender sus funciones, especificaciones, límites, criterios de uso y parámetros de control. Los resultados y conclusiones de estos estudios previos se consideran información y orientación importantes para las posibilidades de explotación, así como también revelan nuevas hipótesis y tendencias para las funcionalidades de los grados variados de caolín.

Halloysite nanotubes are becoming interesting materials for drug delivery. The knowledge of surface interactions is important for optimizing this application. The aim of this work is to perform a computational study of the interaction between 5-aminosalicylic acid (5-ASA) drug and halloysite nanotubes for the development of modified drug delivery systems. The optimization of this nanotube and the adsorption of different conformers of the 5-ASA drug on the internal surface of halloysite in the presence and absence of water were performed using quantum mechanical calculations by using Density Functional Theory (DFT) and methods based on atomistic force fields for molecular modeling, respectively.

Although peroxisome proliferators are considered non-genotoxic agents, most of them, nevertheless, were found to promote and/or induce, hepatocellular carcinoma (HCC) in rodents. The aim of the present study is, first, to investigate whether the peroxisome proliferator perfluorooctanoic acid (PFOA) possesses inherent liver cancer promoting activity, and second, to study the possible mechanisms involved. To acheive these aims two protocols have been applied, a biphasic protocol (initiation by diethyl-nitrozamine (DEN) 200 mg/kg i.p. followed by treatment with 0.005% or 0.02% perflourooctanoic acid (PFOA) for 14 and 25 weeks) and a triphasic initiation, selection-promotion (IS) protocol (initiation by giving 200 mg/kg DEN i.p. followed by a selection procedure for 2 weeks consisting of giving 0.03% 2-acetylaminofluorene (2-AAF) in diet). In the middle of this treatment a single oral dose of carbon tetrachloride (2.0 ml/kg) was given, followed by giving diet containg 0.015% of PFOA for 25 weeks. After applying both protocols, our results showed slight increase in the catalase activity while acyl CoA oxidase activity was markedly increased. Both experiments indicated that PFOA has a liver cancer promoting activity. Other groups of rats were given either basal diet or diet containing 0.02% PFOA. Five or nine weeks later they were sacrificed and the levels of 8-hydroxydeoxyguanosine in the isolated DNA were estimated. The data showed a slight nonetheless insignificant increase in 8-hydroxydeoxyguanosine. From the present data, it is concluded that PFOA is a true liver cancer promoter that may not require extensive initial DNA damage for its promoting activity.
