1. Mediation analysis in longitudinal intervention studies with an ordinal treatment-dependent confounder

Abstract: In interventional health studies, causal mediation analysis can be employed to investigate mechanisms through which the intervention affects the targeted health outcome. Identifying direct and indirect (i.e. mediated) effects from empirical data, however, becomes complicated if the mediator-outcome association is confounded by a variable itself affected by the treatment. Here, we investigate identification of mediational effects under such post-treatment confounding in a setting with a longitudinal mediator, time-to-event outcome and a trichotomous ordinal treatment-dependent confounder. We show that if the intervention always affects the treatment-dependent confounder only in one direction (monotonicity), the mediational effects are identified up to a sensitivity parameter and derive their empirical non-parametric expressions. The monotonicity assumption can be assessed from empirical data, based on restrictions on the conditional distribution of the treatment-dependent confounder. We avoid pitfalls related to post-treatment conditioning by treating the mediator as a functional entity and defining the time-to-event outcome as a restricted disease-free time. In an empirical analysis, we use data from the Finnish Diabetes Prevention Study to assess the extent to which the effect of a lifestyle intervention on avoiding type 2 diabetes is mediated through weight reduction in a high-risk population, with other health-related changes acting as treatment-dependent confounders., Comment: 27 pages, 5 figures

2. Elementary methods provide more replicable results in microbial differential abundance analysis

Abstract: Differential abundance analysis is a key component of microbiome studies. While dozens of methods for it exist, currently, there is no consensus on the preferred methods. Correctness of results in differential abundance analysis is an ambiguous concept that cannot be evaluated without employing simulated data, but we argue that consistency of results across datasets should be considered as an essential quality of a well-performing method. We compared the performance of 14 differential abundance analysis methods employing datasets from 54 taxonomic profiling studies based on 16S rRNA gene or shotgun sequencing. For each method, we examined how the results replicated between random partitions of each dataset and between datasets from independent studies. While certain methods showed good consistency, some widely used methods were observed to produce a substantial number of conflicting findings. Overall, the highest consistency without unnecessary reduction in sensitivity was attained by analyzing relative abundances with a non-parametric method (Wilcoxon test or ordinal regression model) or linear regression (MaAsLin2). Comparable performance was also attained by analyzing presence/absence of taxa with logistic regression.

3. On the distribution of isometric log-ratio transformations under extra-multinomial count data

Abstract: Compositional data arise when count observations are normalised into proportions adding up to unity. To allow use of standard statistical methods, compositional proportions can be mapped from the simplex into the Euclidean space through the isometric log-ratio (ilr) transformation. When the counts follow a multinomial distribution with fixed class-specific probabilities, the distribution of the ensuing ilr coordinates has been shown to be asymptotically multivariate normal. We here derive an asymptotic normal approximation to the distribution of the ilr coordinates when the counts show overdispersion under the Dirichlet-multinomial mixture model. Using a simulation study, we then investigate the practical applicability of the approximation against the empirical distribution of the ilr coordinates under varying levels of extra-multinomial variation and the total count. The approximation works well, except with a small total count or high amount of overdispersion. These empirical results remain even under population-level heterogeneity in the total count. Our work is motivated by microbiome data, which often exhibit considerable extra-multinomial variation and are increasingly treated as compositional through scaling taxon-specific counts into proportions. We conclude that if the analysis of empirical data relies on normality of the ilr coordinates, it may be advisable to choose a taxonomic level where counts are less sparse so that the distribution of taxon-specific class probabilities remains unimodal.

5. Increasing trend of antimicrobial resistance among methicillin-resistant Staphylococcus aureus strains in Southwest Finland, 2007–2016: An analysis of shifting strain dynamics and emerging risk factors

Abstract: Objective: Substantial rise in the annual incidence of methicillin-resistant Staphylococcus aureus (MRSA) was reported in Southwest Finland (12.4–24.9/100,000 people) between 2007 and 2016. To understand the implications of these changes to the management of MRSA, we sought to analyse the antimicrobial resistance (AMR) trends of MRSA in relation with patient characteristics. Methods: Antimicrobial susceptibility was determined for 10 clinically relevant antimicrobials. Strains with resistance to ≥2 antimicrobials were defined multi-resistant. The isolates were spa-typed and clustered. AMR trends and risk factors were identified by associating resistant phenotypes with patient demographics. Results: A total of 983 new MRSA cases were identified between 2007 and 2016. After 2011, significant increasing trends were observed in the proportion of isolates resistant to clindamycin (13.9%–31.5%, P < 0.001), erythromycin (19.4%–35.4%, P < 0.001) and tetracycline (16.7%–32%, P < 0.001). The proportion of multi-resistant isolates more than doubled from 14.8% to 39.2%. The increasing AMR trend was reflected in the increase of new strain types and the decrease of previously dominant, non-multi-resistant strains. Patient risk factors associated with (P < 0.001) the acquisition of multi-resistant strains included community acquisition, livestock contact, hospital care abroad and immigrant status. Conclusions: Notable increasing AMR trends among MRSA isolates were observed in Southwest Finland, 2007–2016. The shift in patient demographics to younger age groups and community acquisition contributed to the increase in multi-resistant strains. Immigration, contact with hospital environment abroad and contact with livestock were identified as essential risk factors of multi-resistance. The increased level of co-resistance has persisted after 2016.

Abstract: Abstract Atomic nuclei serve as prime laboratories for investigations of complex quantum phenomena, where minor nucleon rearrangements cause significant structural changes. 190Pb is the heaviest known neutron-deficient Pb isotope that can exhibit three distinct shapes: prolate, oblate, and spherical, with nearly degenerate excitation energies. Here we report on the combined results from three state-of-the-art measurements to directly observe these deformations in 190Pb. Contrary to earlier interpretations, we associate the collective yrast band as predominantly oblate, while the non-yrast band with higher collectivity follows characteristics of more deformed, predominantly prolate bands. Direct measurement of the $$E0({0}_{2}^{+}\to {0}_{1}^{+})$$ E 0 ( 0 2 + → 0 1 + ) transition and γ-e − coincidence relations allowed us to locate and firmly assign the $${0}_{2}^{+}$$ 0 2 + state in the level scheme and to discover a spherical $${2}_{3}^{+}$$ 2 3 + state at 1281(1) keV with $$B(E2;{2}_{3}^{+}\to {0}_{1}^{+})=1.2(3)$$ B ( E 2 ; 2 3 + → 0 1 + ) = 1.2 ( 3 ) W.u. These assignments are based purely on observed transition probabilities and monopole strength values, and do not rely on model calculations for their interpretation.

Abstract: Excited states in the $T_z = 0, -1$ nuclei $^{62}$Ga and $^{62}$Ge were populated in direct reactions of relativistic radioactive ion beams at the RIBF. Coincident \grays were measured with the DALI2$^+$ array and uniquely assigned to the $A=62$ isobars. In addition, $^{62}$Ge was also studied independently at JYFL-ACCLAB using the ${}^{24}$Mg(${}^{40}$Ca,$2n$)${}^{62}$Ge fusion-evaporation reaction. The first excited $T=1, J^\pi =2^+$ states in $^{62}$Ga and $^{62}$Ge were identified at $979(1)$ and $965(1)$~keV, respectively, resolving discrepant interpretations in the literature. States beyond the first $2^+$ state in $^{62}$Ge were also identified for the first time in the present work. The results are compared with shell-model calculations in the $fp$ model space. Mirror and triplet energy differences are analyzed in terms of individual charge-symmetry and charge-independence breaking contributions. The MED results confirm the shrinkage of the $p$-orbits' radii when they are occupied by at least one nucleon on average., Comment: aceepted Phys. Lett. B

8. Methods for generating and evaluating synthetic longitudinal patient data: a systematic review

Abstract: The rapid growth in data availability has facilitated research and development, yet not all industries have benefited equally due to legal and privacy constraints. The healthcare sector faces significant challenges in utilizing patient data because of concerns about data security and confidentiality. To address this, various privacy-preserving methods, including synthetic data generation, have been proposed. Synthetic data replicate existing data as closely as possible, acting as a proxy for sensitive information. While patient data are often longitudinal, this aspect remains underrepresented in existing reviews of synthetic data generation in healthcare. This paper maps and describes methods for generating and evaluating synthetic longitudinal patient data in real-life settings through a systematic literature review, conducted following the PRISMA guidelines and incorporating data from five databases up to May 2024. Thirty-nine methods were identified, with four addressing all challenges of longitudinal data generation, though none included privacy-preserving mechanisms. Resemblance was evaluated in most studies, utility in the majority, and privacy in just over half. Only a small fraction of studies assessed all three aspects. Our findings highlight the need for further research in this area.

9. Hypothesis-driven mediation analysis for compositional data: an application to gut microbiome

Abstract: Biological sequencing data consist of read counts, e.g. of specified taxa and often exhibit sparsity (zero-count inflation) and overdispersion (extra-Poisson variability). As most sequencing techniques provide an arbitrary total count, taxon-specific counts should ideally be treated as proportions under the compositional data-analytic framework. There is increasing interest in the role of the gut microbiome composition in mediating the effects of different exposures on health outcomes. Most previous approaches to compositional mediation have addressed the problem of identifying potentially mediating taxa among a large number of candidates. We here consider causal inference in compositional mediation when a priori knowledge is available about the hierarchy for a restricted number of taxa, building on a single hypothesis structured in terms of contrasts between appropriate sub-compositions. Based on the theory on multiple contemporaneous mediators and the assumed causal graph, we define non-parametric estimands for overall and coordinate-wise mediation effects, and show how these indirect effects can be estimated from empirical data based on simple parametric linear models. The mediators have straightforward and coherent interpretations, related to specific causal questions about the interrelationships between the sub-compositions. We perform a simulation study focusing on the impact of sparsity and overdispersion on estimation of mediation. While unbiased, the precision of the estimators depends, for any given magnitude of indirect effect, on sparsity and the relative magnitudes of exposure-to-mediator and mediator-to-outcome effects in a complex manner. We demonstrate the approach on empirical data, finding an inverse association of fibre intake on insulin level, mainly attributable to direct rather than indirect effects.

Here, we present the methods and results for the passive register-based surveillance of serious adverse events (SAEs), which replaced conventional SAE reporting during the trial.Patients and Methods: The trial recruited over 33,000 older adults of whom 50% received the QIV-HD while the rest received a standard-dose vaccine (QIV-SD) as a control vaccine. We collected diagnoses related to all acute hospitalizations during the six months following vaccination from national registers. During the blinded phase of the trial, we utilized a cohort study design and compared the incidences of 1811 ICD10 diagnosis groups (SAE categories) between the trial population and older adults vaccinated with the QIV-SD outside the trial, either during the study or the previous influenza season. Based on a real-time probabilistic comparison, we flagged SAE categories with higher incidence in the trial population and then evaluated possible causal associations between each flagged category and the trial intervention.Results: Our novel approach to safety surveillance provided information, which we could evaluate in real-time during the trial. The trial participants experienced 1217 hospitalizations related to any SAE categories, contributed by 941 patients. We flagged 10 SAE categories for further analysis during the study but based on further data review, none presented strong evidence of causality with vaccination.Conclusion: Safety signals can be detected and evaluated in real-time during a pragmatic vaccine trial with register-based follow-up, utilizing passive data collection and population level comparison. Abstract: Information on the $^{38}$S level scheme was expanded through experimental work utilizing a fusion-evaporation reaction and in-beam $\gamma$-ray spectroscopy. Prompt $\gamma$-ray transitions were detected by the Gamma-Ray Energy Tracking Array (GRETINA) and recoiling $^{38}$S residues were selected by the Fragment Mass Analayzer (FMA). Tools based on machine-learning techniques were developed and deployed for the first time in order to enhance the unique selection of $^{38}$S residues and identify any associated $\gamma$-ray transitions. The new level information, including the extension of the even-spin yrast sequence through $J^{\pi} = 8^{(+)}$, was interpreted in terms of a basic single-particle picture as well shell-model calculations which incorporated the empirically derived FSU interaction. A comparison between the properties of the yrast states in the even-$Z$ $N=22$ isotones from $Z=14$ to $20$, and for $^{36}$Si-$^{38}$S in particular, was also presented with an emphasis on the role and influence of the neutron $1p_{3/2}$ orbital on the structure in the region.

Abstract: A ground-state rotational band in the fissile nucleus Rf254 was observed for the first time. Levels up to spin 14 and excitation energy of 1.56 MeV were observed. The Rf254 nuclei were produced using the Pb206(Ti50,2n) fusion-evaporation reaction. It is the weakest reaction channel ever studied using in-beam γ-ray spectroscopic methods. The reaction products were separated from the beam in the Argonne gas-filled analyzer (AGFA). The Rf254 nuclei were implanted into a double-sided Si strip detector at the AGFA focal plane and tagged with subsequent ground-state spontaneous fission decays using temporal and spatial correlations. Prompt γ rays in coincidence with the Rf254 recoils were detected in the Gammasphere array of Ge detectors. In order to identify the ground-state rotational band in Rf254, a method for identifying rotational bands in low statistics γ-ray spectra was developed. The deduced Rf254 kinematic moment of inertia is smaller compared to neighboring even-even nuclei. This is most likely associated with a slightly lower quadrupole deformation and stronger pairing correlations in Rf254. The behavior of the moment of inertia as a function of rotational frequency is similar to that of the lighter N=150 isotones Fm250 and No252.

Abstract: Competing configurations, assigned with three different shapes, are mixed at low angular momentum in the neutron-deficient 188Pb nucleus. Here, we present a simultaneous conversion electron and γ-ray in-beam spectroscopic precision measurement employing the sage spectrometer. The level energy of the first excited state, 02+, has been determined at 591(1)keV through direct measurement of conversion electrons. By using the intensity of the observed 02+→01+ transition to the ground state, the feeding of the 02+ state has been determined for the first time, which suggests the 02+ state is the head of the predominantly prolate band. The compositions of the 42+→41+ and 22+→21+ inter-band transitions have been determined, indicating configuration mixing between the bands.

17. Prognostic factors and overall survival among patients with ovarian cancer in the pre-PARP inhibitor era: the OCRWE-Finland study

Abstract: Background: Despite recent treatment advances in ovarian cancer (OC), more real-world evidence studies investigating patient outcomes are needed. OCRWE-Finland was an observational cohort study investigating OC outcomes in Finland during the pre-PARP inhibitor era. Patients: Patients were diagnosed with OC between 2014 and 2019 in Finland. This analysis reports baseline characteristics of all patients, patients with high-grade serous OC (HGSOC), and overall survival (OS) for patients with HGSOC. Results: Among 1,711 patients diagnosed with OC, 867 (51%) had HGSOC. The absence versus presence of visible residual disease post-debulking surgery was associated with improved OS for patients at stage III (n = 303; median: NR vs. 43 months; p = 0.005), but not stage IV (n = 118; median: 37 months vs. 40 months; p = 0.96). Bevacizumab treatment at any line at stages III/IV improved OS in the short-term only. Receiving versus not receiving bevacizumab at first-line for patients with visible residual disease post-debulking surgery was associated with improved OS at stage III (median: 48 months vs. 36 months; p = 0.003), but not stage IV (median: 42 months vs. 37 months; p = 0.26). Multivariate Cox regression analyses showed that stage IV at initial diagnosis and the presence of R2 classification post-debulking surgery resulted in poorer OS. Interpretation: In the pre-PARP inhibitor era, the absence versus presence of visible residual disease post-debulking surgery was associated with improved OS in stage III, but not stage IV HGSOC. First-line bevacizumab seemed to be beneficial in patients with stage III HGSOC and visible residual disease.

Abstract: Background: As the treatment landscape for advanced ovarian cancer (OC) evolves, it is important to understand patient outcomes in real-world clinical practice. OCRWE-Finland was an observational cohort study investigating OC outcomes, including treatment patterns, time to next treatment 1 (TTNT1), overall survival and healthcare resource utilisation, in Finland during the pre-PARPi era. Materials and methods: Patients included in OCRWE-Finland were diagnosed with OC between 2014 and 2019. Here, we report treatment patterns and TTNT1 outcomes (as a surrogate for progression-free survival) for patients in the high-grade serous ovarian carcinoma (HGSOC) cohort. Results: In OCRWE-Finland, there were 867 patients with HGSOC. Of the 811 patients who received first-line treatment, the most common regimen was surgery and adjuvant chemotherapy (53%), and 227 patients also received first-line bevacizumab. Median TTNT1 among 623 patients with stage III/IV disease was 19 months (95% confidence interval, 18–21 months), with no difference between patients with stage III or IV disease (p = 0.24). The presence versus absence of visible residual disease post-debulking surgery was associated with shorter TTNT1 among patients with stage III tumours (p = 0.031) but showed no impact for stage IV tumours (p = 0.55). Abstract: Understanding the explosion mechanism of a core-collapse supernova (CCSN) is important to accurately model CCSN scenarios for different progenitor stars using model-observation comparisons. The uncertainties of various nuclear reaction rates relevant for CCSN scenarios strongly affect the accuracy of these stellar models. Out of these reactions, the $^{13}$N($\alpha$,$p$)$^{16}$O reaction has been found to affect various stages of a CCSN at varying temperatures. This work presents the first direct measurement of the $^{13}$N($\alpha$,$p$)$^{16}$O reaction performed using a 34.6 MeV beam of radioactive $^{13}$N ions and the active-target detector MUSIC (MUlti-Sampling Ionization Chamber) at Argonne National Laboratory. The resulting total $^{13}$N($\alpha$,$p$)$^{16}$O reaction cross sections from this measurement in the center-of-mass energy range of 3.26 - 6.02 MeV are presented and compared with calculations using the Hauser-Feshbach formalism. Abstract: Background: In Part 1 of the phase III RUBY trial (NCT03981796) in patients with primary advanced or recurrent endometrial cancer (EC), dostarlimab plus carboplatin–paclitaxel (CP) significantly improved progression-free survival and overall survival compared with CP alone. Limited safety data have been reported for the combination of immunotherapies plus chemotherapy in this setting. Objectives: The objective of this analysis was to identify the occurrence of treatment-related adverse events (TRAEs) and immune-related adverse events (irAEs) and to describe irAE management in Part 1 of the RUBY trial. Design: RUBY is a phase III, randomized, double-blind, multicenter study of dostarlimab plus CP compared with CP alone in patients with primary advanced or recurrent EC. Methods: Patients were randomized 1:1 to dostarlimab 500 mg, or placebo, plus CP every 3 weeks for 6 cycles, followed by dostarlimab 1000 mg, or placebo, every 6 weeks for up to 3 years. Adverse events (AEs) were assessed according to Common Terminology Criteria for Adverse Events, version 4.03. Results: The safety population included 487 patients who received ⩾1 dose of treatment (241 dostarlimab plus CP; 246 placebo plus CP). Treatment-emergent AEs were experienced by 100% of patients in both arms. TRAEs occurred in 97.9% of the dostarlimab arm and 98.8% of the placebo arm. The most common TRAEs occurred at similar rates between arms and were mostly low grade. IrAEs occurred in 58.5% of patients in the dostarlimab arm and 37.0% of patients in the placebo arm. Dostarlimab- or placebo-related irAEs were reported in 40.7% of patients in the dostarlimab arm and 16.3% of the placebo arm. Conclusion: The safety profile of dostarlimab plus CP was generally consistent with that of the individual components. Dostarlimab plus CP has a favorable benefit–risk profile and is a new standard of care for patients with primary advanced or recurrent EC. Trial registration: NCT03981796.

Abstract: Excited states have been observed for the first time in the very neutron-deficient odd-odd nucleus 57120La63. The observed γ rays have been assigned based on coincidences with lanthanum X rays measured with the JUROGAM 3 array and with A=120 fusion-evaporation residues measured with the MARA separator. The observed γ rays form a rotational band which decays to the ground state via a cascade of four low-energy transitions. Based on the systematic comparisons with the heavier odd-odd La isotopes we assign spin-parity 4+ to the ground state and a πh11/2⊗νh11/2 configuration to the rotational band. The nuclear shape has been investigated by the cranked Nilsson-Strutinsky model. Two quasiparticle plus triaxial rotor model calculations including the np interaction nicely reproduce the spin of the inversion between the even- and odd-spin cascades of E2 transitions, giving credit to the np interaction as an important parameter responsible for the mechanism inducing the inversion. The position of the Fermi levels, in particular for neutrons, also has a strong impact on the observed inversion in the chain of lanthanum nuclei.

Abstract: Decay spectroscopy of the odd-proton nuclei $^{249}$Md and $^{251}$Md has been performed. High-$K$ isomeric states were identified for the first time in these two nuclei through the measurement of their electromagnetic decay. An isomeric state with a half-life of $2.8(5)$ ms and an excitation energy $\geq 910$ keV was found in $^{249}$Md. In $^{251}$Md, an isomeric state with a half-life of $1.4(3)$ s and an excitation energy $\geq 844$ keV was found. Similarly to the neighbouring $^{255}$Lr, these two isomeric states are interpreted as 3 quasi-particle high-$K$ states and compared to new theoretical calculations. Excited nuclear configurations were calculated within two scenarios: via blocking nuclear states located in proximity to the Fermi surface or/and using the quasiparticle Bardeen-Cooper-Schrieffer method. Relevant states were selected on the basis of the microscopic-macroscopic model with a deformed Woods-Saxon potential. The most probable candidates for the configurations of $K$-isomeric states in Md nuclei are proposed.

Abstract: Introduction A better understanding of the earliest stages of Alzheimer’s disease (AD) could expedite the development or administration of treatments. Large population biobanks hold the promise to identify individuals at an elevated risk of AD and related dementias based on health registry information. Here, we establish the protocol for an observational clinical recall and biomarker study called TWINGEN with the aim to identify individuals at high risk of AD by assessing cognition, health and AD-related biomarkers. Suitable candidates were identified and invited to participate in the new study among THL Biobank donors according to TWINGEN study criteria.Methods and analysis A multi-centre study (n=800) to obtain blood-based biomarkers, telephone-administered and web-based memory and cognitive parameters, questionnaire information on lifestyle, health and psychological factors, and accelerometer data for measures of physical activity, sedentary behaviour and sleep. A subcohort is being asked to participate in an in-person neuropsychological assessment (n=200) and wear an Oura ring (n=50). All participants in the TWINGEN study have genome-wide genotyping data and up to 48 years of follow-up data from the population-based older Finnish Twin Cohort (FTC) study of the University of Helsinki. The data collected in TWINGEN will be returned to THL Biobank from where it can later be requested for other biobank studies such as FinnGen that supported TWINGEN.Ethics and dissemination This recall study consists of FTC/THL Biobank/FinnGen participants whose data were acquired in accordance with the Finnish Biobank Act. The recruitment protocols followed the biobank protocols approved by Finnish Medicines Agency. The TWINGEN study plan was approved by the Ethics Committee of Hospital District of Helsinki and Uusimaa (number 16831/2022). THL Biobank approved the research plan with the permission no: THLBB2022_83.

30. Estimation of marriage incidence rates by combining two cross-sectional retrospective designs: Event history analysis of two dependent processes

Abstract: The aim of this work is to develop methods for studying the determinants of marriage incidence using marriage histories collected under two different types of retrospective cross-sectional study designs. These designs are: sampling of ever married women before the cross-section, a prevalent cohort, and sampling of women irrespective of marital status, a general cross-sectional cohort. While retrospective histories from a prevalent cohort do not identify incidence rates without parametric modelling assumptions, the rates can be identified when combined with data from a general cohort. Moreover, education, a strong endogenous covariate, and marriage processes are correlated. Hence, they need to be modelled jointly in order to estimate the marriage incidence. For this purpose, we specify a multi-state model and propose a likelihood-based estimation method. We outline the assumptions under which a likelihood expression involving only marriage incidence parameters can be derived. This is of particular interest when either retrospective education histories are not available or related parameters are not of interest. Our simulation results confirm the gain in efficiency by combining data from the two designs, while demonstrating how the parameter estimates are affected by violations of the assumptions used in deriving the simplified likelihood expressions. Two Indian National Family Health Surveys are used as motivation for the methodological development and to demonstrate the application of the methods.

Abstract: The low-spin structure of the semimagic 64Ni nucleus has been considerably expanded: combining four experiments, several 0+ and 2+ excited states were identified below 4.5 MeV, and their properties established. The Monte Carlo shell model accounts for the results and unveils an unexpectedly complex landscape of coexisting shapes: a prolate 0+ excitation is located at a surprisingly high energy (3463 keV), with a collective 2+ state 286 keV above it, the first such observation in Ni isotopes. The evolution in excitation energy of the prolate minimum across the neutron N = 40 subshell gap highlights the impact of the monopole interaction and its variation in strength with N.

Abstract: This letter reports on the first observation of an octupole band in the neutron-deficient (N=Z+2) nucleus 110Xe. The 110Xe nuclei were produced via the 54Fe(58Ni,2n) fusion-evaporation reaction. The emitted γ rays were detected using the jurogam 3 γ-ray spectrometer, while the fusion-evaporation residues were separated with the MARA separator at the Accelerator Laboratory of the University of Jyväskylä, Finland. The experimental observation of the low-lying 3− and 5− states and inter-band E1 transitions between the ground-state band and the octupole band proves the importance of octupole correlations in this region. Abstract: The odd-$Z$ $^{251}$Md nucleus was studied using combined $\gamma$-ray and conversion-electron in-beam spectroscopy. Besides the previously observed rotational band based on the $[521]1/2^-$ configuration, another rotational structure has been identified using $\gamma$-$\gamma$ coincidences. The use of electron spectroscopy allowed the rotational bands to be observed over a larger rotational frequency range. Using the transition intensities that depend on the gyromagnetic factor, a $[514]7/2^-$ single-particle configuration has been inferred for this band, i.e., the ground-state band. A physical background that dominates the electron spectrum with an intensity of $\simeq$ 60% was well reproduced by simulating a set of unresolved excited bands. Moreover, a detailed analysis of the intensity profile as a function of the angular momentum provided a method for deriving the orbital gyromagnetic factor, namely $g_K = 0.69^{+0.19}_{-0.16}$ for the ground-state band. The odd-$Z$ $^{249}$Md was studied using $\gamma$-ray in-beam spectroscopy. Evidence for octupole correlations resulting from the mixing of the $\Delta l = \Delta j = 3$ $[521]3/2^-$ and $[633]7/2^+$ Nilsson orbitals were found in both $^{249,251}$Md. A surprising similarity of the $^{251}$Md ground-state band transition energies with those of the excited band of $^{255}$Lr has been discussed in terms of identical bands. Skyrme-Hartree-Fock-Bogoliubov calculations were performed to investigate the origin of the similarities between these bands.

Abstract: An extensive, model-independent analysis of the nature of triaxial deformation in $^{76}$Ge, a candidate for neutrinoless double-beta ($0\nu\beta\beta$) decay, was carried out following multi-step Coulomb excitation. Shape parameters deduced on the basis of a rotational-invariant sum-rule analysis provided considerable insight into the underlying collectivity of the ground-state and $\gamma$ bands. Both sequences were determined to be characterized by the same $\beta$ and $\gamma$ deformation parameter values. In addition, compelling evidence for low-spin, rigid triaxial deformation in $^{76}$Ge was obtained for the first time from the analysis of the statistical fluctuations of the quadrupole asymmetry deduced from the measured $E2$ matrix elements. Abstract: Excited states in the Tz=0,−1 nuclei 62Ga and 62Ge were populated in direct reactions of relativistic radioactive ion beams at the RIBF. Coincident γ rays were measured with the DALI2+ array and uniquely assigned to the A=62 isobars. In addition, 62Ge was also studied independently at JYFL-ACCLAB using the 24Mg(40Ca,2n)62Ge fusion-evaporation reaction. The first excited T=1,Jπ=2+ states in 62Ga and 62Ge were identified at 979(1) and 965(1) keV, respectively, resolving discrepant interpretations in the literature. States beyond the first 2+ state in 62Ge were also identified for the first time in the present work. The results are compared with shell-model calculations in the fp model space. Mirror and triplet energy differences are analyzed in terms of individual charge-symmetry and charge-independence breaking contributions. The MED results confirm the shrinkage of the p-orbits' radii when they are occupied by at least one nucleon on average.

Abstract: The radioactivity of 76160Os84 and 74156W82 that lie at the two-proton drip line has been measured in an experiment performed at the Accelerator Laboratory of the University of Jyväskylä. The 160Os nuclei were produced using fusion-evaporation reactions induced by a beam of 310 MeV 58Ni ions bombarding a 106Cd target. The 160Os ions were separated in flight using the recoil separator MARA and implanted into a double-sided silicon strip detector, which was used to measure their decays. The α decays of the ground state of 160Os (Eα = 7092(15) keV, t1/2 = 97−32+97 μs) and its isomeric state (Eα = 8890(10) keV, t1/2 = 41−9+15 μs) were measured, allowing the excitation energy of the isomer to be determined as 1844(18) keV. These α-decay properties and the excitation energy of the isomer are compared with systematics. The α decays were correlated with subsequent decays to investigate the β decays of the ground state of 156W, revealing that unlike its isotones, both low-lying isomers were populated in its daughter nuclide, 156Ta. An improved value for the half-life of the proton-decaying high-spin isomeric state in 73156Ta83 of 333−22+25 ms was obtained in a separate experiment using the same experimental systems with a 102Pd target. This result was employed to improve the precision of the half-life determined for 156W, which was measured as 157−34+57 ms.

Abstract: In response to the global megatrends of digitalization and transportation automation, Turku University of Applied Sciences has developed a test platform to advance autonomous maritime operations. This platform includes the unmanned surface vessel eM/S Salama and a remote operations center, both of which are detailed in this article. The article highlights the importance of collecting and annotating multi-modal sensor data from the vessel. These data are vital for developing deep learning algorithms that enhance situational awareness and guide autonomous navigation. By securing relevant data from maritime environments, we aim to enhance the autonomous features of unmanned surface vessels using deep learning techniques. The annotated sensor data will be made available for further research through open access. An image dataset, which includes synthetically generated weather conditions, is published alongside this article. While existing maritime datasets predominantly rely on RGB cameras, our work underscores the need for multi-modal data to advance autonomous capabilities in maritime applications.

Abstract: In the study of the odd-$Z$, even-$N$ nuclei $^{243}$Es and $^{249}$Md, performed at the University of Jyv\"askyl\"a, the fusion-evaporation reactions $^{197}$Au($^{48}$Ca,2$n$)$^{243}$Es and $^{203}$Tl($^{48}$Ca,2$n$)$^{249}$Md have been used for the first time. Fusion-evaporation residues were selected and detected using the RITU gas-filled separator coupled with the focal-plane spectrometer GREAT. For $^{243}$Es, the recoil decay correlation analysis yielded a half-life of $24 \pm 3$s, and a maximum production cross section of $37 \pm 10$ nb. In the same way, a half-life of $26 \pm 1$ s, an $\alpha$ branching ratio of 75 $\pm$ 5%, and a maximum production cross section of 300 $\pm$ 80 nb were determined for $^{249}$Md. The decay properties of $^{245}$Es, the daughter of $^{249}$Md, were also measured: an $\alpha$ branching ratio of 54 $\pm$ 7% and a half-life of 65 $\pm$ 6 s. Experimental cross sections were compared to the results of calculations performed using the KEWPIE2 statistical fusion-evaporation code.

We combined epidemiological data and whole genome sequencing (WGS) of a set of 64 (27%, 64/237) t172-MRSA isolates covering 10 years. Isolates represented sporadic and index cases of all identified healthcare-associated outbreaks (HAOs) and family clusters (FCs). Among the included 62 isolates, core-genome MLST analysis revealed eight genomic clusters comprising 24 (38.7%) isolates and 38 (61.3%) non-clustered isolates. Cluster 1 comprised ten and the remaining seven clusters two isolates each, respectively. Two epidemiologically distinct HAOs were linked in cluster 1. FCs were involved in all clusters. All strains were associated with epidemic clonal complex CC59. We were able to confirm the spread of several successful t172-MRSA subclones in regional healthcare and the community. WGS complemented routine surveillance by revealing undetected links between t172-MRSA cases. Targeted, WGS-based typing could enhance MRSA surveillance without the need for routine WGS diagnostics.

Abstract: Abstract Nemaline myopathy (NM) is one of the most common non-dystrophic genetic muscle disorders. NM is often associated with mutations in the NEB gene. Even though the exact NEB-NM pathophysiological mechanisms remain unclear, histological analyses of patients’ muscle biopsies often reveal unexplained accumulation of glycogen and abnormally shaped mitochondria. Hence, the aim of the present study was to define the exact molecular and cellular cascade of events that would lead to potential changes in muscle energetics in NEB-NM. For that, we applied a wide range of biophysical and cell biology assays on skeletal muscle fibres from NM patients as well as untargeted proteomics analyses on isolated myofibres from a muscle-specific nebulin‐deficient mouse model. Unexpectedly, we found that the myosin stabilizing conformational state, known as super-relaxed state, was significantly impaired, inducing an increase in the energy (ATP) consumption of resting muscle fibres from NEB-NM patients when compared with controls or with other forms of genetic/rare, acquired NM. This destabilization of the myosin super-relaxed state had dynamic consequences as we observed a remodeling of the metabolic proteome in muscle fibres from nebulin‐deficient mice. Altogether, our findings explain some of the hitherto obscure hallmarks of NM, including the appearance of abnormal energy proteins and suggest potential beneficial effects of drugs targeting myosin activity/conformations for NEB-NM.

Abstract: A beam containing a substantial component of both the $J^{\pi}=5^+$, $T_{1/2}=162$ ns isomeric state of $^{18}$F and its $1^+$, 109.77-min ground state has been utilized to study members of the ground-state rotational band in $^{19}$F through the neutron transfer reaction $(d$,$p)$ in inverse kinematics. The resulting spectroscopic strengths confirm the single-particle nature of the 13/2$^+$ band-terminating state. The agreement between shell-model calculations, using an interaction constructed within the $sd$ shell, and our experimental results reinforces the idea of a single-particle/collective duality in the descriptions of the structure of atomic nuclei.
